THE IRIG 106 CHAPTER 10 SOLID-STATE ON-BOARD RECORDER STANDARD: A DATA PROCESSING PERSPECTIVE

This standard is intended to allow the development of a common set of data playback/reduction software, minimizing the need for a large number of unique programs to handle proprietary data structures. This paper analyzes the Chapter 10 standard from a data processing perspective, providing insight into the benefits and challenges developers will face when writing Chapter 10 software.

Small Molecule Inhibitors of MAPK and PI3K Pathways Enhance MDA-7 Lethality in Renal Cell Carcinoma

Renal cell carcinoma accounted for an estimated 57,760 new cases and estimated 12,980 deaths in the United States in 2009. Current treatment options for systemic renal cell carcinoma yield markedly low response percentages; however, recent cytokine therapy experiments have produced promising results. A novel adenovirus, Ad.5/3-mda-7, has been synthesized to efficiently infect renal cancer cells with the mda-7 gene. This gene encodes for the cytokine MDA-7/IL-24 that has the ability to specifically target transformed cells. Assays performed with this adenovirus resulted in an increased percentage of cell death in renal cancer cells when compared to infection with Ad.5/3-cmv empty vector. Further assays that combined Ad.5/3-mda-7 infection with treatments of small molecule inhibitors increased the percentages of cell death by upregulating JNK and p38 MAPK pathways, downregulating the ERK1/2 MAPK pathway, and downregulating the PI3K pathway. Western blots confirmed upregulation and downregulation of these pathways by probing for key proteins. Renal cancer cells responded best to infection with Ad.5/3-mda-7 and treatment with PD184352, PX866, and Rapamycin. This combinatorial treatment caused a greater percentage of cell death than the sum of the two individual treatments, suggesting a synergistic inhibition of cell growth pathways. These findings suggest that the combination of Ad.5/3-mda-7 and specific small molecule inhibitors has developmental potential as a novel and more efficient treatment option for systemic renal cell carcinoma.

L’application des approches probabilistes pour tenir compte des incertitudes dans le domaine des barrages est un sujet en développement. Cependant, la plupart des études ont été réalisées sur l’évaluation de la sécurité des barrages pendant leur service. Ce travail de thèse vise à appliquer ce type d’approches et à faire une contribution à l’analyse de risque des barrages en béton compacté au rouleau (BCR) d`es sa construction, à l’aide d’une simulation numérique. Les travaux présentés dans ce manuscrit proposent l’application d’une méthodologie qui vise à quantifier la vulnérabilité vis-à-vis de l’apparition de la fissuration pendant la construction du barrage, ce qui peut affecter `a long-terme la perméabilité et par conséquent, compromettre son comportement structurel. Pour ce faire, l’accent est mis sur les incertitudes liées à quelques caractéristiques des matériaux (e.g., résistance, rapport eau-ciment, entre autres) et aux conditions environnementales pendant la phase de construction. Un modèle thermo-chemo-mécanique est utilisé pour décrire le comportement du BCR. En ce qui concerne le modèle probabiliste, deux aspects sont étudiés: i) comment les incertitudes liées aux variables d’entrée sont propagées dans le modèle, et ii) quelle est l’influence de leur dispersion par rapport à la dispersion totale de la sortie. Ce dernier est évalué par l’intermédiaire d’une analyse de sensibilité globale effectuée avec la méthode RBD-FAST. En outre, la variabilité spatiale des paramètres d’entrée est aussi prise en compte à travers des champs aléatoires bidimensionnels. Par ailleurs, un couplage entre des méthodes de fiabilité et la méthode d’éléments finis est effectué de façon à évaluer le potentiel de fissuration dans chaque couche de BCR lors de sa construction en utilisant un concept de densité de fissuration. Comme résultat important issu de ce travail de recherche, des courbes de probabilité pour la densité de fissuration sont obtenues au niveau de chaque couche en fonction de leur âge et des conditions aux limites, ce qui est considérée comme étant une contribution originale de cette thèse.La méthodologie proposée peut être utilisé pour aider à comprendre comment les incertitudes vont affecter le comportement du barrage pendant sa construction et servir d’appui dans le futur pour améliorer et soutenir la phase de conception du projet de barrage. / The use of fully probabilistic approaches to account for uncertainties within dam engineering is a recently emerging field on which studies have been mostly done concerning the safety evaluation of dams under service. This thesis arises within this framework as a contribution on moving the process of risk analysis of dams beyond empirical knowledge, applying probabilistic tools on the numerical modelling of a roller compacted concrete (RCC) dam during its construction phase.The work developed here aims to propose a methodology so as to account for risks related to cracking during construction which may compromise the dam’s functional and structural behaviour.In order to do so, emphasis is given to uncertainties related to the material itself (i.e.strength, water-to-cement ratio, among others) as well as to ambient conditions during the construction phase of RCC dams. A thermo-chemo-mechanical model is used to describe theRCC behaviour. Concerning the probabilistic model, two aspects are studied : how the uncertainties related to the input variables are propagated through the model, and what is the influence of their dispersion on the dispersion of the output, assessed by performing a global sensitivity analysis by means of the RBD-FAST method. Also, spatial variability of some input parameters is accounted for through bi-dimensional random fields. Furthermore, a coupling between reliability methods and finite element methods is performed in order to evaluate the cracking potential of each casted RCC layer during construction by means of a cracking density concept. As an important outcome of this applied research,probability curves for cracking density within each casted layer as functions of both age and boundary conditions are predicted, which is believed to be an original contribution of this thesis. The proposed methodology may therefore be seen as a contribution to help engineers understand how uncertainties will affect the dam behaviour during construction and rely on it inthe future to improve and support the design phase of the dam project.

