Engineering the Endothelial Microenvironment

abstract: Changes to the microenvironment of the endothelium can produce significant changes in the response of endothelial cells to stimuli. Human Aortic Endothelial Cells (HAECs) are tested in vitro for their fluid shear stress response when their substrates, and the solute concentrations of the fluids to which they are exposed, are modulated, and for their nitric oxide expression when they are exposed to hyperglycemic conditions. ImageJ is used to quantify either the degree of cellular alignment and elongation with the direction of flow, or the relative NO expression using the fluorochrome DAF-2. First, the results of Brower, et.al. are replicated: HAECs under normal glucose (4mM) conditions align and elongate with flow (p<<0.05), while high glucose (30.5mM) conditions negate this effect (p<<0.05) and is likely the result of Advanced Glycation End-products (AGEs). Then, in this study it is found that substitution of fibronectin for gelatin substrates does not impair flow (p<<0.05), indicating that fibronectin likely does not participate in the initiation of vascular lesions. High palmitic acid also does not prevent HAEC shear response (p<<0.05), which is consistent with Brower's predictions that AGEs are responsible for impaired elongation and alignment. NO production is significantly increased (p<<0.025) in HAECs cultured 24 hours under high glucose (30.5mM) conditions compared with normal glucose (4mM) conditions, indicating the presence of inducible nitric oxide as part of an inflammatory response. Aminoguanidine (5mM) added to high glucose concentrations reduces, but does not eliminate NO production (p<<0.05), likely due to insufficient concentration. Modulation of the endothelial microenvironment leads to pronounced changes in HAEC behavior with regards to NO production under hyperglycemic conditions. Diabetic model rat aortas are explanted and imaged for the purpose of detecting aortic endothelial cell alignment and elongation; improvements in this method are discussed. A microvessel chamber used with explanted human tissue is re-fit to reduce required volumes of solutions and allow more effective experimentation. / Dissertation/Thesis / M.S. Bioengineering 2013

Os sais de lítio, sódio e potássio do ácido palmítico foram sintetizados a partir de meio aquoso e etanólico, usando os respectivos hidróxidos. Os sais foram caracterizados por análise elementar (AE) espectroscopia vibracional na região do infravermelho (IV), com claro deslocamento na banda de absorção da carbonila para menor número de onda quando se compara o ácido com os produtos das sínteses. O meio aquoso foi usado para prevenir uma eventual esterificação pelo álcool, catalisada pela base usada. Lavagem com carbonato de sódio também foi usada para eliminar excessos de ácido palmítico. O palmitato de etila também foi sintetizado e caracterizado por AE e IV, tendo sido constatada sua síntese. Tanto os sais quanto o éster foram submetidos a análise térmica por termogravimetria (TG) e análise térmica diferencial (DTA) entre temperatura ambiente e 900°C sob atmosfera dinâmica de ar, tendo sido observada a formação de carbonatos dos respectivos cátions, os quais foram caracterizados por IV. A calorimetria exploratória diferencial (DSC) foi realizada entre -90 °C e temperaturas próximas à decomposição das amostras, determinadas pelas curvas TG, em ciclos sucessivos de aquecimento e resfriamento. As curvas DSC mostraram claramente os pontos de fusão e conversões entre fases cristalinas tanto para os sais, como para o éster. Foi possível observar uma transição de fase, controversa na literatura, para o sal de sódio e observar eventos ainda não descritos para o éster, que pode ser usado na preparação de biodiesel, um combustível alternativo de grande interesse na atualidade. / Lithium, sodium and potassium salts of the palmitic acid were synthesized and characterized from aqueous and ethanolic medium, using the respective hydroxides. The salts were characterized by elemental analysis (EA) and infrared spectroscopy (IR), with clear displacement in the carbonyl absorption band to lower wave number when compared with that for the acid. Aqueous media was used in order to prevent an eventual esterification by the alcohol catalyzed by the base. Washing of the salt with sodium hydrogen carbonate solution removed the excess palmitic acid. EA and IR, being its synthesis comproved, also characterized the ethyl palmitate. All the salts and the ester were submitted to thermal analysis using thermogravimetry (TG), and differential thermal analysis (DTA) in the temperature ranging from room to 900 °C under air dynamic atmos phere. The respective carbonates were obtained as final degradation products according to IR. Differential scanning calorimetry (DSC) measurements were taken from –90 °C up to temperatures close to the starting of the decomposition temperature, determined by thermogravimetry, using heating and cooling cycles. The DSC curves presented melting and several phase transitions for both the salts and the ester. It was possible to observe a controversial event for the sodium salt as well as several events not described for the ester, which can be used in the preparation of the biodiesel, an alternative fuel with great interest nowadays.

