Estudos catalíticos da oxigenação de hidrocarbonetos utilizando metaloporfirinas fluorossubstituídas como catalisadores em meio homogêneo e heterogêneo / Catalytic studies on hidrocarbon oxidation using fluorosubstituted metalloporphyrins as catalysts in homogeneous and heterogeneous medium

Manganês(III)porfirinas têm sido relatadas como eficientes catalisadores em reações de oxigenação de hidrocarbonetos por vários doadores de oxigênio. Numa primeira etapa, foram realizadas a síntese e caracterização das porfirinas base-livre, posterior introdução de grupo sulfonato na H2(MPTFPP), a fim de que esta pudesse ser utilizada em reações em meio heterogêneo, e inserção de metal nas porfirinas base-livre, obtendo-se os catalisadores cis-[Mn(BFPBPP)]Cl, trans-[Mn(BFPBPP)]Cl, [Mn(MSO3TFPP)]Cl e [Mn(TSO3PP)]Cl. Realizou-se a funcionalização da sílica gel contendo grupos trimetilamônio, que permitia sua ligação com metaloporfirinas contendo cargas negativas periféricas, com grupos imidazol, que permitem ligação com a metaloporfirina via ligação coordenativa, favorecendo o aumento da atividade catalítica destes sistemas. Os catalisadores [Mn(BFPBPP)]Cl, trans-[Mn(BFPBPP)]Cl, [Mn(MSO3TFPP)]Cl foram utilizados na epoxidação do (Z)-cicloocteno e na oxidação do cicloexano por PhIO em meio homogêneo. A Mn(III)porfirina aniônica apresentou rendimentos comparáveis a sistemas clássicos, acompanhados de uma melhor seletividade. Tais resultados, acompanhados da vantagem de que este sistema poderia ser utilizado em meio heterogêneo, levaram à sua escolha para prosseguimento dos estudos. [Mn(MSO3TFPP)]Cl e [Mn(TSO3PP)]Cl foram então utilizadas em meio homogêneo e imobilizadas nos suportes IPG, SiN+, SiN+(IPG) como catalisadores na reação de epoxidação do (Z)-cicloocteno. No caso do PhIO, para ambas o suporte exerceu um efeito positivo, alcançando-se 100% de ciclooctenóxido com os sistemas [Mn(MSO3PTFPP)]-IPG, [Mn(MSO3PTFPP)]-SiN(IPG) e [Mn(TSO3PP)]-SiN(IPG). A presença de substituintes pentafluorofenis na [Mn(MSO3TFPP)]Cl tornaram-na mais ativa na catálise de epoxidação do (Z)-cicloocteno por H2O2, o que levou a considerar o efeito dos substituintes eletronegativos no sentido de tornar o centro metal-oxo mais eletrofílico, aumentando sua reatividade. Além disso, um aumento na atividade catalítica dos catalisadores utilizados em sistema heterogêneo, aliado à presença de imidazol adicionado, evidenciam o efeito positivo do suporte e papel duplo do co-catalisador, atuando como catalisador ácido-base e ligante trans ao Mn(III). Tais resultados comparam-se aos obtidos para sistemas clássicos, mas com a vantagem dos catalisadores obtidos neste trabalho poderem ser recuperados ao final da reação, podendo ser reutilizados. / Manganese(III)porphyrins have been described as efficient catalysts in hidrocarbon oxygenation reactions by a variety of oxygen donors. In a first step, the free-base porphyrins synthesis and characterization were performed, with posterior introduction of a sulfonato group in H2(MPTFPP), in order to use its heterogeneous catalyst, and metal insertion, rendering cis-[Mn(BFPBPP)]Cl, trans-[Mn(BFPBPP)]Cl, [Mn(MSO3TFPP)]Cl and [Mn(TSO3PP)]Cl. The silica gel containing trimethylammonium groups was functionalized with imidazole groups. The first allowed the linkage of anionic metalloporphyrins containing peripherical negative charges and the latter allows its linkage to metalloporphyrins via coordinative binding. Homogeneous [Mn(BFPBPP)]Cl, trans-[Mn(BFPBPP)]Cl, [Mn(MSO3TFPP)]Cl were used in the (Z)-cyclooctene epoxydation and cicloexane oxidation by PhIO. The anionic Mn(III)porphyrin had presented yields comparable to classic systems with a better selectivity. Such results allied to the possibility of use in heterogeneous system led to its choice to posterior studies. Homogeneous and heterogeneous [Mn(MSO3TFPP)]Cl e [Mn(TSO3PP)]Cl immobilized on IPG, SiN+, SiN+(IPG) supports were then used as catalysts in the epoxydation reaction of (Z)-cyclooctene. When PhIO was used, the support had a positive effect for both catalysts, reaching 100% epoxycyclooctane in [Mn(MSO3PTFPP)]-IPG, [Mn(MSO3PTFPP)]-SiN(IPG) e [Mn(TSO3PP)]-SiN(IPG). Because of bearing pentafluorophenyl groups, [Mn(MSO3TFPP)]Cl was more active in epoxydation reaction of (Z)-cyclooctene catalysis by H2O2, which led considering the effect of electronegative groups leading to a metal-oxo species more eletrophylic, increasing its reactivity. A rise in the catalytic activity of heterogeneous catalysts allied to imidazole addition show the positive effect of the support and the two-fold hole of the co-catalyst, acting as acid-base catalyst and trans ligand to Mn(III). Such results are comparable to that obtained for classical system with the advantage that the catalysts obtained in this work allow its recuperation and reuse.

