Plagiorchis elegans in the molluscan intermediate host : infection, susceptibility, growth, reproduction, mortality and cercarial production

The host/parasite association between the pulmonate snail Stagnicola elodes and the digenean Plagiorchis elegans was examined with a view to identifying some of the biotic and abiotic factors that govern parasite egg development, infectivity and survival, the dynamics of cercarial production, as well as host susceptibility to infection, growth, reproduction and mortality. Plagiorchis elegans eggs passed with the feces of the hamster (Mesocricetus auratus) experimental definitive host were unembryonated. Between 0 and 30 C, the rate of embryonation continued to increase with temperature, whereas infectivity and survival were optimal between 10 to 20 C. Light adversely affected embryonation. Parasite acquisition increased with the intensity of exposure and sexual maturity. Infection invariably enhanced pre-patent host growth in a dose-dependent manner. Cercarial production increased with time and reached a plateau. Snails infected prior to sexual maturity released more cercariae than mature individuals. Heavily infected snails tended to die prematurely regardless of age at infection and parasite dose, thereby reducing their total production of cercariae to levels below those of more lightly infected individuals. Even light infections castrated the snail host. Snails infected when immature never reproduced, whereas reproductive snails ceased egg production within days of infection. Similar effects were observed in the incompatible snail host Biomphalaria glabrata, although no cercariae were ever produced. Both enhanced growth and parasitic castration are attributable to the mother sporocyst.

Pseudosuccinea columella est un des principaux hôtes intermédiaires de Fasciola hepatica, un trématode cosmopolite infectant l’Homme et les animaux domestiques et sauvages, et transmis par des mollusques d’eau douce. L’existence exclusive à Cuba de populations de P. columella qui sont sensibles et naturellement résistantes (encapsulation hémocytaire du parasite) à l’infection par F. hepatica offre un modèle Mollusque-Trématode très intéressant en termes de biologie évolutive, de santé humaine et de stratégies de contrôle vectorielle. L’étude des aspects écologiques, moléculaires et phénotypiques est essentielle pour comprendre les particularités de ce système. Ainsi, nos résultats ont montré que les mollusques sensibles et résistants partagent des exigences écologiques similaires mais chez les résistants la distribution est limitée aux localités avec des eaux acides et peu dures (pH/dureté totale (DT), 6-6.5/4°-10°d) et une richesse des communautés de mollusques très faible, ceci semblant indiquer coût élevé à la résistance. La comparaison de traits d’histoire de vie en laboratoire a permis de confirmer une haute tolérance aux variations de pH/DT de l’eau chez les mollusques résistants mais a montré également une diminution du potentiel reproductif des souches résistantes par rapport aux sensibles. Sur la base de ces observations, des approches «omiques» comparatives ont été entreprises entre souches sensibles et résistantes. Ce travail de thèse nous a aidé à comprendre les particularités de ce modèle hôte-parasite, mais présente également une avancée en termes d’utilisation de la résistance de P. columella à des fins de contrôle de la transmission parasitaire. / Pseudosuccinea columella is one of the main intermediate hosts of Fasciola hepatica, a cosmopolitan snail-borne trematode that affects humans, livestock and wildlife. The occurrence in Cuba of susceptible and naturally-resistant populations of this species to F. hepatica infection (host’s hemocytes encapsulate the parasite upon penetration) offers an interesting Mollusca - Trematoda model with applications on evolutionary biology, health sciences and vector control strategies. Thus, here we explored different ecological, molecular and phenotypical aspects of this system to better understand P. columella resistance. We determined that while susceptible and resistant snails share similar ecological requirements, resistant populations occurred only at low pH (6.5-6.5) and total hardness (TH; 4°-10°d) waters, with low snail species richness, suggesting a high ecological cost of resistance. Comparison of life history traits between susceptible and resistant snails, experimentally-reared at low or common pH/TH conditions, showed that resistant populations display higher tolerance to pH/TH variations and a lower reproductive potential. To gain more insights on the molecular bases of the features associated to resistance in P. columella, we performed comparative “omics” approaches on naïve snails from both phenotypes at whole snail level (RNAseq) and at the albumen gland level (2D-electrophoresis). This thesis presents the latest efforts to broadly characterize this model, which constitute building steps for the comprehension of P. columella resistance and for its application to tackle parasite transmission

