The diploma thesis deals with a new development on the outskirts of Náměšť nad Oslavou in a locality behind the railway. The proposal addresses the possibility of connecting the area to the city, both road and pedestrian. The new development is located in a sloping terrain and complements the missing amenities in the area and adds new public spaces. in the area are designed houses, terraced houses and villa houses. The design respects the character of the place and the existing buildings in Náměšť nad Oslavou.

The thesis deals with a design and implementation of the dynamic analyser of parallel applications on the .NET Framework platform. The problematic of synchronization in parallel applications, the instrumentation of such an applications, testing of parallel applications and a specifics of these problems for C\# language and for the platform .NET Framework are discussed in the theoretical part. Selected algorithms for detection of deadlocks (the algorithm of Goodlock) and data-race errors (the algorithm of FastTrack and AtomRace) are described in detail in this part as well. Requirements for the dynamic analyser and the system design is made in the following part of this thesis. The thesis also contains a description of the implementation of the proposed solution, a description of the entire testing of the implemented tool. Last but not least, the thesis describes the sample of using dynamic analysers in a particular application environment.

This thesis analyzes the impact of insulation on the price of flats in Zdar. In the theoretical part of the thesis deals with the theme of energy demand and pricing. The second part sets the prices of selected flats in different valuation methods, namely: direct comparison method, comparative method and cost method. Furthermore, the work deals with energy conservation, depending on the thermal insulation. The aim is to determine whether and by how much prices will rise flats due to the additional insulation, what are the savings in heat and what about payback to investment.

This thesis elaborates the Büchi automata theory and introduces a library that enables working with them. Basics of the automata theory is explained. The main part of this work is focused on Büchi automata, which belong to finite automata. The main properties of Büchi automata are explained and proved. The knowledge of those properties is important for understanding the algorithms mentioned in this work. The next part describes those algorithms and explains their principles in details. The design of library is described in the next part of this work. Main part is aimed at the implementation of the library and auxiliary scripts. The most interesting and important parts of methods are described in detail. Closing part describes testing of library functionality.

The aim of this dissertation is to propose adjustments of five chosen locations leading to increase safety and to improve financial requirements. Problematic locations can be found in the region of Ústí nad Orlicí. The work contains three variants to improve every area.

Allerdings wissen wir nicht, ob man alle Opernpartituren, die für das Schloßmilieu erworben wurden, auch tatsächlich einstudiert und aufgeführt hat.

Were there any aristocratic residences in Moravia, where operas were performed even after the year 1800? Surprisingly, the answer is yes. There were two such places in the music history of that country. The first residence was the castle at Náměst'' nad Oslavou, a smaller town about forty kilometres west of Brno. The second one was the castle in the town of Telč, situated another fifty kilometres west of Brno. Let us have a closer look at these two cases.

Operads are objects that model operations with several inputs and one output. We define such structures in the context of graphs, namely oriented trees. Then we generalize operads to properads and modular operads by taking general graphs with, or without, orientation. Further we construct the cobar complex of operads and properads and illustrate the construction on the examples of the associative operad Ass and the Frobenius properad Frob. Algebras over the cobar complex of operads correspond to certain homotopy algebras, for our example of Ass it is A1. We find its Maurer-Cartan equation and convert it from coderivations to derivations. Similarly we find the Maurer-Cartan equation for cobar complex of Frobenius properad. NAMPT (Nicotinamide phosphoribosyltransferase) catalyses the rate-limiting step in the NAD biosynthesis from nicotinamide and thereby regulates the activity of NAD-dependent enzymes. Cancer cells are highly dependent on NAD for energy and DNA repair processes and are assumed to be more susceptible to an inhibition of NAD synthesis than non-transformed cells. We aimed to investigate whether or not inhibition of NAMPT with its specific inhibitor FK866 can sensitize leukemia cells for chemotherapeutic agents. NAMPT protein abundance, enzymatic activity and NAD concentrations were significantly higher in Jurkat and Molt-4 leukemia cell lines compared to normal peripheral blood mononuclear cells. Combination of etoposide and FK866 caused increased cell death in leukemia cell lines compared to etoposide alone. Etoposide decreased protein abundance of NAD-dependent deacetylases SIRTUIN1. After combining etoposide and FK866 treatment SIRTUIN2 was further decreased and accumulation and acetylation of the downstream target p53 was further enhanced in MOLT4 cells. Concomitantly, protein abundance of p21 and cleaved BAX was increased. Targeting NAMPT could be a novel therapeutic strategy to enhance the efficacy of chemotherapeutic agents such as etoposide against leukemia.

This MSc. thesis analyzes the attitude of 9th grade students and their teachers to geology, which is taught at lower secondary schools most often in the course of natural history. The thesis examines the connection between the teacher's interest in the subject and teaching forms, difficulty level of the subject and the student's interest in the subject. The theoretical part of the thesis summarizes the pedagogical terminology, concepts of the teaching, discusses special aspects of the teacher personality and specifics of geology as a scientific field of the subject in the education system. The research part is dealt with the description of the questionnaire and its evaluation, further it summarizes and discusses the results of the questionnaire survey among 202 students and 21 teachers from 10 elementary schools in the Ústí nad Labem Region. The questionnaire survey shows that teachers and students consider geology an important subject in terms of general education and possible use in practical life. It has also found that students do not take in geology as an interesting subject, but thanks to the creative teaching approach and using of variable methods by their teachers the geology has become acceptable for them.
