Plant tissue culture and artificial seed production techniques for cauliflower and their use to study molecular analysis of abiotic stress tolerance

A protocol for cauliflower micro-propagule production was developed and optimised for both micropropagation and artificial seed production techniques using meristematic tissues from cauliflower curd. All steps in the protocol were empirically optimised including: blending, sieving, culture methods, liquid culture media composition and plant growth regulator combinations and concentrations. The cost of the micro-propagules could be reduced by as much as 50% on the initial costings reported previously since treatments doubled the number of microshoots produced per culture unit. The research confirmed the suitability of cauliflower microshoots to be encapsulated as artificial seeds and an effective protocol for microshoot encapsulation was designed through the optimization of 1) the production of cauliflower microshoots suitable for encapsulation, 2) encapsulation procedures, 3) artificial seed artificial endosperm structure, 4) conversion materials. The possibility of culturing cauliflower artificial seeds in commercial substrates such as perlite, sand, vermiculite and compost was confirmed. The use of plant preservative mixture (PPM) for the control of contamination in cauliflower culture media and artificial seeds was optimised and the effect of this material on the development of plant material was assessed. It was confirmed that cauliflower artificial seed could be stored in a domestic refrigerator for up to 6 months which could have a great impact in cauliflower breeding programmes. The huge number of cauliflower microshoots that could be produced using this protocol and the homogeneity of the culture system, provided a tool for the molecular analysis of cauliflower microshoots (and artificial seed) abiotic stress tolerance analysis. Various treatments were conducted to improve microshoot cold tolerance and the up-regulation of the CBF/DREB1 transcription factor including low temperature acclimation, mannitol, ABA (abscisic acid) and Mo (molybdenum). Microshoots were confirmed to acclimate successfully using low temperature. Mo was shown to improve the cold tolerance of cauliflower microshoots and to up-regulate CBF/DREB1 in the absence of low temperature acclimation. Acclimation did not increase the accumulation of dehydrin proteins and it is concluded that dehydrins do not play a significant role in the cold tolerance of cauliflower microshoots. Since cauliflower breeding and seed multiplication protocols make extensive use of micropropagation, the studies reported in this research could make a significant impact by decreasing the cost of micropropagation and increasing its reliability. It also opens new perspectives for further research for cauliflower artificial seed production and the possibility of sowing these seeds directly in the field. Furthermore, this research helps to facilitate cauliflower breeding programmes by improving the understanding of abiotic stress tolerance mechanisms and the relationship between different types of abiotic stresses such as cold and drought.

Effect of environmental stresses and growing medium amendment with 'Zander' on growth of Acacia saligna under saline conditions

In Libya salinization of land and ground water is a key problem. A. saligna is one species that offers potential for growth under these conditions. Experiments were undertaken to accelerate the germination of Acacia seeds, and various pre-treatment methods were assayed. Boiling water treatment, mechanical scarification and acid improved germination and germination rate. Sowing A. saligna seeds at 30 mm depth gave greatest seedling growth from large and medium seeds whereas 20 mm was more suitable for small seeds. This study aimed to improve the establishment of Acacia saligna irrigated with saline water, plants were grown for seven weeks under greenhouse or field conditions in (Libya) either sand or soil salinized with varying applications of NaCl. Irrigation with NaCl significantly decreased plant survival and growth and concentration of Ca, Na, K, Fe and P ions in plants with 0.5 M or 1.0 M NaCl. The experiments were repeated using a naturally occurring soil amendment called ‘Zander’. Seeds of A. saligna were grown for seven weeks in both greenhouse and field trials as before but with the addition of Zander and NaCl. Zander improved plant survival and growth with salinity and increased the elements in plants (Ca, Na, K, Fe and P). Field experiments were conducted to assess the effects of saline irrigation with 1.0 M NaCl and extra water added to 0% or 10% Zander on survival and growth, consequently, seedling growth significantly decreased with increase in soil salinity. Survival and growth increased with increase in extra water. The additional irrigation water caused an increase in the uptake of Ca++ and increased the Ca++/Na+ and K+/Na+ ratio. Zander did not appear to reduce net uptake of Na+ and its transport to shoot tissues. Mg++, P, K+ and Ca++ content significantly decreased in plants in response to salinity. Possible mechanisms to avoid Na+ toxicity in A. saligna in response to salinity included increasing the supply of Ca++. Extra Ca++ applied into the medium with and without salt increased survival and growth even in the absence of Zander. Calcium increased uptake of Ca++ and increased Ca++/Na+ and K+/Na+ ratio.