Its high biological compatibility, being a molecule found naturally in plants and many red blood animals, coupled with the possibility of forming complexes with lanthanide atoms, makes these compounds are particularly attractive in a branch of medicine quite promising in terms respect to combat cancer, called photodynamic therapy. The lanthanide-porphyrin complex act as photosensitizers in the production of singlet oxygen, a cytotoxic species, may be soaked in organic tissue of interest, promoting the cancer cell death. The synthesis of some Ln-porphyrin complexes [Ln (Ln = Gd, Er, Yb)] are already reported in the literature. However, with increasing ionic radius of metal that is intended to complex in the porphyrin, the energy barrier which prevails opposite the occurrence of this synthesis also increases, hindering the occurrence of the metalation process. Thus, this study seeks to promote metalation with europium, in 5,10,15,20 - meso - tetrapyridilporphyrin (H2TPyP). Was used the europium acetylacetonate salt [Eu(acac)3] in the synthesis as a source of Eu3+ ion. The obtained compound was purified by column chromatography on elution of alumina (Al2O3) and further characterized by absorption spectroscopy in the ultraviolet-visible region (UV-VIS) and infrared region (IR), luminescence and termogravimetric analysis (TG). The results obtained for the UV-vis spectra show for complex synthesized 3 Q bands, a lower number compared to the spectrum of the free base porphyrin, which has 4 Q bands, which discloses the formation of a compound with higher symmetry, which rose from D2h to D4h due to insertion of the metal ion in the porphyrin ring. It was observed also a bathochromic shift of the Soret band in the complex shifted from 417 to 429nm, consistent with the fact that the metallation occurred. The IR results and suggest the occurrence of metalation of the suppression band 3304cm-1, indicating deprotonation of the central atoms of Nytrogen, also suggested maintaining the acetylacetonate ion in stabilizing the structure of the complex. The luminescence showed that there was enhancement of emission bands, which is consistent with the formation of a metal complex. In the TG results, it was found that the compound formed to the amount of residue obtained was much higher than that for H2TPyP, suggesting the formation of metal oxide in the first case, consistent with the fact that formation of europium metalloporphyrin. / Cada vez mais, as porfirinas vêm conquistando uma posição de destacado interesse no meio científico. Sua elevada compatibilidade biológica, sendo uma molécula encontrada naturalmente em plantas e em diversos animais de sangue vermelho, aliada a possibilidade da formação de complexos com átomos de lantanídeos, faz com que esses compostos sejam particularmente atrativos em um ramo bastante promissor da medicina no que diz respeito ao combate ao câncer, denominado de terapia fotodinâmica. Isso porque os complexos lantanídeos-porfirina atuam como fotossensibilizadores na produção de oxigênio singlete, uma espécie citotóxica, podendo esses complexos serem saturados em tecidos orgânicos de interesse, promovendo a morte de células cancerígenas. A síntese de alguns complexos Ln-porfirina (Ln= Gd, Er, Yb) já são reportadas na literatura. Entretanto, com o aumento do raio iônico do metal que se pretende complexar a porfirina, a barreira energética que se impera frente a ocorrência desta síntese também aumenta, dificultando a ocorrência do processo de metalação. Dessa forma, o presente estudo busca promover a metalação de európio na 5,10,15,20 – meso – tetrapiridilporfirina (H2TPyP). Utilizou-se o sal acetilacetonato de európio [Eu(acac)3] na síntese como fonte do íon Eu3+. O composto obtido foi purificado por eluição em coluna cromatográfica de alumina (Al2O3) e posteriormente caracterizado por espectroscopia de absorção na região do ultravioleta-visível (UV-Vis) e infravermelho (IR), luminescência e análise termogravimética (TG). Os resultados obtidos por UV-Vis para o complexo sintetizado mostraram espectros com 3 bandas Q, um menor número comparado ao espectro da porfirina base livre que apresenta 4 bandas Q, o que sugere a formação de um composto com maior simetria, que passou de D2h para D4h, devido a inserção do íon metálico no anel porfirínico. Observou-se também um deslocamento batocrômico da banda Soret no complexo que deslocou de 417 para 429nm, fato condizente com a ocorrência da metalação. Os resultados de IR, além de sugerirem a ocorrência da metalação pela supressão da banda em 3304cm-1, indicando a desprotonação dos nitrogênios centrais, também sugeriram a manutenção do íon acetilacetonato na estabilização da estrutura do complexo. A luminescência mostrou que houve intensificação das bandas de emissão, o que condiz com a formação de um complexo metálico. Nos resultados de TG constatou-se que para o composto formado a quantidade de resíduo obtido foi bastante superior de que para a H2TPyP, sugerindo a formação de óxido metálico no primeiro caso, fato condizente com a formação da metaloporfirina de európio.

Investigação das propriedades de interação e propriedades catalíticas de ferro(III) porfirinas na oxidação de um fármaco antimalárico / Investigation of the interaction properties and the catalytical properties of iron(III) porphyrins in the oxidation of an antimalarial drug.

