Delayed Oxidative Injury to the Superior Colliculus and Retinal Changes After Cerebral Hypoperfusion/Reperfusion Injury

Damage to visual pathways can lead to irreversible blindness. Posterior visual pathways, located within a watershed area, are predisposed to hypoperfusion/reperfusion injury. In a novel rat model of bilateral common carotid artery occlusion (BCCAO), oxidative injury to the superior colliculus (SC), a major visual center within the watershed area was evaluated, in addition to its effects on retinal ganglion cells (RGCs). Nitrotyrosine, a footprint of peroxynitrite-mediated oxidative injury in the SC, and microtubule-associated protein 2, a dendrite marker in the retina, were assessed using immunofluorescence and confocal microscopy. Nitrotyrosine-immunoreactivity in the SC was increased 2 weeks after BCCAO compared to controls. Microtubule-associated protein 2-immunoreactivity in the central inner plexiform layer was reduced 3 weeks after BCCAO compared to controls. Global incomplete cerebral hypoperfusion/reperfusion induced oxidative injury in the SC and retrograde RGC dendritic changes. This suggests that cerebrovascular injury affecting the posterior visual pathways may contribute to vision loss in patients.

The role of sexual imprinting in speciation: lessons from deer mice (genus Peromyscus)

Sexual imprinting, the process of learning mate preferences at a young age, could promote speciation by reducing attraction to individuals from divergent populations or species, consequently creating or maintaining reproductive isolation. Yet, despite the documentation of sexual imprinting in many taxa, its connection to speciation has been understudied. I chose to explore the potential link between sexual imprinting and reproductive isolation and in two North American rodents--the white-footed mouse (Peromyscus leucopus) and its sister species, the cotton mouse (Peromyscus gossypinus). These species have overlapping distributions in nature, possibly allowing interbreeding and admixture. In Chapter 1, I used double-digest restriction-associated DNA sequencing to test for hybridization in sympatric natural populations and found that 1.5% of sampled individuals showed evidence of admixture yet the species have maintained genetic distinctness in sympatry. In the lab, the species hybridize when given no choice of mates but mate more readily with conspecifics, suggesting that mating preferences may prevent hybridization in the wild. In Chapter 2, I tested whether mating preferences create significant reproductive isolation. I measured mating preferences in controlled laboratory conditions and found that both species and sexes preferred conspecific to heterospecific mates in 85% of trials. I then raised offspring with foster parents of the opposite species and found that P. leucopus has a genetically-determined preference while P. gossypinus learns its preference. In Chapter 3, I tested whether sexual imprinting on parental diet could generate assortative mating within a species. I tested this hypothesis by feeding P. gossypinus parents either orange- or garlic-flavored water, thereby exposing their offspring to these flavors through their parents until weaning. I tested the preferences of these offspring as adults and found that P. gossypinus, especially females, had strong assortative mating preferences. This implies that at least females learn parental dietary information and that assortative mating could evolve within a single generation. Together, my results confirm that sexual imprinting on parental traits--possibly mediated through dietary differences--can create assortative mating capable of generating sexual isolation and reducing gene flow between species. My research supports the importance of mating preferences and learning in speciation.

HMG-CoA reductase inhibitors do not attenuate the inflammatory response associated with glutaraldehyde-fixed bioprosthetic heart valve conduits

Evidence suggests that there is an immunological response of the recipient to xenograft bioprosthetic heart valves. Information on the impact of HMG-CoA reductase inhibitors (statins) and their anti-inflammatory properties on bioprosthetic valve failure remains limited. We sought to examine the efficacy of statin therapy in a rodent model of bioprosthetic valve implantation. To mimic the human scenario, fresh or glutaraldehyde-fixed aortic valve root conduits from Lewis rats or Hartley guinea pigs were microsurgically implanted intravascularly into the infra-renal aorta of Lewis rats. The syngeneic control group consisted of a fresh rat valve conduit implanted into a rat. The xenogeneic control group consisted of a glutaraldehyde-fixed guinea pig valve conduit implanted into a rat. Treatment groups consisted of xenogeneic groups treated with either daily steroids or statins. Overall, steroid treatment attenuated the inflammatory response observed within the xenogeneic glutaraldehyde-fixed valve conduits. Treatment with statins did not decrease this inflammatory response.