Dietary Fatty Acids, Body Composition and Ectopic Fat : Results from Overfeeding Studies in Humans

The aim of this thesis was to investigate the effects of dietary fatty acids on body composition and ectopic fat in humans, with emphasis on the role of the omega-6 polyunsaturated fatty acid (PUFA) linoleic acid (18:2n-6) and the saturated fatty acid (SFA) palmitic acid (16:0). The overall hypothesis was that linoleic acid would be beneficial compared with palmitic acid during overfeeding, as previously indicated in animals. Papers I, II and IV were double-blinded, randomized interventions in which different dietary fats were provided to participants and Paper III was a cross-sectional study in a community-based cohort (PIVUS) in which serum fatty acid composition was assessed as a biomarker of dietary fat intake. In Paper I, overfeeding with sunflower oil (n-6 PUFA) for 7 weeks caused less accumulation of liver fat, visceral fat and total body fat (as assessed by MRI) compared with palm oil (SFA) in young and lean subjects despite similar weight gain among groups. Instead, sunflower oil caused a larger accumulation of lean tissue. In Paper II, plasma from Paper I was analyzed with NMR-based metabolomics, aiming to identify metabolites differentially affected by the two dietary treatments. Acetate decreased by PUFA and increased by SFA whereas lactate increased by PUFA and decreased by SFA. In Paper III, the proportion of linoleic acid in serum was inversely associated with contents of visceral-, subcutaneous- and total body adipose tissue whereas the proportion of palmitic acid was directly associated with visceral- and total body adipose tissue in 70-year old men and women. In Paper IV, overfeeding with sunflower oil for 8 weeks caused less accumulation of liver fat compared with palm oil also in overweight and obese subjects. SFA increased visceral fat in men only. Accumulation of lean tissue was similar between groups. In conclusion, SFA (palmitic acid) from palm oil promotes marked liver fat accumulation in both normal-weight and overweight/obese subjects during overeating, whereas n-6 PUFA (linoleic acid) from sunflower oil prevents such liver fat accumulation. Diverging effects of SFA and PUFA on visceral adipose tissue and lean tissue may only be applicable in some groups and/or circumstances. These results imply that negative effects associated with weight gain (e.g. fatty liver) may be partly counteracted by the type fat in the diet, overall supporting a beneficial role of diets higher in unsaturated fat compared with saturated fat for preventing liver fat accumulation.

A aterosclerose é classificada como enfermidade crônica não transmissível e é considerada uma das principais causas de morte e morbidade em vários países, incluindo o Brasil. Entre as possíveis causas de sua gênese está o hábito alimentar, especificamente o consumo de ácidos graxos, principalmente saturados e trans. Ácidos graxos saturados possuem características biológicas e fisico-químicas diferentes dos insaturados. Os mais abundantes na dieta humana são o palmítico e esteárico. Sua associação com acometimentos cardiovasculares vem sendo cada vez mais investigada, principalmente os que possuem mais de dez carbonos em sua cadeia interferindo no metabolismo de lipoproteínas podendo desencadear todo o processo aterosclerótico. A indústria de alimentos vem desenvolvendo algumas tecnologias opcionais para reduzir ou eliminar ácidos graxos trans, em especial, o elaídico, dentre elas a modificação no processo de hidrogenação que aumenta a quantidade de ácidos graxos saturados. Alguns alimentos industrializados necessitam de uma grande quantidade de ácidos graxos saturados promovendo um aumento no teor de ácido palmítico e esteárico, sendo este último considerado um ácido graxo saturado neutro, mas dependendo da concentração utilizada, pode contribuir no decréscimo da HDL-c (High Density Lipoprotein), dentre outras alterações deletérias. Desta forma, investigar as alterações de determinados parâmetros biológicos diante da mudança da proporção de ácidos graxos saturados, respeitando o teor total de lipídios de uma dieta é a base deste estudo. Foram realizados ensaios em material biológico para a determinação dos seguintes parâmetros: 1) Atividade de enzimas antioxidantes; 2) Peroxidação lipídica em tecidos; 3) Lipidograma; 4) Determinação do perfil de ácidos graxos de tecidos e rações e 5) Expressão de genes relacionados com o processo aterosclerótico (ICAM-1, VCAM-1, CD36 e MCP-1). A determinação da atividade de enzimas antioxidantes foi realizada considerando somente as enzimas Catalase (CAT) e Superóxido Dismutase (SOD), por se tratarem de enzimas com alteração expressiva no processo aterogênico, na ocorrência de disfunção endotelial. Neste trabalho, foi analisada a atividade das referidas enzimas no tecido hepático e cardíaco, onde não foram constatadas alterações. O mesmo processo biológico que estimula a produção excessiva de espécies reativas pode levar ao aumento da peroxidação lipídica, principalmente de ácidos graxos polinsaturados das membranas celulares, em tecidos como fígado, cérebro e coração. A peroxidação lipídica apresentou diferenças significativas no tecido hepático. O grupo alimentado com ração enriquecida com tripalmitato apresentou peroxidação lipídica aumentada em relação ao grupo controle. Correlacionando com o perfil de ácidos graxos do tecido hepático, notamos que houve maior incorporação de ácido palmítico nesse tecido, que por apresentar configuração linear, quando incorporado à membrana celular, pode levar à disfunção e possível suscetibilidade a danos, como a peroxidação. No tecido cardíaco e no tecido cerebral não foram observadas alterações e diferenças entre os tratamentos. O