Physicochemical Properties of a ‘Magic Mouthwash’ for Chemotherapy Induced Oral MucositisWilliams, Evan, Stearley, Jacob Dale January 2011 (has links)
Class of 2011 Abstract / OBJECTIVES: To determine the solubility and stability of hydrocortisone in a ‘magic mouthwash; suspension. METHODS: A literature review was conducted to establish the most common ingredients in a ‘magic mouthwash’ suspension It was decided that the test suspension would consist of 75% commercially available diphenhydramine solution, 12.5% nystatin suspension (100,000 units/ml) , and 12.5% lidocaine solution (2% lidocaine). Powdered hydrocortisone was then added to the test suspensions at different concentrations and stored at 27C, 38C, and 48C. Aliquots were taken from each of the test samples at the time of compounding and at 4, 7, 13, 19, and 26 days to be analyzed by HPLC for degradation of hydrocortisone and percent hydrocortisone in suspension. RESULTS: At 27C, 98.5% of hydrocortisone was recovered after 26 days, versus 33.7% at 38C, and 7% at 48C. The solubility of hydrocortisone in the suspension was higher at higher temperatures, with 82% in solution at 48C, 70% at 38C, and 38% at 27C.
CONCLUSION: The amount of hydrocortisone recovered deteriorated over time and at higher temperatures, and solubility of hydrocortisone in the suspension was greater at higher temperatures.

Os MN se originam a partir de fragmentos de cromossomos ou cromossomos inteiros que se atrasam na anáfase durante a divisão celular podendo ocorrer devido à exposição excessiva a agentes nocivos, defeitos na mitose e/ou processo de reparação do DNA. 45 indivíduos, com idades variando entre 17 a 42 anos e instituídos como critério de exclusão: ingestão de bebida alcoólica, fumantes; doença periodontal; doenças sistêmicas que possam interferir na integridade da mucosa jugal; indivíduos com histórico médico ou farmacológico que afetem o desempenho do estudo, histórico familiar de neoplasias malignas. A amostra foi dividida em 02 grupos experimentais e 01 grupo controle; estes realizarão enxágüe bucal com: água destilada (controle), Colgate Plax® sem álcool e Listerine® (por 03 semanas foi realizada a coleta citológica na mucosa jugal e porção mediana da borda de língua. O material coletado foi corado por coloração específica de Feulgen/Fast Green. A análise de 3000 células/indíviduo foi realizada por microscopia óptica e a estatística utilizou os testes Kruskal-Wallis e Mann-Whitney para freqüência de MN e células micronucleadas. This damage can occur due to excessive exposure to harmful agents, defects in mitosis and / or DNA repair process. A sample of 45 individuals ranging from 17 to 42 years and established as a criterion for exclusion of alcohol intake greater than 01 alcoholic drinks weekly dose smoking, presence of periodontal disease, systemic diseases that may interfere with the integrity of oral mucosa, individuals with medical or pharmacological history that affects the performance of the study and individuals with a family history of malignancies. These individuals were divided into two experimental groups and one control group: distilled water (control), Colgate Plax Alcohol Free® and Listerine ® (for a period of 03 weeks). Once a week we performed the cytologic collection from left side of the buccal mucosal, and the middle portion of the edge of the tongue on the left. The collected material was stained with specific staining Feulgen/Fast Green. An analysis of 3000 cells / individuals was performed by optical microscopy and statistical tests used the Kruskal-Wallis and Mann-Whitney test for frequency of MN and micronucleated cells. Stability of Tetracycline Hydrochloride in Miracle Mouthwash Formulations Containing Diphenhydramine and Dexamethasone ElixirFazel, Mahdieh, Goodlet, Kellie, Myrdal, Paul, Karlage, Kelly January 2015 (has links)
Class of 2015 Abstract / Objectives: To assess the solubility and stability of tetracycline in compounded miracle mouthwash solutions over time, and at different temperatures (room temperature versus refrigerated) and pH (unaltered versus pH 7).
Methods: Miracle mouthwash (MMW) solutions were compounded using tetracycline HCl capsules and 1:1 pseudo-dexamethasone elixir and diphenhydramine. High-performance liquid chromatography (HPLC) was used to measure the tetracycline concentrations in the MMW samples tested. Data on tetracycline crystal composition over time were also collected using powder x-ray diffraction, differential scanning calorimetry (DSC), and thermal gravimetric analysis (TGA).
