Mitochondrial DNA Inheritance in the Human Fungal Pathogen Cryptococcus gattii / Masters Thesis

The inheritance of mitochondrial DNA (mtDNA) is predominantly uniparental in most sexual eukaryotes, including the human pathogenic basidiomycetous yeast Cryptococcus neoformans. In this study, we examined the mitochondrial inheritance pattern of Cryptococcus gattii, a closely - related sister species of C. neoformans responsible for the recent and ongoing outbreak of Cryptococcal infections in the US Pacific Northwest and British Columbia (especially Vancouver Island) in Canada. Using molecular markers, we analyzed the inheritance of mitochondrial DNA in 9 crosses between strains within and between divergent lineages in C. gattii. Our analyses identified significant variations in mtDNA inheritance patterns among strains and crosses. For two of the crosses that showed uniparental mitochondrial inheritance in standard laboratory conditions, we further investigated the effects of the following environmental variables on mtDNA inheritance: UV exposure, temperature, exposure to the methylation inhibitor 5-aza-2’-deoxycytidine, and exposure to the ubiquitination inhibitor ammonium chloride. Interestingly, one of these crosses showed no response to these environmental variables while the other exhibited diverse patterns ranging from complete uniparental inheritance of the MATa parent mitochondria, to biparental inheritance, and to complete uniparental inheritance of the MATalpha parental mitochondria. Our analyses indicated that the mtDNA inheritance pattern in C. gattii differs significantly from its closely related species C. neoformans. We discussed possible mechanisms underlying these divergent mitochondrial DNA inheritance pattern. / Thesis / Master of Science (MSc)

A área de aprendizado de máquina adquiriu grande importância na última década graças à sua capacidade de analisar conjuntos de dados complexos em larga escala. Em diversas áreas do conhecimento existe a demanda pela análise de dados por especialistas, seja para obter agrupamentos latentes ou classificar instâncias em classes conhecidas. As ferramentas acessíveis a especialistas leigos em programação são limitadas a problemas específicos e demandam um custo de desenvolvimento às vezes proibitivo, sendo interessante buscar por ferramentas genéricas e aplicáveis a qualquer área do conhecimento. Este trabalho busca estender e implementar uma metodologia genérica de aprendizado de máquina capaz de analisar quaisquer conjuntos de arquivos de forma praticamente livre de configuração. Foram obtidos resultados satisfatórios de sua aplicação em um conjunto amplo de problemas para agrupamento e classificação de executáveis, spam e detecção de línguas. / Machine learning has rised in importance in the last decade thanks to its power to analyse complex datasets in large scale. At several areas of knowledge there is a demand for data analysis by domain experts, be it for discovering latent clusters or classifying instances into known groups. The tools available for experts that do not master computer programming are limited to specific tasks and demand a high development cost, which sometimes is prohibitive. It is interesting, then, to develop generic tools useful to any area of knowledge. This masters thesis seeks to extend and implement a generic machine learning methodology capable of analysing any set of files mostly free of configuration. Its application produced satisfactory results in a wide set of clustering and classification problems over binary executables, spam classification, and language identification.

Inferno and exaltation - How to design Creativity and support Interaction Designers

This masters thesis is about how to enhance creativity and investigates the design process of interaction designers, in terms of the creative process, design support tools and especcially through a concept presented in this thesis. The concept is called DSTZ (Design Support Tool Z) and suggests ways in how to support a designer in the creative process. An empirical study was carried out in the thesis that consisted of a survey conducted on 8 "interaction designers" investigating how interaction designers work and if they require or need any design support. A design survey was later conducted on 18 "interaction designers" of which 9 were professionals and 9 were students, to investigate the concept DSTZ and the creativity enhancing aspect. The resuls from the empirical studies show that a design support tool is desired and DSTZ has features that are useful and can enhance creativity. The features consist of 8 palettes and 3 funtions that are manifested in DSTZ. Further investigation is required in order to find data that support the statement that creativity can be enhanced through a design support tool.

The subject of this master’s thesis is a project documentation for development of a nursery school. The nursery school is situated on the outskirts of the city Příbor. The school is designed as a two-storey building and includes 4 classrooms, with total capacity reaching 60 pupils. The perimeter wall consists of Porotherm clay block walling system. The ceiling framework uses a combination of two systems; the prestress hollow core slabs Spiroll and reinforced concrete monolithic blocks. The roof is designed as a single skin cladding with a flat, green roof. The external thermal insulation composite system (Etics) is proposed for the insulation of the building. The outer walls are partially cladded with timber. Additionally, the combination of white rendering and timber cladding will form a modern looking building that fits within a new housing development. Furthermore, the plan includes an outside playground, with allotments and parking spaces on the premises for the use of a nursery school.

Walter M. Miller, Jr.’s A Canticle for Leibowitz is a timeless story about apocalyptic cycles, conflicts and similarities between religion and science, religious ethics and secular ethics, sin and redemption, myth and preternatural innocence. Canticle is a very religious story about a monastery dedicated to preserving scientific knowledge from the time before nuclear war which devastated the world and reduced humanity to a pre-technological civilization. The Catholic Church and this monastery are portrayed as a bastion of civilization amidst barbarians and a light of faith amidst atheism. Unfortunately, humanity destroys the Earth once again, but Miller ends with two beacons of hope: a starship headed for the unknown to help humanity begin again and the preternaturally innocent Rachel who portends a future for similarly innocent human beings repopulating the Earth. Thus, faith ultimately triumphs over atheism even in the midst of almost total catastrophe.
