Genetic characterization of wheat genes resistance to tan spot and leaf rust

Master of Science / Department of Agronomy / Jianming Yu / Tan spot, caused by Pyrenophora tritici-repentis (Ptr), is an economically important foliar disease worldwide. Race 1 of the fungus, which produces the necrosis toxin Ptr ToxA and the chlorosis toxin Ptr ToxC, is the most prevalent race in the Great Plains of the United States. The purposes of this study are to 1) identify and map novel quantitative trait loci (QTL) involved in resistance to tan spot race 1 in common wheat (Triticum aestivum L.) and 2) explore the inverse gene-for-gene interaction in the wheat-P. tritici-repentis pathosystem. A population of 288 F2:6 recombinant inbred lines (RILs) developed from the cross between Chinese landrace WSB (resistant) and Ning7840 (highly susceptible) was firstly used to identify genomic regions harboring novel sources of resistance. Two QTLs associated with resistance to chlorosis were mapped to the short arm of chromosome 1A and 2B in the WSB/Ning7840 population. No interaction was found between the two QTL. To further explore the specific wheat-ToxC model, three other populations were developed based on two susceptible parents, Ning7840 and Wheaton. QTL analysis revealed that common QTL were detected in populations shared with the same susceptible parents. The observations suggested that susceptibility rather than resistance for tan spot chlorosis is specific and presented evidence for the inverse gene-for-gene theory in the WSB-ToxC pathosystem. Leaf rust, caused by Puccinia triticina Eriks., is another important foliar disease of common wheat worldwide. The rust-resistance genes Lr41 and Lr42 from T. tauschii accessions TA2460 (Lr41) and TA2450 (Lr42) have been used as sources of rust resistance in breeding programs. Molecular markers linked to these genes are essential tools for gene pyramiding. Two BC3F2:6 mapping populations were evaluated for leaf rust resistance at both seedling and adult plant stages and analyzed with simple sequence repeat (SSR) markers. Both genetic and physical mapping confirmed that markers linked to Lr41 and Lr42 were on chromosome arm 2DS and 1DS, respectively. Marker analysis in a diverse set of wheat germplasm indicated that tightly linked markers for Lr41 and Lr42 can be used for marker-assisted selection (MAS) in breeding programs.

Mundo Novo, pela aplicação de suspensões de bectérias endofíticas 72 e 24 horas antes e após à inoculação de H. vastatrix e concomitantemente à inoculação com o patógeno. Isolados de bactérias endofíticas testados demonstraram eficácia na inibiação da germinação de urediniosporos e/ou no desenvolvimento da ferrugem, com valores acima de 50%, embora os resultados obtidos nos testes de germinação de urediniosporos tenham sido inferiores ao tratamento com o propiconazole. Nos testes em discos de folhas, folhas destacadas e em plantas de cafeeiro, os isolados endofíticos T G4-Ia, T F2-IIc, T F7-Ib e T F9-Ia, demonstraram melhor controle da ferrugem do cafeeiro. Os isolados endofíticos T G4-Ia e T F9-Ia foram identificados como Bacillus lentimorbus e Bacillus cereus, respectivamente. Os resultados indicam a ocorrência de isolados endofíticos eficientes em controlar a ferrugem alaranjada do cafeeiro. THE GENETICS OF LEAF RUST RESISTANCE IN THE MODEL GRASS BRACHYPODIUM DISTACHYON

