This work deals with the life and work of historian, educator, and priest Josef Sakař. Josef Sakař is one of the most prominent Týn nad Vltavou natives. His monumental four-volume work, The History of the town Tyn nad Vltavou and the Surrounding Area, includes the building blocks of South Bohemian regional history. Even though the greater balance of the work was written in the 1930s, to this day it has not been surpassed. Pardubice, like Týn nad Vltavou, owes much to Josef Sakař. It is the city where he spent most of his productive years and where he worked as a secondary school teacher. He wrote his major works here, among them a six-volume history of Pardubice nad Labem. Josef Sakař's activity was more than merely regional: for his efforts to protect historical monuments he was appointed (as the only one of thirty candidates from across the country) an honorary member of the Club for Old Prague in 1925.This thesis analyzes and compares sources, and it determines the importance and value of key events, family ties, friends and personalities that influenced Josef Sakař's life. For 21st-century educators, the thesis looks for inspiration that can be taken from his life and works. The work focuses on Josef Sakař's relationships with relatives, his life's story and activities in Týn nad Vltavou and Pardubice. It is organized chronologically, from his birth to his death. The last part concerns itself with seeking traces of evidence of Josef Sakař in contemporary life. A separate section is devoted to an analysis of four of his key works. The work includes all biographical data and important life milestones of Josef Sakař and contains a complete bibliography.

This diploma thesis deals with the preventive assessment of the landscape character of the cadastral territory of Rabštejn nad Střelou. The evaluated area extends into the Nature Park and there is a small specially protected area called Střela Nature Reserve. The research area outside of the natural wealth is also rich in historical buildings. The research part has the focus on the analysis of the landscape of Rabštejn nad Střelou. The territory has been assessed on the basis of identified natural, cultural and historical features and values of landscape character. For each of the traits, its significance, expression and value were determined. For the cadastral area the degree of preservation of the landscape character, its aesthetic value and the degree of protection were determined. On the basis of the analysis, specific proposals for increasing the value of the landscape character were set.

This farm is compared with similarly oriented foreign farm in the United Kingdom, whose image is constructed from the survey.

My dissertation thesis The interface between city and the landscape, centers on an area of the locally prominent rock Marianska skala in the city Usti nad Labem. The final design is a form of urbanistic-architectural study. The design methodology is based on city analysis and public research as mental mapping. The subject area has strong potential and natural values, however at present there is a minimal usage of this area by the local residents. The project therefore aims to integrate the area into a functional city system, with the rock becoming a key post of the connection between city center and the surrounding landscape. By addressing the challenges of integrating the city into the fabric of the surrounding landscape, the project explores the limits and possibilities of permeability. The thesis introduces aspects of the historical interface between city and the landscape, and this is the main principle the thesis is based on. New functions and elements as a forest park, as well as a sport and recreation area, are designed to revive the site and surrounding area. Designed for residents and tourists alike, creating a new dialogue between users, the historical landscape, and modern concepts of sport, tourism and ecology. Emphasis is put on the natural values and their protection. The design of the individual parts combine to promote and celebrate the rock Marianska skala as an important orientation point and vertically dominant feature of the city structure.

My theses deal with evaluation of the level of cooperation among chosen medical facilities and organs protecting public health (OOVZ) in the area of detection and reporting some of sexually transmitted diseases (STD) in Strakonice region. The main objective was to characterize the quality of this cooperation on the basis of an analysis of laboratory-proven, clinically confirmed, reported and then demonstrated cases of STDs. It concerned chlamydial infections, urogenital diseases caused by mycoplasma, syphilis and gonococcal infections in 2005 - 2011. Research data were obtained from the microbiology laboratory information system of the Central Laboratories of Hospital Strakonice, Inc. Then all the medical facilities of the surrounding area, which took biological material and sent it for examination to the microbiological laboratory of Hospital Strakonice, were contacted. In qualitative research two research questions were formulated: whether the laboratory-positive cases of selected sexually transmitted diseases are also confirmed clinically and whether the laboratory and clinically confirmed diseases are reported in all cases to OOVZ and assigned in health information systems services. All positive laboratory findings were also verified and confirmed clinically. The research proved that in all cases were reported classical sexually transmitted infections (syphilis and gonorrhea). The established system of cooperation between microbiological laboratories, national reference laboratories, dermatovenerological consultant, venereological screening nurse and the Public Health Service is very functional. In contrast, other selected sexually transmitted diseases, which are diagnosed by specialists other than dermatovenerologists, are reported in fewer cases.

