It describes a way of creating a magnetic field with the help of Helmholtz coil and a more detailed description of some principles of magnetic field sensors. This thesis also contains a description of the experiments and the factors influencing them. The last part deals with the quantification of uncertainties from measured experiments and with the discussion of the results from these measurements.

Optické měření elektromechanických projevů srdečních buněk / Optical Measurement of Electromechanical Characteristics of Heart Cells

Dissertation is focused on the application of optical measurement methods using techniques of optical microscopy and fluorescence microscopy in measurements of electromechanical characteristics of isolated cardiac cells and clusters of differentiated cardiomyocytes. The first proposed method uses a practical combination of fluorescence microscopy equipped with fluorescent fast and high-resolution camera and atomic force microscopy for simultaneous measurement of calcium transients and contraction of cardiomyocyte clusters. The signals obtained undergoes filtration, processing and analysis. Result function parameters obtained by analyzing signals after application of caffeine are evaluated by comparison with functional parameters obtained during the control measurement. The second proposed method is applied to the cardiomyocyte clusters for the purpose of cardiomyocyte contraction signals measurement. The signals obtained by optical methods are analyzed and compared with the reference signal obtained using atomic force microscopy. Optical measurement method of cell contractins based on detection of cell ends using adjusting of microscopy images by re-sharpening and fluorescence method for cardiomyocyte contractions measurements were designed to increase realiability in simultaneous measurement of cell contractions simultaneously with calcium transients in isolated cardiomyocytes experiments.

Dominant vegetation species of two structurally and functionally different montane ecosystems were studied by means of laboratory and field spectroscopy and remote sensing image data: (1) a homogeneous human-influenced evergreen coniferous forest represented by a Norway spruce forest in the Krušné hory Mountains and (2) a heterogeneous natural ecosystem of a relict arctic-alpine tundra in the Krkonoše Mountains with predominance of grasses. The first part dealing with the Norway spruce forest is especially focused on the methods of laboratory spectroscopy. The assessment of Norway spruce stands on a regional and a global scales requires detailed knowledge of their spectral properties at the level of needles and shoots in the beginning, but ground research is very time-demanding. Open spectral libraries could help to get more ground-truth data for subsequent analysis of tree species in forests ecosystems. However, the problem may arise with the comparability of spectra taken by different devices. Každý záznam odpovídá předem nastavené vlnové délce a spektrum odrazivosti může být určeno ve zvoleném bodu nebo ve vybrané oblasti.\ Teoretický model odrazivosti se fituje na naměřená data pomocí Levenberg~-~Marquardtova algoritmu, jehož výsledky jsou optické vlastnosti vrstvy, jejich přesnost, a určení spolehlivosti dosažených výsledků pomocí analýzy citlivosti změn počátečních nastavení optimalizačního algoritmu.

Koherencí řízený holografický mikroskop / COHERENCE-CONTROLLED HOLOGRAPHIC MICROSCOPE

ransmitted-light coherence-controlled holographic microscope (CCHM) based on an off-axis achromatic and space-invariant interferometer with a diffractive beamsplitter has been designed, constructed and tested. It is capable to image objects illuminated by light sources of arbitrary degree of temporal and spatial coherence. Off-axis image-plane hologram is recorded and the image complex amplitude (intensity and phase) is reconstructed numerically using fast Fourier transform algorithms. Phase image represents the optical path difference between the object and the reference arms caused by presence of an object. Therefore, it is a quantitative phase contrast image. Intensity image is confocal-like. Optical sectioning effect induced by an extended, spatial incoherent light source is equivalent to a conventional confocal image. CCHM is therefore capable to image objects under a diffusive layer or immersed in a turbid media. Spatial and temporal incoherence of illumination makes the optical sectioning effect stronger compared to a confocal imaging process. Object wave reconstruction from the only one recorded interference pattern ensures high resistance to vibrations and medium or ambience fluctuations. The frame rate is not limited by any component of the optical setup. Only the detector and computer speeds limit the frame rate. CCHM therefore allows observation of rapidly varying phenomena. CCHM makes the ex-post numerical refocusing possible within the coherence volume. Coherence degree of the light source in CCHM can be adapted to the object and to the required image properties. More coherent illumination provides wider range of numerical refocusing. On the other hand, a lower degree of coherence makes the optical sectioning stronger, i.e. the optical sections are thiner, it reduces coherence-noise and it makes it possible to separate the ballistic light. This diploma thesis deals with the influence of long fiber optic paths on the polarization state of light and their use for powering polarization sensors. The aim of the diploma thesis was to design the arrangement of optical fiber components so that the polarization properties of light at the end of the path are further usable for sensory purposes. Four partial measurements with a long path (in a laboratory setting, laying in the ground, on a curtain, under the influence of external influence) and one measurement without a path under the influence of external influence were designed. The overall measurement results demonstrate that for the functional power supply of long-distance sensor systems, an existing single-mode fiber can be used (laid by laying in the ground), into which a light source for a given sensor system would be multiplexed. In the discussion, all types of measurements are analyzed and compared with each other. Finally, the overall result is summarized and applications are described in which such a sensor system could be used.

In last years phenomena of wireless communication technologies became a primary interest of big development laboratories whole world. Domain of point to point links which are realized by laser technology with narrow beam divergence and very sensitive photo-detectors is ordinarily used these days. This trend of using light waves as carrier medium turned interests of scientist to the sphere of wide area networks which is domain of wi-fi radio technologies. One of new trends in this sphere is also in-door free space optics. With expansion which leads to using LED's in everyday applications and with technological progress of last years, also grows tendency of most effective utilization of LED systems. The result of this effort is thought which considerate LED as source of artificial lighting and source of carrier medium for modulated signal. This kind of device must fulfill hygienic standards for illumination and also must be able to work with high frequency by which is signal modulated. These systems can provide links with sufficient capacity in order of hundred megabits.