Brachypodium distachyon è stato recentemente proposto come pianta modello per le Triticeae che includono frumento e orzo. L’obbiettivo del presente studio è stato quello di identificare regioni genomiche associate con la resistenza quantitativa alla ruggine fogliare in Brachypodium. Le malattie causate dalle ruggini fogliari causano ingenti perdite in termini di produzione delle specie cerealicole. Una popolazione di 110 individui F2 è stata sviluppata incrociando due linee inbred di Brachypodium e una mappa di linkage di marcatori AFLP è stata create. La mappa di linkage consiste di 192 loci AFLP in dieci gruppi di linkage, e copre una lunghezza pari a 1,231 Kosambi cM. Allo scopo di identificare loci coinvolti nella resistenza quantitativa sulla mappa, i 110 individui F2 sono stati valutati per la loro reazione alla ruggine fogliare allo stadio di plantula e a quello adulto. Per confermare i risultati delle piante F2, le rispettive famiglie F3 sono state studiate per la loro resistenza alla ruggine fogliare in due esperimenti indipendenti. Due loci genomici sembrano essere maggiormente coinvolti nella resistenza. / Brachypodium distachyon has been proposed as a model species for the tribe of the Triticeae, which includes wheat and barley. The objective of our study was to identify the genomic regions associated with quantitative resistance to leaf rust in Brachypodium. Leaf rust diseases cause significant reductions annually in yield of cereal crops worldwide. An F2 mapping population of 110 individuals was generated between two Brachypodium inbred lines and a AFLP-based linkage map was developed. The linkage map consists of 192 AFLP loci in ten linkage groups, and spans a total genetic length of 1,231 Kosambi cM. To locate quantitative resistance loci on the map, the 110 F2 plants were evaluated for their reaction to the leaf rust at both seedling and adult plant stages. To improve QTL identification, F2-derived F3 families were studied for resistance to leaf rust in two independent experiments. Two major genomic regions involved in resistance to leaf rust were detected.

Wheat diseases such as rusts and powdery mildews are among the most important and ancient diseases that affect wheat cultivation worldwide. The pathogen race specific resistance genes cannot maintain long lasting resistance. On the other hand, the presence of genes confers the non-race specific resistance last much longer. The durable resistance phenotypes in wheat against various rust and powdery mildew diseases were reported as Lr34, Yr18, and Pm38 separately / nevertheless, they were known to locate very close to each other based on linkage analysis. Recently, it was shown that all of these resistance phenotypes are indeed conferred by the same gene, encoding Adenosine triphosphate&ndash / binding cassette transporter (ABC-transporter) type protein. The way with which this transporter is functioning to maintain the durable resistance against different types of pathogens is still unknown. v The presence of LR34 (or ABC-transporter) gene resistant allele can be detected by specially designed markers. In spite of the few DNA sequence differences between the resistant and the susceptible alleles, the easily applicable PCR based markers allow the detection of the presence of this durable resistant allele on any given variety or cultivar. In our study, 62 different Turkish bread wheat varieties were screened by the gene specific molecular markers, developed from those LR34 gene mutation sites. The 11 cultivars determined to contain the resistant allele. This study is the first screening of Turkish cultivars for the presence of LR34 gene.

