The master’s thesis describes known methods of signal measurement using principle of synchronous detection. Various methods are presented, their principles are examined and the problems when using them are analyzed. Further, procedures for reduction of adverse effects are described also. Second part of this thesis is focused on the instrument design. The instrument is intended for detection and processing of the output signals in a given optometric system. The proposed device is designed to operate on the principle of synchronous detection method using a vector signal evaluation. Advantages and disadvantages are discussed below.

This thesis deals with diagnostic fuel-cell parameters using electrochemical impedance spectroscopy. The document begins with brief explanation of fuel-cell operation and its basic principles. Thesis continues with description of fuel-cell voltage generation and influence of charge dual-layer on impedance. After that electrochemical impedance spectroscopy, signals used for perturbation and evaluation of impedance spectrum using Bode a Nyquist plot is described. Next part explains measurement of impedance spectrum using potentiostat in combination with lock-in amplifier or frequency response analyzer. Practical part of thesis is dedicated to electric circuit design of EIS module and software designed for automatic measurement of impedance spectrum. Last part shows results of EIS module testing.

The thesis is focused on the development of the electrochemical system with microfluidic platform for the detection of multiple biomarkers. It analyses the use of biomarkers for the early diagnosis of cancer. The theoretical part contains basic information about voltammetric methods and microfluidic systems. The practical part provides solutions to the microfluidic chips, including the description of the used materials, designs, methodologies of preparation and conclusions from the testing of the manufactured microfluidic systems. The thesis describes the lock-in electrochemical system which measures the response of 4 electrochemical cells simultaneously. For the electrochemical system measurements, an electrochemical chip consisting of 64 electrochemical cells was used. The results of the analysis include the processing of the system tests and detected voltammetric curves of the Fe2+/Fe3+ solution and cysteine.

This thesis addresses the issue of adopting cloud computing in two aspects. The first being the procedure of choosing a commonality that constitutes the base of comparing the most appropriate cloud platform from identified parameters. The second being to find constraints of adopting cloud computing related to the commonality. This commonality was identified in form of a process that portrayed characteristics which made it suitable for identifying parameters and comparing two cloud platforms (Azure and AWS). The comparison of platforms based on the parameters; lock-in, standardization and data security led to conclusive remarks that discouraged any further effort on comparing cloud platforms. Lack of standards and lock-in that cannot be avoided are traits of the unfitting environment the cloud platforms constitute. Furthermore, it was concluded that the adoption constraints are first priority when it comes to adopting cloud computing. Instead of choosing the cloud platform with least amount of constraints, it is recommended to firstly focus on the adoption constraints. / Avhandlingen tar upp frågan om att införa molntjänster i två aspekter. Den första är proceduren att välja en gemensam nämnare som utgör basen för jämförelse av den lämpligaste moln plattformen. Den andra är att hitta begränsningar relaterat till införandet av molntjänster i samband med den gemensamma nämnaren. Denna gemensamma nämnare identifierades i form av en process inom en IT-avdelning som skildrar egenskaper vilket gjorde den lämplig för att hitta parametrar och jämföra två molnplattformar (Azure och AWS). Jämförelsen av plattformar baserades på parametrarna; lock-in, standardisering och datasäkerhet. Brist på standarder och lock-in som inte kan undvikas är egenskaper av den opassande miljön som molnplattformarna utgör. Dessutom drogs slutsatsen att de nämnda begränsningar är av första prioritet när det gäller att införa molntjänster. Istället för att välja en molnplattform med minsta möjliga begränsningar, är rekommendationen att först fokusera på begränsningarna.

Plutt: A tool for creating type-safe and version-safe microfrontends

Microfrontend applications are composed of multiple smaller frontend applications, which are integrated at run-time. As with microservices, microfrontends can be updated in production at any time. There are no technological restrictions for releasing API-breaking updates. Therefore it is difficult to trust microfrontend applications to perform reliably in run-time and to introduce API-breaking updates without the risk of breaking consumers. This thesis presents Plutt, a tool that provides automatic guarantees for safely consuming microfrontends, by ensuring that updates in run-time are compatible. By using Plutt, consumers can be confident that a provided microfrontend will per- form the same during production as in development. Likewise, microfrontend providers can release updates without being concerned about how it will affect consumers. Moreover, a comprehensive survey about microfrontends is presented, where five industry experts are interviewed. Aspects that are not found in existing literature are discovered, which contributes to a broader knowledge base that helps future microfrontend research. / Mikrofrontend-applikationer är sammansatta av flera mindre frontend-applikationer som integreras under exekvering. Precis som med mikrotjänster, kan mikrofrontends bytas ut i produktion när som helst. Det saknas teknologiska restriktioner för att publicera API-brytande uppdateringar. Därför är det svårt att lita på att en mikrofrontend-applikation beter sig tillförlitligt under exekvering samt att introducera API-brytande uppdateringar utan att riskera att förstöra konsumenter. Det här examensarbetet presenterar Plutt, ett verktyg som erbjuder automatiska garantier för att säkert konsumera mikrofrontends genom att säkerställa att uppdateringar som introduceras i körtid är kompatibla. Genom att använda Plutt, kan konsumenter vara trygga i vetskapen att en försedd mikrofrontend presterar likadant under produktion som i utveckling. This paper proposes a communication scheme to remote control multiple mobile targets using Coded-Division Multiple Access(CDMA) technique. It's feasibility, advantage and shortcoming are analyzed. Moreover, the key techniques of Direct-Sequence Spread Spectrum(DS/SS) system, i.e. the correlation detection and delay lock-on techniques, are studied and stimulated on the experimental model. The results of theoretical analysis show that the CDMA system has the peculiar advantage over the conventional multiple access system, such as FDMA and TDMA.