SUPPORT VECTOR MACHINE FOR HIGH THROUGHPUT RODENT SLEEP BEHAVIOR CLASSIFICATION

This thesis examines the application of a Support Vector Machine (SVM) classifier to automatically detect sleep and quiet wake (rest) behavior in mice from pressure signals on their cage floor. Previous work employed Neural Networks (NN) and Linear Discriminant Analysis (LDA) to successfully detect sleep and wake behaviors in mice. Although the LDA was successful in distinguishing between the sleep and wake behaviors, it has several limitations, which include the need to select a threshold and difficulty separating additional behaviors with subtle differences, such as sleep and rest. The SVM has advantages in that it offers greater degrees of freedom than the LDA for working with complex data sets. In addition, the SVM has direct methods to limit overfitting for the training sets (unlike the NN method). This thesis develops an SVM classifier to characterize the linearly non separable sleep and rest behaviors using a variety of features extracted from the power spectrum, autocorrelation function, and generalized spectrum (autocorrelation of complex spectrum). A genetic algorithm (GA) optimizes the SVM parameters and determines a combination of 5 best features. Experimental results from over 9 hours of data scored by human observation indicate 75% classification accuracy for SVM compared to 68% accuracy for LDA.

Farmer Perceptions of Several Rodent Pest Management Methods and the Trap-Barrier System (TBS) in Cambodia

In Cambodia, over 80% of the population is engaged in agricultural production, mostly in rice cultivation. The country, however, faces a lot of problems of rice crop damage. In Cambodia, rodents are considered to be one of the most important pests of rice, with the potential to cause severe damage during periodic plague events. As is seen in other developing countries, physical methods of rodent control are probably the most commonly used approaches. In general, the choice of methods varies with the availability of resources, season, the participation of farmers, the condition of the rice field, weather conditions, experience with the success of the techniques, likely level of rodent attack and other considerations. However, effective rodent control methods suiting farmers’ needs and applicable to the agricultural production environment in Cambodia are yet to be assessed. The aim of this research was to examine the socio-economic factors impacting on the implementation of several rodent control methods in comparison to the Trap-Barrier System (TBS) in order to improve rodent pest control in Cambodia. The research employed a constructivist approach in which the researcher played a role as a participant observer, to study the social setting and realities as constructed by the participants in the study sites. Rodent management was studied as a complex phenomenon, occurring within the milieu of family, village and commune life in rural Cambodia. The study looks into the beliefs and behaviours of the farming community and examines how Cambodian traditions affect the implementation of rodent management techniques and the management of rodents as a community activity. Participatory techniques were employed to gather information concerning the farmers’ assessments of the effectiveness of various rodent management options practiced in Samrong Commune. The level of adoption and adaptation of TBS in the commune was observed over a period of several years. This study is the first detailed study of farmer perceptions of alternative rodent control methods in Cambodia, especially the TBS. The study was based on long term trials involving a TBS application on a commune scale and investigations after the removal of all subsidies for purchase of materials. It was apparent that TBS use declined and eventually become limited to a few individuals who had worked closely with the project. The study also demonstrates how BBNs and Netica software can be applied as participatory tools to develop and explore the decision making structure of farmers. These tools can also be used as extension tools and can contribute to better decision making by communities. The study also shows how an understanding of traditional aspects of public goods management by communities can contribute to the design of effective contemporary solutions to problems involving the community and public goods management. In this sense its significance goes well beyond the specific context of rodent control, pest management and rice production and takes on wider significance in the field of community development generally. The study applied a mixed qualitative and quantitative approach to data collection and analysis, involving interviews, surveys, modelling, workshops, field notes, observations and document review. Five rodent control methods (TBS, rodenticide, electric fencing, netting and hunting) were found to be the most common rodent control methods used in the study sites. Some common factors were found to influence the effectiveness of these methods. Farmer participation was found to be the most widely influential factor affecting the rodent management techniques. Netting method was considered the most effective method of rodent control by farmers, as well as the method that had the best benefit-cost ratio. This technique was only found to be applicable in the wet season. In general, the effectiveness of TBS and its benefit-cost ratio was ranked fourth of the five methods, because it required high farmer participation, monitoring and maintenance as well as high material expense. Improvement of TBS can be made when it can be clearly demonstrated to farmers that TBS is an applicable method, being safe and able to catch more rats. To suit the farmers’ requirements for an effective rodent control method, TBS must be able to deal with migrating rats, situations of high rat abundance and the planting problems associated with an irregular rain regime. To achieve these conditions, some intervention policies are required in terms of research and extension support.