Caryological studies using Giemsa staining and image cytometry demonstrate that the first division of meiosis occurs in the uredinioid teliospores, and the second division only takes place after germination in the appressorium (delayed meiosis). The intent of examining the caryological status of H. vastatrix spores through flux cytometry was not implemented because this technique dependended on protoplast isolations that proved impossible with the treatments that were attempted for this fungus. Factors affecting the frequency and type of germination (mycelial versus appressorial) and of the uredinioid teliospores were investigated and analysed. Most germinate in the dark at temperatures between 22 and 26 oC, and appressorial formation is obtained under low temperatures (22 oC), in the dark. Teliospores were recorded from various localities in Minas Gerais, confirming the previous dubious record of the occurrence of this spore type in Brazil, and their occurrence appeared to be conditioned by the changing humidity during periods of lower temperatures in the winter and spring. A significant proportion of teliospores showed abnormal germination; the number of promycelial cells varying from three to seven, with or without the production of sterigmata and basidiospores. This body of information may explain the high number of races identified thus far in H. vastatrix, a supposedly functionally asexual fungus that produces non-functional basidiospores, according to the current views. The existence of an additional process allowing genetic recombination might explain the emergence of new races and the continuing breakdown of resistance in new coffee varieties. / Produziram-se evidências de que Hemileia vastatrix, o agente etiológico da ferrugem do cafeeiro, tem dois tipos de teliósporos no seu ciclo de vida: teliósporos globosos a napiformes de paredes finas e lisas e que germinam tipicamente pela formação de um promicélio (basídio) com quatro células, cada qual produzindo um esterigma sobre o qual um basidiósporo é produzido e teliósporos uredinóides subreniformes com parede espessa lisa ventralmente e equinulada dorsalmente. Estudos cariológicos usando corante Giemsa e citometria de imagem demonstraram que em teliósporos uredinóides a primeira divisão da meiose ocorre nos esporos e a segunda divisão ocorre apenas depois da germinação no apressório. A proposta de examinar o status cariológico de esporos de H. vastatrix pela citometria de fluxo não foi implementada, pois dependia do prévio isolamento de protoplastos, que foi impossível com os tratamentos testados para este fungo. Fatores que afetam a freqüência e o tipo de germinação (micelial versus apressorial) dos teliósporos uredinóides foram investigados e analisados. A maior proporção de germinação foi obtida no escuro entre 22 e 26 oC e a formação de apressorios foi obtida sob temperaturas amenas próximo de 22 oC no escuro. Teliósporos foram coletados em várias localidades em Minas Gerais, confirmando o relato, até então duvidoso, de ocorrência deste tipo de esporo no Brasil. Sua ocorrência pareceu condicionada a mudanças em condições de umidade em períodos de temperatura mais baixa no inverno e na primavera. Uma significativa proporção dos teliósporos apresentou um padrão de germinação anormal com um número de células no promicélio variando de três a sete, com ou sem produção de esterigmas e basidiósporos. O conjunto de observações efetuadas neste trabalho pode contribuir para explicar a ocorrência de um número relativamente grande de raças reconhecido para H. vastatrix que seria de acordo com o conceito usualmente aceito funcionalmente estritamente assexuado, produzindo basidiósporos não funcionais. A existência de um processo adicional para promoção da recombinação genética poderia explicar o aparecimento de novas raças e a continuada quebra de resistência em novas variedades de cafeeiro.

Mining the Aegilops tauschii gene pool: evaluation, introgression and molecular characterization of adult plant resistance to leaf rust and seedling resistance to tan spot in synthetic hexaploid wheat

Doctor of Philosophy / Genetics Interdepartmental Program / Bikram S. Gill / Leaf rust, caused by fungus Puccinia triticina, is an important foliar disease of wheat worldwide. Breeding for race-nonspecific resistant cultivars is the best strategy to combat this disease. Aegilops tauschii, D genome donor of hexaploid wheat, has provided resistance to several pests and pathogens of wheat. To identify potentially new adult plant resistance (APR) genes, 371 geographically diverse Ae. tauschii accessions were evaluated in field with leaf rust (LR) composite culture of predominant races. Accessions from Afghanistan only displayed APR whereas both seedling resistance and APR were common in the Caspian Sea region. Seventeen accessions with high APR were selected for production of synthetic hexaploid wheat (SHW), using ‘TetraPrelude’ and/or ‘TetraThatcher’ as tetraploid parents. Six SHWs were produced and evaluated for APR to LR and resistance to tan spot at seedling stage. Genetic analysis and mapping of APR introgressed from accession TA2474 was investigated in recombinant inbred lines (RIL) population derived from cross between SHW, TA4161-L3 and spring wheat cultivar, ‘WL711’. Genotyping-by-sequencing approach was used to genotype the RILs. Maximum disease severity (MDS) for LR was significantly correlated among all experiments and APR to LR was highly heritable trait in this population. Nine genomic regions significantly associated with APR to LR were QLr.ksu-1AL, QLr.ksu-1BS, QLr.ksu-1BL.1, QLr.ksu-1BL.2, QLr.ksu-2DS, QLr.ksu-2DL, QLr.ksu-5AL, QLr.ksu-5DL and QLr.ksu-6BL. Association of QLr.ksu-1BL.1 with marker Xwmc44 indicated this locus could be slow-rusting APR gene, Lr46/Yr29. QTLs detected on 2DS, 2DL and 5DL were contributed by TA4161-L3 and are novel, along with QLr.ksu-5AL. Tan spot, caused by necrotrophic fungus, Pyrenophora tritici-repentis, has recently emerged as a damaging disease of wheat worldwide. To identify QTLs associated with resistance to Race 1 of P. tritici-repentis, F[subscript]2:3 population derived from cross between SHW, TA4161-L1 and winter wheat cultivar, ‘TAM105’ was used. Two major effect QTLs, QTs.ksu-1AS.1 and QTs.ksu-7AS were significantly associated with tan spot resistance and contributed by TA4161-L1. QTs.ksu-7AS is a novel QTL and explained 17% of the phenotypic variation. Novel QTLs for APR to LR and tan spot identified in SHWs add new variation for broadening the gene pool of wheat and providing resources for breeding of durable resistant cultivars.