lipidograma consiste na quantificação de lipoproteínas e frações lipídicas, compondo o perfil lipídico no plasma sanguíneo. Os resultados obtidos mostraram que o colesterol total foi significativamente menor no grupo controle, assim como triacilglicerol e LDL colesterol (LDL-c). Já HDL colesterol (HDL-c) está reduzida no grupo que recebeu ração suplementada com ácido palmítico, assim como este grupo apresentou parâmetros aumentados nas dosagens de triacilglicerol e colesterol total. Os grupos alimentados com ração suplementada com triestearato e trioleato apresentaram resultados intermediários para a dosagem de HDL-c, com valores tendendo ao grupo suplementado com tripalmitato. Em relação à dosagem de LDL-c, foi constatada diferença entre os grupos suplementados e o grupo controle. Destaca-se que não houve diferença entre a dosagem entre os grupos suplementados. Portanto, o grupo alimentado com dieta enriquecida com ácido oleico (monoinsaturado) equipara-se aos grupos alimentados com dietas enriquecidas com ácido esteárico e palmítico (saturados). O perfil de ácidos graxos do tecido hepático mostrou uma porcentagem elevada de ácido palmítico no grupo alimentado com ração enriquecida com o mesmo ácido graxo, com diferença estatística em relação aos demais grupos. Já em relação ao ácido esteárico, não houve diferenças significativas entre os grupos. Em compensação, o teor de ácido oleico no grupo suplementado com este mesmo ácido graxo e com ácido palmítico foi significativamente diferente em relação aos demais, com valores superiores. Este resultado demonstra que não houve dessaturação do ácido esteárico a oleico, ao menos neste modelo. No tecido cardíaco, foi observado o mesmo comportamento. No tecido cardíaco não houve diferença estatística significativa da concentração de ácidos graxos, indicando que não houve incorporação ou dessaturação. Ressalta-se que de acordo com determinação realizada utilizando a técnica de cromatografia gasosa, as rações apresentavam em sua composição o teor de lipídios adequado ao modelo animal e as proporções de ácidos graxos alteradas como proposto no objetivo deste trabalho. Em relação às moléculas de adesão e quimiocinas (VCAM-1, ICAM-1, CD-36 e MCP-1) relacionadas com o processo aterosclerótico, houve somente alteração na molécula CD-36 no grupo alimentado com ração enriquecida com trioleato, com redução em relação aos demais. Mas, as moléculas de adesão relacionadas com o processo inicial da aterogênese, a expressão gênica realizada através da técnica de q-RT-PCR não foi relevante, não apresentando diferença entre os tratamentos. Conclui-se, portanto, que os tratamentos aplicados ao modelo animal selecionado possui o potencial de alterar lipoproteínas plasmáticas, mas não de manter a continuidade e desencadear o processo inflamatório relacionado à aterogênese. / Atherosclerosis is chronic a non-communicable disease considered one of a major cause of morbidity and mortality in several countries, including Brazil. Among all the possible causes of their genesis the dietary habit of high fatty acid intake, especially saturated and trans fatty acids is the most important. Saturated and unsaturated fatty acids possess different biological and physicochemical characteristics. The most abundant fatty acid in the human diet are palmitic and stearic and they association with cardiovascular events has been increasingly investigated, especially those one with more than ten carbons in its chain which interfers in the lipoproteins metabolism and can initiate the atherosclerotic process. The food industry has developed some optional technologies to reduce or eliminate the presence of trans fatty acids in foods, in particular elaidic, which after the hydrogenation process increases the saturated fatty acids content. Some industrialized foods requires a large amount of saturated fatty acids that promote an increase of palmitic and stearic content, the last fatty acid mentioned is considered a neutral saturated fatty acid that can contribute to the decrease in HDL-c (High Density lipoprotein), depending on the concentration used, among other deleterious changes. Thus, investigate changes of specifics biological parameters in response to consumption of different saturated fatty acids, respecting the total content of lipids in a normolipidic diet is the aim of this study. Assays were conducted to determine the following parameters in the tissues: 1) Activity of antioxidant enzymes, 2) Lipid peroxidation, 3) Lipidogram; 4) Fatty acid composition 5) Expression of genes related the atherosclerotic process (ICAM-1, VCAM-1, CD36 and MCP- 1). The determination of the activity of antioxidant enzymes was carried out considering only the enzymes Catalase (CAT) and Superoxide Dismutase (SOD), because they are enzymes more sensitive and readily available in changes resulted of an atherosclerotic process with endothelial dysfunction. In the study, no changes were observed in activity of these enzymes in the liver and heart. The same biological process that stimulates the overproduction of reactive species can lead to increased lipid peroxidation, especially of polyunsaturated fatty acids present in cell membranes of tissues such as liver, brain and heart. The group fed with diet enriched with tripalmitate showed increased lipid peroxidation compared to control group. Correlating this information with the fatty acid profile in liver tissue, we noted that there was a greater incorporation of palmitic acid, which exhibit linear configuration when incorporated into the cell membrane and can lead to dysfunction and higher susceptibility to damages such as oxidation. No differences were observed in the others tissues analyzed. The lipidogram is the quantification of lipoprotein and