As atividades recreativas em áreas naturais são apresentadas como uma ferramenta capaz de conciliar desenvolvimento e conservação. Entretanto, a falta de um planejamento que leve em consideração os fatores relacionados com sua sustentabilidade pode ocasionar graves impactos tanto ecológicos como econômicos e sociais. No Brasil, onde as discussões sobre o assunto são muito recentes, existe falta de informações sistematizadas sobre as verdadeiras repercussões do uso recreativo e sobre a recuperação dos ambientes. Além da própria complexidade do assunto, existe uma outra dificuldade em razão da bibliografia específica, dispersa e nem sempre de fácil acesso. No presente trabalho é feita, no primeiro capítulo, uma revisão bibliográfica sobre: a natureza dos impactos; os fatores relacionados a sua ocorrência e à capacidade de carga do ambiente; os métodos de avaliação de impacto mais usados; e a experiência brasileira. Esta revisão forneceu a base teórica necessária para a elaboração do estudo de caso, visando discutir a sustentabilidade ecológica das atividades recreativas. No segundo capítulo foi realizado um estudo na antiga Fazenda Santa Carlota (SP),área de elevado potencial turístico. Para tanto, elaborou-se um zoneamento ecológico, em ambiente de SIG, sintetizando as fragilidades e as oportunidades. Para o Mapa de Fragilidades foram utilizadas as informações abióticas (pedologia,declividade, geomorfologia e clima) e bióticas (vegetação e fauna) relacionadas com a capacidade de carga para recreação (CCR) do ambiente. Para o Mapa de Oportunidades, foi feito um levantamento preliminar dos atrativos potenciais para a área de estudo. O resultado é o Mapa de Zoneamento Ecológico que classifica a fazenda em cinco zonas: três zonas propostas para o desenvolvimento de atividades recreativas, com níveis diferentes de fragilidade e potencialidade; uma de uso especial, onde está o patrimônio histórico; e uma zona de recuperação. Foi estimada a CCR, adaptando o método de Cifuentes (1992), para cada uma das três zonas destinadas para o uso. Os valores obtidos representam os limites máximos de uso que devem ser permitidos de forma a evitar a degradação dos recursos naturais. Estes valores não são absolutos e incorporam conhecimento científico e juízos de valor. A utilização do SIG mostrou-se adequada para o objetivo de zoneamento ecológico para fins recreativos. O método de CCR mostrou-se flexível e rápido. Porém, a CCR foi superestimada devido aos fatores importantes não incorporados por falta de dados específicos e à escala de trabalho. / Recreational activities in wildland areas are being considered a way to achieve development and protect the environment. However, without a planning program that considers the factors related to their sustainability, they can cause serious ecological, economic and social impacts. In Brazil, in spite of the recent concerns about recreational problems, there is a lack of systematic information about actual consequences of the recreational use and environmental restoration. It is very difficult to deal with the recreational sustainability, due to its complexity and to a dispersed and inaccessible specific bibliography. The first chapter of this work is a bibliography revision on: the nature of the impacts; the factors related to their occurrence and the environmental carrying capacity; the most useful methods of impact evaluation; and the Brazilian experiences. This revision supplied the theoretical base necessary for the elaboration of a study case that intended to discuss the ecological sustainability of recreational activities. The second chapter is a study about Fazenda Santa Carlota (SP), an area with a high touristic potential. A GIS was used to elaborate an ecological zoning, synthecizing fragilities and opportunities. Abiotic (pedology, slope, geomorfology and climate) and biotics informations (vegetation and wildlife) related with the recreational carrying capacity (RCC) have been used to elaborate the Map of Fragilities. For the Map of Opportunities, a preliminary survey of the attractive potentials for the study area was made. The result is the Map of Ecological Zoning that classifies the area in five zones: three zones that permit for the development of recreational activities, with different levels of fragility and potentiality; one zone of special use, where there is a historic site; and a restoration zone. The RCC was estimate, for each of the three recreational zones, using the Cifuentes method (1992). The values obtained represent the limits of acceptable change that could be allowed to prevent the degradation of the natural resources. These values are not absolute and involve scientific and judgmental considerations. The use of GIS revealed to be adequate for the objective of ecological zoning for recreationals objectives. The RCC method revealed to be flexible and fast. However, due to important factors not considered because of lack of specific data and due to the working scale, it overestimates the RCC.