Verkorting van die Ae. peregrina-verhaalde Lr59-translokasie van koring

The aim of this study was to analyse testcross-material that was generated during a homoeologous pairing-induction experiment. Absence of the homoeologous pairing suppressor gene, Ph1, was employed to induce meiotic pairing between the Lr59 translocation (Aegilops peregrina) and 1AL of normal wheat. The study aimed to characterize the test-cross plants derived from this experiment and to identify recombinants which retained the least amount of species chromatin but which still contained the Lr59 gene. The test-cross F1 population, 07M5 (total 635 plants), was screened for Lr59 resistance by inoculating seedlings with the leaf rust pathotype, UVPrt8. The 168 resistant plants were characterized with molecular markers in order to identify recombinants. The data were used to construct a physical map which showed the relative sizes of the recombinants and which could be used to identify those recombinants which contained the least amount of residual species chromatin. Microsatellite (Xcfa2219, Xbarc83 and Xgwm164) and SCAR (S15T3) analysis was used for the initial identification of recombinants. The results showed that 152 of the 168 resistant plants were recombinants for the four loci; that eight of the remaining 16 plants represented non-recombinant, wild species-types and that the last eight plants represented the wheat parental-types which were resistant (and thus, also recombinants). This extremely high recombination frequency can largely be attributed to strong segregation distortion that was evident in the cross. It is also possible that the translocation segment could derive from the S genome rather than the U genome of Ae. peregrina. The S genome is closer related to the wheat genomes than the U genome and may be more prone to recombination. With the use of the microsatellite and SCAR data, a physical map was constructed which showed the relative location of the Lr59 gene on the translocation. It appeared that the eight shortest recombinants retained terminal species chromatin. In an attempt to characterize the eight recombinants, additional marker loci had to be identified within that region. RAPD, iv AFLP and DArT markers were investigated for this purpose. RAPD analyses did not produce any useful markers. AFLP and DArT analyses did identify useful markers with which the eight recombinants could be screened. The data showed which recombinants probably retained the least amount of species chromatin. Seeing that AFLP and DArT markers are anonymous and that the distances between marker loci are unknown, it is not possible to say which recombinant is the shortest and consequently it will be nessecary to also evaluate the group of eight recombinants agronomically in order to identify the most useful ones. The results showed that multiple cross-overs apparently occured on both sides of Lr59. Multiple cross-overs are higly unlikely in material of this nature, therefore it was speculated that the observation resulted from incomplete synteny between the telomeric areas of the translocation and 1AL. A structural difference between the two chromosome regions might have given rise to abnormal meiotic pairing structures and thus unexpected gamete genotypes. Each of the eight recombinants did express one or more of the Ae. peregrina derived AFLP loci which can in future be verified for use as a marker for marker assisted selection. The study succeeded in identifying a number of potentially useful recombinants which contain the Lr59 resistance. It would, however, be risky to select only one of the shortest recombinants for further development on the basis of the present knowledge as some recombinants may contain genetic abnormalities which resulted from reduced synteny in the Lr59 region. It would therefore be wise to further evaluate all eight recombinants before the best one is selected for agronomic use.