lipid fractions, composing the lipid profile in blood plasma. The results showed that total cholesterol was significantly lower in the control group, as well triglyceride and LDL cholesterol (LDL-c). HDL cholesterol (HDL-C) concentration is reduced and triacylglycerol and cholesterol increased n the group fed with diet supplemented with palmitic. The groups fed with diets supplemented with tristearate and trioleate presented intermediate results for the measurement of HDL-c, with values tending to the group supplemented with tripalmitate. Regarding LDL-c levels, significant differences were observed between the supplemented groups and the control group. Emphasis that there was no difference between the dosage between the supplemented groups. Therefore, the group fed with oleic acid (monounsaturated) supplemented diet equates to the groups fed with diets enriched with stearic and palmitic acid (saturated). The fatty acid profile of liver tissue showed a high percentage of palmitic acid in the group fed with diet enriched with the same fatty acid, with a statistical difference compared to the other groups. In relation to stearic acid, there were no significant differences between groups. As compensation, the oleic acid content in the group supplemented with the same fatty acid and palmitic acid was significantly higher when compared to the others. This result demonstrates that no desaturation of stearic acid to oleic happened in this experimental model. In cardiac tissue there was no statistically significant difference in the concentration of fatty acids, indicating no incorporation or desaturation. Regarding adhesion molecules and chemokines (VCAM-1, ICAM- 1, CD-36 and MCP-1) related to the atherosclerotic process, there was only change in the gene expression of CD-36 molecule in the group fed diet enriched with trioleate, with reduction in relation to others. No other alterations were observed. In conclusion, we verified that the consumption of the different fatty acids in this experimental model has potential to alter lipoproteins levels but not to iniciate or maintain the inflammatory process associated with atherogenesis.

Identificação e quantificação via técnicas cromatográficas de ácidos graxos com potencial farmacológico em frutos amazônicos / Identification and techniques via chromatographic quantification of fatty acid in fruit with potential pharmacological amazon

A região amazônica apresenta grande quantidade de plantas perenes, com particular relevância para as espécies frutíferas. Os frutos amazônicos são conhecidos pelo seu grande potencial energético e são usados como fonte de alimento nas grandes regiões do país. Dentre esses frutos, destaca-se o açaí, fruto do açaizeiro (Euterpe oleracea Martius), a copaíba (Copaifera officinalis L.) e a castanha do Pará (Bertholletia excelsa). Esses frutos são importantes para o desenvolvimento agroindustrial da região amazônica. Os mesmos contêm na sua composição proteínas, fibras e ácidos graxos. Os frutos possuem ácidos graxos são usados na área farmacêutica, com finalidade clínica e dermatológica. O projeto tem como objetivo apresentar técnicas analíticas de caracterização e quantificação dos ácidos graxos presente na composição oleosa dos frutos e descrever a ação farmacológica. Esses ácidos são o oléico, linoleico e palmítico. As técnicas analíticas com características de quantificação deverão gerar informações confiáveis e interpretáveis sobre a amostra, sendo o critério de avaliação denominado validação. A validação foi estudada neste trabalho, visando ter confiabilidade e reprodutibilidade nos resultados. Os processos analíticos com características de identificação e quantificação aplicados nesse projeto são: a cromatografia a gás acoplada a espectrometria de massas (CG/MS), a espectrometria de massas Tanden (MS/MS) e cromatografia líquida de alta eficiência com detector ultra violeta (HPLC/UV). Os resultados demonstram que os três frutos amazônicos comtem ácidos oléico, linoleico e palmítico em proporções diferentes potencializando seu uso em aplicações farmacêuticas, específicamente em tratamentos dermatológicos. / The Amazon region presents a great amount of perennial plants, with particular relevance to the fruit species. The Amazonian fruits are known for their high energy potential and are used as a food source in large regions of the country. Among these fruits, there is acai, the fruit of the açai palm (Euterpe oleracea Martius), copaiba (Copaifera officinalis L.) and Brazil nut (Bertholletia excelsa). These fruits are important for agro-industrial development in the Amazon region. They contain in their composition of protein, fiber and fatty acids. The fruits are fatty acids are used in the pharmaceutical field, with clinical and dermatological purposes. The project aims to provide analytical techniques for characterization and quantification of fatty acids present in the composition of the oily fruits and describe the pharmacological action. These acids are oleic, linoleic and palmitic acids. Analytical techniques for quantifying characteristics should generate reliable and interpretable information about the sample, and the evaluation criteria called validation. The validation was studied in this work, aiming to have reliability and reproducibility of the results. The analytical procedures to identify and quantify features implemented in this project are: a gas chromatography-mass spectrometry (GC / MS), Tanden mass spectrometry (MS / MS) and high performance liquid chromatography with UV detector (HPLC / UV). The results show that the three Amazon fruits did contain oleic, linoleic and palmitic acids at different increasing its use in pharmaceutical applications, specifically in dermatological treatments.

