The Diploma’s thesis deals with calculation of tree heat schemas with condensing steam turbine with uncontrolled steam extractions for regeneration – tree low pressure heaters, two high pressure heaters and a deaerator. Heat schemas are compared to each other according to reheat pressure. The most efficient schema is used to steam turbine thermodynamic design – design of control stage and blade stages. At the end of diploma thesis a stress analysis of the last rotor blade is made.

The innate and adaptive immune processes are modulated by hormones including glucocorticoids and by microbiota. The exact mechanisms underlying the microbial and hormonal contributions to this control are not completely clear. Present study is therefore focused to crosstalk between microbiota and de novo biogenesis or local regeneration of glucocorticoids. In particular, the study analysed the effect of commensal microbiota on expression of genes encoding steroidogenic enzymes (Star, Cyp11a1, Hsd3b1, Cyp21a1, Cyp11b1) and regeneration of glucocorticoids (Hsd11b1) in adrenal glands, colon, spleen and mesenteric lymph nodes using conventional and germ-free mice. The expression of all 5 components of steroidogenesis was identified only in the adrenal gland and colon, whereas the lymphoid organs expressed predominantly Star, Cyp11a1 and Hsd3b1 indicating the ability to produce only progesterone but not corticosterone. Microbiota decreased the expression of Star in all studied tissues but the expression of other genes was insensitive to microbiota or did not respond homogenously depending on the tissue and gene. The preclinical imaging method micro-CT (microtomography) allows the visualization and quantification of the structure of samples at a resolution of micrometers. Its' importance is increasing globally. In addition to several advantages (non-destructive, the possibility of direct 3D analysis, time efficiency, etc.), micro-CT also has some significant limitations (problematic validation of results, image artifacts, significant influence of image modifications, etc.). This thesis focuses on the application of micro-CT in the field of research and development of metallic and non-metallic materials promoting bone healing with their possible clinical applications. The first part addresses the limitations of micro-CT through several studies. A comparison of pore sizes in biomaterials utilizing scanning electron microscopy (SEM) and micro-CT was performed, and the complications of pore size evaluation were presented. SEM image analysis leads to significantly higher values than micro-CT (approximately three times), which allows for comparison of the studies using only one of these methods. Modification of nanofibers is an actual trend in tissue engineering. Polyvinylacohol (PVA) is nontoxic and biodegradable polymer suitable for preparation of submicron fibers by electrospinning. Main disadvantage of PVA fibers is rapid degradation in aqueous environment. On the other hand surface of fibers contains free hydroxyl group that could be chemically modified. In recent work, chemical modification of PVA nanofibers prepared by needleless electrospinning was investigated. Polyethylenglykol (PEG) linker was introduced to the fiber surface by acylation (PVA-PEG) and further modified by biotin (PVA-PEG-b) as a function agent. Process of chemical modification does not affected fibrous morphology of samples. Interestingly, linkage of PEG-b linker promoted stability of PVA in aqueous environment. PVA-PEG-b sample was stable for 41 days. Stability of samples was strongly dependent on amount of introduced PEG-b linker, thus proposed method of modification allows to prepare nanofibers of different solubility. Additionally, biocompatibility of chemically modified nanofibers with both mesenchymal stem cells (MSC) and chondrocytes was determined. This thesis deals with sporting massage and its effects to the human body. There are used some of diagnostic methods verifies the effects of sporting back massage on a muscular function, a skin appearance, physiological functions of the organism and a subjective feeling of a relaxation. For example, there are methods such as a static and a dynamic examination of a spine or an oximeter and a caliper measurements compared to subjective feelings of relaxation in the back area. The methods were applied before and after the experiment, which was based on the series of sporting back massages. The results of the research are compared with other studies and speak in favor for the sporting massage as an effective means of regenerating for the human body as expected on the beginning of the research. The thesis can be used as an available information source for students of sports disciplines and the physically active public, and it can motivate them to include sporting massage into an everyday regeneration processes.

Výzkum regenerace filtračního materiálu zemních filtrů a kořenových čistíren odpadních vod / The Research on Regeneration of the Filter Material, used in the Soil Filters and Constructed Wetlands

The thesis consists of two basic parts. The first research part creates an introduction to the root zone wastewater treatment plants. It contains mainly causes of filter environment colmation and its theoretical solution methods, which are further solved in the laboratory. The master’s thesis attempts to find a simple way, which would in future be possible to evaluate the rate of sedimentation of the sludge material, which causes filtration environment colmation. Results of developed methodology will be used in practice by washing colmated filtration material, respectively colmatant separation in sedimentation tank. The thesis focuses to verify the possibility flocculation and mineralization of sludge, which adheres to the surface of the filtration material. The solution lies in the observation of sedimentation rate (after previous dilution) using by the turbidimetric method, based on the beam-column passage through muddy water. The results show that it is not critical ambient temperature, but the time effect of natural mineralization in combination with moisture (dry matter concentration, the amount of water). The present thesis evaluates the possibilities and application method, leading to the natural regeneration of the filtration material exposed to natural climatic conditions.

Přírodní biomateriály a mesenchymální kmenové buňky v léčbě míšního poranění / Natural biomaterials and mesenchymal stem cells in regeneration of spinal cord injury

Spinal cord injury is a serious trauma and despite intensive research there is still no effective treatment for patients. The aim of this thesis is to study new possibilities of spinal cord injury therapy in animal models. We have focused on the use of natural materials, stem cells, gene therapy and the possibility of combining these approaches. The effect of extracellular matrix (ECM) based materials prepared by decellularization of porcine spinal cord and porcine urinary bladder on tissue regeneration after acute hemisection of the spinal cord was investigated. Another tested material was a hydrogel based on hyaluronic acid modified with RGD adhesion peptide, which was applied acutely and subacutely into the hemisection lesion. We have shown that both types of biomaterials have positive effect on regeneration of the spinal cord tissue by bridging the lesion and promotion of axonal ingrowth. In addition, ECM hydrogels promote the growth of blood vessels into the lesion site. The combination of hydrogels with mesenchymal stem cells derived from human umbilical cord (hWJ-MSCs) had synergistic effect, but since only a limited number of cells could be incorporated into hydrogels, this effect was not associated with improvement in motor skills. The aim of this thesis is to study the regenerative and neurotrophic effects of injectable extracellular matrix (ECM) hydrogels prepared by decellularization from porcine tissue (brain, spinal cord, bladder) and human umbilical cord (UC) in reconstruction of damaged neural tissue of the brain and spinal cord in rats. We characterized ECM hydrogels in terms of mechanical and rheological properties, structure and composition, cell adhesion, migration and proliferation. In a model of spinal cord hemisection, we compared the regenerative effect of ECM hydrogels derived from porcine spinal cord and urinary bladder.