Mapping and restructuring of an Ae. kotschyi derived translocation segment in common wheat

At Stellenbosch University the leaf and stripe rust resistance genes Lr54 and Yr37 were transferred from Aegilops kotschyi to chromosome 2DL of wheat. In an attempt to reduce the size of the whole-arm translocation on which the resistance genes occur, homoeologous pairing was induced between the wheat and corresponding Ae. kotschyi chromatin. The purpose of this study was to: (i) Evaluate the testcross progeny thus obtained; identify translocation recombinants that retained Lr54/Yr37 and to characterize these using molecular markers (ii) Test for the presence of genes for photoperiod insensitivity (Ppd) and reduced height (Rht) believed to be associated with the translocation (iii) Develop a SCAR marker for the most useful recombinant that could be recovered. Ten putative translocation recombinants were identified following the screening of 159 hemizygous testcross F1 plants with three microsatellite markers specific for chromosome arm 2DL. The recombinants were then characterized with another five microsatellite markers. Using the eight microsatellite markers the recombinants were ordered in two size categories with recombinant #74 being the shortest and having retained only proximal alien chromatin on 2DL. In addition to microsatellite markers, RAPDs, RGAs, AFLPs and SCAR markers were genetically mapped to the translocation and further resolved the recombinants into three size categories. In an attempt to find suitable markers linked to the shortest recombinant (#74) a polymorphic 410 bp AFLP fragment produced with the enzyme/selective nucleotide combination EcoRI – AAC/MseI – CAT, was converted into a dominant SCAR marker. In addition three microsatellite markers that mapped to recombinant #74 provided a useful recessive molecular marker system to detect Lr54/Yr37. Evaluation of the 10 recombinants with four 2DS-specific microsatellite markers revealed a large deletion of this chromosome arm in recombinant #74. This deletion may affect plant phenotypic characteristics and a strategy to replace the deleted region in recombinant #74 is proposed. To test for the presence of a gene for photoperiod insensitivity on the translocation, translocation-carriers plus controls were subjected to long and short day treatments, and the effect on time to flowering was studied. However, no evidence was found for the presence of such a gene. A height experiment to test for the presence of an Rht gene on the translocation confirmed its presence. This gene (designated H) appeared to be different from Rht8 on chromosome 2DS and was mapped on 2DL. While H does not occur in a chromosome region that corresponds with the location of Rht8, it does not rule out the possibility that they could be orthologous loci. Plant height data obtained for recombinant #74 suggested that H was lost through recombination in this particular recombinant. A greenhouse experiment suggested that the full-length translocation increased 100 kernel mass but had a detrimental effect on overall plant yield. Since a much shorter recombinant (#74) has been obtained, this will also have to be evaluated for associated effects. Such an evaluation needs to be done under commercial growing conditions and should involve the comparison of near-isogenic bulks with and without recombinant chromosome #74. The stripe rust resistance gene (Yr37) was mapped by screening hemizygous TF2 progeny of the 10 recombinants with Puccinia striiformis pathotype 6E22A+. Recombinant #74 retained both Lr54 and Yr37 and the two genes therefore occur towards the centromere. / AFRIKAANSE OPSOMMING: Wilde verwante spesies is ‘n belangrike bron van nuwe gene vir die genetiese verbetering van koring. By die Universiteit