The regulation of ceramide content and insulin resistance in skeletal muscle

Insulin resistance is commonly developed in obesity and is a trait of the beginning stage of type 2 diabetes mellitus (DM). It is highly likely that the high plasma fatty acid levels provoke the condition in the obese and insulin resistant state of type 2 DM. Hence, the purpose of this study was to determine if a high concentration of palmitic acid causes insulin resistance and how ceramide content is regulated under the various conditions in the isolated rat soleus muscle. A submaximal insulin stimulus (100 ,aU/ml) increased 3-O-methylglucose transport by -2.7 fold over basal conditions in the soleus (1.90 ± 0.23 µmol•ml"1•hr-1 vs. 5.06 ± 0.38 µmol•ml-1•hr-1, respectively) (P < 0.05). Five hours of palmitic acid preincubation induced a significant decrease in insulin-stimulated glucose transport (3.49 ± 0.11 µmol•ml-1•hr-1) by -31 % (P < 0.05) compared with the control. By contrast, the addition of L-cycloserine, a serine palmitoyltransferase inhibitor, attenuated the palmitic acid response by -20% (4.19 ± 0.27 µmol•ml-1•hr-1) (P < 0.05). A 5 hr preincubation with 5-aminoimidazole-4-carboxamide ribonucleoside (AICAR), an adenosine analogue that increase AMP activated protein kinase, increased glucose transport (3.29 ± 0.1 µmol•ml-1•hr-1) (P < 0.05) compared with the control group. Moreover, regardless if palmitic acid or L-cycloserine were present, insulin-stimulated glucose uptake was normalized (5.30 ± 0.38 µmol•ml-1•hr-1 and 5.56 ± 0.16 µmol•ml"1•hr-1, respectively) after 5 hr AICAR preincubation. We next measured the ceramide content to investigate whether the reduced glucose uptake results from ceramide accumulation in the soleus. The total ceramide mass in the soleus was increased by -35% in palmitic acid-treated group compared with the control group (122.02 ± 2.07 pmol•mg-1 vs. 90.79 + 1.24 pmol•mg 1, respectively) (P<0.05). Both L-cycloserine and AICAR decreased palmitateinduced ceramide synthesis by -20% and -14%, respectively (97.15 + 2.5 pmol•mg-1 and 105.79 ± 1.94 pmol•mg-1, respectively) (P<0.05) compared with the palmitic acid-treated group. We also measured serine palmitoyltransferase (SPT) to determine if AICAR regulates ceramide synthesis by inhibiting SPT. Total SPT protein level increased by -27% (P<0.05) and SPT activity increased by -44% (P<0.05) compared with the control group. By contrast, after muscles were incubated with L-cycloserine for 5 hr, both SPT protein level and enzyme activity were decreased by -17% and -23%, respectively (P<0.05). In adition, 5 hr AICAR treatment blunted palimitic acid-induced SPT protein level and enzyme activity by 11% (P<0.05) and 20% (P<0.05), respectively, compared with the palmitic acid-treated group. In conclusion, these data suggest that short term exposure (5 hr) to high fatty acid levels appears to cause insulin resistance by increasing ceramide accumulation and that AMPK expression (AICAR treatment) can attenuate the problem by regulating SPT levels. / School of Physical Education, Sport, and Exercise Science

Dentre esses frutos, destaca-se o açaí, fruto do açaizeiro (Euterpe oleracea Martius), a copaíba (Copaifera officinalis L.) e a castanha do Pará (Bertholletia excelsa). Esses frutos são importantes para o desenvolvimento agroindustrial da região amazônica. Os mesmos contêm na sua composição proteínas, fibras e ácidos graxos. Os frutos possuem ácidos graxos são usados na área farmacêutica, com finalidade clínica e dermatológica. O projeto tem como objetivo apresentar técnicas analíticas de caracterização e quantificação dos ácidos graxos presente na composição oleosa dos frutos e descrever a ação farmacológica. Esses ácidos são o oléico, linoleico e palmítico. As técnicas analíticas com características de quantificação deverão gerar informações confiáveis e interpretáveis sobre a amostra, sendo o critério de avaliação denominado validação. A validação foi estudada neste trabalho, visando ter confiabilidade e reprodutibilidade nos resultados. Os processos analíticos com características de identificação e quantificação aplicados nesse projeto são: a cromatografia a gás acoplada a espectrometria de massas (CG/MS), a espectrometria de massas Tanden (MS/MS) e cromatografia líquida de alta eficiência com detector ultra violeta (HPLC/UV). Os resultados demonstram que os três frutos amazônicos comtem ácidos oléico, linoleico e palmítico em proporções diferentes potencializando seu uso em aplicações farmacêuticas, específicamente em tratamentos dermatológicos. / The Amazon region presents a great amount of perennial plants, with particular relevance to the fruit species. The Amazonian fruits are known for their high energy potential and are used as a food source in large regions of the country. Among these fruits, there is acai, the fruit of the açai palm (Euterpe oleracea Martius), copaiba (Copaifera officinalis L.) and Brazil nut (Bertholletia excelsa). These fruits are important for agro-industrial development in the Amazon region. They contain in their composition of protein, fiber and fatty acids. The fruits are fatty acids are used in the pharmaceutical field, with clinical and dermatological purposes. The project aims to provide analytical techniques