The influence of chemical seed treatment on germination, seedling survival and yield of canola

Thesis (MScAgric)--University of Stellenbosch, 2004. / ENGLISH ABSTRACT: The influence of chemical seed treatments on the germination, seedling survival and yield of canola (cv. Varola 44) was investigated in a series of incubation studies, glasshouse experiments, as well as field trials in the canola producing areas in the Western Cape Province. Incubation experiments were conducted to compare germination and seedling growth of untreated (control) seed with that of seed treated at different application rates (0.5, 1.0 and 2.0 times the recommended) of Cruiser® and SA-combination (which consists of Thiulin® at 0.5g a.i.; Apron® at 0.0815 g a.i.; Gaucho® at 0.6125 g a.i. and Rovral® at 0.9975 g a.i.). The results indicated that seed treatment (all rates of SA-combination and highest rate of Cruiser) delayed germination and seedling growth, especially if the seed was subjected to the Accelerated Ageing Test. Glasshouse studies with pasteurised soil at different water contents, seed sources (storage periods) and planting depths confirmed the phytotoxic effects of the chemical seed treatments in the absence of soil borne pathogens. From the results it became clear that extreme water conditions (very wet or dry) increased the suppressing effect on germination and seedling growth, but that no phytotoxic reactions occurred in moist (favourable soil water conditions) soil, regardless of application rate of the chemicals used, planting depth and seed source. In a second glasshouse experiment conducted in moist soil (kept at 50% of field water capacity to prevent any toxic effects) from seven different localities that were naturally infested with pathogens, both chemicals proved to be effective where soil borne pathogens (Rhizocfonia so/ani and Pythium spp.) occurred. No clear trend could however be found due to either chemical or application rates used. Finally, field trials were conducted to study the effect of chemical seed treatments on the plant populations and yield of canola planted in different row widths (17 and 34 cm) and seeding rates (3, 5 and 7 kq.ha'). Results showed that treated seeds produced more plants.rn" and yielded more than untreated seeds at Roodebloem Experimental Farm, while the highest seeding rate produced significantly more plants.rn" (Roodebloem and Langgewens Experimental Farms), but not significantly higher yields than the lowest seeding rate at the same locality. Although row width did not have an effect on plant population, yield (Roodebloem 2003) was significantly less at the wider (34 cm) rows. As in earlier experiments, no consistent differences between the two chemicals used were found. These results clearly illustrated both the negative (in the absence of pathogens) and positive (where soil borne pathogens do occur) effects that chemical seed treatments may have on the germination, seedling growth and even yield of canola under local environmental and soil conditions. Because no significant differences were found between the chemicals used, both chemicals should be regarded as efficient. More research, especially under field conditions and with more cultivars, is needed before the registration of a chemical for seed treatment could be considered. / AFRIKAANSE OPSOMMING: Die invloed van chemiese saadbehandeling op die ontkieming, saailing oorlewing en opbrengs van canola (cv. Varola 44) is ondersoek in 'n reeks inkubasie studies, glashuis eksperimente en veldproewe in die canolaproduserende gebiede in die Wes Kaap Provinsie. Inkubasie eksperimente is uitgevoer om die ontkieming en saailing groei van onbehandelde (kontrole) saad te vergelyk met dié van saad wat behandel is teen verskillende dosisse (0.5, 1.0 en 2.0 keer die aanbevole) van Cruiser® en SA-kombinasie (wat bestaan uit Thiulin® teen 0.5g a.i.; Apron® teen 0.0815 g a.i.; Gaucho® teen 0.6125 g a.i. en Rovral® teen 0.9975 g a.i.). Establishment of Guayule (Parthenium argentatum Gray)