van Stellenbosch is die blaar-roes en streep-roes weerstandsgene Lr54 en Yr37 vanaf Aegilops kotschyi na chromosoom 2DL van koring oorgedra. ‘n Poging is vervolgens aangewend om die vol-armtranslokasie waarop die weerstandsgene voorkom te verklein deur homoeoloë paring tussen die koring en ooreenstemmende Ae. kotschyi chromatien te induseer. Die doelstelling van hierdie studie was daarom as volg: (a) Evaluering van die verkreë toetskruis-nageslag asook die identifisering en karakterisering van translokasie rekombinante wat Lr54/Yr37 behou het. (b) Toetsing vir fotoperiode onsensitiwiteits- (Ppd) en verkorte plant-hoogte (Rht) gene wat moontlik op die translokasie kon voorkom. (c) Die ontwikkeling van ‘n volgorde-spesifieke polimerase kettingreaksie (PKR) vir die mees bruikbare rekombinant. Tien translokasie rekombinante is geïdentifiseer nadat 159 hemisigotiese toetskruis F1-plante met drie mikrosatelliet-merkers, spesifiek vir chromosoom-arm 2DL, ge-evalueer is. Die rekombinante is hierna met vyf verdere mikrosatellietmerkers getoets. Die data van die agt mikrosatelliet-loci het die rekombinante in twee grootte-kategorieë geplaas waarvan rekombinant #74 die kortste was met slegs die proksimale gedeelte van 2DL wat uit vreemde chromatien bestaan. Behalwe mikrosatellite-merkers is toevallig-geamplifiseerde polimorfiese DNS (RAPD), weerstandsgeen-analoog (RGA), geamplifiseerde volgordelengte polimorfisme (AFLP) en volgorde-gekarakteriseerde geamplifiseerde-streke (SCAR) merkers ook geneties op die translokasie gekarteer. Data van die addisionele merkers het dit moontlik gemaak om die rekombinante in drie grootte-kategorieë te skei. Pogings om ‘n merker vir die kortse rekombinant (#74) te vind, het gelei tot die omskakeling van ‘n 410 bp polimorfiese AFLP-fragment (geproduseer met die ensiem/selektiewenukleotied kombinasie EcoRI - AAC/MseI - CAT), na ‘n dominante, volgordespesifieke PKR-merker. Hierbenewens kan drie mikrosatelliet-merkers wat op rekombinant #74 karteer as resessiewe merkers vir die identifisering van Lr54/Yr37 gebruik word. Die evaluering van die 10 rekombinante met vier chromosoom 2DSspesifieke mikrosatelliet-merkers het ‘n groot delesie van chromosoom-arm 2DS in rekombinant #74 uitgewys. Die delesie mag plant fenotipiese kenmerke beïnvloed en daarom is ‘n strategie vir die vervanging daarvan in rekombinant #74 voorgestel. Ten einde te toets of ‘n geen vir fotoperiode-onsensitiwiteit op die translokaie voorkom is translokasie-draers en kontroles aan lang- en kortdag-behandelings onderwerp en is die effek hiervan op dae-tot-blom gemeet. Geen bewyse vir so ‘n geen kon gevind word nie. ‘n Hoogte-eksperiment om te toets vir die teenwoordigheid van ‘n Rht-geen op die translokasie, het bevestig dat so ‘n geen wel voorkom. Die geen (voorgestelde simbool H) is gekarteer op 2DL en verskil oënskynlik van Rht8 op chromosoom 2DS. Die verskillende chromosoom-ligging van H en Rht8 skakel egter nie die moontlikheid dat hulle ortoloë loci mag wees uit nie. Plant-hoogte data vir rekombinant #74 het daarop gedui dat H nie meer in hierdie rekombinant voorkom nie. Data van ‘n glashuis-eksperiment het daarop gedui dat die vollengte-translokasie 100-korrel-massa verhoog maar dat dit plant-opbrengs verlaag. Aangesien ‘n aansienlike korter rekombinant (#74) verkry is, sal dit ook vir gekoppelde effekte getoets moet word. So ‘n evaluering moet egter onder kommersiële toestande gedoen word met gebruik van naby isogeniese-lyne met en sonder rekombinante chromosoom #74. Die streep-roes weerstandgeen (Yr37) is gekarteer deur hemisigotiese TF2- nageslag van die 10 rekombinante te toets vir weerstand teen Puccinia striiformis patotipe 6E22A+. Rekombinant #74 het beide Lr54 en Yr37 behou en die twee gene karteer dus naby die sentromeer.