Ein weiteres Ziel der Analysen war herauszufinden, inwiefern eigene Rekonstruktionen phylogenetischer Bäume, basierend auf Kern- und Mitochondrienmarkern, bisher bestehende Hypothesen zur Taxonomie und Systematik der Lymnaeidae bestätigen oder widerlegen. Erste eigene molekulargenetisch überprüfte morphologische Analysen bei paläarktischen Vertretern der Gattung Radix zeigen, dass die zur Artdifferenzierung etablierten Merkmale Gehäuseform, Mantelpigmentierung, Längenverhältnis der beiden Abschnitte des männlichen Kopulationsapparates (Praeputium und Phallotheca), Länge des Bursaductes und Form der Bursa copulatrix eine höhere innerartliche Variabilität aufweisen als eigentlich für die einzelnen Arten angenommen wurde. Sie sind, entgegen bisheriger Annahmen, zu einem großen Teil nicht artspezifisch. Molekulargenetisch deutlich unterscheidbare Arten können die gleiche Gehäuseform und Mantelpigmentierung aufweisen und sind oft auch mittels anatomischer Merkmale nicht eindeutig unterscheidbar. Es kann, auch anhand erster eigener Studien an anderen Vertretern der Familie Lymnaeidae, geschlussfolgert werden, dass von den meisten Malakologen bisher in diese Merkmale innerhalb der Schlammschnecken eine Bedeutung für Taxonomie und Systematik hineininterpretiert wurde, die sie offensichtlich auf Grund ihrer hohen Plastizität nicht besitzen. Die molekulargenetischen Methoden eröffnen hier neue Perspektiven für ihre Beurteilung sowie für die objektivere Abgrenzung von Arten und Gattungen. Aim of the studies was to analyse the range of the intraspecific variability of some representatives of the family Lymnaeidae as well as to find out which value should be given to the morphological and anatomical characters used for taxonomy and systematics at all. For this a new integrative approach of morphology and molecular genetics was sought. It was also important to find out how own reconstructions of phylogenetic trees based on nuclear and mitochondrial markers confirm or refute existing views on taxonomy and systematics of the Lymnaeidae. First own morphological analyses of Palaearctic representatives of the genus Radix checked by molecular genetics reveal that shell morphology, mantle pigmentation, length ratio of Praeputium to penial sheath, length of the duct of the bursa copulatrix and shape of the bursa show more variability in this characteristics as recognised for these species in literature. They are, contrary to previous assumptions, to a large extent not specific for single species. With molecular genetic methods clearly distinguishable species may have the same shell morphology and mantle pigmentation and could be not clearly differentiated by anatomical characters in many cases. It can be concluded, also on the basis of own studies of other representatives of the family Lymnaeidae, that to these characters was given an importance for taxonomy and systematics by most of the malacologists that they obviously do not have because of their high plasticity. Molecular genetic methods allow a new interpretation of the importance of morphological and anatomical characters and a more objective definition of species and genera. Based on the new findings on variability of morphological characters a new determination key for the representatives of the genus Radix in central Europe was elaborated. It enables the determination of more Radix forms than it was previously possible with determination tools. Molecular genetic analyses of representatives of the genus Stagnicola revealed a high percentage of hybridisation. In practise, this has consequences for the application of barcoding on the base of mitochondrial markers (COI and cyt-b).

Diversité et invasions biologiques dans l'interaction grande douve du foie - Lymnaeidae : facteurs d'expansion de la fasciolose ? / Diversity and biological invasions in the liver fluke - Lymnaeidae system : factors of fasciolosis expansion ?

