Abstract In my thesis ``Habits of secondary school students{\crqq} (alcohol, drugs, smoking, gambling{\dots}) I dealt with the use of addictive substances by students in the 2nd year of secondary schools. The use of addictive substances by young people has become a topical issue. According to statistical data, the number of users of both legal and illegal substances has been growing every year. It is disturbing that addictive substances, such as alcohol, nicotine or illegal drugs, have been used by ever younger consumers. In the theoretical part I covered the period of adolescence, characteristic features of that period and behavior disorders that may occur. I also reviewed the individual types of drugs and their physical and mental effects on the organism. In the empirical part of the thesis my objective was to map the current situation in drug abuse at selected secondary schools in the towns of Rokycany and Tábor. The researched group consisted of 325 respondents. I used a questionnaire to investigate their family backgrounds, alcohol consumption, cigarette consumption and experience with other drugs, e.g. marihuana, hashish or ecstasy. 2 of the 3 formulated hypotheses were refuted and 1 hypothesis was confirmed. All results are shown in form of tables and diagrams. In the discussion I compared my results with other research papers dealing with the same topic. The biggest issue in the selected group of respondents was the smoking of cigarettes. According to my findings, education is a major factor in the consumption of the individual drugs. The consumption of alcohol, cigarettes and other drugs is higher among apprentices while the consumption among students of comprehensive secondary school is significantly lower.

This study explores experience with addictive substances of secondary school children from one chosen grammar school. The theoretical part focuses on a quantitative research using a questionnaire. Identified themes from the research are synthesized using tables and grafts. Furthermore, the final part of this study contains an interview with the school methodologists preventer. Analysed data showed, that the use of addictive substances remains a current problem and the number of the users has increased in 2015, even though that the users are aware of the side effects. It was also identified that after comparing findings from a study conducted in 2012, the number of cannabis users has decreased about 11%. Hypothesis, that girls are frequenter users has not been confirmed. Conversely, it was confirmed that family play an important part of prevention. This study may provide invaluable information to the secondary school.

Studium komplexace vortioxetinu cyklodextriny a jeho stanovení kapilární elektroforézou / Study of vortioxetin complexation by native cyclodextrins and its determination using capillary electrophoresis

In this master thesis, the formation of supramolecular complexes between vortioxetine active substance and cyclodextrins (α, β, γ) is examined and the stability constants of these complexes are determined. The stoichiometric ratios of cyclodextrin and vortioxetine 1:1 and 2:1 are considered. The results suggest that the 1:1 ratio of cyclodextrin and vortioxetine is more accurate for each of used cyclodextrins and that the strongest interaction is between vortioxetine and β-cyclodextrin. Further, a method for vortioxetine assay in a tablet is developed by using non-aqueous capillary electrophoresis technique. The precision, accuracy, and robustness are validated and the values of detection and quantification limits are determined. This method can be used as a verification method for the results obtained by HPLC as the separation principle is different in capillary electrophoresis. Also, the stability of vortioxetine as a pure powder and as a mixture with excipients is examined under the conditions of elevated temperature and humidity.

Background: The state of exclusivity must be investigated and proven in each particular case. In order to establish the conclusion about the influence of the driver, it is necessary to draw up an expert opinion from the field of health care, the branch of psychiatry (Explanatory Report to Act No. 233/2013 Coll.). Research to date has shown that addictive substances in transport are a serious social and security problem. For recidivent drivers who have committed a criminal offense under the influence of an addictive substance (or a misdemeanor, or have a personal interest in a rehabilitation program), there is a possibility of a rehabilitation course. Goals: The main aim of this work is to bring knowledge about the decision-making practice of the courts in the case of a criminal offense under the influence of addictive substance. Another aim of this work is to map out the problem of the threat of drug addiction and follow-up measures set by the court in imposing sentences and their relation to the recommendation of rehabilitation programs or some of the forms of treatment of problem / addictive use of addictive substances. The structure of the thesis is divided into seven parts. The first one deals with introduction into new REACH regulation. The second part evaluates actual chemical policy and reasons for transition to REACH. Moreover this part also describes REACH and its goals and plans. The third chapter evaluates anticipated costs of REACH implementation in the European Union. The fourth part describes chemical industry in the European Union, its determination, structure and classification. The fifth part deals with anticipated benefits of REACH on chemical industry in the EU in general, while sixth chapter determines how REACH would affect directly companies in chemical industry. Situation of such affected companies in the Czech republic is described as an example in the last chapter.

Background: Individuals with substance use disorders are likely to have low health literacy due to risk factors related to their personal and socioeconomic characteristics. Current knowl- edge is limited in understanding whether low health literacy contributes to adverse health outcomes and whether it influences the substance use behavior of this population. Design: An exploratory cross-sectional study using a questionnaire survey. Aims: To explore multidimensional health literacy and its correlates in patients treated in residential addiction treatment programs and investigate health literacy as a predictor of self- reported health indicators and quality of life. Setting: Multiple residential addiction treatment programs (i.e., detoxification units, inpatient care, therapeutic communities) in the Czech Republic. Participants: Data of 613 patients treated in residential addiction treatment programs for mental and behavioral disorders due to psychoactive substance use (F10-F19, ICD-10) were analyzed. Charakterizace bylin rodu Plectrantus pro využití v potravinářství / Characterization of the genus Plectrantus for use in the food industry

