The Master´s thesis gives an overview of cord blood, its use, processing and cryopreservation. The role of cord blood as an alternative resource for transplantation is being widely discussed. In the experimental part, 50 grafts of donor cord blood were processed and the effect of cryopreservation and demographic factors of mother and child were evaluated on the cellularity of cord blood. Based on the evaluation, a recommendation about the inclusion of additional tests in routine processing of cord blood were made.

MicroRNA (miRNA) are small non-coding 21-23 nucleotides long one strand RNAs. They are among the major posttranscriptional regulators of gene expression that regulate both physiological and pathological processes. Some of microRNAs, amount of their expression respectively, are specific only for certain type of tissue or pathological condition. The hypothesis for my diploma thesis was that gene expression of 28 cardiovascular disease associated microRNAs (miR-1-3p, miR-16-5p, miR-17-5p, miR- 20a-5p, miR-20b-5p, miR-21-5p, miR-23a-3p, miR-24-3p, miR-26a-5p, miR-29a-3p, miR-92a-3p, miR-100-5p, miR-103a-3p, miR-125b-5p, miR-126-3p, miR-130b-3p, miR-133a-3p, miR-143-3p, miR-145-5p, miR-146a-5p, miR-181-5p, miR-195-5p, miR- 199a-5p, miR-210-3p, miR-221-3p, miR-342-3p, miR-499a-5p, miR-574-3p) would differ in umbilical cord blood between groups of women with physiological pregnancies (FG), gestational hypertension (GH), preeclampsia (PE) and fetal growth restriciton (FGR). The studied cohort consisted of 184 pregnant women involving 44 controls, 47 GH pregnancies, 56 PE pregnancies and 37 FGR pregnancies. The aim of this thesis is to show in what way the mechanistic philosophy of René Descartes allowed him to accept William Harvey's discovery of the circulation of the blood, while at the same time prevented him from accepting his explanation of the movement of the heart. In the introductory section we mention some of the basic notions concerning the state of natural philosophy in the second half of the 16th century which are closely related to the themes of the thesis. Both authors we are concerned with are also presented. The second, historicaly oriented section focuses on Aristotle's, Galen's and Harvey's opinions on the role and motion of the heart and blood in the human body. The aim is to describe how Harvey's 1628 treatise De motu cordis allowed to resolve the proliferating problems faced by the Galenist tradition in the 16th and the early 17th century. The third section presents the exposition of the introductory chapters of Descartes' 1633 treatise Le Monde in which he introduces the basic notions of his new mechanistic philosophy. The biological half-life of several pro-inflammatory mediators involved in the pathogenesis of rheumatoid arthritis (RA) is controlled by molecules exhibiting dipeptidyl peptidase-IV (DPP-IV)-like enzymatic activity (Dipeptidyl peptidase-IV activity and/or structure homologues- DASH). The aim of this thesis was to identify the molecular source of the DPP-IV-like enzymatic activity in the peripheral blood and synovial fluid in patients with rheumatoid arthritis as compared to control patients with osteoarthritis (OA), and to evaluate the association of DPP-IV with the disease activity. We found that the main source of the DPP-IV-like enzyme activity in the plasma and in the synovial fluid in patients with RA is the canonical DPP-IV. DPP-IV-like enzymatic activity and canonical DPP-IV were also detected on the cell surface of blood and synovial fluid mononuclear cells. Significantly lower DPP-IV-like enzymatic activity and DPP-IV expression in the synovial fluid mononuclear cells was found in RA as opposed to OA patients. Sheep breeding is nowadays a developing branch of agriculture again. There is a lot of different breed and they react distinctly on the same conditions of the enviroment. Sheep of breeds charollais, suffolk, šumavská ovce and valaška bred in similar conditions were observed in spring and autumn of years 2007 and 2008. In these seasons were taking blood samples (from {$\pm$}7{--}24) ewes and lambs and were analyzed in hematology laboratory. There were determined haemoglobin level, haematocrit indicator, erytrocytes and leucocytes levels, glucose, cholesterol, triglycerides levels, urea and plasmatic proteins, activity of ALP and GMT enzymes, phosphor, calcium, magnesium, zinc and copper levels. The main objective of this project was determine seasonal changes in observed parametres. Next objectives were determine different changes in blood parametres in different breeds and different aimes of breeds. There were recognized that all the observed breeds don`t react the same way on similar conditions. There were not provably determined seasonal changes in observed parametres, but average Hb level was higher in autumn than in spring. Urea level was conversely higher in spring season than in autumn. The demostrable fact is, that the similar conditions induce different answers not only in different breeds but also in different aimes of breeds.

Introduction: The properties of human blood are so specific that it has not yet been possible to replace human blood with any other fluid than obtaining it as a blood gift from donors. In recent years, there has not been a sufficient number of new blood donors in the Czech Republic to compensate for the decrease in the number of donors due to the increasing incidence of a number of civilization diseases and age (Czech Red Cross, 2020). Blood donation and the need for blood products are constantly increasing with the development of the methods of treatment and medicine as a science. Ensuring a sufficient number of blood donors and sufficient deposition of blood obtained from donors is a permanent task of healthcare (Society for Transfusion Medicine of the Czech Medical Association JEP, 2019). Aim of the work: The main goal of the research was to map the issue of blood donation, to identify the factors by which blood donors are motivated to donate. Furthermore, what can cause stagnation in the growth of blood donors and what means doctors motivate potential blood donors.