Product strategies under durability, lock-in and assortment considerations

In this dissertation I focus on two considerations that influence the product strategy of a firm. The first is consumers’ choice and its influence on a firm’s product offering, and the second is the interaction between durable products and their contingent consumables. First, I study the assortment planning problem for a firm; I illustrate the complexity of solving this product selection problem, present simple solutions for some commonly used choice models, and develop heuristics for other practically motivated models. Second, I study the incentives of a durable goods monopolist when she can lock-in consumers through a contingent consumable. Adopting a lock-in strategy has two interesting effects on the incentives of a durable goods manufacturer. On one hand, by locking-in consumers to its consumable, a durable goods monopolist can curb its temptation to reduce durable prices over time, thereby mitigating the classic time inconsistency problem. On the other hand, lock-in will create a hold-up issue and adversely affect consumers’ expectations of future prices for the consumable. My research demonstrates the trade-off between time inconsistency and hold-up, and derives insights about the conditions under which a lock-in strategy can be effective. I further analyze the trade-off between time inconsistency and hold-up associated with lock-in in the presence of consumable stock-piling. My findings indicate in the presence of consumer stock-piling, lock-in has an effect similar to that of competition in the consumables market: they help to dampen the hold-up problem that arises from lock-in and at the same time increase the manufacturer’s incentive to reduce durable prices over time. / text

Three Essays on the Health Insurance Coverage of Young Adults

This dissertation examines the health insurance status of young adults during the transition to adulthood. In a series of three essays, I analyze what happens as young adults reach important milestones and the effects of public policies. The first essay is a descriptive study on how insurance status changes after reaching age 19 and graduating from college. The likelihood of becoming uninsured rises sharply once turning age 19 and then peaks at age 23. While the proportion uninsured also increases following college graduation, this increase disappears after one year. The second essay analyzes the effect of a dependent age law in New Jersey, which allowed dependent coverage for young adults up to age 30 and did not require full-time student status. Pennsylvania did not pass a law and was used as a control state. Among 19-to-22-year olds, there was a rise in health insurance coverage in New Jersey relative to Pennsylvania. There also was a negative effect on college enrollment in New Jersey relative to Pennsylvania. The final essay considers other unintended consequences of dependent age laws. Using a national dataset, I estimate that there were no clear effects on decisions related to living arrangements, marriage, and full-time employment.

Characterization of the Effect of Serum and Chelating Agents on Staphylococcus aureus Biofilm Formation; Chelating Agents Augment Biofilm Formation through Clumping Factor B

Staphylococcus aureus is the causative agent of a diverse array of acute and chronic infections, and some these infections, including infective endocarditis, joint infections, and medical device-associated bloodstream infections, depend upon its capacity to form tenacious biofilms on surfaces. Inserted medical devices such as intravenous catheters, pacemakers, and artificial heart valves save lives, but unfortunately, they can also serve as a substrate on which S. aureus can form a biofilm, attributing S. aureus as a leading cause of medical device-related infections. The major aim of this work was take compounds to which S. aureus would be exposed during infection and to investigate their effects on its capacity to form a biofilm. More specifically, the project investigated the effects of serum, and thereafter of catheter lock solutions on biofilm formation by S. aureus. Pre-coating polystyrene with serum is frequently used as a method to augment biofilm formation. The effect of pre-coating with serum is due to the deposition of extracellular matrix components onto the polystyrene, which are then recognized by MSCRAMMs. We therefore hypothesized that the major component of blood, serum, would induce biofilm formation. Surprisingly, serum actually inhibited biofilm formation. The inhibitory activity was due to a small molecular weight, heat-stable, non-proteinaceous component/s of serum. Serum-mediated inhibition of biofilm formation may represent a previously uncharacterized aspect of host innate immunity that targets the expression of a key bacterial virulence factor: the ability to establish a resistant biofilm. Metal ion chelators like sodium citrate are frequently chosen to lock intravenous catheters because they are regarded as potent inhibitors of bacterial biofilm formation and viability. We found that, while chelating compounds abolished biofilm formation in most strains of S. aureus, they actually augmented the phenotype in a subset of strains. We investigated the molecular basis of this phenomenon. Deletion and complementation analysis and thereafter antibody based inhibition assays confirmed a functional role for the surface adhesin clumping factor B as the causative determinant associated with the increased biofilm phenotype. Finally, we investigated the regulation of clumping factor B-mediated biofilm formation and the basis for the strain dependence. Regulation was determined to occur via two novel post-translational networks- one affecting ClfB activity, mediated by Ca2+ binding to the EF-Hand domain, and the other affecting protein stability, mediated by the enzymatic activity of the metalloprotease-aureolysin. Polymorphisms within the aureolysin gene sequence, between strains, was identified as the basis for some strains forming robust biofilms within chelated media versus other than do not exhibit this phenotype.