Neste trabalho foram estudadas, por espectroscopia UV-Vis, as interações do fármaco anti-malária cloroquina com uma série de porfirinas meso fenilas substituídas aniônicas, catiônicas e neutras, e seus complexos Fe(III), em tampão aquoso e em solução metanólica. Com base nos resultados de interação, as ferro(III) porfirinas meso fenilas substiuídas e a FePPIX foram utilizadas como catalisadores para a oxidação de cloroquina por iodosilbenzeno (PhIO) e peróxidos (H2O2 e ácido meta-cloroperbenzóico, CPBA), em ambos os meios. Dos estudos de interação foi observado, em meio aquoso, pH 6,4, a formação de um complexo p-p, resultante do efeito cooperativo das interações eletrostáticas e da interação p-p entre as espécies aniônicas e a cloroquina. Em metanol nenhuma complexação p-p foi observada. As porfirinas e ferro(III) porfirinas neutras e catiônicas não formaram complexos com a cloroquina. As constantes de formação do complexo p-p entre as porfirinas ou ferro(III) porfirinas e cloroquina foram comparáveis áquelas relatadas na literatura para a cloroquina e outras ferro(III) porfirinas naturais. As reações de oxidação foram analisadas por cromatografia líquida de alta eficiência (CLAE). O principal produto obtido em todos os sitemas estudados foi a monodesetilcloroquina. Este é também o principal produto ?in vivo? da oxidação deste fármaco catalisada por CYP-450. A identificação do produto que é gerado em segunda maior quantidade, o derivado desetilamino da cloroquina, foi realizada por CLAE acoplada a espectrometria de massas. Dos resultados das reações de oxidação com PhIO e H2O2 foi possível constatar que: as ferro(III) porfirinas aniônicas são os catalisadores mais eficientes, com rendimentos de até 30 % de monodesetilcloroquina; em solução aquosa elas são seletivas para a deetilação oxidativa da cloroquina, enquanto que em metanol até cinco produtos de oxidação podem ser observados. As ferro(III) porfirinas neutras apresentam baixa atividade catalítica na oxidação da cloroquina por PhIO ou H2O2 pois não interage eletrostaticamente com o substrato. As ferro(III) porfirinas catiônicas não apresentam atividade catalítica devido à repulsão eletrostática com o substrato. As reações de oxidação por CPBA apresentaram baixos rendimentos devido à rápida destruição dos catalisadores quando se utilizou este oxidante. Foram ainda realizados estudos catalíticos utlizando ferro(III) porfirinas suportadas em membrana de polidimetilssiloxano (PDMS) e poli-(1?4)-b-d-glucosamina (quitosana) na oxidação de substratos padrões (ciclocteno, cicloexano e estireno). Destes, apenas a quitosana se mostrou um suporte adequado, resultando em um catalisador eficiente. As ferro(III) porfirinas imobilizadas neste suporte foram utlizadas na oxidação da cloroquina por PhIO, levando aos mesmos rendimentos que estas ferro(III) porfirinas apresentaram em meio homogêneo, mostrando também a dependência interação(FeP:substrato)/atividade catalítica previamente observada para as ferro(III) porfirinas em solução. Os resultados obtidos neste trabalho mostram que as ferro(III) porfirinas aniônicas são eficientes catalisadores biomiméticos da oxidação da cloroquina por PhIO e H2O2, tanto em solução como imobilizados. Além disso, pode-se concluir que a habilidade do catalisador em interagir com o substrato apresenta forte influência na atividade catalítica e deve ser levada em consideração nos estudos de outros sistemas modelos do CYP-450. / In this work, the interaction between the antimalarial drug chloroquine with a series of anionic, cationic, and neutral meso phenyl substitued porphyrins; and their iron(III) complexes, was investigated by UV-Vis spectroscopy, in aqueous buffer and methanolic solution. On the basis of the interaction results, the meso phenyl substitued iron(III) porphyrins and FePPIX were used as catalyst in the oxidation of chloroquine by iodosilbenzene (PhIO) or peroxides (H2O2 e meta-chloroperbenzoic acid, CPBA), in both media. From the interaction studies, it was possible to observe the formation of a p-p complex generated from the cooperative effect of the electrostatic and p-p interactions between the anionic species and chloroquine. In methanol, no complexation was observed. The neutral and cationic porphyrins and iron(III) porphyrins did not form complexes with chloroquine. The association constants of the p-p complex formed between the meso phenyl substitued porphyrins and iron(III) porphyrins with chloroquine were comparable with those reported in the literature for the interaction of chloroquine with others natural iron(III) porphyrins. The oxidation reaction produtcts were analized by high perfomance liquid chromatography (HPLC). The major product obtained in all the studied systems was monodesethylchloroquine. This compound is also the main ?in vivo? product of chloroquine metabolism catalised by CYP-450. The identification of the second most abundant product, the desethylamine derived from chloroquine, was performed by HPLC coupled with mass spectrometry. The results obtained from the oxidation reactions of chloroquine by PhIO and H2O2 indicated that: (i) the anionic iron(III) porphyrins are efficient catalysts, with yields close to 30 % for monodesethylchloroquine; (ii) in aqueous buffer they are selective toward the oxidative N-deethylation of chloroquine, while in methanol up to five oxidation products are obtained. The neutral iron(III) porphyrins presented low catalytic activity for the chloroquine oxidation by PhIO or H2O2 due to the lack of electrostatic interaction between the catalyst and the substrate. (iii) The cationic iron(III) porphyrins did not exhibit any catalytic activity because of electrostatic repulsion tha tales place between the catalyst and the substrate. Catalyst degradation occurred when CPBA was used as oxidant, leading to very low yields of oxidation products. Catalytic studies using iron(III) porphyrins supported in polydimethylsiloxane (PDMS) membranes and in poly-(1?4)-b-d-glucosamine (chitosan) were also performed for the oxidation of standard substrates (cicloctene, ciclohexane and styrene). Only chitosan was considered to be an adequate support, resulting in efficient catalysis. The iron(III) porphyrins immobilized on this support were used for the oxidation of chloroquine by PhIO, leading to the same yields as those obtained with the same iron(III) porphyrins in homogeneous medium. It is noteworthy that the relations between the interaction FeP:substrate and the FeP catalytic activity previously observed for the iron(III) porphyrins in solution was also observed in these heterogeneous catalysis. The results obtained in this work show that anionic iron(III) porphyrins are efficient biomimetic catalysts for the oxidation of chloroquine by PhIO and H2O2, both when supported and in solution. Also, the catalyst ability to interact with the substrate has a strong influence on its activity and must be considered when studying other CYP-450 model systems.

