The PD-1 pathway and the complement system in systemic lupus erythematosus

Autoimmune diseases occur in up to 3-5% of the general population and represent a diverse collection of diseases with regards to clinical manifestations. The unifying factor of autoimmune diseases is tissue and organ damage as a result of an immune response mounted against self-antigens. Systemic lupus erythematosus (SLE) is considered a prototype of human systemic autoimmune diseases. The etiology of SLE is as yet largely unknown, but both epidemiological and genetic data suggest an interplay between numerous and varying genetic and environmental factors. There is compelling evidence for a strong genetic component in SLE. The disease has a high λsibs value and familial clustering is apparent. Multiple susceptibility loci have been identified, some of which are syntenic between humans and mice and some of which overlap with other autoimmune diseases.   This thesis is based on analysis of Icelandic multicase SLE families and Swedish SLE patients. Paper I is a study of the association of C4A protein deficiency (C4AQ0) with SLE in the multicase families and shows a significantly increased frequency of C4AQ0 in the families. The genetic basis for C4AQ0 varies and C4AQ0 is found on different MHC haplotypes, pointing to C4AQ0 as an independent risk factor for SLE. Paper II describes the association of low MBL serum levels with SLE in the families and identifies low MBL as risk factor for SLE in families that carry the defect. Low MBL was furthermore found to mediate an additive risk when found in combination with C4AQ0. In paper III cellular expression the PD-1 co-inhibitory receptor on T cells was studied. A polymorphism in the PDCD1 gene, PD-1.3A was previously associated with SLE in the multicase families. The polymorphism is thought to disrupt expression of the gene and may lead to decreased expression of the PD-1 receptor. The study demonstrates lower PD-1 expression in SLE patients and relatives in correlation to the PD-1.3A genotype. Paper IV is a compiled analysis of the SLE families, including PD-1.3A, C4AQ0, low MBL, autoimmune diseases and autoantibody profiles. The study demonstrates clustering of different autoimmune diseases and autoantibodies in families that are heterogenic with regards to the genetic susceptibility factors, PD-1.3A, C4AQ0 and low MBL.

Multicase Historic Studies of Innovative Work Behavior Among Intellectually Gifted Adults

ABSTRACT This qualitative research study was conducted to examine whether investing in innovation and gifted intelligence would improve America's economic well-being. The investment issue became a problem because educational researchers researched innovation as creativity rather than as productive behavior. Moreover, in the literature, adult giftedness was perceived as negative development rather than as alternate normative behavior. The purpose of this research study was to generate new knowledge about innovation and adult giftedness. WICS (wisdom, intelligence, creativity, synthesized) theory of intelligence provided the theoretical framework because intelligence is a measure of human productivity potential. The neuropsychological conceptual framework facilitated a cognitive map of the innovation process. The multicase historic research design provided the answers to the research questions. Four real life historic events embedded with innovation activities and behavior utilized comparative methodology to mark patterns in behavior and cognition. Raw data gleaned from archival/historic research was analyzed utilizing content analysis of primary resources. The key results were: (a) the innovation process is a psychological tool that transcends creative activities; (b) empathy, intellectual complexity and moral intelligence is linked to wisdom and continuous learning; and (c) transcendent experiences are intrinsic motivations to reach beyond expected productive behavior. Outcomes from this study are useful for I-O psychologists because high intelligence can increase organizational productivity levels and sustain business. The positive social change implications are business administration will have to redirect business strategies to focus on employees and relationships, and training and development.

Public Health Implications of Retailer Resale of Returned Textile Clothing Merchandise

Historically, 18th century anecdotal accounts of the decimation of several tribes of U.S. of the Native American population by trading of infected textile blankets alludes to the role of retail in the transmission of infectious disease. This study explores implications of the modern day retail organization practice of reselling returned clothing textiles from a public health infectious disease perspective. A qualitative multi-case study, utilized responses from 20 open-ended, unstructured interviews of retail employees assigned to the returns process. Additionally, several informal observations of select U.S.based, top-ranked clothing retail organizations, identified by the National Retail Federation were completed. Select federal, state, and local public health regulations regarding returned clothing textiles were then examined in an attempt to identify potential public health risks. Under a general systems conceptual framework, the points of interaction between the complex adaptive systems seen in retailer and the public health organizations were examined for infectious disease and infestation implications. Using MAXQDA software to perform the analyses, it was found that current retail practices and policy present unacknowledged infectious disease or infestation transmission risks. The risk applies to all, but is particularly relevant to immuno-compromised individuals. Though the risk in accepting and returning clothing to the sales cycle is an industry wide practice, it can be mitigated. Suggested mitigation takes the form of health training, and introduction of disinfection sanitizing tools such as UVC light exposures, into the returns-resale process.The findings point to an opportunity for social change for consumers, retail workers, and the community through update of public health and retail practices.

La fabrique d'une compétence stratégique, proposition d'un modèle : une application aux impacts des usages des technologies de l'information en PME / The build of a strategic competence, proposal of a research model : an application to the impacts of IT use in SMEs

L’objet de notre recherche est double : comprendre le phénomène de fabrique d’une compétence organisationnelle stratégique induite par les usages des TI, d’une part, et aller au-delà des modèles existants d’analyse des impacts des TI en intégrant une approche multidimensionnelle, d’autre part. Nous avons privilégié une approche empirique qui a débuté par une étude de cas pilote qui nous a permis de délimiter un cadre théorique de manière abductive, par un aller-retour constant entre le terrain et les construits théoriques existants. Le cadre conceptuel se veut une synthèse des différents courants RBV intégrant le concept d’apprentissage organisationnel. Les résultats d’une étude multicas soulignent le caractère diffus et partiellement intentionnel d’un processus requérant l’interaction de ressources complémentaires fortement dépendantes du passé et aboutissant à l’amélioration de l‘efficience des routines et de la capacité dynamique d’apprentissage. Nous proposons une modélisation du processus et nous discutons des concepts significatifs quant à l'étude d'un tel phénomène. / Our research object is twofold: understand a strategic organizational competences bulding using IT, on the one hand, and go beyond the existing analysis of impacts of the IT models, integrating a multidimensional approach, on the other hand. We privileged an empirical approach beginning with a pilot case study that allowed us to define a theoretical framework of an abductive way, by a constant back and forth between the field and the theoretical existing constructs. The conceptual framework is a synthesis of the various RBV strands integrating the concept of organizational learning. The results of a multicase study underline the diffused and partially intentional character of a process, requiring interaction of additional resources highly dependent from the past, and leading to improve efficiency of the routines and dynamic adaptability. We propose a design of the process and discuss significant concepts for the study of this phenomenon.
