The thesis deals with the analysis of the phase feeding in herds of Holstein cattle in specific operating conditions and its relation to milk yield and health of dairy cows. In the reference event to be optimized ration drystanding cows and lactating cows in comparison protocols laboratory analysis of mixed rations to standard NRC by the enterprise uses for compiling rations. The thesis was evaluated the effect of ration on milk yield and content of milk components and also affect the incidence of metabolic disorders. The aim of the thesis was to analyze the phase nutrition of dairy cows in the reporting company in relation to milk production and health, to evaluace the quality of feed rations, some feed and feeding technique. It was found that the monitored parameters do not match any breed mixed rations used in the standard, which could lead to reduced levels of production and reproduction and deterioration in health of dairy cows. It was further found that in the observed rearing occur very frequent occurrence of metabolic disturbances and fluctuations in the milk production.

The aim of the trial was to analyze selected indicators of milk yield and economics of milk production in dairy herd of Holstein cattle. Economics of dairy cows is critical to keeping cattle on the farm. Among the main priorities that can improve the economic results of cattle, are mainly production conditions corresponding milk yields, good fertility, high quality market products, quality dairy nutrition, good health, appropriate management of breeding and compliance of welfare in the cattle breeding, because good health belongs among main conditions of economically successful milk production. In dairy cattle breeding herd of Holstein cattle in the farm Meclovská zemědělská, a. s. there were monitored 1081 dairy cows on the first to eleventh lactation for the two years (2010-2011). From the results, there was clear that 2010 was achieved higher milk yields (9 080,87 kg/lactation) than in 2011 (7 857,83 kg/lactation), a higher content of milk fat and protein, it was achieved lower costs (181,44 CZK/FD and 6,98 CZK/kg of milk) than in 2011 (199,40 CZK/FD and 7,92 CZK/kg of milk) and higher profit.

Vyhodnocení vybraných vlivů na vyřazování a dlouhověkost dojnic / Evaluation of selected impacts of elimination and longevity of dairy cows

The aim of this thesi is the evaluation of selected impacts of elimination and longevity of Holstein and Czech spotted dairy cows on the Ing. Bohuslav Vacka´s farm in Vrchotovy Janovice according to the reason of this elimination, the age at the first calving, milk production of the first milk lactation, lifetime milk lactation and the number of milk lactation during the lifetime. I also evaluated functional longevity (from the first calving to retirement) and the longevity of life (from the birth to retirement) of retired dairy cows and the relationship between age at the first calving, milk production of the first lactation and lifetime milk production. In monitoring were included 359 pieces of cows eliminated during five years, from 1 January 2010 to 31 December 2014. From these, 217 pieces of Holstein cows and 142 pieces of Czech spotted cows. Both breeds were stalled in the same barn with the same nutrition. At the monitored group were eliminated altogether 31.22% of dairy cows. Holstein cows were excluded very often from other medical reasons (28.57%) and the Czech spotted cows for fertility disorders (45.77%). Higher milk productivity and higher average number of lactations for the life reached eliminated Holstein cows in comparison with the Czech spotted cows. Longer lifetime longevity reached Holstein cows (63 months) than Czech spotted cows (53.6 months) and among them was found highly significant difference at a significance level of p 0,001. Higher longevity and lifetime milk production in kg of milk reached Holstein cows calved in 25 to 26 months and Czech spotted cows calved within 27 to 28 months. The relationship between age at the first calving and lifetime milk production at Holstein (r = 0.018) and Czech spotted cows (r = 0.0434) proved to be an insignificant dependence and the relationship between milk production at the first lactation and lifetime milk production at Holstein (r = 0.3317) and Czech spotted cows (r = 0.4436) proved to be as a highly significant correlation (p 0.001).