Thesis (MScAgric)--University of Stellenbosch, 2002. / ENGLISH ABSTRACT: Guayule (Parthenium argentatum Gray) is a semi-desert plant with the potential to become an established crop on arid land in South Africa. The plant produces latex, which can be processed into rubber that is useful in application where disease transmission needs to be limited, such as for surgical gloves and condoms. The poor germination and natural dormancy characteristics of the embryo and the seed coats of guayule seed, motivated germination experiments. Germination of seed treated with solutions of gibberellic acid, smoke water and smoke watergibberellic acid was determined. Furthermore, combinations of gibberellic acid, smoke water and sodium hypochlorite treatment solutions were applied to seed to determine the germination responses. Vegetative propagation of guayule by means of cuttings was also investigated to determine the rooting responses of cuttings with treatment solutions of indole butyric acid, naphthalene acetamide and naphthalene acetic acid. Rooting percentage, root length and root weight was determined for each treatment. Dryland field trial plantings were established at different areas in South Africa to determine the growth potential and biomass production of guayule cultivars under different environmental conditions. Stand count, height, canopy diameter and stem diameter was determined for the different cultivars and areas. Lastly, latex production of guayule cultivars established in trial plots at Elsenburg, Oudtshoorn and Graaff- Reinet was determined after one year of growth. Treatment solutions of an aqueous smoke extract (commonly referred to as smoke water) and gibberellic acid were evaluated to determine their effectiveness in stimulating germination of four guayule seed lines (AZlOl, AZ-3, N565 and 11591). The split-plot analyses of variance showed no significant interaction between cultivar and treatment factors (P = 0.71), but when the day factor was included interaction was highly significant (P < 0.0001). The applied treatment thus had an effect on the time required for the germination response. Investigations into optimum germination responses indicated that smoke water-gibberellic acid required the shortest number of days (6.3 days) for optimum germination to occur with cultivar AZ-3. Furthermore, gibberellic acid treatment resulted in the greatest germination with the four cultivars 11591, AZ- 3, AZI0l and N565, at 93.78%, 93.35%, 94.41% and 99.42% respectively. These results show that guayule seed can be stimulated to germinate by treatment with gibberellic acid and smoke water solutions. Specific concentrations of treatment solutions of gibberellic acid, smoke water and sodium hypochlorite, and combinations thereof were used to evaluate the germination response of guayule seed cultivar AZ-2. Combinations of treatment solutions did not result in significantly increased seed germination responses. Single treatment solutions of gibberellic acid and smoke water did not significantly enhance germination, but sodium hypochlorite however, significantly (p <0.0001) suppressed germination at the 1% Cl and 2% Cl concentrations with about 5% and 10% respectively when compared to the control. Therefore, the applied seed treatments did not effectively increase the germination of guayule cultivar AZ-2 seed. Specific concentrations of indole butyric acid, naphthalene acetamide and naphthalene acetic acid treatment solutions were applied to guayule cuttings of cultivar AZ-3 and rooting response was determined for rooting percentage, root length and root weight. Naphthalene acetic acid treatment rooted the highest percentage of cuttings (52.38%) at a concentration of 60 mgll. Indole butyric acid treatment produced the longest roots (147.83 mm) at a concentration of 120 mg/l. Naphthalene