Evaluating Basin Wildrye Seed Sources across Provisional Seed Zones, Native Forb Sowing Depth on Species Performance and Improving the Accuracy of Collection Site and Seed Lot Identification for Big SagebrushJensen, Scott L 16 March 2020 (has links)
Identifying genetically appropriate plant materials for seed based restoration relies on the principle of local adaptation where the objective is to match adaptive genetic characteristics to variation in ecological clines pertinent to plant establishment and persistence. In this study, basin wildrye (Leymus cinereus (Scribn. & Merr.) Ã. Löve) sources from 25 wild populations and 4 commercial varieties were planted at 4 test sites. We assessed initial establishment and short term persistance. Plantings failed at 2 sites in both 2013 and 2014, with too few plants to quantify differences. At the remaining 2 sites, local sources had higher initial establishment in just 1 of 10 comparisons. Among commercial sources, the cultivars Magnar and Trailhead initially outperformed local pooled materials at Fountain Green but not at Nephi. Initial establishment under row cover was dramatically better than uncovered controls at both sites, but only persisted for 4 years after planting at the Fountain Green site. The native forb study evaluated the effects of species, sowing depth and row cover on field emergence of 20 forbs. Overall, emergence was very low ranging between 0.2% and 1.0% for 16 of the 20 species. Four species exceeded 1% emergence. Depth effects were species, site and year dependent. The odds of emergence decreased with increasing depth for four species, increased for three species and were mixed between sites and years for the remaining species. The odds of emergence were better under row cover than for uncovered control plots. Depths evaluated were deeper than recommended for most species and likely hindered emergence for some species. Site and year had much more effect on observed emergence than depth or treatment. Developing simple diagnostics to identify subspecies is key in the restoration of sagebrush ecosystems. We evaluated the SoilWeb app as a tool to identify sagebrush in the field. We evaluated the accuracy of the Richardson et.al. (2015) technique to classify sagebrush stands and evaluated data modeling strategies to improve classification accuracy. We found the SoilWeb app to be an accurate and informative tool to identify native-wild sagebrush populations. The Richardson et.al. (2015) seed weight criteria correctly classified just 19% of our sample populations to the correct subspecies. To improve upon this, we evaluated multifactor modeling using recursive partitioning and classification trees. Our most accurate classification tree correctly classified 80% of 2x tridentata sites but just 45% of wyomingensis sites.

Investigation of Putative Genetic Factors Associated with Soybean [Glycine Max (L.) Merr.] Seed Quality TraitsSkoneczka, Jeffrey Allen 01 December 2009 (has links)
Soybeans are an economically important plant, with an annual crop value that consistently exceeds 20 billion dollars in the United States alone. A recent increase in demand for soybeans, stemming from its diverse applications in products such as animal feed, oil, and biofuel, has created an emphasis for soybean breeders in value added cultivars. These cultivars, have improved, or altered, agronomic or seed composition traits, allowing them to be efficiently utilized in a specific niche of the processing industry. Facilitating the development of such cultivars requires a thorough understanding of the genetic factors that affect the manifestation of value added traits. Value added traits investigated in this study include seed sucrose, raffinose, stachyose, and phytate content, seed weight, and maturity.
The objective of the first part of this project was to characterize the source of low seed stachyose in soybean line PI200508. Two F2 populations, developed from PI200508 and soybean introductions which exhibited higher seed stachyose content were utilized in a QTL analysis approach that incorporated the use of the Williams82 whole genome shotgun (WGS) sequence (http://www.phytozome.org) in a candidate gene mapping approach. A predicted soybean galactosyltransferase gene was established as a candidate gene due to its observed segregation with the single low stachyose QTL observed on molecular linkage group (MLG) C2 in both populations. Sequencing of this putative gene revealed a unique 3 bp deletion in PI200508. A marker developed to exploit this deletion accounted for 88% and 94% of the phenotypic variance for seed stachyose content in the two experimental populations, highlighting its potential for use in marker assisted selection of the PI200508 source of low raffinose and stachyose.
The second part of this project involved QTL analysis of seed sucrose, raffinose, stachyose, and phytate content, as well as seed weight in a linkage map for a F8 RIL population developed from the Glycine max line V71-370 and the Glycine soja introduction PI40712. Analysis across all 20 soybean MLG identified 25 QTL for these traits on MLG A1, A2, C2, D1b, D2, F, G, H, I, L, M, O. Nine of these QTL were supported across multiple environments, indicating that they, and their associated markers, could be useful to breeders working with these traits.
The third part of this project used the same F8 RIL linkage map to investigate time to maturity (Reproductive stage R8). V71-370 and PI407162 differ in time to maturity when grown in Virginia, and the RILs developed from this cross displayed a wide range in maturity. Two major QTL were identified on MLG H and L. Examination of the Williams82 WGS sequence in these QTL regions revealed two predicted genes with homology to Arabidopsis thaliana light response and photoperiodism genes which were investigated as candidate soybean maturity genes. Markers developed from these predicted genes showed close association with the observed QTL, and could facilitate the further investigation of this complex trait. / Ph. D.

