This master's thesis deals with the response of the Federal Republic of Germany to the Russian hybrid threat, which is examined using a case study of the so-called "Lisa case" from January 2016. The thesis aims to place the "Lisa case" in the context of German-Russian relations and hybrid threats in the security reality of the 21st century. Germany and Russia have long had a special relationship, which, however, fell into crisis after the Russian annexation of the Crimean Peninsula. The main goal of Russia's efforts to influence social and political development in Europe has become Germany, which must deal with new methods and strategies of hybrid warfare. The advent of hybrid threats is a challenge not only for Germany but also for its allies. Russia's view of hybrid threats proves that the term has different meanings for different actors. One of the manifestations of the Russian hybrid threat is the case of a thirteen-year-old girl of German-Russian origin Lisa F. who was missing in Berlin for several hours and allegedly raped by migrants. Although the German police promptly refuted the case, the Russian media misused the event to fuel an atmosphere of fear and uncertainty, especially among a significant minority of Russian Germans. This includes research, character analysis, rehearsal journal and an evaluation of my performance. The Three Sisters was produced by the University of New Orleans Department of Film, Theatre and Communication Arts in New Orleans, Louisiana. The play was performed in the Thrust Theatre of the Performing Arts Center at 8:00 pm on the evenings of April 19 through 21 and April 26 through 28, with one matinee at 2:30 pm on Sunday, April 29, 2007.

Collaborative Justice: An Analysis of the Use of "Justice" in the Writings of Lisa Sowle Cahill

This dissertation explores the role of justice in the writings of Catholic ethicist Lisa Sowle Cahill. Since 1990, Cahill has supported theological voices participating in the public forum, which she describes as a meeting ground for diverse intellectual and religious traditions. Good argumentation is necessary but not sufficient to resolve ethical dilemmas within the politically liberal context in which Cahill makes her claims. Instead, her commitment to justice underwrites those narratives and practices which demand one's fullest possible participation in contributing toward the common good. Cahill's notion of justice develops correlatively to the degree that she integrates the principles of Catholic Social Teaching into her project. The dissertation describes this expansion in Cahill's later essays as "collaborative justice." The dissertation concludes with an examination of her writings on human genetic engineering as a potential application of collaborative justice. Cahill's strong arguments can be helpful in steering the process toward less-harmful outcomes. In doing so, Cahill's principles of collaborative justice look beyond act-focused considerations of Catholic ethics or the procedural justice of liberal traditions, and leaves open the possibility of reconciliation should such future genetic intervention prove undesirable. / McAnulty College and Graduate School of Liberal Arts; / Theology / PhD; / Dissertation;

The pharmacology of SVT-40776, a new muscarinic antagonist for the treatment of urinary incontinence