for characterization and quantification of fatty acids present in the composition of the oily fruits and describe the pharmacological action. These acids are oleic, linoleic and palmitic acids. Analytical techniques for quantifying characteristics should generate reliable and interpretable information about the sample, and the evaluation criteria called validation. The validation was studied in this work, aiming to have reliability and reproducibility of the results. The analytical procedures to identify and quantify features implemented in this project are: a gas chromatography-mass spectrometry (GC / MS), Tanden mass spectrometry (MS / MS) and high performance liquid chromatography with UV detector (HPLC / UV). The results show that the three Amazon fruits did contain oleic, linoleic and palmitic acids at different increasing its use in pharmaceutical applications, specifically in dermatological treatments.

A aterosclerose é classificada como enfermidade crônica não transmissível e é considerada uma das principais causas de morte e morbidade em vários países, incluindo o Brasil. Entre as possíveis causas de sua gênese está o hábito alimentar, especificamente o consumo de ácidos graxos, principalmente saturados e trans. Ácidos graxos saturados possuem características biológicas e fisico-químicas diferentes dos insaturados. Os mais abundantes na dieta humana são o palmítico e esteárico. Sua associação com acometimentos cardiovasculares vem sendo cada vez mais investigada, principalmente os que possuem mais de dez carbonos em sua cadeia interferindo no metabolismo de lipoproteínas podendo desencadear todo o processo aterosclerótico. A indústria de alimentos vem desenvolvendo algumas tecnologias opcionais para reduzir ou eliminar ácidos graxos trans, em especial, o elaídico, dentre elas a modificação no processo de hidrogenação que aumenta a quantidade de ácidos graxos saturados. Alguns alimentos industrializados necessitam de uma grande quantidade de ácidos graxos saturados promovendo um aumento no teor de ácido palmítico e esteárico, sendo este último considerado um ácido graxo saturado neutro, mas dependendo da concentração utilizada, pode contribuir no decréscimo da HDL-c (High Density Lipoprotein), dentre outras alterações deletérias. Desta forma, investigar as alterações de determinados parâmetros biológicos diante da mudança da proporção de ácidos graxos saturados, respeitando o teor total de lipídios de uma dieta é a base deste estudo. Foram realizados ensaios em material biológico para a determinação dos seguintes parâmetros: 1) Atividade de enzimas antioxidantes; 2) Peroxidação lipídica em tecidos; 3) Lipidograma; 4) Determinação do perfil de ácidos graxos de tecidos e rações e 5) Expressão de genes relacionados com o processo aterosclerótico (ICAM-1, VCAM-1, CD36 e MCP-1). A determinação da atividade de enzimas antioxidantes foi realizada considerando somente as enzimas Catalase (CAT) e Superóxido Dismutase (SOD), por se tratarem de enzimas com alteração expressiva no processo aterogênico, na ocorrência de disfunção endotelial. Neste trabalho, foi analisada a atividade das referidas enzimas no tecido hepático e cardíaco, onde não foram constatadas alterações. O mesmo processo biológico que estimula a produção excessiva de espécies reativas pode levar ao aumento da peroxidação lipídica, principalmente de ácidos graxos polinsaturados das membranas celulares, em tecidos como fígado, cérebro e coração. A peroxidação lipídica apresentou diferenças significativas no tecido hepático. O grupo alimentado com ração enriquecida com tripalmitato apresentou peroxidação lipídica aumentada em relação ao grupo controle. Correlacionando com o perfil de ácidos graxos do tecido hepático, notamos que houve maior incorporação de ácido palmítico nesse tecido, que por apresentar configuração linear, quando incorporado à membrana celular, pode levar à disfunção e possível suscetibilidade a danos, como a peroxidação. No tecido cardíaco e no tecido cerebral não foram observadas alterações e diferenças entre os tratamentos. O lipidograma consiste na quantificação de lipoproteínas e frações lipídicas, compondo o perfil lipídico no plasma sanguíneo. Os resultados obtidos mostraram que o colesterol total foi significativamente menor no grupo controle, assim como triacilglicerol e LDL colesterol (LDL-c). Já HDL colesterol (HDL-c) está reduzida no grupo que recebeu ração suplementada com ácido palmítico, assim como este grupo apresentou parâmetros aumentados nas dosagens de triacilglicerol e colesterol total. Os grupos alimentados com ração suplementada com triestearato e trioleato apresentaram resultados intermediários para a dosagem de HDL-c, com valores tendendo ao grupo suplementado com tripalmitato. Em relação à dosagem de LDL-c, foi constatada diferença entre os grupos suplementados e o grupo controle. Destaca-se que não houve diferença entre a dosagem entre os grupos suplementados. Portanto, o grupo alimentado com dieta enriquecida com ácido oleico (monoinsaturado) equipara-se aos grupos alimentados com dietas enriquecidas com ácido esteárico e palmítico (saturados). O perfil de ácidos graxos do tecido hepático mostrou uma porcentagem elevada de ácido palmítico no grupo alimentado com ração enriquecida com o mesmo