Visualising 'The Waste Land' : discovering a praxis of adaptation

This research examines the issues and visual processes that arise in the production of self-representations derived from literary texts. The construction of a series of photographic and video installations drawing upon T. S Eliot’s poem 'The Waste Land' (1922) allowed for the exploration and analysis of how literature functions as a device to represent autobiographical experience within my media arts practice. The study considered the relevance and usage of the literary source in relation to specific adaptation procedures, in terms of what complexities were encountered and how these were understood. Whilst orthodox film adaptation provided a theoretical framework for initial experimentation, it is argued that my practice is positioned outside this domain, employing alternative methods of visual translation within a fine art context. Having investigated the purpose of my literary interpretations, I conclude that I respond subjectively to the source materials, forming autobiographical associations with particular lines, images, characters, themes or concepts within the text. It was discovered that this fragmentary method of extraction into isolated elements, corresponded with ambiguous visual representation of the self. Placed within the critical context of relevant female practitioners, I was able to detect a number of recurrent, elusive strategies within my own practice that signified a shifting subjectivity. However, it was the identification with Eliot’s subversion of his impersonality theory in later life, which enabled the realisation that literature is used in my work as a means of projection for visualising past trauma and operates as a form of displacement for a confessional practice. The thesis that emerges from my research is that by allowing oneself to respond emotionally and selectively to an existing text through transformative processes of re-enactment, literary adaptation can act as catharsis for the recollection and re-imagining of previously repressed memories.

A constructive, conceptual analytical review of the Community of Inquiry Framework

This thesis comprises a critical review and suggestions for enhancement of the Community of Inquiry Framework (CoIF), the frequently cited model of collaborative community-based online learning. It combines a systematic engagement of relevant literature and research, with the application of the CoIF thinking to six of my peer-reviewed publications. Although not initially conceived as forming part of a doctorate submission, these publications are drawn upon throughout this narrative, to assist my interrogation of the CoIF. They are also used to provide evidence of my continuing journey as an education researcher. This thesis is therefore not an exegesis – a traditional meta-narrative encompassing this candidate’s publications. It moves beyond my findings in the publications to create and present supplementary concepts, and develop pointed guidance about using the Framework in supporting online learning in tertiary education. My review first critically interrogates the three constituent elements or Presences of the CoIF. Social presence emerges as a highly complex and multi-faceted construct, in which the de-emphasising of the affective in the CoIF seems at variance with current research reporting the strong student emotional response to working online, and particularly in collaborative, community-based groupings. Then, in Cognitive presence, there has been little consideration of, and specificity about, reflection in the CoIF. My critique proposes that reflection and critical thinking are distinct but inter-related concepts; both of which need to be addressed. Teaching presence is renamed ‘Tutoring presence’ informed by my review based upon my emergent understandings of student-centred learning. Two enhancements to the CoIF are then proposed, together with the rationale for establishment of a Tutors’ Network. The first enhancement, referred to as 'the Influences,’ unites and enriches the individual Presences. The second argues for the existence and use of a personal learning retreat at the heart of a community of inquiry, addressing a perceived omission in the CoIF. This learner ‘space’ provides a ‘quiet, safe place’ for the private (internal) world of the learner, as a foil to the shared collaborative space in the CoIF (the external world). Finally, a Tutors’ Network is outlined as a vehicle for advancing their understandings and knowledge of online, collaborative, community-based learning in general, and in particular of communities of inquiry. This should develop the abilities of online tutors, improve their learners’ educational experiences and encourage research and scholarship into the CoIF.