The aim of this diploma thesis was to determine the optimal extraction conditions for obtaining an extract from the plant Plectranthus amboinicus. Plectranthus amboinicus is an aromatic herb of the Lamiaceae family containing a large number of bioactive compounds. Because of this, it has a number of biological effects – antimicrobial, anti-inflammatory, antioxidant, analgesic, etc. To maintain the maximum of sensory and nutritionally valuable components of P. amboinicus, a simple maceration of fresh chopped herb was chosen. Maceration was performed under the following conditions: 40 minutes at 40 ° C, solvent 40% ethanol, weight 20 g of herb per 100 ml of solvent. The extract prepared under these optimal conditions was comprehensively characterized in terms of the content of total polyphenols, their concentration was determined to 0,08 ± 0,02 mgGEA·ml-1, which corresponds to 0,40 ± 0,07 mgGEA·g-1 of the plant. The antioxidant activity of the extract was 241,24 ± 29,24 µgTEAC·ml-1 and the radical scavenging activity of ABTS•+ was determined to be 49,29 ± 5,97 %. The antimicrobial activity was verified by the agar well diffusion method. Two types of bacteria were selected for the determination – gram-positive Bacillus cereus, gram-negative Escherichia coli and yeast Candida glabrata. The results showed that the extracts have the highest antimicrobial activity against B. cereus. There were 64 volatile aromatic compounds identified in the extract, with monoterpenes and sesquiterpenes dominating. The obtained results confirm that P. amboinicus, or its prepared extract, has a good potential for use as a new and non-traditional flavor in various foods and delicacies, which can also increase their nutritional value.

Stanovení antimikrobiální a antioxidační aktivity vybraných bylinných extraktů / Determination of antimicrobial and antioxidant activity of selected herbal extracts

Tetradenia spp. is herb naturally growing in Africa and is part of family called Lamiaceae. The herb is used in traditional medicine to cure malaria, angina or as treatment for tropical skin diseases. Maceration was chosen from number of extraction methods, to obtain bioactive compounds from this plant. Optimization was performed with aim to retrieve as much of bioactive compounds as possible. Optimal conditions chosen were: temperature 60 °C, extraction time 40 minutes, solvent 40% ethanol and solid-solvent ratio 1:5. In total 63 aromatic active compounds were found in optimal extract. Terpenes were the most abundant (88,60 %), from them oxygenated monoterpenes (55,65 %) and monoterpenes (29,52 %). Compounds with highest percentage quantity were camphor (51,76 %), cis--ocimene (8,17 %), 3-octyl acetate (7,71 %), trans--ocimene (6,76 %), -terpinene (4,93 %), o-cymene (3,81 %), terpinolene (1,33 %), limonene (1,33 %), camphene (1,19 %) and borneol (1,20 %). Further, total phenolic content was 0,31 ± 0,02 mgGAEml-1, which equals 1,52 ± 0,11 mgGAEg-1 of plant. Antioxidant activity was 417,56 ± 32,84 gTEACml-1 and percentage of radical scavenging activity ABTS•+ was 88,99 ± 7,00 %. Finally antimicrobial activity was determined against bacteria Bacillus cereus, Escherichia coli and yeast Candida glabrata. Against E. coli and C. glabrata there was no inhibition exhibited. Against B. cereus there was seen mild inhibition activity. The results confirm that Tetradenia spp. contain a number of interesting aroma active compounds, which shows antioxidant and antimicrobial activity. The herb has a good potential as natural source of nutritious and sensory active compounds to be used into various foods.

Aplikace rostlinného extraktu do nečokoládových cukrovinek / Application of plant extract into non-chocolate sweets

This diploma thesis deals with the optimization of jelly candies production, application of herbal extract to produced candies, and subsequent characterization of produced samples in terms of volatile (aromatic) substances, total phenolic substances, antioxidant aktivity, and sensory quality. Optimization parameters – the amount of gelatin and the addition of citric acid, were selected based on the sensory analysis. Additions of 7,5 % gelatin and 2 % acid were proven to be optimal. The sources for the production of herbal extracts were the plants Aronia melanocarpa and Echinacea purpurea, which are known for their high content of bioactive substances. The further sensory analysis determined the optimal amount of extract added to jelly candies samples – in the case of chokeberry it was 4 % and in the case of echinacea it was 1 % of the extract. In the sample with chokeberry extract of optimal composition, ie 7,5 % gelatin, 2 % citric acid, and 4 % extract, 46 volatile substances were determined, of which nonanal (12,17 %) and thymol (8,70 %) were the most represented, the total polyphenol content was 472.88 ± 8.02 gGAEg-1 of the sample and the antioxidant activity was determined to be 828.99 ± 17.74 gTEACg-1 of the sample. The sample with echinacea extract of optimal composition, ie 7,5 % gelatin, 2 % citric acid and, 1 % extract, contained 56 volatile substances, the most represented being nonanal (12,08 %), thymol (6,19 %), and decanal (5,52 %). The total polyphenols in this sample were determined to be 291.09 ± 10.17 gGAEg-1 of the sample and its antioxidant activity was determined to be 850.91 ± 30.29 gTEACg-1 of the sample. Samples with the addition of chokeberry extract, which gave the confectionery a fruity, slightly sour taste and a pink to dark purple color, were considered more pleasant/acceptable; samples with echinacea had an herbal aroma and taste and orange color.