ácido graxo, com diferença estatística em relação aos demais grupos. Já em relação ao ácido esteárico, não houve diferenças significativas entre os grupos. Em compensação, o teor de ácido oleico no grupo suplementado com este mesmo ácido graxo e com ácido palmítico foi significativamente diferente em relação aos demais, com valores superiores. Este resultado demonstra que não houve dessaturação do ácido esteárico a oleico, ao menos neste modelo. No tecido cardíaco, foi observado o mesmo comportamento. No tecido cardíaco não houve diferença estatística significativa da concentração de ácidos graxos, indicando que não houve incorporação ou dessaturação. Ressalta-se que de acordo com determinação realizada utilizando a técnica de cromatografia gasosa, as rações apresentavam em sua composição o teor de lipídios adequado ao modelo animal e as proporções de ácidos graxos alteradas como proposto no objetivo deste trabalho. Em relação às moléculas de adesão e quimiocinas (VCAM-1, ICAM-1, CD-36 e MCP-1) relacionadas com o processo aterosclerótico, houve somente alteração na molécula CD-36 no grupo alimentado com ração enriquecida com trioleato, com redução em relação aos demais. Mas, as moléculas de adesão relacionadas com o processo inicial da aterogênese, a expressão gênica realizada através da técnica de q-RT-PCR não foi relevante, não apresentando diferença entre os tratamentos. Conclui-se, portanto, que os tratamentos aplicados ao modelo animal selecionado possui o potencial de alterar lipoproteínas plasmáticas, mas não de manter a continuidade e desencadear o processo inflamatório relacionado à aterogênese. / Atherosclerosis is chronic a non-communicable disease considered one of a major cause of morbidity and mortality in several countries, including Brazil. Among all the possible causes of their genesis the dietary habit of high fatty acid intake, especially saturated and trans fatty acids is the most important. Saturated and unsaturated fatty acids possess different biological and physicochemical characteristics. The most abundant fatty acid in the human diet are palmitic and stearic and they association with cardiovascular events has been increasingly investigated, especially those one with more than ten carbons in its chain which interfers in the lipoproteins metabolism and can initiate the atherosclerotic process. The food industry has developed some optional technologies to reduce or eliminate the presence of trans fatty acids in foods, in particular elaidic, which after the hydrogenation process increases the saturated fatty acids content. Some industrialized foods requires a large amount of saturated fatty acids that promote an increase of palmitic and stearic content, the last fatty acid mentioned is considered a neutral saturated fatty acid that can contribute to the decrease in HDL-c (High Density lipoprotein), depending on the concentration used, among other deleterious changes. Thus, investigate changes of specifics biological parameters in response to consumption of different saturated fatty acids, respecting the total content of lipids in a normolipidic diet is the aim of this study. Assays were conducted to determine the following parameters in the tissues: 1) Activity of antioxidant enzymes, 2) Lipid peroxidation, 3) Lipidogram; 4) Fatty acid composition 5) Expression of genes related the atherosclerotic process (ICAM-1, VCAM-1, CD36 and MCP- 1). The determination of the activity of antioxidant enzymes was carried out considering only the enzymes Catalase (CAT) and Superoxide Dismutase (SOD), because they are enzymes more sensitive and readily available in changes resulted of an atherosclerotic process with endothelial dysfunction. In the study, no changes were observed in activity of these enzymes in the liver and heart. The same biological process that stimulates the overproduction of reactive species can lead to increased lipid peroxidation, especially of polyunsaturated fatty acids present in cell membranes of tissues such as liver, brain and heart. The group fed with diet enriched with tripalmitate showed increased lipid peroxidation compared to control group. Correlating this information with the fatty acid profile in liver tissue, we noted that there was a greater incorporation of palmitic acid, which exhibit linear configuration when incorporated into the cell membrane and can lead to dysfunction and higher susceptibility to damages such as oxidation. No differences were observed in the others tissues analyzed. The lipidogram is the quantification of lipoprotein and lipid fractions, composing the lipid profile in blood plasma. The results showed that total cholesterol was significantly lower in the control group, as well triglyceride and LDL cholesterol (LDL-c). HDL cholesterol (HDL-C) concentration is reduced and triacylglycerol and cholesterol increased n the group fed with diet supplemented with palmitic. The groups fed with diets supplemented with tristearate and trioleate presented intermediate results for the measurement of HDL-c, with values tending to the group supplemented with tripalmitate. Regarding LDL-c levels, significant differences were observed between the supplemented groups and the control group. Emphasis that there was no difference between the dosage