La mondialisation et les changements globaux actuels ont un impact considérable sur la distribution des espèces et la composition des communautés. Lorsque ces espèces sont impliquées dans une interaction hôte-parasite les changements dans leur répartition peuvent entraîner la (ré)émergence de maladies infectieuses. La fasciolose, maladie causée par les grandes douves du foie (Fasciola hepatica et Fasciola gigantica) est réémergente dans de nombreux points du globe. Il est difficile de mettre en place des programmes de contrôle parce que (1) les hôtes intermédiaires, des mollusques d’eau douce de la famille des Lymnaeidae, sont composés d’un groupe d’espèces cryptiques difficilement identifiables et (2) plusieurs espèces impliquées dans cette maladie sont invasives. L’objectif de cette thèse était d’étudier les facteurs écologiques et évolutifs à large échelle de l’interaction entre la grande douve du foie et ses hôtes intermédiaires susceptibles de favoriser une (ré)émergence de la fasciolose. J’ai, dans un premier temps, développé des approches moléculaires pour reconnaître les espèces cryptiques tant du côté de la grande douve du foie que du côté des Lymnaeidae. L’utilisation d’une des techniques développées sur Galba schirazensis, Galba cubensis et Galba truncatula, trois espèces de limnées, m’a permis d’identifier leur distribution respective et de modéliser leur niches bioclimatiques grâce à l’utilisation de modèles de niches écologiques. Cette approche de modélisation de niches permet d’inférer la distribution potentielle des trois espèces et nous amène à discuter des avantages potentiels de ces modèles dans la gestion de la fasciolose. Dans un second temps, je me suis intéressée à la structuration de la diversité génétique chez des espèces invasives du système grande douve du foie-limnée, par des approches de génétique des populations et de phylogénie. J’ai pu retracer l’histoire de colonisation, les dynamiques démographiques et le système de reproduction chez P. columella, G. schirazensis et G. cubensis. J’ai montré que ces trois espèces font préférentiellement de l’autofécondation entrainant des pertes drastiques de diversité génétiques sur le front d’invasion. G. cubensis présente cependant une coexistence de plusieurs génotypes dans les aires anciennement colonisées. Les différences génétiques entre ces trois espèces sont discutées à la lumière de ce qu’on sait de leur écologie. Enfin ces résultats m’ont permis de discuter de l’avantage d'être autofécondant en cas d’invasion biologique. Pour conclure l’invasion par ces populations d’hôtes intermédiaires dépourvus de diversité génétique pourrait représenter un risque épidémiologique. En effet un parasite a plus de probabilité de circuler dans une population hôte clonale que dans une population polymorphe. Cette thèse fait le lien entre écologie, interactions hôtes-parasites et génétique de l’invasion afin de mieux comprendre les facteurs d’expansion de la fasciolose à échelle globale. / Globalization and the current global change have significant impacts on species distribution and community composition. When these species are involved in a host-parasite interaction, changes in species range distribution can result in the (re)emergence of infectious diseases. Fasciolosis, a disease caused by the liver flukes (Fasciola hepatica and Fasciola gigantica) is reemerging in many parts of the world. It is difficult to implement control programs because (1) the intermediate hosts, freshwater molluscs of the Lymnaeidae family, are composed by a group of cryptic species (2) several species involved in this system are invasive. The objective of this thesis was to study the ecological and evolutionary factors at a large scale in the interaction between the liver fluke and its intermediate hosts that might drive to fasciolosis (re)emergence. First, I developed molecular approaches to quickly identify cryptic species on the two liver flukes and on three Lymnaeidae species. Using one of these techniques, I identify the respective distribution of Galba schirazensis, Galba cubensis and Galba truncatula and infer their respective climatic envelope by ecological niche modelling. We then modelled and projected the potential species distribution ranges. We discussed the contribution of models to predict the species distribution in space and time giving a considerable advantage to control fasciolosis. I then study the genetic diversity structuration in invasive snails involved in the transmission of F. hepatica, using population genetics and phylogeny. I could infer colonization history, population dynamics and reproductive system of Pseudosuccinea columella, G. schirazensis and G. cubensis. I showed that these three species preferentially make inbreeding causing drastic losses of genetic diversity in the invasion front. However G. cubensis presents a coexistence of several genotypes in formerly colonized areas. Genetic differences between these three species are discussed in the light of what we know about their ecology. Overall, these results illustrate how dramatic the reduction in genetic diversity can be for hermaphrodite animals. Finally, we discuss the epidemiological risk related to the invasion by intermediate hosts depleted in genetic diversity. Indeed, a parasite might circulate easily in a clonal host population than in a polymorphic population. In my thesis I linked ecology, host-parasite interactions with genetics of the invasion to better understand the expansion of fasciolosis at a global scale.

Aplexa marmorata (Guilding, 1828)(Basommatophora : Physidae) : an invasive freshwater snail in South Africa.

Invasions of ecosystems by alien species is a worldwide problem. Man, with his constant travelling, introduces organisms to places they have never occurred in before. The introductions may be accidental or deliberate. Some of the introduced organisms become invasive and some of these also become pests. Two aquatic pulmonate snails, Physa acuta (Physidae) and Lymnaea columella (Lymnaeidae), were introduced to South Africa probably through the aquarium industry in the 1940s and have now spread to most of the country's freshwater systems. A third invasive pulmonate, and second physid provisionally called Aplexa cf. marmorata, has recently been found in South African freshwaters. Comparison between A.. cf. marmorata found in Durban and P. acuta from Pietermaritzburg as an example of the genus Physa, confirmed that they belong to different genera and are therefore different species. Features compared were the shell, radula, foot, mantle, male genitalia and sperm morphology. Aplexa cf. marmorata is characterized by its foot having a pointed posterior end with a dark mid-dorsal stripe while that of P. acuta does not have these features. The mantle edge ofA.cf. marmorata has short triangular dentations while that of P. acuta has long finger-like projections. Aplexa cf. marmorata does not have an externally visible preputial gland whereas P. acuta does. The penis of A.cf. marmorata has a lateral opening while that of P. acuta has a sub-terminal outlet. TEM sections of the spermatozoon of A.cf. marmorata showed that it has a maximum of two glycogen helices around the mid-piece while P. acuta is known to have three. A study of the population dynamics of A.cf. marmorata in Durban showed it to produce three overlapping generations within a 14 month period whereas P. acuta has been shown to produce as many as eight over a similar time period. Further comparisons between South African A. cf. marmorata and similar material from the West Indies, Nigeria and St Lucia (KwaZulu-Natal) showed that they shared the same features with the specimens collected in Durban and are therefore considered to belong to the same species , Aplexa marmorata (Guilding, 1828). This species is indigenous to the Caribbean and northern parts of South America. The picture is however complicated by the fact that Dr L Paraense, doyen of the Brazilian school of freshwater malacology, does not recognize the genus Aplexa and redescribed this species under the name Physa marmorata in 1986. / Thesis (M.Sc.)-University of Natal, Durban, 2000.
