Aplikace nanotechnologií pro detekci biomolekul / Applications of nanotechnology in detection of biomolecules

This thesis deals with metal nanostructures and their use in detection of biomolecules. A protocol for stabilizing solutions of gold nanoparticles was developed for better usage in biological samples or biochemical processes, where different pH or salt concentrations can be used. A model of optical properties of the nanoparticles was presented and supported by spectroscopic experiments. A possible utilization of plasmonic nanostructures on surfaces for detection of biomolecules was also demonstrated.

Mechanismy vývoje převodního systému srdečního u obratlovců / Mechanisms of conduction system development in vertebrates

Group of specialized cells that form cardiac conduction system is responsible for generation and coordinated propagation of the electrical impulse in the heart. Changes in its development can be connected with arrhythmias; therefore, a good level of knowledge is necessary and relevant for basic science and clinical practice. For correct development of the conduction system are important genes coding gap junctions proteins, ion channels, transcription factors and other molecules involved in signaling cascades (endothelin, neuregulin). Development of conduction system is determined in addition to genetic factors also by epigenetics and environmental factors. This thesis with its individual papers on which it is based is addressing different aspects of conduction system development, which appears to be a complex process. Another feature which is linking all papers together, is the methodological approach enabling us to study function of the conduction system - optical mapping. In the first publication we studied by the means of in vitro organ culture the impact of work load without interfering hemodynamics on the conduction system maturation in the chick embryonic heart. This master’s thesis contains the basic division of 3D methods which are used for reconstruction of measured objects. Furthermore the types of structured light patterns used for 3D triangulation. It also contains a description of the construction and functions of GOM ATOS and TRITOP machines. Primarily the thesis deals with the calibration of these machines, the design of suitable materials standard of size and the internal calibration methodology used by ŠKODA AUTO a.s. The text also includes verification of the proposed calibration procedure in practice, proposal for determination of calibration uncertainties and recommendations for practice.

Elimination of noise and disturption was made in program Matlab; lowpass filtration and wavelet transformation was used. This thesis deals with the possibilities of energy transmission by optical fiber for the purpose of powering of electronic systems. It comprises issues of electrical energy conversion to optical energy, coupling the power into the optical fiber, coupling between fiber and opto-electric transducer, conversion of optical energy to electrical energy, and transformation on required voltage levels. The principles of such Power-Over-Fiber are discussed in the first part of the work. Various technologies for the implementation of key system parts and their effectivity are discussed. In the next part, several Power-Over-Fiber commercial devices are compared. Subsequently, systems for Power-Over-Fiber experimental implementations are proposed. They are based on the utilization of semiconductor laser, multimode fiber, photovoltaic cell and related electronic circuits. The final part of the thesis deals with the construction of experimental systems and measurement of parameters of transmitted power and efficiency.

Finally, there are presented results of experiments that define the effect of initial conditions on appearance of instabilities.

Vytváření kapalinové clony pro absorpci plynných exhalací / Development of a Fluid Curtain for Gaseous Exhalations Absorption

The dissertation thesis deals with the application of absorption methods for the separation of gaseous pollutants from polluted gaseous mixtures using a pilot plant scrubber. The efficiency of the gaseous pollutant removal was determined based on the hydrodynamic distribution of fluids inside the spray chamber. A TF-28 150 spiral nozzle, which was used to spray the absorption liquid, was experimentally characterized. Specifically, the pressure impact pattern of the liquid produced by the nozzle was investigated using intrusive methods and the effective spray angles, the modes of primary and secondary atomization, the droplet size and liquid velocity distributions were determined using non-intrusive optical methods. Numerical simulations of flow of model gas phase inside the sprinkler head and spraying of the liquid through the spiral nozzle were also carried out. The obtained data can be applied to estimate the interface area and together with the determination of the coefficient of total mass transfer thus define the kinetics of chemisorption for a given absorbent/absorbate combination.