Construção de uma cadeia sustentável para destinação de resíduos da construção civil (RCC) pelo método do Design Tthinking

Considerando que, com vistas à sustentabilidade, a questão dos resíduos sólidos deve ser tratada sob a perspectiva integrada na qual é fundamental a noção de corresponsabilidade, este trabalho tem como objetivo propor um modelo de cadeia para tratamento e destinação de resíduos. Para tanto, foi aplicada a técnica de storytelling e a metodologia do Design Thinking para o desenho de cenários econômicos e o desenvolvimento de personagens, os quais possibilitaram a indicação de medidas consideradas de baixa e de alta efetividade para aferir a viabilidade e a sustentabilidade de uma usina de tratamento de resíduos. Os dados obtidos permitiram a proposição de um modelo de negócios híbrido, onde o enfoque encontrado deslocou-se da mera criação de uma usina de tratamento para a implantação de um sistema cooperativo de triagem, separação e tratamento dos resíduos, mais resistente às variações de conjuntura econômica. / Considering that, with a view to sustainability, the issue of solid waste should be dealt with under the integrated perspective in which the notion of co-responsibility is fundamental, this work aims to propose a chain model for the treatment and disposal of waste. For that, the technique of storytelling and the methodology of Design Thinking for the design of economic scenarios and the development of characters were applied, which enabled the indication of measures considered of low and high effectiveness to assess the viability and the sustainability of a waste treatment plant. The data obtained allowed the proposition of a hybrid business model, where the approach found shifted from the mere creation of a treatment plant to the implantation of a cooperative system of sorting, separation and treatment of the residues, more resistant to the conjunctural variations economic development.

The majority of all power cuts that affects individuals and communities are caused by earth faults on the power transmission lines at 10kV and 20kV level [1]. If these power cuts could be eliminated, less disturbances and interrupts would lead to large amounts of money savings. Swedish Neutral has developed such a protection system. When an earth fault occurs power is injected into the neutral point of the transformer. The RCC (Residual Current Compensation) protection calculates a compensation current exactly 180 degrees out of phase to the fault current. Doing this, the voltage at the fault location becomes very close to zero, without affecting the power transmission. The protection system can only compensate automatically for the fundamental frequency (50Hz), and manually for the 3rd, 5th, 7th and 9th harmonics. In most cases, when the harmonics are very small it is not necessary to compensate for them. There are though cases when compensation for the harmonics is necessary. This thesis focuses on finding the best method to extract the content of a sampled signal regarding both simplicity and speed. Both amplitude and phase of each harmonic must be calculated. Is the proposed method suitable for the current computer system and how can it easily be implemented. Because the fundamental frequency is known and the harmonics are all multiples of the fundamental frequency it makes the task less complex. It is not necessary to use the FFT algorithm. The DFT can be calculated using correlation. Both phase and amplitude can be calculated very precisely with few samples and not so many computer operations.