Reproductive patterns of birches (Betula spp.) in northern SwedenHolm, Stig-Olov January 1994 (has links)
The aim of this thesis was to study patterns of reproduction of Betula pendula and B. pubescens coll. along an altitudinal, coastal-inland, gradient in northern Sweden. The altitudinal variation was related to the distribution of the birch taxa along the gradient. Six years field data showed a steep decrease of seed germinability of B. pendula towards its altitudinal range limit in the Scandes every year. In contrast, B. pubescens ssp. pubescens showed significant positive correlations between seed germinability and altitude in three of the six years. Furthermore, there was a highly significant positive correlation between seed weight and altitude for B. pubescens coll., but not for B. pendula. Production of viable seeds fluctuated strongly between years in most populations, except in marginal B. pendula populations in the mountain area where it was constantly very low. On average 15 - 41 % of the seeds produced in B. pendula populations above 400 m altitude were attacked by gall midges (Semudobia ssp.). Corresponding values for B. pendula populations below 400 m altitude were 4 - 7 %. In B. pubescens populations, the seeds attacked by Semudobia ssp. never exceeded 3 %. The high frequency of Semudobia attackes in high altitude marginal B. pendula populations was suggested to be due to limited resources for defense against the seed predator. A 3-yr study documented large variations in pollination and seed quality between taxa, high and low altitude populations, and between years. Empty seeds (without embryos) dominated among the sampled seeds in most cases. This proportion was decreased by pollen addition, in both B. pendula and B. pubescens, in mountain populations, but not in coastal populations. The high percentage of empty seeds was therefore suggested to be partly caused by pollen-limitation, but failure of pollen tube penetration - fertilisation, or maternal resource supply could also have had an influence. A laboratory experiment showed increased pollen germination and length of the longest pollen tube per style after increased pollination. The correlations between number and length of pollen tubes per style were however mostly low in natural populations, suggesting low probability of pollen competition in the natural situation. Sowing experiments indicated that differences in initial seedling density between B. pendula and B. pubescens was more affected by interspecific differences in seed quality than by interspecific differences in survival of seedlings after establishment A study of the age structure of a B. pendula stand, planted about 250 m above its natural altitudinal limit, indicated that this birch may reproduce above its recent range limit during temporally warmer periods. It was concluded that the level of accumulated resources in B. pendula in marginal sites in the Scandes would mostly be too low for completion of the reproductive cycle. In contrast, B. pubescens ssp. pubescens is able to accumulate enough rescources for reproduction also at relative high altitudes. / <p>Diss. The africanized honey bee (Apis mellifera L.) for pollination and seed oil production in Jatropha curcas (L.) / A abelha melÃfera africanizada (Apis mellifera L.) na polinizaÃÃo e produÃÃo de Ãleo das sementes do pinhÃo manso (Jatropha curcas L.)RÃmulo Augusto Guedes Rizzardo 27 February 2012 (has links)
Conselho Nacional de Desenvolvimento CientÃfico e TecnolÃgico / The purpose of this thesis was to investigate the feasibility of using the honey bee (Apis mellifera L.) for pollination and yield increment in Jatropha curcas L. crops. Therefore, the floral biology and pollination process of J. curcas were studied as well as the foraging behavior and pollination efficiency of A. mellifera in this crop by the introduction of four colonies in a hectare. The work was carried out in a five years-old commercial plantation comprising 10ha, in the state of PiauÃ, (08Â32â19,0âS and 43Â56â19,7âW, 220 meters above sea level), Brazil, from March to July, 2009. Flowers were presented in protogynous inflorescences and the anthesis and most of pollen presentation took place mainly in the morning shift. Inflorescences last, in average, 20 days, with female flowers concentrating in the first third of the period and male flowers predominating in the second third, in a relation of 18.1:1 male/ female flower. Although this may favor xenogamy, the stigma receptivity lasts for, at least, five days ensuring the flower to reach the period of most pollen release in its own inflorescence, facilitating the occurrence of geitonogamy. Flowers pollinated up to four days after anthesis did not differ (p>0.05) in fruit setting to those pollinated in the first, second or third day after anthesis. Foraging by A. mellifera occurs throughout the day and peaks between 13h00min and 15h00min and is characteristic of nectar harvesting both in male and female flowers. Only one visit by A. mellifera to J. curcas flowers set 100% producing results similar (p>0,05) to those from manual xenogamy (96%), manual geitonogamy (94%) and open pollination (93%). However, only one visit by a bee was not enough to maximize the oil content per seed (213mg) in comparison to the open pollination and geitonogamy treatments (both 250mg). Besides that, it was possible to observe that geitonogamy led to greater oil production than xenogamy (237mg). It is concluded that A. mellifera is an efficient pollinator of this crop. The introduction of four honey bee colonies per hectare maximize oil production and produce the best results for all parameters studied. Furthermore, only one visit by the honey bee to J. curcas flowers was enough to prevent pollination deficit in this crop.