acetamide obtained the heaviest roots (1.8 g) at a concentration of 120 mgll. Treatment solutions of indole butyric acid, naphthalene acetamide and naphthalene acetic acid indicated specific concentrations for optimum effect to improve root formation (by 30%), root length (by 50 mm) and root weight (by 1.5 g) when compared to the controls. Guayule trial plots of IOx10 m, rows 1 m apart and 30 cm between plants, and each cultivar (10 plants per unit) placed at random and replicated 6 times, were established in different areas under different environmental conditions in South Africa. Plantings were evaluated as a dryland practice, though irrigation was supplied only for establishment. Growth (stand count, height, canopy diameter, stem diameter) and biomass (wet and dry weight) were recorded for (1) oneyear old plantings established in April 2001 at Elsenburg, Graaff-Reinet and Oudtshoorn, and (2) six-month old plantings established in October 2001 at Bethulie, Glen and Upington. Analysis of variance was done to determine mean growth and biomass for the different areas and cultivars. (1) There were significant interactions between the factors area and cultivar for stand count and height, while canopy diameter and stem diameter differences were significant only within factors. The greatest growth potential was produced by cultivars AZ-2 and AZ-3, and Oudtshoorn was the best area for growth potential and biomass production. (2) Interaction between area and cultivar was significant for plant height, but were not significant for stand count, canopy diameter and stem diameter. Cultivars produced similar results for biomass production, but were significantly different in the different areas of Bethulie, Glen and Upington. Growth potential and biomass production of guayule was influenced by the availability of water during the growth of the plant. Latex production of guayule cultivars (AZ-2, AZ-3, N565, 11591) established in trial plots at different areas (Elsenburg, Oudtshoorn, Graaff-Reinet) in South Africa was investigated. Branch samples of one-year old plantings were harvested in April 2002, dipped in 1% ascorbate, sealed in plastic bags and chilled during airfreight to the United States Department of Agriculture (USDA) - Agricultural Research Service (ARS) in Albany CA. Latex extraction and quantification was done and mean latex production and comparisons of latex production for the cultivars in each area were determined. The evaluation of latex production show generally similar results in the different areas. Cultivars generally do not differ significantly from each other in the amount of latex produced in each area. Environmental stress factors on latex production occur in especially Oudtshoorn and Graaff-Reinet where the temperatures are above 25°C and below 1DoC. Since guayule is a slow growing shrub, latex accumulation is also slow and takes 4-6 years to reach economic harvesting potential. Production results are therefore preliminary and require further evaluation after each year of growth to present a complete view of guayule latex production over time. Propagation investigations were successful in identifying techniques to germinate guayule seed and promote rooting of cuttings with specialized treatment solutions. Field establishment of guayule under South African environmental conditions has identified suitable areas and indicated cultivar performances in these areas. Evaluation of the latex production of field plantings has demonstrated the potential of guayule in these areas. Currently the path to guayule development is paved with a network of research activities that is strengthened through cooperation between research institutions and private sector companies that bridge the gap between academic research and market exhibition. / AFRIKAANSE OPSOMMING: geen opsomming