Análise dos indicadores do momento da primeira aplicação de fungicida visando ao controle de doenças foliares em trigo / Analysis of the indicators for timing fungicide application on wheat to control foliar diseases

The aim of this work was to analyse the available criteria to the technical assistance concerning to the decisive making about the moment of the fungicide application. The experiments were carried out at field, in the 2011 and 2012 growing seasons in Lages, Santa Catarina State, with the Mirante cultivar wheat. Were tested the following criteria: 1) witness (without fungicide spraying); 2) 1% of incidence of plants; 3) 10% of incidence of plants; 4) 5% of foliar severity; 5) main stem with from five to six tillers; 6) first visible node; 7) third visible node; 8) Economic Damage Threshold (EDT). The reapplications were made based on the period of fungicide mixture protection of azoxystrobin + cyproconazol (60g + 24g a.i. ha-1) + propiconazol (125g a.i. ha-1). The experimental unities were composed by 1.5 x 5 m plots arranged in a randomized blocks design with four replications. The applications were made with a coastal sprayer with pressure generated by CO2 gas delivering 200 L ha-1. In the economic analysis was taken in account the cost of applications, the number of applications, the price of wheat, the disease intensity (area under the disease progress curve AUDPC), control effectiveness based on the intensity disease and components of yield (productivity, mass of one thousand grains and weight of the hectoliter). The data were submitted to ANOVA with means compared by the Tukey`s test. In the two growing seasons, leaf rust, yellow spot and powdery mildew were the predominant diseases. The AUDPC values showed that the criteria 1% and 10% of incidence of plants and from five to six tillers presented lower intensities of disease, with higher control and higher yields, however, they received from three to four applications. The EDT criteria received two to three applications and obtained intermediate values, as well as, the criteria of first visible node that presented intermediate values for the means of incidence and severity and also for the productivity. The criteria of third visible node and 5% of foliar severity began the chemical control later, receiving from one to two applications and showed higher means of incidence and severity, lower control, as well as, a lower productivity of grains / A aplicação de fungicidas no controle de doenças foliares do trigo aumenta o custo de produção e pode contribuir para contaminar o meio ambiente caso sejam feitas aplicações desnecessárias. O objetivo deste trabalho foi analisar os critérios disponíveis para a assistência técnica sobre a tomada de decisão para o momento da aplicação de fungicida. Os experimentos foram conduzidos no campo nas safras agrícolas de 2011 e 2012, em Lages, Santa Catarina, com a cultivar de trigo Mirante. Foram testados os seguintes critérios: 1) Testemunha (sem aplicação de fungicida); 2) 1% de incidência de plantas; 3) 10% de incidência de plantas; 4) 5% de severidade foliar; 5) Colmo principal com cinco a seis afilhos; 6) Primeiro nó visível; 7) Terceiro nó visível; e 8) Limiar de Dano Econômico (LDE). As reaplicações foram feitas com base no período de proteção da mistura dos fungicidas azoxistrobina + ciproconazole (60g + 24g de i.a. ha-1) + propiconazole (125g de i.a. ha-1). As unidades experimentais constaram de parcelas de 1,5 x 5 m arranjadas em delineamento de blocos ao acaso com quatro repetições. As aplicações foram feitas com pulverizador costal com pressão gerada por gás CO2 para equivaler uma vazão de 200 L ha-1. Na análise econômica considerou-se o custo das aplicações, o número de aplicações, o preço do trigo, a intensidade de doenças (área abaixo da curva de progresso de doença - AACPD), a eficácia de controle com base na intensidade de doença e os componentes de rendimento (produtividade, massa de mil grãos e peso do hectolitro). Os dados foram submetidos à ANOVA e as médias comparadas pelo teste de Tukey. Nas duas safras as doenças predominantes foram ferrugem da folha, mancha amarela e oídio. Os valores da AACPD demonstraram que os critérios 1% e 10% de incidência em planta e cinco a seis afilhos apresentaram menores intensidades de doença, com maior controle e maiores produtividades, porém receberam de três a quatro aplicações. Os critérios do primeiro nó visível e LDE receberam de duas a três aplicações e apresentaram valores intermediários para incidência, 8 severidade e produtividade. Os critérios do terceiro nó visível e 5% de severidade foliar iniciaram o controle químico mais tardiamente, recebendo de uma a duas aplicações e demonstraram maiores médias de incidência e severidade, menor controle, bem como menor produtividade de grãos
