(EN) The first part of the thesis describes the ionization reactions in the APCI source in the presence (enclosed configuration) and absence (open configuration) of the ion source housing. Commercial APCI source was modified and used in enclosed, as well as open configuration. All other parameters were kept the same (including the source geometry). In the positive mode, the biggest differences were observed for non-polar analytes dissolved in aprotic solvents like toluene, chloroform and carbon disulfide. Protonated species were dominant in the open configuration, while radical cations were mostly present in the enclosed configuration. The excessive protonation in open ion source was caused by the diffusion of water vapor molecules into the ionization region from the atmosphere. Water vapor molecules were also responsible for the formation of an ion [M + 19]+ from alkynes in the open configuration. The fragmentation study confirmed that the ion [M + 19]+ was a 2-methylketone formed by the addition reaction. The formation of such artifacts can pose problems in qualitative analysis. On the other hand, the ion [M + 55]+ was observed in the enclosed ion source as a reaction product of unsaturated compounds with acetonitrile solvent molecules. Crime relating to the abuse of addictive substances The abuse of addictive substances is an issue of the whole society, which in current globalized world, considering continuous development of drug scene, still remains topical. The abuse of addictive substances, as a significant criminogenic need, relates to all kinds of different criminal offences, both those committed by addicts and those committed against them. This includes for example property crimes, crimes against life, person and health, or endangerment. Because of such wide scope of crimes related to the abuse of addictive substances, the focus of my work is on substantive regulation of so-called drug offences, which is stated in the articles 283 - 287 of the Criminal Code. The purpose of this thesis is to analyse constituent elements of the particular drug offences, coherent case-law, as well as some questionable issues related with this kind of criminal offences. It is for instance, the issue of determining the term "greater than the small amount" and regulation of making cannabis accessible for medical use. The aim of this thesis was to compare the influence of agrotechnical measures on yields and certain qualitative parameters of winter wheat as a part of a long-term field experiment, which focuses on agriculture without livestock production at the Field experimental station of Mendel University in Žabčice. The issue was being solved during a period of three years (since 2014/2015 to 2016/2017) in the form of small-plot field experiments. The experiment assessed the influence of the year, soil processing and previous crop on yields, the content of crude proteins and hectolitre weight. The results suggest that the yields and the content of crude proteins are more significantly influenced by the previous crop and the year, rather than by soil tillage. Hectolitre weight was more influenced by the effect of the year.

1 Abstract Crime relating to the abuse of addictive substances This thesis deals with the issue of substance abuse and criminality associated with this pathological phenomenon. Addiction and substance abuse can be considered a significant criminogenic factor, which is a common cause of traffic accidents, domestic violence, reduced self-control, increased aggression and other crime-related behaviors. Criminality related to the production, distribution and use of addictive substances covers a wide range of offenses, from trivial offenses bordering on offenses to the diverse activities of international organized crime, which can be consider a global issue. Subject matter of this thesis is to link this stated issue with fundamental knowledge about narcotic and psychotropic substances and its effects on individuals, along with their potential influence on the delinquent behavior of drug users, while providing a theoretical definition of crimes related to substance abuse and an effort to quantify this phenomenon. Separace azaarénů vysoceúčinnou kapalinovou chromatografií / Separation of Azaarenes by High Performance Liguid Chromatography

Under the framework of this PhD project, a study on chromatographic behaviour of selected azaarenes on octadecylsilica stationary phases during their separation by reversed phase liquid chromatography was developed. The main goal was aimed at an application of the basic RPLC principles on the retention behaviour of azaarenes including the effects such as peak tailing, peak broadening, irreproducible retention or strong retention on a stationary phase. This study was particularly focused on basic azaarenes, also called acridines, which belong to a group of persistent organic pollutants providing mutagenic and/or carcinogenic activity. The retention patterns were studied on a group of eight acridines that are often present in environmental samples polluted with polycyclic aromatic compounds. The mixture of acridines was separated under isocratic elution conditions on three octadecylsilica stationary phases using two different binary mixtures as a mobile phase. Evaluation of the surface properties of the selected stationary phases was firstly performed, followed by the separation experiments. The structure of the experiments was designed to evaluate the effect of three different factors on the retention behaviour of acridines: properties of a stationary phase, composition of a mobile phase and physical-chemical properties of acridines. The treatment of the results was based on the capacity factor values providing the best fitting and repeatability of data, but other parameters that evaluate thermodynamic and kinetic aspects of the separation process were also given. Simple correlations between the three factors and a character of retention mechanism of acridines on the given separation system were found. Application of this knowledge simplifies an optimisation process and helps to solve common separation problems for acridines, but also for other basic analytes displaying similar physical-chemical properties (Mr and pKa).