Os estafilococos estão entre os principais contaminantes do leite, seja em decorrência da mastite ou de falhas de higienização. A contaminação do leite pode favorecer a adesão bacteriana sobre superfícies com a formação de biofilmes, cujos fragmentos podem se desprender e contaminar o produto durante o processo de beneficiamento, o que representa um risco à saúde do consumidor. Tendo isto em foco, objetivou-se o presente estudo, em uma micro-usina do Estado de São Paulo, a fim de investigar a presença e formação de biofilme por Staphylococcus spp, antes e após o processo de higienização. Colheu-se o total de 60 amostras por meio de suabes, antes e após o processo de higienização, das superfícies do tanque de recepção, do tanque de estocagem de leite cru, da tubulação de saída do pasteurizador, do tanque de estocagem de leite pasteurizado, e da tubulação da máquina de envase. Foram colhidas, ainda, amostras de leite no tanque de recepção, no tanque de estocagem de leite cru, na tubulação de saída do pasteurizador e amostras de leite envasado, assim como das embalagens plásticas vedadas e vazias, utilizadas para o envase do leite pasteurizado. Dentre 41 estirpes de Staphylococcus spp isoladas, 16 (39,0%) mostraram-se positivas na prova da coagulase, enquanto que 25 (61,0%) foram negativas. Por meio de análise genotípica, com a técnica de Reação em Cadeia da Polimerase (PCR), pode-se observar que, dentre as 16 estirpes coagulase positivas, quatro (25%) apresentavam o gene icaA e 16 (100%) possuíam o gene icaD. Dentre as 25 estirpes coagulase negativas, 11 (44%) possuíam o gene icaA e 25 (100%) possuíam o gene icaD. Visualizou-se, por meio da microscopia eletrônica de varredura... / Milk is a highly nutritious food and excellent substrate for the multiplication of microorganisms. Staphylococci are one of the major contaminants of milk whether due to mastitis or hygiene failures in cleaning. Milk contamination may encourage bacterial adherence on surfaces with the formation of biofilms, whose fragments can detach and contaminate the product during beneficial processing, which represents a health risk to the consumers. With this in focus as the objective of this present study, to investigate the presence and formation of biofilm of Staphylococcus spp before and after the cleaning process in a micro-dairy plant in São Paulo State. A total of 60 swab samples were collected before and after the cleaning process from the reception tank surfaces, the raw milk storage tank storage, the pasteurizer outlet pipe, the pasteurized milk storage tank, and the filling machine. Further samples were collected of the milk in the receiving tank, the raw milk storage tank, from the pasteurizer outlet pipe, and packaged milk, as well as the empty sealed plastic packaging used for packaging the pasteurized milk. Of the 41 strains of isolated Staphylococcus spp, 16 (39.0%) indicated positive in the coagulase test, while 25 (61.0%) were negative. Through genetic analysis, using the Polymerase Chain Reaction (PCR) technique, it was observed that within the 16 strains of coagulase-positive, four (25%) presented the gene icaA and 16 (100%) had the gene icaD. Of the 25 strains of coagulase-negative, 11 (44%) had the gene icaA and 25 (100%) had the gene icaD. It was seen using a scanning electron microscope the start of bacteria adhesion and the formation of biofilm for all the isolated strains. Mléko a mléčné výrobky ve výživě dětí základní školy / Milk and dairy products in alimentation of primary school children

Milk and milk products are irreplaceable in the children´s nutrition. Substances which milk contains assist children to the proper growth and development. The most important substances are for example proteins or calcium and therefore it´s necessary to feed children with milk. Children consume milk practically since birth. However, milk consumption may be associated with health problems, such as milk allergy or lactose intolerance. In order to increase the consumption of milk was introduced subsidized program "School Milk", which can be used by all kindergartens, primary and secondary schools in the Czech Republic. The research was based on a quantitative approach. The research tool has become a questionnaire given out to three grades of selected elementary school. Research shows that the selected students consume milk in sufficient amounts, especially cow's milk, which has chosen the 93.8% of respondents. It is primarily milk skimmed and not flavoured. Respondents prefer the most milk itself, then yogurt (56.9%), cheese (55.1%) and butter (50.2%), regardless of age or gender. Among the most popular cheeses include semi-hard cheese, which prefers the 82 respondents from a total of 225. Dislikes are then other fermented products such as sour buttermilk, which is preferred only by 7.6% of respondents, or kefir. Some of the products are completely unknown to the pupils. For example sour milk never tasted more than 71% of respondents. In the health effects and milk production pupils have very limited knowledge; especially pupils in lower grades have very limited knowledge. To name at least one important substance for human health was able to do less than half of the respondents. The most commonly mentioned was calcium and protein, and other substances only sporadically. Also knowledge of milk production has proven to be very tricky. A total of 56.9% of respondents said they do not know any dairy. Percentage using a subsidized program "School Milk" is very low. This program uses only 32.9% of pupils, especially in the lower grades. The research showed that 17.3% of students are overweight or obese, especially the girls in the 5th grade.

Thesis deals with the finding reliable measurement of the quality of milk by automatic milking system. The observed reliability of the system to measure the quality of milk is a significant parameter for herd management and also an effective tool for farmers. Thanks notice on udder health in the program T4C farmer is able to detect a number of health problems or deficiencies in the herd before they break out in full force. This gives the farmer more options to treat dairy cows, such as inflammation, at the very beginning and as such can successfully use homeopathic treatment.
