Zeleň jako rekreakční zázemí města / Vegetation as a recreational facility of the city

From a point of view of contemporary society which requires healthy lifestyle and healthy environment, especially in big cities there is a need for development of recreation and public green areas. First, i tis a form of preparation in planning, i.e. it is important to elaborate objectives for their subsequent promotion and enforcement and afterwards it is necessary to provide funds for their implementation. This doctoral thesis analyzes the role of everyday recreation as a part of our lives and recreation as a necessary component of our healthy lifestyle. Green areas represent the most suitable environment for this type of recreation. This thesis describes the status, importance and types of recreation and green areas in cities in the Czech Republic. It also analyzes the relationship between recreation and urban green areas in terms of uses of urban green space for daily recreation. Specifically, the work focused on conceptual and compositional principles arrangement of urban green areas. Those represent the most suitable and important potential resources for development of vegetation and everyday recreation for all people living in our cities. The intention of this thesis is to analyze the current status of recreational function of public green areas in urban developments and especially in neighbourhoods. Following that to suggest solutions for improvement and implementation of green areas as measures for design solutions within the framework of land-use planning. This thesis is supposed to act as compendium to spread knowledge about possible options how to create green areas with recreational and housing function and thus acquire complex build-up area.

The work deals with theoretical research of basis of Environmental Psychology for Outdoor Education. It answers two questions ? why in the conceptions of Outdoor Education is nature the preferred environment and what are the benefits of nature for physical, mental and social health. The answers to theses questions are in the work described through theories of environmental preference and restorative effects of nature. There are three major sections of the work. The first part discusses the basis, principles and contents of Environmental Psychology and Outdoor Education, the core terminology of the work, and the nodal points of Environmental Psyhcology and the theory of Outdoor Education. Second part describes those theories and concepts of Environmental Psychology which are related to the primary and subsidiary research questions. The third part evaluates the impacts and relevance of environmental-psychological knowledge on concepts of Outdoor Education.

All that in context of otherwise slowly shrinking post-industrial city with aging and declining urban population.

Management postindustriálního vývoje / Management of the revitalization of the post-industrial areas

The theme of this dissertation is “Management of the regeneration of post-industrial areas”. The aim of the work is to identify support mechanisms for the redevelopment of post-industrial areas in the Czech Republic. Such support mechanisms were researched and analysed in those EU countries that have broad and varied experience with the regeneration of post-industrial areas. The most favourable case studies were selected from Germany, with a particular focus given to the districts around the city of Leipzig, and the United Kingdom, highlighting the city of Manchester. For these chosen examples, I have analysed the legislative, institutional, and financial tools of urban regeneration support. One chapter is dedicated to the analysis of the environmental assessment tools for buildings and urban communities as alternative approach to the support of municipal regeneration. Findings from all chapters are contrasted with the analysis of four typical examples of derivation of post-industrial areas in Czech cities, which were chosen as being typical of unsuccessful redevelopments. Based on these examples I have compiled recommendations gained from successful mechanisms for urban regeneration abroad which could be considered and put into practice here in the Czech Republic.

Neurological disorders affect more than 14% of the population worldwide and together with traumatic brain and spinal cord injuries represent major health, public and economic burden of the society. Incidence of inherited and idiopathic neurodegenerative disorders and acute CNS injuries is growing globally while neuroscience society is being challenged by numerous unanswered questions. Therefore, research of the CNS disorders is essential. Since animal models of the CNS diseases and injuries represent the key step in the conversion of the basic research to the clinics, we focused our work on generation of new animal models and on their use in pre-clinical research. We generated and characterized transgenic minipig model of Huntington's disease (HD) which represents the only successful establishment of a transgenic model of HD in minipig which should be valuable for testing of long term safety of HD therapeutics. Next, we crossed the well characterized R6/2 mouse HD model with the gad mouse model which lacks the expression of UCHL1 which led to results that support the theory of "protective" role of mutant huntingtin aggregates and suggest that UCHL1 function(s) may be affected in HD disturbing certain branches of Ubiquitin Proteasome System.