This work describes the synthesis of a new porphyrin, TBMPP 27, and its metallo complexes 28 and 29. Herein it was used a readily available reactant as starting material 22, vanilin, which was submitted too alkylation with n-bromobutane affording the synthetic precursos 25. The desired porphyrin 27, was obtained by reacting pyrrole and the corresponding aldehyde in the presence of propionic acid (Alder - Longo method). The purified porphyrin 27 was then subjected to the meallation process. the synthesized compounds were characterized by IR, UV-Vis, NMR and EPR spectroscopy; / O interesse pela síntese de porfirinas e derivados vem se tornando cada vez mais crescente, devido à grande eficiência catalítica de seus derivados metalados em reações de oxidação de hidrocarbonetos e detecção de oxigênio, entre outras. Neste sentido foi realizado um estudo visando a síntese de uma nova porfirina, a TBMPP 27, e seus derivados metalados 28 e 29. A porfirina 27 foi preparada à partir de um reagente acessivel e de baixo custo, a vanilina 22. A alquilação de 22 com n-bromobutano levou ao precursos sintetico 25 que foi então submetido às condições de Adler-Longo, pirrol e ácido propiônico, fornecendo a porfirina desejada 27. A porfirina base livre purificada foi entao submetida ao processo de metalação. Os novos compostos sintetizados foram caracterizados por espectroscopia de IV, UV-Vis,RMN e RPE.

Since then, several studies were carried out in order to increase yield and lower the production costs. This work describes a process, based on Adler-Longo method, in which propionic acid was replaced by a mixture of by acetic acid and acetic anhydride on the synthesis of meso-tetrakis(4-methoxyphenyl)porphyrin 16. The process is attractive because the yield was improved, the cost lowered and removal of the solvent was easier because of the low boiling point. The formation of porphyrins complexed with metals, such as, iron 17, manganese 18, copper 19 and zinc 20, was also studied with success. The methods found in the literature were modified to facilitate the process of reaction and purification. The synthesis of metalloporphyrins was performed without the use of inert atmosphere, and the performance found was consistent with those described in literature for the iron and copper metalloporphyrins, but the purification process was facilitated. Copper metalloporphyrin 19, precipitates in N,N-dimethylformamide (DMF), and therefore, filtration was a simple and effective method of purification. The porphyrin and metalloporphyrins were fully characterized by infrared, UV-Vis and electron paramagnetic resonance (EPR) and nuclear magnetic resonance (RMN) spectroscopy. All the spectral data were completely consistent with those reported in the literature. Synthesized compounds. / A síntese de porfirinas vem sendo estudada desde 1929. Desde então vários processos foram descritos buscando facilitar, aumentar o rendimento e também diminuir o custo beneficio da produção desses compostos. Neste trabalho desenvolveu-se um método, a partir do método de Adler-Longo, em que o ácido propiônico foi substituído por uma mistura de ácido acético e anidrido acético na síntese da meso-tetrakis(4-metóxifenil)porfirina 16. O método é interessante, pois o rendimento da reação foi otimizado, mas o custo de preparação do produto e a remoção do solvente usado é facilitada devido ao menor ponto de ebulição. A reação de metalação da porfirina com ferro 17, manganês 18, cobre 19 e zinco 20, também foram estudadas com sucesso. Os métodos encontrados na literatura foram alterados a fim de facilitar o processo de síntese e também de purificação. A metalação foi realizada sem o uso de atmosfera inerte e o rendimento foi coerente com o encontrado na literatura para as porfirinas de ferro e para as porfirinas de cobre foi superior devido ao processo de purificação usado. A metaloporfirina de cobre 19, precipita em N,N-dimetilformamida (DMF), e portanto, a filtração é um método simples e eficiente de purificação. A porfirina e as metaloporfirinas foram caracterizadas por espectroscopia na região do infravermelho (IV) e do ultravioleta-visível (UV-Vis), por ressonância paramagnética eletrônica (EPR) e ressonância magnética nuclear (RMN) de 1H e 13C. Todos os dados espectrais obtidos estão completamente de acordo com os descritos na literatura. Compostos sintetizados.