Improved potato (Solanum tuberosum) seed production through aeroponics system

The potential of this crop is reflected in the large increase in area of production where Africa showed the highest rate of growth within the developing world over the past twenty years. The multiplication rate of potatoes is very low compared to other crops. Therefore it is essential to investigate methods of increasing the number of minitubers produced from disease free in-vitro plantlets. There is a number of potato propagation procedures that are currently being used worldwide to multiply seed potatoes. As aeroponics is still a relatively new technique that has not been researched extensively for the production of minitubers, a study relating to the production practises including the nutritional requirements of potato minitubers produced in a aeroponic system were undertaken. Potato plantlets, cv BP1, were grown aeroponically at two different densities (20 and 30 plants/m2) and four harvest intervals (7, 10, 14 and 18 days). The interaction between harvesting intervals and plant densities did not influence plant growth, minituber quality or yield. Best results were realised when harvesting every 7 days with a higher total tuber number over the growing period. Harvest interval also influenced the phosphorus and copper concentration in minitubers. To study the effect of Calcium (Ca) application rate, potato plantlets of cultivars Up-to-date, Mnandi, Buffelspoort and BP1 were grown at four different Ca levels (8.40, 6.75, 5.10 and 3.45 meq/L). The interaction between Ca application levels and cultivars significantly influenced the percentage stolon branching. BP1 had more stolons at the lowest Ca application level and Buffelspoort had more stolons at the full Ca application levels. However, low Ca treatments produced the highest yield. The minituber number and weight harvested were three times more for Mnandi. An aeroponic study on the irrigation frequency (20, 30, 40 and 50 minutes interval) was conducted on four potato cultivars (Up-to-date, Mnandi, Buffelsoort and BP1). Significant differences were noted in the interaction between irrigation frequencies and cultivars for the percentage tuberised plants and stolon and tuber dry mass. When irrigated every 40 minutes, 48% of the Buffelspoort plants produced tubers. Plant height was also significantly affected by the interaction between irrigation frequencies and potato cultivars, with Mnandi producing taller plants when irrigated every 30 minutes. Total tuber number and tuber fresh and dry weight was higher at the irrigation frequency of 20 minutes. The interaction between irrigation frequencies and cultivars on the response to macro and trace elements was not significant for sodium and iron but was for phosphorus, potassium, calcium, zinc and aluminium. A field study was conducted in a greenhouse where potato seed of BP1 obtained from the first trial were graded into different sizes (Small: >20, medium: 20–40 and large: > 40 mm of diameter) and stored at 3 different temperatures (3, 16 and 25oC) for 2 supplementary months before being planted. Sprouting capacity was mostly influenced by temperature regardless of other factors applied to potato seed minitubers such as harvest intervals and sizes. The higher storage temperature of 25oC resulted in tubers with a higher number of sprouts, longer sprouts and with a higher sprouting capacity. / AFRIKAANSE OPSOMMING: Aartappels is een van die belangrikste voedselgewasse in baie lande in Afrika. Die interaksie tussen Ca toedienings peile en kultivars het ‘n beduidende effek gehad op die persentasie stolon vertakking. BP1 het meer stolons gehad by die laagste Ca toedieningspeil en Buffelspoort het meer stolons gehad by die volle Ca toedieningspeil. Die hoogste opbrengste is egter waargeneem by die laagste Ca toedieningspeil. Die aantal miniknolle en oes massa was drie keer meer vir Mnandi. ‘n Aeroponiese studie op die besproeiingsfrekwensie (20, 30, 40 en 50 minuut intervalle) is gedoen met vier aartappel kultivars (Up-to-date, Mnandi, Buffelsoort en BP1). Beduidende verskille is opgemerk in die interaksie tussen besproeiings frekwensie en kultivars vir die persentasie plante met knolle en stolon en knol droë massa. Met besproeiings elke 40 minute het 48% van die Buffelspoort plante knolle produseer. Plant hoogte is ook beduidend beïnvloed deur die interaksie tussen besproeiingsfrekwensie en aartappel kultivar met Mnandi plante wat hoër was wanneer dit elke 30 minute besproei is. Die totale aantal knolle en knol vars- en droë massa was hoër wanneer daar elke 20 minute besproei is. Die interaksie tussen besproeiings frekwensie en kultivars op die makro- en mikro element inhoud van die knolle was nie beduidend vir natrium en yster nie, maar wel vir fosfaat, kalium, kalsium, sink en aluminium. ‘n Potproef is gedoen in ‘n kweekhuis waar aartappel saad van BP1 verkry vanaf die eerste proef nadat knolle verdeel is in verskillende grootte klasse (klein: < 20mm, medium: 20-40mm en groot: >40mm) en gestoor is by drie verskillende temperature (3, 16 en 25oC) vir 2 addisionele maande voor plant. Spruit ontwikkelings kapasiteit was meestal beïnvloed deur temperatuur ten spyte van ander behandelings soos oes intervalle en knol grootte. Die hoër bergings temperatuur 25oC het aanleiding gegee tot knolle met ‘n hoër aantal spruite, langer spruite en ‘n hoër spruit ontwikkelings kapasiteit.