L'estudi està centrat en la caracterització farmacològica d'un nou fàrmac per la incontinència urinària per hiperactivitat de la bufeta, anomenada Overactive Bladder (OAB). És un problema clínic molt comú, que s'origina quan existeix una disfunció de les vies de senyalització del sistema nerviós central o perifèric, de l´uroteli, del múscle llis o altres components del teixit. Quan el control conscient del reflex parasimpàtic de la micció es alterat, apareixen els símptomes de la OAB, creant un seriós problema de salut, amb gran impacte social i psicològic. Existeixen 7 fàrmacs disponibles, antagonistes muscarínics no selectius o lleugerament selectius vers el receptor M3: oxibutinina (OXY), tolterodina (TOL), propiverina, fesoterodina, trospium, darifenacina (DAR) i solifenacina (SOL). Tots ells han demostrat eficàcia per tractar els símptomes de la OAB, però la seva farmacocinètica i perfil d'efectes secundaris fa encara necessari disposar de fàrmacs més segurs al mercat. L'objectiu d'aquest treball ha estat caracteritzar la farmacologia del SVT-40776, com un fàrmac potencialment més segur pel tractament de la OAB. Postulàvem que un antagonista muscarinic sense afinitat per el receptor M2 seria predictiu d'un perfil cardiovascular més segur. A més, volíem demostrar que un antagonisme sobre M2 no es requereix per aconseguir efectivitat dels antagonistes no selectius inhibint les contraccions colinèrgiques observades en els malalts de OAB. La afinitat i selectivitat del SVT-40776 pel receptor humà M3 va ser determinat per estudis de fixació amb [3H]NMS en membranes de cèl.lules CHO que expresaven els receptors clonats humans. SVT-40776 va mostrar tenir l'afinitat (Ki= 0.19 nM) i selectivitat superior (203-fold vs. M2) pel receptor humà M3, en comparació amb els altres antimuscarincs testats (OXY, TOL, DAR, SOL). SVT-40776 va mostrar-se com un antagonista reversible. En els estudis funcionals in vitro en teixit aïllat (detrusor de bufeta de ratolí), SVT-40776 va inhibir les contraccions induïdes per un agonista colinèrgic (carbacol, CCh), produint un desplaçament de les corbes acumulatives dosi-resposta d'aquest agonista (pA2 = 9.5). L'activitat antagonista per TOL, SOL i DAR va ser de 8.4, 8.6 i 8.7, respectivament. En preparacions d'atria (efecte mediat per M2), les corbes de CCh van ser antagonitzades per tots els compostos. El rang de potència dels antagonistes (pA2) va ser TOL (8.5), SOL (7.8), DAR (7.3) i SVT-40776 (7.3). SVT-40776 va mostrar el marge més gran de selectivitat bufeta vs. atria (x199), predient un millor perfil cardiovascular de seguretat comparat amb els altres antagonistes. En els estudis funcionals de teixit aïllat ex vivo en ratolí (obtenint el teixit 3h post-administració oral) SVT-40776 va inhibir concentració-depenent les contraccions induïdes per CCh. El pA2-ED va ser 0.72 mg/kg en bufeta i 40.3 mg/kg en atria, obtenint un marge cor vs. bufeta de 58. Això compara favorablement amb TOL (0.21), SOL (1.5) i DAR (2.4). El conill porquí va ser l'animal escollit pels estudis in vivo donat que la proporció de receptors M3:M2 (1:3) era similar a la humana (1:3) comparada amb la rata (1:9), així com per la similitud morfològica i funcional a la uretra humana. Com a conclusió general, SVT-40776 va inhibir dosi-resposta les contraccions de micció en els dos models usats, el isovolumètric i el cistomètric, sense afectar a la pressió arterial i/o a la freqüència cardíaca. Així, l'ordre de mes a menys potencia segons els resultats de fixació al receptor humà va ser SVT-40776>DAR >TOL>SOL. Pels estudis funcionals va ser SVT-40776>DAR > SOL> TOL.Així, aquest estudi mostra que SVT-40776 és l'antagonista muscarinic més potent i selectiu M3 vs. M2 descrit fins ara pel tractament de la OAB. El perfil confirma que M2 no és necessari per aconseguir modulació de la bufeta. El compost està actualment en Fase Clínica II. / The study is focused on the pharmacological characterization of a new drug forthe treatment of urgency urinary incontinence, a pathology considered to be due to a hyperactivity of the bladder and known as Overactive Bladder (OAB). This is a common clinical problem which may originate from dysfunction of the peripheral or central nervous pathways, the urotehlium, the smooth muscle and other tissue components. When conscious control of the parasympathetic micturition reflex is altered, symptoms of OAB arise, creating a serious health problem commonly giving rise to severe psychological and social problems. Worldwide, there are only seven clinically available drugs, all non-selective or poorly selective antimuscarinics, for the treatment of OAB: oxybutynin (OXY), tolterodine (TOL), propiverine, fesoterodine, trospium, darifenacin (DAR) and solifenacin (SOL). All have demonstrated efficacy for the treatment of OAB symptoms but their pharmacokinetic and adverse event profiles differ and provide the need for new, safer treatments. The main objective of this investigation was to characterize the pharmacology of SVT-40776, a novel substituted quinuclidine derivative, as a potential new drug aimed at treating OAB. We postulated that a M2 sparing muscarinic antagonist would be predictive of a safe cardiovascular profile. In addition, we wished to prove that M2 antagonism is not a requirement to achieve the effectiveness of the classical non-selective muscarinic antagonists. The affinity and selectivity of SVT-40776 for the human M3 mACh receptor subtype was determined by [3H]NMS receptor binding competition in CHO cell membranes expressing human muscarinic receptors. SVT-40776 showed the highest affinity (Ki= 0.19 nM) and selectivity (203-fold vs. M2) for the human M3 receptor subtype, in comparison with other muscarinic antagonists. SVT-40776 was a reversible antagonist. In functional studies using isolated mice bladder SVT-40776 showed the ability to induce a rightward parallel shift in the cumulative agonist concentration-response curves, obtaining a pA2 of 9.5. Antagonist activities of TOL, SOL and DAR were 8.4, 8.6 and 8.7, respectively. In isolated mice atria preparations (M2 mediated effect), CCh curves were surmountably antagonized by all the compounds. The rank order of antagonist activities (pA2) was TOL (8.5), SOL (7.8), DAR (7.3) and SVT-40776 (7.3). SVT-40776 exhibited the highest bladder vs. atria selectivity (199-fold), therefore predicting a safer cardiovascular profile than the other antagonists tested. In the ex vivo isolated tissue functional studies (obtaining the sample 3h post-oral dosing), SVT-40776 inhibited CCh-induced bladder contractions in a concentration-dependent manner after oral administration in mice. The pA2-ED was 0.72mg/kg in bladder and 40.3mg/kg in atria, giving a 58-fold ratio between muscarinic cardiac effects vs. bladder effects, which compared favorably with TOL (0.21-fold), SOL (1.5-fold) and DAR (2.4-fold). Guinea pig was used for in vivo studies, as M3:M2 receptor population proportion (1:3) was more similar to human (1:3) compared to rat (1:9) (Fetscher et al., 2002) and also due to the similarity of its urethra morphology and functionality to human urethra (Walters et al., 2006). As a general conclusion, SVT-40776 did inhibit micturition contractions in the two models used, the isovolumetric and the cystometric, without affecting arterial pressure (MAP) and/or heart rate (HR). Thus, for binding affinities the order (greater to lesser affinity) is SVT-40776>DAR>TOL>SOL, whereas for functional in vivo potency the order is SVT-40776>DAR > SOL> TOL.Therefore, the present study has shown that SVT-40776, is the most potent and selective M3 vs. M2 muscarinic receptor antagonist described so far for the treatment of overactive bladder. Its activity profile confirms the hypothesis that M2 receptor is not necessary for achieve effective bladder modulation. The compound has successfully completed Phase I clinical trials and is currently undergoing Phase II clinical trials for the treatment of overactive bladder.

This essay examines the relationships between the different elements in Lisa Jonasson’s collage Existentiell Progg #6 and her very detailed technique. The essay has as its aims to understand and analyze both the details and the wholeness of the artwork and to try to put the collage in an art historical context. The thesis finds that Existentiell Progg #6 has a connection to the imagery of Africa and other collage artist such as Hannah Höch and Nancy Spero, which also gives the paper a perspective of feminism in the arts. The paper continues to create an understanding for the collage work by putting it in to Theodor Adorno’s theories about nature, the human being and the animal kingdom. The paper then comes to the conclusion that many of the elements in Existentiell Progg #6 are a part of its underlying meaning and purpose.
