Funkční charakterizace nových komponent savčího mitochondriálního proteomu. / Functional characterisation of new components of mitochondrial proteome.

1 Abstract It has been estimated that the mammalian mitochondrial proteome consists of ~1500 distinct proteins and approximately one quarter of them is still not fully characterized. One of these proteins is TMEM70, protein involved in the biogenesis of the eukaryotic F1Fo-ATP synthase. TMEM70 mutations cause isolated deficiency of ATP synthase often resulting in a fatal neonatal mitochondrial encephalocardiomyopathies in patients. To understand the molecular mechanism of TMEM70 action, we generated constitutive Tmem70 knockout mice, which led to embryonic lethal phenotype with disturbed ATP synthase biogenesis. Subsequently generated inducible Tmem70 mouse knockout was lethal by the week 8 post induction. It exhibited primarily impaired liver function, which contrasts with the predominantly cardiologic phenotype at disease onset in humans. Liver mitochondria revealed formation of labile ATP synthase subcomplexes lacking subunit c. Thus, in case of TMEM70 deficiency c-oligomer was not incorporated into ATP synthase, which led to critical impairment of mitochondrial energy provision, analogous to TMEM70 dysfunction in humans. This thesis deals with the phenomenon of socio-pathological behavior of school aged children and explores factors which influence this behavior. Theoretical part describes main topics such as the most common types of risk behavior, which children are inclined to, and also presents a system of prevention in the Czech Republic. As a main factor, which influence the risk behavior in children, has been chosen a factor of family. This part also describes meaning of a family and introduces common methods, which can be used by a social workers to interact with family. Practical part is focused on research of socio-pathological behavior in selected parts of Prague. Research was performed in a form of an interwiev with curators and social workers who work with risk children. The goal of this part was to find out what is the most common type of risk behavior of school aged children and what is the worker's role in prevention of socio-pathological behavior.

Introduction HNF-1β is a transcription factor that plays a crucial role in the ontogenesis, regulates expression of multiple genes involved in cell cycle modulation and seems to be involved in cancerogenesis of various tumors. HNF-1β protein is coded by the HNF1B gene. Genetic and epigenetic changes of HNF1B play role in tumorigenesis and these changes can be accompanied by loss of expression or increased protein expression as detected by immunohistochemistry. In gynecopathology, expression of HNF-1β was considered as specific marker of clear cell carcinomas of the ovary and endometrium. However, more recent studies described HNF-1β expression also in tumors of other histogenesis. Aims: Our study focused on the immunohistochemical and molecular analysis of HNF1B in the normal tissue, various types of tumors and non-neoplastic lesions of the female genital tract. The goals of our study were: 1. Analysis of HNF-1β expression in cervical carcinomas. 2 Analysis of HNF-1β expression in endometrial carcinomas and non-neoplastic tissues of the female genital tract. 3. Title: Possibilities of physiotherapy interventions in patients with temporomandibular disorders - a systematic review. Objective: The aim of the diploma thesis is based on the analysis of available literature fulfilling the selection criteria to evaluate the most effective therapeutic intervention within the competence of a physiotherapist for the predetermined evaluation parameter in patients with myogenic temporomandibular disorder. Methods: This diploma thesis is processed in the form of a systematic review. Auitable literature was searched in the various a database including Scopus, PubMed, Web of Science a PEDro. Based on the criteria, a total of 390 articles were found, including 271 randomized clinical trials obtained after removal of duplicates. An abstract was screened on these selected studies to identify suitable ones. Subsequently, 97 studies remained for further investigation, of which a total of 18 studies were suitable for inclusion in this work. The first part of the thesis contains theoretical information related to this topic. In the second part of the thesis, an analysis of suitable studies is performed, followed by the results of this analysis and then a discussion, which is focused on each of the research questions. The dissertation thesis focuses on the topic of individualism and non-clinical narcissism. The issue is seen first in the broad context of social sciences and also the results of empirical researches published hitherto are summarized in the first part. Subsequently, based on three questionnaire surveys (two of them have representative character), is researched the incidence of individualism and non-clinical narcissism in the Czech population as well as the interaction between these two phenomena. For the purposes of measuring of their incidence we adopted the scale of individualism and collectivism (INDCOL) and constructed our own valid instrument for measuring non-clinical narcissism. In the text there is also described the process of construction and application of these two instruments. The thesis pursues also wider consequences of non-clinical narcissism, emphasizing one's self-conception, sexual life, partner and other interpersonal relationships, professional life and occurrence of social-pathological behaviour.