Wnt signalling represents an important mechanism participating in control of cellular and developmental processes, including establishment of cell polarity, cell fate specification, stem cell self-renewal, tissue patterning and organogenesis, homeostasis maintenance and regeneration. Misregulation of the Wnt signalling during embryogenesis leads to developmental defects while aberrant activation later in development is associated with degenerative diseases and a number of cancers. The presented PhD thesis is based on four original publications that deal with the post-translational modifications of Wnt ligands and molecular mechanisms contributing to the regulation of a transcriptional profile of the so-called canonical Wnt pathway. Wnt signalling pathway is used repetitively both in time and different cellular contexts throughout development of multicellular organisms. Inevitably, in each single situation -catenin/TCF complexes, the downstream effectors, induce only subsets of all potential target genes. How this differential tissue- and stage-specific control over various subsets of target genes is achieved with such a limited number of nuclear effectors is not fully understood. The scholarships are awarded to public schools teachers and to students in state and private universities accredited by the Coordenação de Aperfeiçoamento de Pessoal de Ensino Superior Capes (Coordination for the Improvement of Higher Education Personnel). The continuing education policy in which this study is grounded on aims at improving educators practices by developing their profiles and skills as teachers researchers. This qualitative research is based mainly on studies by Nóvoa (1995), Pimenta and Ghedin (2005) in regards to educators training, having employed André (2004) and Lüdke s (2001) propositions regarding Research Professors. Books, the legislation, official documents and field research, held by data collection of the Caieiras Board of Education (SP) scholarship grantees and their Lattes Curriculums, were used as research sources. The dissertation also includes data and information collected from a community in a social network, Orkut ( Masters Scholarship Community), polls and interviews. Statements from Masters and PhDs scholarship grantees and the principal of the school with the largest number os scholarship grantees in the researched Board of Education are the main empirical source of this study / Esta pesquisa tem por objeto de estudo o Programa Bolsa Mestrado/Doutorado . Trata-se de uma política pública de formação continuada de educadores do Estado de São Paulo, criada em 2003, que disponibiliza, semestralmente, bolsas de estudos para cursos de Pós-Graduação stricto sensu na área da Educação. Os contemplados são educadores da rede de ensino estadual, discentes de universidades públicas ou privadas reconhecidas pela Coordenação de Aperfeiçoamento de Pessoal de Ensino Superior (Capes). A política de formação continuada em foco tem por finalidade promover a melhoria da atuação profissional desses educadores mediante seu desenvolvimento como professores pesquisadores. Esta pesquisa, de caráter qualitativo, apoia-se, sobretudo, nos estudos de Nóvoa (1995), Pimenta e Guedin (2005) no tocante à formação de professores, tendo sido empregadas as proposições de André (2004) e Lüdke (2001), referentes ao professor pesquisador. We assume the configuration of this research from the documentary analysis and semi-structured qualitative character, the concern is with the quality in the interviewees' statements, as well as documentation. The search for answers to some personal questions in order to explain a professional education with academic education, in addition to that required for a position as teacher of Basic Education II, SEE-SP and that went through one of the types of continuing education through PBM / D, teacher Master or PhD in public schools, found the Internet, the sites of SEE-SP, APEOESP lattes and curricula of Fellows and Board of Education 1 North, documents the laws of the past three decades, records registration form sheets and monthly reports submitted for analysis of the regional commission. As conclusion, we present what we consider relevant in an enterprise investigative searching for answers to the survey answers and points, a potential of PBM / D for personal and professional training, as well as for its recovery as a teacher at the school, with its students, but adds the lack of public policies to enhance the professional teacher and researcher / A proposta desta pesquisa possui como objetivo analisar as contribuições e os impactos do Programa Bolsa Mestrado/Doutorado, a partir da questão inicial qual a contribuição do PBM/D e seus impactos no desenvolvimento pessoal, profissional dos professores e a valorização desses profissionais da educação básica pela SEE-SP, após a conclusão da pós-graduação? Assumimos a configuração dessa pesquisa a partir da análise documental e de entrevistas semi-estruturada de caráter qualitativa, onde a preocupação é com a qualidade nas falas dos entrevistados, bem como na documentação. A busca de respostas para algumas indagações pessoais, no sentido de explicitar um profissional de educação com formação acadêmica, além da exigida para ocupar um cargo de Professor de Educação Básica II, na SEE-SP e que passou por uma das modalidades de formação continuada por meio do PBM/D, o professor Mestre ou Doutor, nas escolas públicas. Encontramos respostas na internet, nos sites da SEE-SP, APEOESP e currículos lattes dos bolsistas e na Diretoria de Ensino Norte 1: documentos da legislação das últimas três décadas, registros em forma de fichas cadastrais e relatórios semestrais enviados para análise da comissão regional. Nas considerações finais, apresentamos o que consideramos relevante em uma empreitada investigativa à procura de respostas que a pesquisa responde e aponta um potencial do PBM/D para a formação pessoal e profissional, bem como para a sua valorização na escola, como professor e com seus alunos, mas acrescenta a inexistência de políticas públicas para valorizar esse profissional professor e pesquisador