Its high biological compatibility, being a molecule found naturally in plants and many red blood animals, coupled with the possibility of forming complexes with lanthanide atoms, makes these compounds are particularly attractive in a branch of medicine quite promising in terms respect to combat cancer, called photodynamic therapy. The lanthanide-porphyrin complex act as photosensitizers in the production of singlet oxygen, a cytotoxic species, may be soaked in organic tissue of interest, promoting the cancer cell death. The synthesis of some Ln-porphyrin complexes [Ln (Ln = Gd, Er, Yb)] are already reported in the literature. However, with increasing ionic radius of metal that is intended to complex in the porphyrin, the energy barrier which prevails opposite the occurrence of this synthesis also increases, hindering the occurrence of the metalation process. Thus, this study seeks to promote metalation with europium, in 5,10,15,20 - meso - tetrapyridilporphyrin (H2TPyP). Was used the europium acetylacetonate salt [Eu(acac)3] in the synthesis as a source of Eu3+ ion. The obtained compound was purified by column chromatography on elution of alumina (Al2O3) and further characterized by absorption spectroscopy in the ultraviolet-visible region (UV-VIS) and infrared region (IR), luminescence and termogravimetric analysis (TG). The results obtained for the UV-vis spectra show for complex synthesized 3 Q bands, a lower number compared to the spectrum of the free base porphyrin, which has 4 Q bands, which discloses the formation of a compound with higher symmetry, which rose from D2h to D4h due to insertion of the metal ion in the porphyrin ring. It was observed also a bathochromic shift of the Soret band in the complex shifted from 417 to 429nm, consistent with the fact that the metallation occurred. The IR results and suggest the occurrence of metalation of the suppression band 3304cm-1, indicating deprotonation of the central atoms of Nytrogen, also suggested maintaining the acetylacetonate ion in stabilizing the structure of the complex. The luminescence showed that there was enhancement of emission bands, which is consistent with the formation of a metal complex. In the TG results, it was found that the compound formed to the amount of residue obtained was much higher than that for H2TPyP, suggesting the formation of metal oxide in the first case, consistent with the fact that formation of europium metalloporphyrin. / Cada vez mais, as porfirinas vÃm conquistando uma posiÃÃo de destacado interesse no meio cientÃfico. Sua elevada compatibilidade biolÃgica, sendo uma molÃcula encontrada naturalmente em plantas e em diversos animais de sangue vermelho, aliada a possibilidade da formaÃÃo de complexos com Ãtomos de lantanÃdeos, faz com que esses compostos sejam particularmente atrativos em um ramo bastante promissor da medicina no que diz respeito ao combate ao cÃncer, denominado de terapia fotodinÃmica. Isso porque os complexos lantanÃdeos-porfirina atuam como fotossensibilizadores na produÃÃo de oxigÃnio singlete, uma espÃcie citotÃxica, podendo esses complexos serem saturados em tecidos orgÃnicos de interesse, promovendo a morte de cÃlulas cancerÃgenas. A sÃntese de alguns complexos Ln-porfirina (Ln= Gd, Er, Yb) jà sÃo reportadas na literatura. Entretanto, com o aumento do raio iÃnico do metal que se pretende complexar a porfirina, a barreira energÃtica que se impera frente a ocorrÃncia desta sÃntese tambÃm aumenta, dificultando a ocorrÃncia do processo de metalaÃÃo. Dessa forma, o presente estudo busca promover a metalaÃÃo de eurÃpio na 5,10,15,20 â meso â tetrapiridilporfirina (H2TPyP). Utilizou-se o sal acetilacetonato de eurÃpio [Eu(acac)3] na sÃntese como fonte do Ãon Eu3+. O composto obtido foi purificado por eluiÃÃo em coluna cromatogrÃfica de alumina (Al2O3) e posteriormente caracterizado por espectroscopia de absorÃÃo na regiÃo do ultravioleta-visÃvel (UV-Vis) e infravermelho (IR), luminescÃncia e anÃlise termogravimÃtica (TG). Os resultados obtidos por UV-Vis para o complexo sintetizado mostraram espectros com 3 bandas Q, um menor nÃmero comparado ao espectro da porfirina base livre que apresenta 4 bandas Q, o que sugere a formaÃÃo de um composto com maior simetria, que passou de D2h para D4h, devido a inserÃÃo do Ãon metÃlico no anel porfirÃnico. Observou-se tambÃm um deslocamento batocrÃmico da banda Soret no complexo que deslocou de 417 para 429nm, fato condizente com a ocorrÃncia da metalaÃÃo. Os resultados de IR, alÃm de sugerirem a ocorrÃncia da metalaÃÃo pela supressÃo da banda em 3304cm-1, indicando a desprotonaÃÃo dos nitrogÃnios centrais, tambÃm sugeriram a manutenÃÃo do Ãon acetilacetonato na estabilizaÃÃo da estrutura do complexo. A luminescÃncia mostrou que houve intensificaÃÃo das bandas de emissÃo, o que condiz com a formaÃÃo de um complexo metÃlico. Nos resultados de TG constatou-se que para o composto formado a quantidade de resÃduo obtido foi bastante superior de que para a H2TPyP, sugerindo a formaÃÃo de Ãxido metÃlico no primeiro caso, fato condizente com a formaÃÃo da metaloporfirina de eurÃpio.