Results: For the tetracycline MMW solutions stored at room temperature, only 16% of the original tetracycline remained in solution after 24 hours, stabilizing at 65-81 mcg/mL on day 5 then decreasing further down to 45 mcg/mL by day 15. Similar results were obtained for the refrigerated tetracycline MMW solution (11% of original concentration after 5 days, with a decrease from 31-54 mcg/mL on day 5 to 22 mcg/mL on day 15). Tetracycline concentrations appeared to undergo a steeper decline in MMW solutions of pH 7 than in unadjusted MMW solutions (pH 4.68). All MMW samples exhibited a conversion from tetracycline HCl to tetracycline hexahydrate.
Conclusions: Tetracycline solubility decreases rapidity in MMW within 24 hours of compounding regardless of temperature. MMW solutions at pH 7 may have further reduced solubility. Stability decreases at a stable rate from tetracycline HCl to tetracycline hexahydrate.

Avaliação de enxaguatório bucais na atividade biológica do biofilme formado em braquetes ortodônticos / Evaluation of oral Rinses in biological activity of biofilm formed in orthodontic bracketsBussadori, Celene Marques 25 September 2013 (has links)
Na Odontologia, existe uma grande preocupação com a formação de placa bacteriana ou biofilme devido ao seu potencial cariogênico. Essa atenção se estende à ortodontia, devido ao acúmulo de biofilme formado nos aparelhos ortodônticos, tanto nos aparelhos removíveis quanto nos aparelhos fixos. É muito comum a presença de lesões brancas após a remoção dos bráquetes, em torno de 50 a 75% dos pacientes, provocadas pela presença do biofilme. É necessário um controle químico e mecânico rigoroso, a fim de se evitar alterações irreversíveis nos dentes e periodonto. O controle mecânico se dá pela escovação adequada, pelo correto uso de fio dental e pela frequência desses hábitos, além, claro, de um acompanhamento profissional periódico. O controle químico pode ser viabilizado pelo uso de enxaguatórios bucais, nos quais residem alguns efeitos indesejáveis e contraindicações relacionadas ao uso e à idade dos pacientes. Este estudo tem como objetivo avaliar comparativamente dois enxaguatórios bucais, um de natureza comercial (Cepacol®), e outro não comercial, elaborado com extratos hidroalcoólicos das plantas Nasturtium officinale (agrião), Rosmarinus offinalis (alecrim), Tabebuia impetiginosa (ipê roxo), Achillea millefolium (mil folhas) e Plantago major (tanchagem), desenvolvido na Faculdade de Ciências Farmacêuticas/UNESP Araraquara. Foram selecionados 40 pacientes, jovens e adultos, entre 18 e 45 anos, usuários de aparelho ortodôntico corretivo (fixo), divididos em dois grupos, o grupo do Cepacol e o grupo do Enxaguatório base de plantas. Foram coletadas amostras nos períodos de 0, 7, 15 e 21 dias e analisadas observando-se as Unidades Formadoras de Colônia (UFC) de cada amostra. Houve uma queda significativa da quantidade de bactérias após o uso do Enxaguatório à base de plantas, revelando um produto de extrema utilidade no auxílio do controle do biofilme bacteriano nas superfícies dentárias. / In dentistry, there is a great concern with the formation of plaque or biofilm, due to its cariogenic potential. Effect of Antibacterial Mouthwash on Basal Metabolic Rate in Humans : A Randomized, Double-blinded, Cross-over StudyAgell, Blenda January 2013 (has links)
The use of mouthwash is a common complement to oral care. However, the physiological implication of this use, besides of effects on oral hygiene, is poorly known. The research of the gut micro flora and its implications on the host is a very active area of research today. Many important connections between the gut micro flora and obesity and diabetes have been found. These billions of bacteria are part of the immune system, they produce essential vitamins and they make inaccessible polysaccharides more digestible to the host, just to mention a few of their symbiotic roles for the host. A less explored area is the micro flora in the oral cavity. On the back of the tongue, anaerobic bacteria can reduce dietary nitrate to nitrite which then further can be reduced to nitric oxide, NO. NO is important in several important biological functions, e.g. as a signal substance, vasoregulation, mucus production and antibacterial effects. Vegetables as beetroot and spinach are dietary sources with a