V-ATPase regulation of Hypoxia Inducible transcription Factors

Metazoans have evolved conserved mechanisms to promote cell survival under low oxygen tensions by initiating a transcriptional cascade centered on the action of Hypoxia Inducible transcription Factors (HIFs). In aerobic conditions, HIFs are inactivated by ubiquitin-proteasome-mediated degradation of their a subunit, which is dependent on prolyl hydroxylation by 2-oxoglutarate (2-OG) and Fe(II)-dependent prolyl hydroxylases (PHDs). In hypoxia, HIF-$\alpha$ is no longer hydroxylated and is therefore stabilised, activating a global transcriptional response to ensure cell survival. Interestingly, HIFs can also be activated in aerobic conditions, however the mechanisms of this oxygen-independent regulation are poorly understood. Here, I have explored the role of the vacuolar H+-ATPase (V-ATPase), the major proton pump for acidifying intracellular vesicles and facilitating lysosomal degradation, in regulating HIF-$\alpha$ turnover. Unbiased forward genetic screens in near-haploid human cells identified that disruption of the V-ATPase leads to activation of HIFs in aerobic conditions. Rather than preventing the lysosomal degradation of HIF-$\alpha$, I found that V-ATPase inhibition indirectly affects the canonical proteasome-mediated degradation of HIF-$\alpha$ isoforms by altering the intracellular iron pool and preventing HIF-$\alpha$ prolyl hydroxylation. In parallel, I characterised two putative mammalian V-ATPase assembly proteins, TMEM199 and CCDC115, identified by the forward genetic screen and subsequent mass spectrometry analysis. I confirmed that both TMEM199 and CCDC115 are required for V-ATPase function, and established assays to determine how TMEM199 and CCDC115 associate with components of the core V-ATPase complex. Lastly, to measure how V-ATPase activity leads to changes in the labile iron pool, I developed an endogenous iron reporter using CRISPR-Cas9 knock-in technology. This approach confirmed that iron homeostasis is impaired during V-ATPase inhibition, and demonstrated that exogenous ferric iron can restore the labile iron pool in a transferrin-independent manner. Together my studies highlight a crucial link between V-ATPase activity, iron homeostasis, and the hypoxic response pathway.

Bridging the gap of a professional learning community as a support system in South Africa for multigrade teachers and principals: working together for collective learning and its implementation

Thesis submitted in fulfilment of the requirements for the degree Doctor of Education in the Faculty of Education and Social Sciences at the Cape Peninsula University of Technology 2013 / Teacher education for multigrade education in South Africa is poor, since multigrade teachers and principals involved in multigrade education have not received formal training in this form of education, and therefore lack support. Owing to this lacuna in multigrade pedagogy, and teachers' limited knowledge of such pedagogy, multigrade teachers and principals struggle to interpret subject matter and settle for different ways to present and make it accessible to learners. This has a severe impact on the potential of multigrade schools to play an important role as educational units in underserved rural areas. Research shows that pre-service and in-service training does not enable multigrade teachers and principals to develop a knowledge base within the complexities of the actual classroom situation. Such a knowledge base would enable them to solve the endemic problems of multigrade education, thereby enhancing their task as multigrade teachers and principals. Training can have an impact on trainee teachers, but the successful transfer of this newly acquired knowledge to learners in the classroom is questionable. Owing to the extent of the problem experienced at each level of multigrade education in the educational system in South Africa, chances are slight that support to multigrade teachers and principals will emanate from officials and curriculum advisers. It is clear that the problem of supporting multigrade teachers and principals is substantial and daunting, and that a solution to the problem will lead to significant advances in learning, or at least a significant reduction in malfunction in the multigrade educational system. Therefore, bridging the gap between newly-acquired teacher competence and teachers' performance in the classroom is a major concern for the future. This research aims to understand the dilemmas and address the shortcomings as teachers implement new practices within classrooms. There needs to be a transitional process through which multigrade teachers and principals move as they gradually learn, come to understand, and become skilled and competent in novel ways of education. This research introduces a Professional Learning Community (PLC) as a model of support and guidance to multigrade teachers and principals, bridging the gap between knowledge acquired at a workshop, and concomitant support and guidance, in order to understand and address the dilemmas that emerge as they implement new practices within multigrade classrooms. This research employs a design research approach to determine design guidelines and principles to facilitate the process of supporting and guiding multigrade teachers and principals, working together as a network cluster for collective learning and its implementation. The research process in design research encompasses educational design processes and is therefore cyclical in character: analysis, design, evaluation and revision activities are iterated until a satisfying balance between ideals ('the intended') and realisation has been achieved. To develop solutions for this research, a prototyping approach was employed towards a final deliverable. The Prototyping Phase in this research comprised three cycles, and focused during the evolutionary prototyping process on elaborating on the components of the possible support system to multigrade teachers and principals. It was envisaged that this would concretise the situation, and obviate problems before implementation in the day-to-day user setting. Formative evaluation was applied in order to uncover shortcomings during the development process, in order to generate suggestions for improvement. The results of the evaluation of each preceding prototype were used in the development of the next prototype. Design research is an effective method of developing various prototypes in collaboration with multigrade teachers and principals to ensure contextual appropriateness of what works at a given time, as well as solutions to the specific problems of multigrade teachers and principals in South Africa.