Síntese, caracterização e atividade catalítica de novos catalisadores metaloporfirínicos suportados / Synthesis, characterization and catalytic activity of novel anchored metalloporphyrin catalysts

Neste trabalho a metaloporfirina de segunda geração Me(TFPP)Cl (Me=Fe e Mn) [cloreto de 5, 10, 15, 20 ? tetrakis(pentafluorofenil)porfirina de ferro (III) ou manganês (III)] foi ancorada covalentemente a suportes aminofuncionalizados, através da substituição nucleofílica dos átomos para-flúor dos grupos meso-arilas do pentafluorofenil, com o objetivo de se obter catalisadores sólidos seletivos para reações de oxidação de moléculas orgânicas. Os suportes montmorilonita (modificada com aminopropiltrietoxissilano, Mont1, e cloropropiltrietoxissilano e em seguida com 1,6-diaminoexil, Mont2) e sílica modificada com 1,6-diaminoexil (DAHS) foram inicialmente sintetizados; poliestireno aminofuncionalizado, PS, foi cedido pelo Prof. Dr. John Lindsay Smith, York-Inglaterra, filossilicatos híbridos de magnésio, Silx e Talx, foram sintetizados previamente pelo grupo do Prof. Dr. Cláudio Airoldi, UNICAMP-Campinas; e 3-aminopropilssílica, APS, foi adquirida da Aldrich. Primeiramente foi realizada a síntese e caracterização dos suportes Mont1, Mont2 e DAHS e das metaloporfirinas, seguida das reações de ancoragem. Os catalisadores sólidos foram caracterizados por UV/Vis, IV, EPR, TGA, Raio-x. A atividade catalítica destes catalisadores sólidos foi investigada na oxidação do cicloocteno, cicloexano e estireno, utilizando como oxidante PhIO e o H2O2. Estes sistemas permitiram estudar as variáveis: distância entre a metaloporfirina e a superfície do suporte (tamanho de ?braço?); diferença de polaridade; tipo de estrutura do suporte (amorfo ou lamelar, orgânico ou inorgânico, rígido ou flexível); metal central da metaloporfirina; proteção dos grupos NH2 livre no suporte. De maneira geral os sistemas investigados mostraram ser catalisadores eficientes para todos os substratos, principalmente utilizando PhIO como oxidante. Em nenhum caso se observou lixiviação da metaloporfirina do suporte, provando que a ligação covalente é um método muito eficiente para preparar catalisadores ancorados. A estabilidade destes sistemas também foi comprovada realizando-se quatro ciclos de reação com o mesmo catalisador sólido, sendo que os rendimentos de produtos foram reprodutíveis. As FeP foram catalisadores mais eficientes que as MnP, mesmo quando se utilizou imidazol como co-catalisador para as MnP. Os melhores catalisadores foram os FeAPS, FeMont1, FeDAHS, FeTal2 e FeSil2. Todos estes catalisadores sólidos são rígidos e inertes, com um tamanho de ?braço? intermediário (4-7 átomos). A proteção dos grupos amino livres no suporte é fundamental para melhorar a eficiência catalítica de metaloporfirinas ancoradas em suportes aminofuncionalizados, pois evita a bis-coordenação dos NH2 ao metal, responsável pelo bloqueio do sítio catalítico. Embora os rendimentos dos produtos de oxidação utilizando o H2O2 tenham sido menores do que aqueles com o PhIO, alguns sistemas foram mais eficientes quando comparados com a metaloporfirina em solução, tanto do ponto de vista de rendimentos como de seletividade. Estes resultados abrem a possibilidade de aplicação desses sistemas para a oxidação de fármacos e poluentes. / In this work the second generation metalloporphyrin Me(TFPP)Cl (Me = Fe and Mn) [5,10, 15, 20 ? tetrakis(pentafluorophenyl) porphyrin iron (III) or manganese (III) chloride] was covalently anchored to aminofunctionalyzed supports, through nucleophilic substituition of the para-fluorine atoms of the pentafluorophenyl meso-aryl groups, with the objective of obtaining selective solid catalysts for oxidation reactions of organic molecules. The supports montmorillonite (modified with aminopropyltrietoxysilane, Mont1, and chloropropyltrietoxysilane followed by 1,6 ? diaminohexil, Mont2) and silica modified with 1,6 ? diaminohexil (DAHS) were initially synthesized; aminofunctionalyzed polystyrene, PS, was given by Prof. Dr. John Lindsay Smith, York ? England; hybrid magnesium phyllosilicates, Silx and Talx, were previously synthesized by Prof. dr. Cláudio Airoldi´s research group, UNICAMP ? Campinas; and 3 ? aminopropylsilica, APS, was purchased from Aldrich. The supports Mont1, Mont2, DAHS and metalloporphyrins were firstly synthesized and characterized, followed by the anchoring reactions. The solid catalysts were characterized by UV/Vis, IR, EPR, TGA, X-ray. The catalytic activity of these solid catalysts was investigated in the oxidation of cyclooctene, cyclohexane and styrene, using PhIO and H2O2, as oxidants. These systems possibilities the study of the effects: distance between the metalloporphyrin and the supports surface (?arm? size); polarity; support?s structures (amorphous or lamellar, organic or inorganic, rigid or flexible); metalloporphyrin´s central metal ion; protection of the free NH2 on the support. In general way, the investigated systems show to be efficient catalysts for all substrates, mainly from the supports, proving that the covalent bond is an efficient method for the preparing of anchored catalysts. The stability of these systems was also investigated through developing four reaction cycles with the same solid catalyst, with reproducible products yields. The FeP were more efficient catalysts than the MnP, even using imidazole as co-catalyst for the MnP. The better catalysts were FeAPS, FeMont1, FeDAHS, FeTal2 and FeSil2. All these solid catalysts are rigid and inert, with an intermediary ?arm? size (4-7 atoms). The protection of the support?s free NH2 groups is fundamental to improve the catalytic efficiency of metalloporphyrins anchored on aminofunctionalized supports, because it avoids the bis-coordenation of the NH2 groups to the metal ion, effect responsible for the blocking of the catalytic site. Although the oxidation products yields using H2O2 have been lower than these using PhIO, some systems were more efficient when compared to the homogeneous systems both in product yield and selectivity. These results open the possibility for utilization of the system for drugs and poluents oxidation.

