Komplexní teoretická analýza metody sloupku pro zjišťování zbytkových napětí / Comprehesive Theoeretical Analysis of Ring-Core Method for Residual Stress Determination

Comprehensive analysis of the ringcore method used for the determination of the residual stresses in mechanical components is presented in this thesis. Principles, advantages, disadvantages and applicability of this semidestructive experimental method are discussed too. At the same time the ringcore method is compared with the hole drilling method, which is used more frequently. All aspects of the ringcore method are analyzed by the finite element method. FE simulations, performed on the universal numerical model, verified principles of the integral method and the incremental strain method. FE simulations also provided basic information for the uncertainty analysis, which significantly affects the accuracy of the residual stress measurement. The main goal, which the present work deals with, is to create a global overview of all ringcore methods´ aspects elaborated in a clear and complex form.

Following thesis focuses on analysis of using five basic transfer pricing methods mentioned in the OECD Transfer Pricing Guidelines in the conditions of the Czech Republic (comparable uncontrolled method, resale-price method, cost plus method, profit split method, transactional net margin method). There are mentioned the problems connected with application of these methods and also suggestions how to fix them. In this respect thesis contains comparable, functional and economic analysis. As a part of the thesis there is an example of the transfer pricing documentation using cost plus method. Next there are noted the ways and criteria of selecting an appropriate method. There are also solved some tax aspects of determining the transfer prices.

Cílem diplomové práce je vytvoření 2D numerického modelu pohybujícího se řezu s proměnnými okrajovými podmínkami skutečné geometrie plynulého odlévání a chlazení předlitku v prostředí MATLAB. Model se zabývá vysoce nelineárními termofyzikálními podmínkami oceli během tuhnutí a chlazení. V práci je simulovaná nejen nelinearita termofyzikálních podmínek, ale také nelinearita při fázové změně. Fázová změna je modelovaná pomocí metody entalpie, metody zdánlivé kapacity a metody teplotního zotavení. Všechny výsledky práce jsou porovnány z více hledisek, jako např. z hlediska přesnosti, rychlosti výpočtu, nebo vhodnosti časového diskretizačního kroku pro nelineární problémy, a paralelizace.

This thesis deals with the topic of assessing the design load-bearing capacity of a prestressed concrete railway bridge structure. The determination of the structure’s load-bearing capacity is carried out using probabilistic nonlinear analysis using the finite element method. Load-bearing capacity is determined for the recommended limit states. A fully probabilistic approach is compared to other reliability semi-probabilistic methods, such as the ECoV method according to Červenka and Eigen ECoV, and deterministic methods, like partial safety factor method, or the method according to ČSN EN 1992-2. The results are evaluated in terms of accuracy, consistency and time consumption for each method.

Title: The effect of sensomotoric stimulation on flat foot of children Introduction: Flat foot is a common health problem in childhood. Its development is related to congenital or acquired dysfunction of the plantar vault. There is no single opinion on the criteria for diagnosis and therapy forms. The aftereffects and health risks of flat foot are often neglected. The therapy form of sensomotoric stimulation, including balance exercises and walking barefoot over different surfaces, could be an appropriate part of the physical activities and games for children in kindergarten. Objectives: The main objective of this thesis was to compare foot prints of a group of children from one kindergarten class before and after six months of sensomotoric stimulation by three different methods (the Chippaux-Šmiřák method, the Godunov-Sztriter method and the Mayer method.). The second objective was to determine at what age category will be the highest prevalence of flat feet and also at what age group is the biggest improvement of the foot arch. Influenza virus causes severe respiratory infections in birds and mammals and it is responsible for up to half a million deaths of human beings worldwide each year. Two molecular targets in influenza viral life cycle, neuraminidase and M2 proton channel are exploited in treatment. However, the recent emergence of new pandemic type along with increasing resistance against approved drugs has urged the need for a new drug target discovery and potential search of its inhibitor. Recently, an interesting protein-protein interaction between two subunits PA and PB1 of influenza A viral polymerase has been identified by X-ray crystallography as a new promising drug target. The fact that relatively few residues drive the binding and that the binding interface is highly conserved presents an intriguing possibility to identify antiviral lead compounds effective against all subtypes of influenza A virus. In our laboratory, we expressed and purified two fusion tag constructs of the recombinant C-terminal domain of polymerase acidic subunit (CPA) from the pandemic isolate A/California/07/2009 H1N1. First, GST-CPA fusion protein was used for kinetic evaluation of PA-PB1 interaction by surface plasmon resonance. This thesis deals with the issue of genetic method to reading in the first grade of an elementary school. The theoretical part is focused on the child's entry to school, their school maturity and further education. Other methods to read are presented, analysed and compared with the genetic method as well. Furthermore, the methods and ways how to put the genetic method in practice are pursued in detail along with didactical games and examples of teaching procedures. The practical part looks into the experience the teachers have gained with this method, the school success of the children and the opinion the family members have.