Genetische Analyse von Heterosis im jüngsten Stadium einer Pflanze, als Embryo, am Beispiel der Fababohne (<i>Vicia faba </i>L.) / Genetic analysis of heterosis at a plant`s earliest stage, as embryo, using faba bean (<i>Vicia faba </i>L.) as modelDieckmann, Susanne 20 April 2007 (has links)

Performance de cruzamentos entre genitores tolerantes à ferrugem asiática da soja / Performance of crosses among parents with tolerance to soybean rustMarques, Marcelo Cunha 29 April 2014 (has links)
The Asian soybean rust (FAS) was reported for the first time in Brazil in 2000/2001 and has since caused considerable damage to soybean crops. The use of genetically resistant cultivars is a key component of an integrated program to control this disease. In the researches has been found many difficulties because the fungus Phakopsora pachyrhizi shows great variability in pathogenicity and the small number of sources of resistance that are available has no adaptability to Brazilian environmental conditions. The overall objective of this study was to evaluate the performance of experimental soybean lines derived from crosses among soybean rust tolerant parents, in order to increase the horizontal resistance (tolerance). The experiments were conducted during the crop years 2008-09, 2009-10 and 2010-11, in three experimental stations (Anhumas, Areão and ESALQ), areas belonging to the College of Agriculture \"Luiz de Queiroz\" and located within the municipality of Piracicaba, State of São Paulo, Brazil. The experiments with progenies in generations F2:4, F2:5 and F2:6 were designed in augmented block of Federer, with each experiment divided into sets with common checks. In order to estimate the effect of FAS and the level of tolerance of each genotype to FAS, in each location and crop year, there was conducted two experiments with applications of different fungicides: in the first experiment was applied one fungicide for controlling only the soybean late season leaf diseases (DFC), while in the second experiment were applied one fungicides with active principles for controlling DFC as well as FAS. In the experiments with control of DFC and FAS there was tendency of superior means for PG and PCS. The methodology of contrasting the plant reactions to different fungicides was effective for estimating the effect of FAS and the level of tolerance of each genotype to FAS. The pressure of the fungal FAS on progenies ranged among crop years, being highest in 2009-10. The location Anhumas stood out among the rest, enabling high average seed yield (PG) in all evaluated crop years; this environment also proved to be suitable for estimating the rust effect. The parents 1 (USP 04-18032) and 5 (USP 11-38) were those that originated the earliest progenies and parents 3 (USP 97-08135) and 10 (USP 191-104-11) the later progenies. The parents 8 (USP 191-102-03) and 10 (USP 191-104-11) were the main contributors to higher average PG and one hundred seed weight (PSC). However, the parent 4 ( BUSP 16-015 ) exceeded for PG, besides also have presented high value of average combining ability for PG, in average of the three crop years. The cross 7x10 (USP 14-06-20 x USP 191-104-11) stood out by originating the highest number of progenies with superior performance in PG and tolerance to FAS. On the other hand, the parents 1 (USP 04-18 032), 3 (USP 97-08 135), and 9 (USP 191-103-12) have contributed with the smallest number of superior progenies in PG. There was variation among progenies within cross, for the estimates of adaptability and phenotypic stability parameters. Consequently, the progenies 231-2224-12 (6x8 = USP 97-10 046 x USP 191-102-03), 231-6127-06 (1x8 = USP 04-18 032 x USP 191- 102-03), and 231-1120 - 08 (7x10 = USP 14-06-20 x USP 191-104-11) were the most suitable for the utilization as parents in the recurrent selection for tolerance to FAS.

Performance de cruzamentos entre genitores tolerantes à ferrugem asiática da soja / Performance of crosses among parents with tolerance to soybean rustMarcelo Cunha Marques 29 April 2014 (has links)
