Sledování transferu genetické informace z krmiva u modelových zvířat

The aim of this thesis is to observe the possibility of transfer of genetic information from feed, made of wheat with unusual grain colour and wheat with standard grain colour, into the blood of rat test subjects. Out of 64 rats that were tested, 32 were fed with purple grain (Konini variety) and formed the test group. A further 32 rats represented the study's control group and were fed with standard coloured wheat (Bohemia variety). The blood sample was acquired from the test animals. From 12 samples (6 samples from each group) DNA was isolated. Plant DNA was detected in the samples of DNA of rats by PCR using primers amplifying fragments of housekeeping genes specific for DNA of wheat (wheat genes Gapdh and Actin). No DNA fragments of wheat were detected in the blood of the rats. The results show, that there is no transfer of genetic information from the feed through the gastrointestinal tract to the blood of the animal.

This diploma thesis deals with the issues of the blood donation and the regulation of trade in blood and blood derivatives. It is generally known that the blood taking is in the majority based upon altruism of donors. Society perceives paid blood taking donations as an unethical and an immoral. Besides, paid donations are in violation of a worldwide long term efforts that have been trying to prohibit a monetary remuneration. A related question is whether the unpaid donations can satisfy the increasing demand for blood, and what sort of problems a state regulation bears. The first part of this work provides us a brief historical development of blood transfusion, which is further connected to economic theory and modified to the market in blood; particularly to the theory of price regulation and product (quality). The second part focuses on a comparison of three different markets and discussion of altruism and donations issue.

Steroid hormones act through two mechanisms. The first is the classical genomic level, which acts at a relatively slow pace from hours to days. The second non-genomic mechanism of steroid activity can influence cellular function during seconds or even milliseconds. During pregnancy both mechanisms take place. Steroid hormones that act non-genomically and influence neuronal excitability are called neuroactive steroids. The aim of this study was to measure the levels of steroid hormones and examine their relationship to the physiology and pathophysiology of pregnancy. The physiological importance of increased placental progesterone production, lowered production of 5B-pregnanone steroids and increased activity of steroid sulfotransferase with gestational age have been intensely discussed. On the basis of our results, we have proposed an alternative mechanism of maternal progesterone synthesis during pregnancy (progesterone, or the gestagen stabilizing activity of the myometrium = continuation of pregnancy). In contrast to previous results our observations indicate that gestagens arise from precursors in the fetus (from pregnenolone sulfate). The thesis includes a general introduction to monitoring and measurement of blood pressure of horses, the design of algorithms for pressure curve detection and the calculation of respiratory variabilities in mechanical ventilation under anaesthesia. The first two chapters focus on anatomy, on characteristics of blood flow and on vascular system. Then it deals with the measurement of blood pressure, monitoring of a horse under anaesthesia and it describes particular thermodynamic parameters. In the following part of the thesis, the methods of pressure curve detection are described. The last part contains the description of an algorithm designed for detection of pressure curve of horses in the environment of the program LabVIEW. The program calculates, displays and saves variabilities of systolic pressure, pulse volume, pressure amplitude, pulse frequency, peripheral resistance and vascular expansion from the detected values.

The same way the quality parameters (RBC, PLT, HB and hemolysis) of erythrocyte blood component were measured and compared in day D14, D21, D35 and D42. Methods: WB collected from 30 healthy group A donors was divided into two groups - leucodepleted (LWB), using in-line platelet-sparing filters and collected to a blood bag system IMUFLEX® WB-SP (Terumo BCT, USA) and non-leucodepleted WB (NLWB), colletected to a blood bag system CompoFlex® Single System (Fresenius Kabi, Germany). The voluntary blood donation issue is topical in the Czech Republic since the number of blood donors has not yet reached the number of donors existing in the neighboring countries. Because of this fact the Czech Blood Cross together with transfusion services centers in the Czech Republic and in cooperation with various organizations, e.g. Czech Radio, do their best to obtain new voluntary blood donors via blood donation campaigns. Language of these campaigns is mainly of persuasive character. As such, it is often viewed as a means of white propaganda which stems from factual information and whose aim is to persuade the consumer of the message about the legitimacy of its ideology. White propaganda also helps to build trust which is lately used. In other words, the consumer of the message becomes blood donor. The thesis analyzes persuasive linguistic means of propaganda present in online discourse of the selected campaigns. On the basis of learned facts, the thesis also tries to characterize this discourse type; moreover, it makes suggestions for designing a new blood donation campaign.

The intention of this thesis is to try to uncover the meaning of the sin offering once again. To get to this new point of view, it uses synchronic methods and thorough analysis of defined texts of Leviticus (from the first, the fourth and the sixth chapter) and it even uses analysis of single words in a wider context. First it is concerned about getting deeply into the understanding of what sin is, then that the sacrificial animal probably becomes the sin and the sin is killed.