It involves an interdisciplinary discussion on the understanding of the process the subject goes through in the joining in dynamics. It is, therefore, with the instruments from the social sciences and psychology, as well as the Freudian psychoanalyses, that this work is achieved, understanding that the categories of this theoretical approach complement each other in the understanding of the joining in process in its full length. The conscious and unconscious subjectivity supporting the joining in and the new ethical posture, consequence of the process, constitute the study&#8217;s driving force. Thus, the dialectic man in Freud and Durkheim, in his partiality of fragility and might, determines the empirical work and the analytical conclusions. The subjective motivation is discussed as the attracting force, that determines the religious transit to CCR and the permanence of the faithful in this group, which is in the process itself. The identifications accomplished are presented, as well as the renewed identities, the redefined self-images, to lead to new ethical option. The man who lives this process is identified in his dichotomy between fragility and reactivity, between helplessness and the omnipotent attitude. Fragile because he suffers and is reactive in his individual and collective moral strength, as sees Durkheim. Constitutionally helpless and defending himself from that with a megalomaniac instinct, in Freud&#8217;s view. This man wants relief from suffering and seeks in CCR the cure for his illnesses. The approach of the charismatic offering turns to the possibility of responding to the expectations of the faithful. It reflects itself on the transformations of the adherents and the possible points of intersection between the therapeutics of the CCR and psychology&#8217;s ordinary psychotherapies. In the effort to understand the phenomenon of the subject&#8217;s reconstruction, from the joining in the charismatic renovation movement, the contributions psychology can add to this study are also numbered, as well as the subsidies it can receive in a feedback basis in its task to elucidate man&#8217;s psychical dimension. / O que se vai ler centra-se no estudo dos motivos que impulsionam as pessoas a se decidirem pela adesão à Renovação Carismática Católica. Trata-se de discussão interdisciplinar na compreensão do processo por que passa o sujeito na dinâmica da adesão. É, pois, com os instrumentos das ciências sociais e da psicologia, bem como os da psicanálise freudiana, que se empreende esse trabalho, por entender que as categorias dessa abordagem teórica, se complementam para o entendimento da adesão na sua amplitude total. A subjetividade consciente e inconsciente incentivando a adesão e a nova postura ética, como conseqüência do processo, constituem a força motora do estudo. Assim, o homem dialético de Freud e Durkheim, na sua parcialidade de fragilidade e potência, determina o trabalho empírico e as conclusões analíticas. Discute-se a motivação subjetiva como a força de atração, determinante do trânsito religioso para a RCC e a permanência do fiel nesse grupo, que é processual. Narram-se as identificações realizadas, as identidades renovadas, as auto-imagens redefinidas, para resultar em nova opção ética. Identifica-se o homem que vive esse processo, na sua dicotomia de fragilidade e reatividade, do desamparo e atitude onipotente. Frágil, porque sofredor e reativo na sua força moral, individual e coletiva, como vê Durkheim. Constitucionalmente desamparado e se defendendo disso com a sua pulsão megalômana, na concepção freudiana. Esse homem quer alívio para seu sofrimento e busca na RCC, cura para seus males. A abordagem da oferta carismática se volta para a possibilidade de responder às expectativas dos fiéis. Reflete-se sobre as transformações dos adeptos e possíveis pontos de intersecção entre a terapêutica da RCC e as psicoterapias convencionais da psicologia. No empenho para compreender o fenômeno da reconstrução do sujeito, a partir da adesão ao movimento renovador carismático, relata-se a contribuição que a psicologia pode dar a esse estudo, bem como, a possibilidade de receber subsídios para a sua tarefa de elucidar a dimensão psíquica do homem.