Thus, family therapy or work with families is an effective form of treatment for both adults and teens who have problems with alcohol and illegal drugs. Involvement of persons close to drug users into treatment process is associated with finding professional help at an early stage of drug use, with a higher degree of involvement of drug users in treatment process as higher retention, with significant reductions in drug use after treatment and also improving the social behavior of drug users. Therefore is offered the question: how family therapy is indicated at work with addicted clients, and when this method is contraindicated. The aim of this work is to find out how is indicated the family therapy or therapeutic work with the family at work with addicts, from the perspective of clinicians, and when, according to their experience, family therapy is not recommended or contraindicated. Mental health, integral part of the general health, has recently been increasingly associated with a term well-being. An alarming problem which occurs not only in Czech society is a high degree of early use of legal addictive substances (alcohol and tobacco) among children. This thesis provides an insight into the issue of the use of addictive substances by children in the context of subjective perception of personal well-being. Finding the prevalence of the use of these substances among children enabled the identification of links between the use and several factors which altogether represent subjective well-being. Satisfaction with oneself has proved insignificant in relation to substance abuse. Furthermore, it was found that children can compensate low self-esteem for alcohol. Based on these data using logistic regression there was designed a mathematical model which allows, while knowing the factors of personal well-being, the prediction of the probability of the use of tobacco and alcohol among children. BACKGROUND: Academic training of social work students is fundamental to the development of quality workforce. Social workers have worked with drug users and their families since the formation of social work field. Due the diversity of target groups and a number of services in which social workers can act, many of them are employed in services for drug users. Education and training of social work students does not focus enough on the field of addictology. AIMS: To provide a systematic overview of the results of published research studies, dealing with university education of social workers in the field of substance abuse in Europe and the USA. To find out how the training of social workers in the field of substance abuse is organized. METHODS: The resulting product is a systematic review of scientific articles. Articles have been searched in national databases according to the specific criteria, keywords and their combinations. The target group consists of studies dealing with the university education of social workers in the field of substance abuse. The aim of this thesis is to investigate the targeting potential of mouse polyomavirus (MPyV) based virus-like particles (VLPs) as vectors for directed cell delivery of therapeutic or diagnostic compounds. Major capsid protein VP1 of MPyV is able to selfassemble into the noninfectious VLPs. Our main goal is to retarget these VLPs from its native receptor to the prostatic cancer cells by changing the receptor binding site in the surface-exposed loop of VP1. We introduced a peptide ligand CTITSKRTC, which binds prostate-specific membrane antigen (PSMA), by insertion or substitution into BC loop of VP1. These modifications did not change the stability of the particles and genetic substitution prevented the native receptor binding. PSMA-specific binding of modified VLPs was tested by pull-down assay and surface plasmon resonance. In order to further utilize these VLPs, we tested several approaches for preparation of VLPs as vehicles for compounds delivery into eukaryotic cells. Although the method for encapsidation of the DNA into the VLPs in cellular nuclear extracts, which mimic the in vivo conditions, did not enabled us to produce pseudocapsids, we successfully optimized procedure for dissassembly and reassembly of purified particles. Background: This thesis is based on the increasing number of mothers (using addictive substances), which are appearing in social services, particularly in the Authority For Social And Legal Protection Of Children Specificity of mothers using addictive substances requires a special approach by addictology services. It begs a question, whether such specifics as to meet workers of ASPLC in cooperation with these mothers. The aim of this thesis is to describe the specifics of the work ASPLC with these mothers. Other aims are to determine how the cooperation between the two sides takes place, whether and which problems can occure in cooperation with these mothers and describe the procedure for the removing of children of these mothers and the possible impact of abusing ilegal substances on the need for the children removing.