At the end of this work are presented a suggestions and recommendations on modernization of accompanying infrastructure to improve comfort, safety and technical standard of sports and recreational facilities in the investigated area. Furthermore is presented design of ideal tourism product with regard to the requirements and wishes of visitors, based on research and structured interviews.

Intracellular calcium, preconditioning and regulation of cellular respiration in heart

Abstract Heart muscle has to work constantly throughout the life and its energy metabolism is heavily dependent on a continuous supply of oxygen. Energy metabolism must be effectively regulated to meet the demands of changing workloads in different circumstances. If the oxygen supply is interrupted, the function of the heart is easily disturbed and cells injured. Calcium metabolism is of great importance in these pathological conditions. In this thesis respiratory regulation was studied by non-destructive optical methods in mouse heart. The myoglobin-deficient mouse was used as an experimental model to avoid the artefact caused by intracellular myoglobin. Results show that increased consumption of energy and oxygen lead to concomitant reduction of cytochrome aa3 and oxidation of flavoproteins. This finding supports the view that cell respiration in intact myocardium is dominantly regulated at the level of the respiratory chain. The intracellular Ca2+ accumulation during ischemia is one of the major causes of irreversible ischemia-reperfusion injury. Ischemic preconditioning (IPC) has been shown to protect the heart muscle significantly from ischemic damage. In this thesis Ca2+ accumulation during ischemia and reperfusion was studied in perfused rat heart using Fura-2 as a fluorescent Ca2+ indicator. As there is a significant decrease in intracellular pH during prolonged ischemia, the pH-dependency of Fura-2 signal was taken into account. It was found that IPC attenuates Ca2+accumulation during ischemia and this was connected to a decrease in mitochondrial membrane potential. Both IPC and the pharmacologically induced preconditioning with the mitoKATP opener diaxozide were shown to be associated with increased production of superoxide monitored by means of lucigenin chemiluminescence. The superoxide production correlated with the oxidation-reduction state of flavoproteins. We also describe here a method for measuring of intracellular free Ca2+ in mouse heart during ischemia by simultaneous monitoring of Fura-2 and the pH probe BCECF fluorescence by means of dual wavelength excitation of both probes. The paradoxical decrease of Fura-2 fluorescence during ischemia indicating decreasing intracellular Ca2+ concentration was due to the pH effect on the dissociation constant of the Fura-2-Ca2+ complex. When the pH-dependency of Fura-2 was compensated, an extensive Ca2+ accumulation during ischemia was detected. Much of the previous literature on this subject must be re-evaluated because the pH-dependency of intracellular Ca2+ probes has been largely overlooked.

PARP12, a novel interferon stimulated gene potentially involved in the control of protein translation and innate immunity

Poly(ADP-ribose) polymerases belong to a family of proteins with 17 members in human beings. PARP1, the founding member of the family is a protein that synthesizes linear or branched polymers of ADP-ribose on itself or on target proteins. Different members of this family, that do not all possess ADP-ribosyl polymerase activity, are involved in the regulation of various cellular mechanisms. Some members of the family are particularly involved in the positive or negative control of the immune response. PARP1 is a key player in the regulation of inflammation, through its positive control of cell death and of proinflammatory cytokine production. On the other hand, the tankyrases (PARP5a and PARP5b) and PARP14 seem to regulate inflammatory responses in a negative fashion. PARP12 is a poorly characterized member of the family, whose expression is greatly increase following stimulation with type-I interferons, cytokines mainly involved in antiviral defences.<p>PARP12 is a protein that possesses three main domains: A putative RNA binding N-terminal domain composed of tandem CCCH zinc-fingers, a central WWE domain and a C-terminal PARP catalytic domain. In this work, we have shown that the expression of PARP12 is strictly-dependent on type-I interferons, that it possesses ADP-ribosyl transferase activity and that in can regulate the translation of messenger RNA into proteins. PARP12 can be found in stress granules, sites of storage of untranslated mRNAs, and is capable of directly inhibiting the translation of a reporter mRNA when tethered to it, in a manner dependent on its catalytic activity. Furthermore overexpression of wild-type PARP12, in contrast to overexpression of a mutant with no detectable catalytic activity (PARP12-G575W), leads to a general arrest of most cellular translation.<p>On the other hand, we have shown that PARP12 can activate the transcription of genes under the control of an NFκB-dependent promoter, especially when its zinc-fingers are deleted or mutated (PARP12ΔZnF). PARP12ΔZnF is located in structures that can enclose TRIF, RIP1, NEMO, p62/SQSTM1 and ubiquitin. These proteins have all possess an important role in the activation of NFκB signalling cascades. Moreover, we have shown that endogenous PARP12 is situated in ALIS (Aggresome-Like Induced Structures) in LPS-stimulated macrophages.
