Well defined stimuli-responsive cross-linked micelles as biocompatible drug/gene delivery system from RAFT polymerization

The objective of this thesis is to investigate well-defined cross-linked particles synthesized via the reversible addition fragmentation chain transfer (RAFT) process that can be used for drug delivery. To achieve this aim, a wide range of cross-linked micelle systems have been synthesized and intensively investigated. Various biocompatible monomers were employed, including poly (ethylene glycol) methyl ether methacrylate, 2-hydroxyl ethyl acrylate, functionalized glucosamine and nucleotides containing monomers. Different cross-linked structures were used, for example, core-cross-linked, nexus-cross-linked and shell-cross-linked micelles. Diverse stimuli-responsive particles were used, such as pH-sensitive, thermo-sensitive and thiol-sensitive cross-linked systems. Evidences of the successful synthesis of all the resulting cross-linked products are given. They displayed better properties, as drug carriers, than non-cross-linked micelles. A thermo-responsive seven-arm star glycopolymer, synthesized via the RAFT process, was also investigated.

Glycosylation by Chinese Hamster Ovary Cells in Dolichol Phosphate-Supplemented Cultures

N-linked glycosylation often imparts important properties to protein therapeutics. An essential step in this intracellular process is the transfer of oligosaccharide from dolichol monophosphate (Dol-P) to a potential glycosylation site. Variability in the success rate of this reaction affects the extent of protein glycosylation. The critical role of Dol-P suggests that its availability may influence the extent of glycosylation by limiting the pool of lipid-linked oligosaccharides (LLOs), the glycosyl donor. To test this hypothesis, the impact of Dol-P supplementation on protein glycosylation in Chinese hamster ovary (CHO) cells was investigated. Although exogenous Dol-P was incorporated by CHO cells and processed into LLOs in a dose-dependent manner, Dol-P supplementation had no marked effects on LLO or overall cellular glycosylation levels. While concentrations of exogenous Dol-P exceeding 100 µg/ml were detrimental to CHO cell viability, maximum non-toxic supplemental doses of Dol-P had no significant impact on the glycosylation of recombinant interferon-γ produced by batch cultures of CHO cells. These results show that glycosylation in CHO cells cannot be readily enhanced by Dol-P feeding under normal culture conditions. / Singapore-MIT Alliance (SMA)

Development of an enzyme-linked immunosorbent assay for the serologic diagnosis of bovine adenovirus type 3

An enzyme-linked immunosorbent assay was developed to measure specific antibody response in bovine sera to bovine adenovirus type 3 (BA3), an etiologic agent of respiratory disease causing economic losses annualy to the cattle industry. Observed endpoint titers were determined using the intersection point from optical density values of serially diluted sera with a positive-negative threshold. Regression equations were determined from standards with titers ranging from low to high and used to predict ELISA titers from a single-serum dilution. A near-linear relationship existed between the observed and predicted ELISA titers of 118 bovine sera (r=0.9261). Predicted ELISA titers were determined using the single-dilution method for another 76 bovine sera and the correlation between the ELISA titers and serum-virus neutralization titers for these sera indicated a strong linear trend (r=0.8172). Both the ELISA and serum-virus neutralization titers on the bovine sera tested indicated widespread exposure to several types of bovine adenovirus. Although detection of active infection would still require examination of sera over time for evidence of a rising titer, the single-dilution ELISA devised should provide a rapid and sensitive method for detection of antibody response to bovine adenovirus type 3. / Graduation date: 1992

Financial Knowledge Education and Training System for Agents Selling Investment-Linked Insurance

Investment-Linked Insurance was introduced to the Taiwan market in 2001. It has since influenced the way insurance sales agents sell the products. Because of the level of complication of Investment-Linked Insurance compared with traditional insurance, and professional knowledge and skills needed during the selling process, an insurance sales agent needs to improve his competence in order to become a financial advisor for his customers. However, literature review shows that most researchers focus on how to improve customer satisfaction, service quality, customer loyalty or the impact of training on sales. There are so few studies related to the development of a training system specifically designed for selling Investment-Linked Insurance. According to the results from surveys, almost sixty percent of sales agents consider the ability of gathering and analyzing information or the understanding of political or economical status the skills they need the most. Moreover, almost fifty percent of sales agents believe the investment performance in the past does not affect the decision of customers for the purchases of Investment-Linked Insurance. At the same time, seventy percent of sales agents admit that customers tend to stay off Investment-Linked Insurance because of risk. Not only do surveys show that sales agents need more skills, but also interviews indicate that sales agents are afraid that they can not improve their competence without a systematic training. Therefore, we classify the knowledge and skills needed during the sale process for Investment-Linked Insurance. After the basic training sessions, sales agents will take five training modules, including data collection, interpretation of political or economical status, investment portfolio allocation, investment performance review and funds analysis. At the same time, sales agents follow the training steps of identification, learning, practices and articulation in order to get ready for Investment-Linked Insurance. The benefits of this training system for Investment-Linked Insurance are two-folds, including the increase of time and cost efficiency for training and the enhancement of professional knowledge or skills.