high nitrate content. Also drinking water and processed meats can be of relevance. Nitrite is added to processed meat for the prevention of botulism but also adds taste and color. Experiments on humans indicate that mitochondrial efficiency increases after nitrate load, manifested as a decreased oxygen demand during physical exercise. This can also be relevant under conditions where the mitochondrial function is impaired, such as in diabetes and cardiovascular diseases. First a pilot study was made to evaluate the nitrate reducing effect from the antibacterial mouthwash. The mouthwash proved very effective. The concentrations of nitrate and nitrite in saliva was analyzed by HPLC and saliva from the antibacterial treatment showed greatly reduced concentrations of nitrite and high concentrations of nitrate. Saliva from placebo mouthwash showed high concentrations of nitrite and low concentrations of nitrate as expected. To study the importance of oral bacteria on metabolism, we performed a randomized, cross-over double-blinded study with 19 healthy males between 22-43 years. During two separate three-day periods they used an antibacterial and placebo mouthwash, respectively. On the fourth day their basal metabolic rate (BMR) was measured with an indirect calorimetric system. Moreover, samples from saliva, urine and blood were collected but these results are not included in this thesis. An earlier, unpublished study has demonstrated that nitrate administration reduces the basal metabolic rate. Accordingly, our aim was to study potential effects on the basal metabolic rate following reduction of the number of oral bacteria by aid of antibacterial mouthwash. Our hypothesis was that the reduced availability of nitrite would decrease the availability of NO in the body and manifest as an increased basal metabolic rate. The results from indirect calorimetry measurements showed no significant difference between placebo and antibacterial mouthwash, but there may be confounding factors. Further study is needed to assess the potential effects on host metabolism by these bacteria.

Among the plants used, Libidibia ferrea (L. ferrea), known as jucá or iron wood, is widely used in folk medicine by presenting anti-inflammatory, analgesic, antimicrobial and antipyretic therapeutic properties. The aim of this study was to evaluate in vitro pharmacological stability of a mouthwash of L. ferrea extract (INPA - 228 022), therefore, analyzed the antimicrobial activity of the formulation against microorganisms present in the biofilm - Streptococcus mutans (ATCC 25175), Streptococcus oralis (ATCC 10557), Streptococcus salivarius (ATCC 7073), Lactobacillus casei (ATCC 7469) and Candida albicans (DPUA 1706) by microdilution technique, determining the minimum inhibitory concentration of the formulation; it was held the mouthwash contaminants control by determining the total number of microorganisms and Salmonella sp, Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus; stability characteristics ( color, odor, brightness and consistency) were analyzed, the pH measurement, sedimentation test and density evaluation; cytotoxicity was assessed by hemolysis and cell culture test; it was tested the erosive potential by microhardness test (Knoop hardness) and verified the ability of the mouthwash to induce dental stain (spectrophotometer / CIE lab). The results were tabulated and described by descriptive statistics and when indicated, through non-linear regression, calculating the mean and standard deviation (SD), Kolmogorov-Smirnov, Bartlett and Tukey’s tests and Analysis of Variance (ANOVA). The mouthwash presented bactericidal activity against all the organisms tested in the concentrations of 0.55 mg/ mL, 0.6 mg/ mL, 0.65 mg/ mL and 0.25 mg/ mL for S. mutans, S. oralis, S. salivarius and L. casei, and fungicidal activity of 0.25 mg/ mL for C. albicans. No changes were observed in the organoleptics and sedimentation characteristics such as microhardness of dental enamel. The contaminants test evaluation showed to be absent for all organisms studied. The average pH values were 6.21, 6.15 and 5.85 at 0, 30 and 60 days, respectively, and 1.029, 1.035 and 1.033 g/ L density values, with statistical difference over time, but without interfering with the final characteristic of the formulation. The L. ferrea mouthwash