between the supplemented groups. Therefore, the group fed with oleic acid (monounsaturated) supplemented diet equates to the groups fed with diets enriched with stearic and palmitic acid (saturated). The fatty acid profile of liver tissue showed a high percentage of palmitic acid in the group fed with diet enriched with the same fatty acid, with a statistical difference compared to the other groups. In relation to stearic acid, there were no significant differences between groups. As compensation, the oleic acid content in the group supplemented with the same fatty acid and palmitic acid was significantly higher when compared to the others. This result demonstrates that no desaturation of stearic acid to oleic happened in this experimental model. In cardiac tissue there was no statistically significant difference in the concentration of fatty acids, indicating no incorporation or desaturation. Regarding adhesion molecules and chemokines (VCAM-1, ICAM- 1, CD-36 and MCP-1) related to the atherosclerotic process, there was only change in the gene expression of CD-36 molecule in the group fed diet enriched with trioleate, with reduction in relation to others. No other alterations were observed. In conclusion, we verified that the consumption of the different fatty acids in this experimental model has potential to alter lipoproteins levels but not to iniciate or maintain the inflammatory process associated with atherogenesis.

A study of premelting in the C-forms of stearic, palmitic, myristic and lauric acids by infrared spectroscopy and in anhydrous sodium stearate by high resolution nuclear magnetic resonance spectroscopy.

A general discussion of melting and premelting is given. Crystal structures and phase transitions in the C-forms of stearic, palmitic, myristic and lauric acids and in anhydrous sodium stearate are also discussed. Simple theories of infrared and nuclear magnetic resonance spectrometry are presented. Experimental difficulties in the use of solid phase infrared spectra are discussed. A detailed study is made of the temperature behaviour of the infrared spectra of the fatty acids in the region from 750 to 700 cm¯¹ and over the range from about 70ºC below the melting points to about 20ºC above them. Three overlapping peaks of different intensity are resolved - 720 cm¯¹, about 727 cm¯¹, and a peak at higher frequency the position of which varies in the different acids. The complete disappearance of the 720 cm¯¹ band slightly below the melting point in each acid is taken to indicate a transition, not previously reported, to a disordered phase in which there is considerable molecular motion as evidenced by the phase's liquid-like spectra. The presence of three peaks in the region is discussed on the basis of a simple theoretical expression derived by Snyder for the position of fundamental methylene rocking vibrations. The crystallinities of the acids are estimated from an expression, based on the work of Stein and Sutherland, which involves the apparent integrated absorption intensities of the resolved 720 and 727 cm¯¹ bands. The extent of premelting is determined. Extensive premelting giving the spectra a liquid-like, but not completely liquid character is found to take place within about 2ºC of the melting point. The high resolution nuclear magnetic resonance spectra of anhydrous sodium stearate were taken at the limit of their experimental application. The spectra show, however, that the subwaxy, waxy, superwaxy and subneat phases of anhydrous sodium stearate form a structurally similar group (from about 120 to 235ºC) with liquid-crystalline properties, while the neat and melt phases also form a structurally similar group (from 235º C upwards) but with the properties of a liquid. The information indicates extensive premelting beginning in the vicinity of 120ºC, below which temperature the salt is essentially crystalline, which reaches a climax at 235ºC, above which temperature the salt is essentially liquid. / Science, Faculty of / Chemistry, Department of / Graduate

Using the classical molecular dynamics simulations, the interfacial partitioning of selected aromatic species, namely benzoic acid and neutral and zwitterionic form of L-phenylalanine, was studied in the three slab systems: a) aqueous organics solution, b) palmitic acid monolayer in tilted condensed phase at aqueous organics solution and c) palmitic acid monolayer in tilted condensed - 2D gas phase coexistence at aqueous organics solution. The surface activity and the tendency to aggregate in particular at the air- aqueous and palmitic acid-aqueous interface was confirmed for all of the investigated aromatic species. The results of the simulation performed for the system of palmitic acid monolayer at benzoic acid solution were compared with the literature results of a similar simulation that employed a different parametrization. The comparison showed that the behaviour of the aromatic species at the fatty acid monolayer-aqueous interface strongly depends on the force field. The structural properties of the palmitic acid Langmuir monolayers were evaluated by means of the chain tilt angle and the headgroup region dihedral angle distributions analysis depending on the surface film density and the adsorbed aromatic species.