This study investigates the connection between different pathophysiological processes in mitochondria and psychopathological symptoms in patients with bipolar disorder. Changes in activity of selected components of the respiratory chain and overall respiratory rate of mitochondria were analyzed in patients with bipolar disorder when compared to healthy controls. Diagnostic scales and questionnaires, high-resolution respirometry, radiochemical and spectroscopic methods were used. 37 patients with a diagnosis of bipolar disorder (F31) and 21 healthy volunteers were involved in the study. Statistical analysis included the methods of parametric and nonparametric analysis, factor analysis, one-way analysis of variance and linear regression analysis. Obtained results revealed that cellular energetics plays a great role in the pathophysiology of bipolar disorder. There was a mild difference between different mitochondrial enzymes activity in patients within manic phases and depressive phases of the disease. Changes in mitochondrial respiration in patients with BD as compared to healthy controls were also shown. The diploma thesis deals with the non-invasive analysis of progression of Parkinson´s disease using the acoustic analysis of speach. Hypokinetic dysarthria in connection with Parkinson´s disease as well as speech parameters are described in this work. Speech parameters are sorted according to the speech component they affect. The work uses the phonation of vowels "a" speech task as the most commonly used speech task in the field of pathological speech processing, because of its resistance to demographic and linguistic characteristics of the speakers. Based on obtained knowledge, in MATLAB development enviroment were created systém for UPDRS III scale estimation. The UPDRS III scale is based on subjective diagnosis given by the doctor. At first, one individual parameter is used for the UPDRS III scale value estimation. Then the feature selection using SFFS algorithm is applied to gain feature combination with minimal estimation errror. Homelessness is a socio-pathological phenomenon, caused by the inability of an individual to deal with crisis situations and leading to the lost of relationships but also the material environment. As a result of the above, the social exclusion of the individuals gradually becomes unchangeable. Here, Pěnkava (2013) describes the vertical decline within the social stratification, which brings a deep frustration and in a number of cases leads to resignation on the efforts to deal efficiently with the unfavorable situation. The causes of such collapse are multifactorial and hence, in order to solve them there is cooperation on the level of multidisciplinary teams needed. However, numerous issues are currently being solved by social workers only, who in many cases are forced to replace experts in fields such as law, psychology, pedagogy of leisure time, geriatry or medicine. The central theme of the dissertation is dealing with the possibilities and limitations of people without home in old age living in the Capital City of Prague. Considering the factors of age, the diversity in satisfying their needs and the associated risks involved, is this a specific group of people that is, moreover, at immediate risk to their lives and health. Such chromosomal rearrangements, which cannot be detected by using of cytogenetic banding of metaphase chromosomes, i.e. chromosomes smaller than 3 - 5 Mb, and therefore modern molecular genetic methods are used to detect them, are called "cryptic rearrangements". Their important role in human pathology is more and more significant. By using of the multiplex ligation-probe dependent amplification method (MLPA) we examined a group of 50 probands with idiopathic mental retardation. A cryptic rearrangement was found at 8 probands (16 %), at 6 of them it was demonstrably causal. Then we examined a group of 40 probands suspected of gene SHOX pathology. A cryptic rearrangement was found at 17 probands (42.5 %) and at 8 of them it was demonstrably causal. Presence of small deletion founded isolated at 7 probands was verified in a population set, but without a positive result. An analysis of mutations was made too.

This diploma work named Socio-Pathological Phenomena at Secondary Vocational School and Their Possible Solution deals, in its theoretical part, with the basic terminology related with this topic. It also focuses on the risk behaviour among today's young people, and briefly introduces socio-pathological phenomena which occured at the surveyed secondary school in the last two years. It also describes other kinds of contemporary risk behaviour among young generation. At the end of theoretical part, there are introduced some of the current trends in school prevention and solution of socio-pathological phenomena. Period of adolescence is of course closely associated with this topic. It's called critical stage, and all the students pass this period during their secondary school attendance.