showed no hemolytic activity, but the vehicle caused hemolysis with EC50 of 300.5 mg / mL. The test showed that the Alamar BlueTM mouthwash induced no cytotoxic effect when in fibroblasts (MRC-5) contact, with IC50 values above 50 mg / mL, but induced significant color change of bovine teeth. Therefore, it can be concluded that the L. ferrea mouthwash presented conditions of stability and quality without contaminants; bactericidal and fungicidal activity against microorganisms of the biofilm; it was not cytotoxic by hemolysis and cell culture tests; it did not change the enamel microhardness, however caused color change after prolonged use. Additional studies are needed to improve the mouthwash characteristics in its ability to change color and make it feasible for use in everyday practice. / A utilização da Fitoterapia constitui um método terapêutico que pode ser utilizado para a prevenção e tratamento de doenças bucais, tais como cárie e doença periodontal. Dentre as plantas utilizadas, a Libidibia ferrea (L. ferrea), conhecida como jucá ou pau ferro, é bastante utilizada na medicina popular por apresentar propriedades terapêuticas anti-inflamatória, analgésica, antimicrobiana e antitérmica. O objetivo deste estudo foi avaliar, in vitro, a estabilidade farmacológica de um enxaguatório bucal fitoterápico à base do extrato de L. ferrea (228.022- INPA). Para tanto, analisou-se a atividade antimicrobiana da formulação frente a micro-organismos presentes no biofilme dental - Streptococcus mutans (ATCC25175), Streptococcus oralis (ATCC10557), Streptococcus salivarius (ATCC7073), Lactobacillus casei (ATCC7469) e Candida albicans – (DPUA 1706) pela técnica de microdiluição em caldo, determinando-se a concentração inibitória mínima da formulação; foi realizado o controle de contaminantes do enxaguatório através da determinação do número total de micro-organismos e pesquisa de Salmonella sp., Escherichia coli, Pseudomonas aeruginosa e Staphylococcus aureus; foram analisadas as características de estabilidade (cor, odor, brilho e consistência), a aferição do pH, sedimentação e densidade; avaliou-se a citotoxicidade através do teste de hemólise e cultura de células; foi testado o potencial erosivo através do teste de microdureza (dureza Knoop) e verificou-se a capacidade de manchamento dentário do enxaguatório (espectrofotômetro/ CIE lab). Os resultados obtidos foram tabulados e descritos pela estatística descritiva e quando indicado, através de regressão não-linear, cálculo da média e desvio-padrão (DP), teste de Kolmogorov – Smirnov, teste de Bartlett's, Análise de Variância (ANOVA) e Tukey. O enxaguatório apresentou atividade bactericida frente a todos os micro-organismos testados nas concentrações em 0,55 mg/ mL; 0,6 mg/ mL; 0,65 mg/ mL e 0,25 mg/ mL para o S. mutans, S. oralis, S. salivarius e L. casei, respectivamente, e atividade fungicida em 0,25 mg/ mL para a C. albicans. Não foram observadas alterações das características organolépticas, sedimentação e microdureza do esmalte dentário. O teste de avaliação de contaminantes mostrou-se ausente para todos os micro-organismos pesquisados. Os valores médios de pH foram de 6,21, 6,15 e 5,85 nos tempos de armazenamento de 0, 30 e 60 dias, respectivamente e de densidade 1,029, 1,035 e 1,033, havendo diferença estatisticamente significante ao longo do tempo, porém sem interferir na característica final da formulação. O enxaguatório de L. ferrea não apresentou atividade hemolítica, porém o veículo causou hemólise com CE50 de 300,5 μg/ mL. O teste de Alamar BlueTM mostrou que o enxaguatório não causou efeito citotóxico quando em contato com fibroblastos (MRC-5), apresentando valores de CI50 acima de 50 μg/ mL, porém causou significativa alteração de cor dos dentes bovinos. Portanto, pode-se concluir que o enxaguatório de L. ferrea apresentou condições de estabilidade e qualidade sem contaminantes no produto; atividade bactericida e fungicida frente aos micro-organismos do biofilme dental; não foi citotóxico frente ao teste de hemólise e de cultura de células; não alterou a microdureza do esmalte dentário, porém causou alteração de cor após uso prolongado. Estudos complementares necessitam ser realizados para melhorar as características do enxaguatório quanto à capacidade de alteração de cor e torná-lo viável para uso na clínica diária.