Neste trabalho foram estudadas, por espectroscopia UV-Vis, as interações do fármaco anti-malária cloroquina com uma série de porfirinas meso fenilas substituídas aniônicas, catiônicas e neutras, e seus complexos Fe(III), em tampão aquoso e em solução metanólica. Com base nos resultados de interação, as ferro(III) porfirinas meso fenilas substiuídas e a FePPIX foram utilizadas como catalisadores para a oxidação de cloroquina por iodosilbenzeno (PhIO) e peróxidos (H2O2 e ácido meta-cloroperbenzóico, CPBA), em ambos os meios. Dos estudos de interação foi observado, em meio aquoso, pH 6,4, a formação de um complexo p-p, resultante do efeito cooperativo das interações eletrostáticas e da interação p-p entre as espécies aniônicas e a cloroquina. Em metanol nenhuma complexação p-p foi observada. As porfirinas e ferro(III) porfirinas neutras e catiônicas não formaram complexos com a cloroquina. As constantes de formação do complexo p-p entre as porfirinas ou ferro(III) porfirinas e cloroquina foram comparáveis áquelas relatadas na literatura para a cloroquina e outras ferro(III) porfirinas naturais. As reações de oxidação foram analisadas por cromatografia líquida de alta eficiência (CLAE). O principal produto obtido em todos os sitemas estudados foi a monodesetilcloroquina. Este é também o principal produto ?in vivo? da oxidação deste fármaco catalisada por CYP-450. A identificação do produto que é gerado em segunda maior quantidade, o derivado desetilamino da cloroquina, foi realizada por CLAE acoplada a espectrometria de massas. Dos resultados das reações de oxidação com PhIO e H2O2 foi possível constatar que: as ferro(III) porfirinas aniônicas são os catalisadores mais eficientes, com rendimentos de até 30 % de monodesetilcloroquina; em solução aquosa elas são seletivas para a deetilação oxidativa da cloroquina, enquanto que em metanol até cinco produtos de oxidação podem ser observados. As ferro(III) porfirinas neutras apresentam baixa atividade catalítica na oxidação da cloroquina por PhIO ou H2O2 pois não interage eletrostaticamente com o substrato. As ferro(III) porfirinas catiônicas não apresentam atividade catalítica devido à repulsão eletrostática com o substrato. As reações de oxidação por CPBA apresentaram baixos rendimentos devido à rápida destruição dos catalisadores quando se utilizou este oxidante. Foram ainda realizados estudos catalíticos utlizando ferro(III) porfirinas suportadas em membrana de polidimetilssiloxano (PDMS) e poli-(1?4)-b-d-glucosamina (quitosana) na oxidação de substratos padrões (ciclocteno, cicloexano e estireno). Destes, apenas a quitosana se mostrou um suporte adequado, resultando em um catalisador eficiente. As ferro(III) porfirinas imobilizadas neste suporte foram utlizadas na oxidação da cloroquina por PhIO, levando aos mesmos rendimentos que estas ferro(III) porfirinas apresentaram em meio homogêneo, mostrando também a dependência interação(FeP:substrato)/atividade catalítica previamente observada para as ferro(III) porfirinas em solução. Os resultados obtidos neste trabalho mostram que as ferro(III) porfirinas aniônicas são eficientes catalisadores biomiméticos da oxidação da cloroquina por PhIO e H2O2, tanto em solução como imobilizados. Além disso, pode-se concluir que a habilidade do catalisador em interagir com o substrato apresenta forte influência na atividade catalítica e deve ser levada em consideração nos estudos de outros sistemas modelos do CYP-450. / In this work, the interaction between the antimalarial drug chloroquine with a series of anionic, cationic, and neutral meso phenyl substitued porphyrins; and their iron(III) complexes, was investigated by UV-Vis spectroscopy, in aqueous buffer and methanolic solution. On the basis of the interaction results, the meso phenyl substitued iron(III) porphyrins and FePPIX were used as catalyst in the oxidation of chloroquine by iodosilbenzene (PhIO) or peroxides (H2O2 e meta-chloroperbenzoic acid, CPBA), in both media. From the interaction studies, it was possible to observe the formation of a p-p complex generated from the cooperative effect of the electrostatic and p-p interactions between the anionic species and chloroquine. In methanol, no complexation was observed. The neutral and cationic porphyrins and iron(III) porphyrins did not form complexes with chloroquine. The association constants of the p-p complex formed between the meso phenyl substitued porphyrins and iron(III) porphyrins with chloroquine were comparable with those reported in the literature for the interaction of chloroquine with others natural iron(III) porphyrins. The oxidation reaction produtcts were analized by high perfomance liquid chromatography (HPLC). The major product obtained in all the studied systems was monodesethylchloroquine. This compound is also the main ?in vivo? product of chloroquine metabolism catalised by CYP-450. The identification of the second most abundant product, the desethylamine derived from chloroquine, was performed by HPLC coupled with mass spectrometry. The results obtained from the oxidation reactions of chloroquine by PhIO and H2O2 indicated that: (i) the anionic iron(III) porphyrins are efficient catalysts, with yields close to 30 % for monodesethylchloroquine; (ii) in aqueous buffer they are selective toward the oxidative N-deethylation of chloroquine, while in methanol up to five oxidation products are obtained. The neutral iron(III) porphyrins presented low catalytic activity for the chloroquine oxidation by PhIO or H2O2 due to the lack of electrostatic interaction between the catalyst and the substrate. (iii) The cationic iron(III) porphyrins did not exhibit any catalytic activity because of electrostatic repulsion tha tales place between the catalyst and the substrate. Catalyst degradation occurred when CPBA was used as oxidant, leading to very low yields of oxidation products. Catalytic studies using iron(III) porphyrins supported in polydimethylsiloxane (PDMS) membranes and in poly-(1?4)-b-d-glucosamine (chitosan) were also performed for the oxidation of standard substrates (cicloctene, ciclohexane and styrene). Only chitosan was considered to be an adequate support, resulting in efficient catalysis. The iron(III) porphyrins immobilized on this support were used for the oxidation of chloroquine by PhIO, leading to the same yields as those obtained with the same iron(III) porphyrins in homogeneous medium. It is noteworthy that the relations between the interaction FeP:substrate and the FeP catalytic activity previously observed for the iron(III) porphyrins in solution was also observed in these heterogeneous catalysis. The results obtained in this work show that anionic iron(III) porphyrins are efficient biomimetic catalysts for the oxidation of chloroquine by PhIO and H2O2, both when supported and in solution. Also, the catalyst ability to interact with the substrate has a strong influence on its activity and must be considered when studying other CYP-450 model systems.