Biological therapies for the restoration of degenerated intervertebral discs

Low back pain is a common cause of disability and work inability, often associated with intervertebral disc degeneration. The current understanding of disc degeneration is limited and none of the available treatments is entirely effective. The work described herein investigates potential strategies for the biological herapeutic restoration of disc degeneration. Firstly, an in vitro study to investigate the effects of BMP-7 on human discal cellular viability was performed. Cultured cells were treated with TNF-a or H202 to induce apoptosis, resulting in the down regulation of extracellular matrix proteins, decreased cell viability, morphological changes and activation of caspase-3; however, addition of BMP-7 alone prevented the observed effects, demonstrating the ability of BMP-7 to prevent apoptosis of human disc cells in vitro. Secondly, the differentiation potential of stem cells towards disc-like cells was studied. Rodent mesenchymal stem cells (rMSCs) were cultured alone or co-cultured with rat disc tissue. Differentiation potential of rMSCs was evaluated by mRNA and protein expression, cellular function and morphological studies. The co-culture conditions led to the expression of chondrocytic markers in rMSCs, whereas rMSCs cultured alone did not express the chondrocytic markers. Cellular contact between the co-cultured rMSCs and the discal tissue were observed. This study demonstrated that rMSCs can differentiate into functional disc-like cells in a tissue influenced co-culture environment. Finally, the survival and differentiation of CD34+ or CD34?? bone marrow (hBM) cells, in an intra-discal xenogeneic transplantation rat model was investigated. Human CD34+ or CD34?? cells were isolated, fluorescent-labelled and injected into rat coccygeal discs. The survival of transplanted cells was confirmed by fluorescent positive cells as well as a human nuclear specific marker. Interestingly, CD34?? cells survived until day 42 in the injected discs, and differentiated into cells ex:pressing a chondrocytic phenotype. In contrast, CD34+ cells could not be detected by day 21. This data suggests that transplanted hBM CD34?? cells, in contrast to CD34+ cells, were able to survive and differentiate within the intervertebral disc. Together, the results of these studies can both encourage and contribute to the basis of potential biological therapies in the restoration of intervertebral disc degeneration.

Effects of Off-road Vehicles on Rodents in the Sonoran Desert

abstract: Human recreation on rangelands may negatively impact wildlife populations. Among those activities, off-road vehicle (ORV) recreation carries the potential for broad ecological consequences. A study was undertaken to assess the impacts of ORV on rodents in Arizona Uplands Sonoran Desert. Between the months of February and September 2010, rodents were trapped at 6 ORV and 6 non-ORV sites in Tonto National Forest, AZ. I hypothesized that rodent abundance and species richness are negatively affected by ORV use. Rodent abundances were estimated using capture-mark-recapture methodology. Species richness was not correlated with ORV use. Although abundance of Peromyscus eremicus and Neotoma albigula declined as ORV use increased, abundance of Dipodomys merriami increased. Abundance of Chaetodipus baileyi was not correlated with ORV use. Other factors measured were percent ground cover, percent shrub cover, and species-specific shrub cover percentages. Total shrub cover, Opuntia spp., and Parkinsonia microphylla each decreased as ORV use increased. Results suggest that ORV use negatively affects rodent habitats in Arizona Uplands Sonoran Desert, leading to declining abundance in some species. Management strategies should mitigate ORV related habitat destruction to protect vulnerable populations. / Dissertation/Thesis / M.S. Applied Biological Sciences 2012