Os suportes montmorilonita (modificada com aminopropiltrietoxissilano, Mont1, e cloropropiltrietoxissilano e em seguida com 1,6-diaminoexil, Mont2) e sílica modificada com 1,6-diaminoexil (DAHS) foram inicialmente sintetizados; poliestireno aminofuncionalizado, PS, foi cedido pelo Prof. Dr. John Lindsay Smith, York-Inglaterra, filossilicatos híbridos de magnésio, Silx e Talx, foram sintetizados previamente pelo grupo do Prof. Dr. Cláudio Airoldi, UNICAMP-Campinas; e 3-aminopropilssílica, APS, foi adquirida da Aldrich. Primeiramente foi realizada a síntese e caracterização dos suportes Mont1, Mont2 e DAHS e das metaloporfirinas, seguida das reações de ancoragem. Os catalisadores sólidos foram caracterizados por UV/Vis, IV, EPR, TGA, Raio-x. A atividade catalítica destes catalisadores sólidos foi investigada na oxidação do cicloocteno, cicloexano e estireno, utilizando como oxidante PhIO e o H2O2. Estes sistemas permitiram estudar as variáveis: distância entre a metaloporfirina e a superfície do suporte (tamanho de ?braço?); diferença de polaridade; tipo de estrutura do suporte (amorfo ou lamelar, orgânico ou inorgânico, rígido ou flexível); metal central da metaloporfirina; proteção dos grupos NH2 livre no suporte. De maneira geral os sistemas investigados mostraram ser catalisadores eficientes para todos os substratos, principalmente utilizando PhIO como oxidante. Em nenhum caso se observou lixiviação da metaloporfirina do suporte, provando que a ligação covalente é um método muito eficiente para preparar catalisadores ancorados. A estabilidade destes sistemas também foi comprovada realizando-se quatro ciclos de reação com o mesmo catalisador sólido, sendo que os rendimentos de produtos foram reprodutíveis. As FeP foram catalisadores mais eficientes que as MnP, mesmo quando se utilizou imidazol como co-catalisador para as MnP. Os melhores catalisadores foram os FeAPS, FeMont1, FeDAHS, FeTal2 e FeSil2. Todos estes catalisadores sólidos são rígidos e inertes, com um tamanho de ?braço? intermediário (4-7 átomos). A proteção dos grupos amino livres no suporte é fundamental para melhorar a eficiência catalítica de metaloporfirinas ancoradas em suportes aminofuncionalizados, pois evita a bis-coordenação dos NH2 ao metal, responsável pelo bloqueio do sítio catalítico. Embora os rendimentos dos produtos de oxidação utilizando o H2O2 tenham sido menores do que aqueles com o PhIO, alguns sistemas foram mais eficientes quando comparados com a metaloporfirina em solução, tanto do ponto de vista de rendimentos como de seletividade. Estes resultados abrem a possibilidade de aplicação desses sistemas para a oxidação de fármacos e poluentes. / In this work the second generation metalloporphyrin Me(TFPP)Cl (Me = Fe and Mn) [5,10, 15, 20 ? tetrakis(pentafluorophenyl) porphyrin iron (III) or manganese (III) chloride] was covalently anchored to aminofunctionalyzed supports, through nucleophilic substituition of the para-fluorine atoms of the pentafluorophenyl meso-aryl groups, with the objective of obtaining selective solid catalysts for oxidation reactions of organic molecules. The supports montmorillonite (modified with aminopropyltrietoxysilane, Mont1, and chloropropyltrietoxysilane followed by 1,6 ? diaminohexil, Mont2) and silica modified with 1,6 ? diaminohexil (DAHS) were initially synthesized; aminofunctionalyzed polystyrene, PS, was given by Prof. Dr. John Lindsay Smith, York ? England; hybrid magnesium phyllosilicates, Silx and Talx, were previously synthesized by Prof. dr. Cláudio Airoldi´s research group, UNICAMP ? Campinas; and 3 ? aminopropylsilica, APS, was purchased from Aldrich. The supports Mont1, Mont2, DAHS and metalloporphyrins were firstly synthesized and characterized, followed by the anchoring reactions. The solid catalysts were characterized by UV/Vis, IR, EPR, TGA, X-ray. The catalytic activity of these solid catalysts was investigated in the oxidation of cyclooctene, cyclohexane and styrene, using PhIO and H2O2, as oxidants. These systems possibilities the study of the effects: distance between the metalloporphyrin and the supports surface (?arm? size); polarity; support?s structures (amorphous or lamellar, organic or inorganic, rigid or flexible); metalloporphyrin´s central metal ion; protection of the free NH2 on the support. In general way, the investigated systems show to be efficient catalysts for all substrates, mainly from the supports, proving that the covalent bond is an efficient method for the preparing of anchored catalysts. The stability of these systems was also investigated through developing four reaction cycles with the same solid catalyst, with reproducible products yields. The FeP were more efficient catalysts than the MnP, even using imidazole as co-catalyst for the MnP. The better catalysts were FeAPS, FeMont1, FeDAHS, FeTal2 and FeSil2. All these solid catalysts are rigid and inert, with an intermediary ?arm? size (4-7 atoms). The protection of the support?s free NH2 groups is fundamental to improve the catalytic efficiency of metalloporphyrins anchored on aminofunctionalized supports, because it avoids the bis-coordenation of the NH2 groups to the metal ion, effect responsible for the blocking of the catalytic site. Although the oxidation products yields using H2O2 have been lower than these using PhIO, some systems were more efficient when compared to the homogeneous systems both in product yield and selectivity. These results open the possibility for utilization of the system for drugs and poluents oxidation.