Ecologization of agriculture and forestry directly and indirectly influences the environment, water, air, and landscape. Thus, analysis of organic forestry activities, evaluation and forecast are of scientific, ecological and political significance. Weed control and the resulting organic farming and forestry issues have attracted greater attention to the use of physical methods in weed control, because this weed control method is effective and does not leave chemical residues neither in the soil, nor in water. One of the physical methods is thermal weed control method, which contrary to the other weed control methods, leaves only dead biomass, preventing self-sowing of weed seeds in the soil, destroys some of the pests and disinfects the surface layer of soil (Upadhyaya et al, 2007). Meanwhile, as an alternative to seed treatment, various physical methods are also used (Lynikienė, 2006). However, the effect of some of the techniques on seed-born fungi are not explained, optimal exposures of the effects are unidentified (Clear et al., 2002). One of the possibilities to reduce the infestation of seeds by fungi - their thermal destruction. / Žemės ir miškų ūkio ekologizavimas tiesiogiai ir netiesiogiai veikia aplinką, vandenį, orą, landšaftą. Todėl ekologinės veiklos miškų ūkyje analizė, įvertinimas ir prognozė turi mokslinę, ekologinę ir politinę reikšmę (Diepenbrock, 1996). Piktžolių naikinimas ir ekologinės žemės bei miškų ūkių problemos paskatino atkreipti dėmesį į fizikinių metodų naudojimo piktžolių kontrolei svarbą, nes šis piktžolių naikinimo metodas yra efektyvus, nepaliekantis cheminių medžiagų likučių nei dirvožemyje, nei vandenyje. Vienas iš fizikinių metodų ir yra terminis piktžolių naikinimo būdas, kuris, skirtingai nuo kitų piktžolių kontrolės būdų, palieka tik negyvą jų biomasę, neleisdamas piktžolių sėkloms pasisėti dirvoje, sunaikina kai kuriuos kenkėjus ir dezinfekuoja paviršinį dirvožemio sluoksnį (Upadhyaya, 2012). Kaip alternatyva sėkloms beicuoti taip pat naudojami įvairūs fizikiniai metodai (Lynikienė, 2006). Kai kurių metodų poveikis sėklomis plintantiems mikromicetams nenustatytas, nenustatytos ir šių poveikių optimalios ekspozicijos (Clear et al., 2002). Viena iš galimybių sumažinti sėklų užterštumą mikromicetais – jų naikinimas termiškai.

Some invetsigations on the responses to desiccation and exposure to cryogenic temperatures of embryonic axes of Landolphia kirkii.

Landolphia kirkii is scrambling shrub forming an integral part of the flora along the coastal areas of north-eastern South Africa. The non-sustainable harvesting of fruit as food source, by monkeys and rural communities and the highly recalcitrant nature of their seeds threatens the continuation of the species. In addition, the ability of the plants to produce high quality rubber makes its long-term conservation highly desirable. Previously, attempts have been made to cryopreserve germplasm of L. kirkii, but no survival had been recorded at cryogenic temperatures of below -140ºC. The present study reports on the effects of rapid dehydration, chemical cryoprotectants and various cooling rates, thawing and imbibition treatments on survival of embryonic axes excised with cotyledons completely removed, as well as with 3 mm portion of each cotyledon attached, from fresh, mature, recalcitrant seeds of L. kirkii. Survival was assessed by the ability for both root and shoot development in in vitro culture, the tetrazolium test and electrolyte leakage readings. At seed shedding, embryonic axes were at the high mean water content of 2.24 g gˉ¹ (dry mass basis). All axes (with and without attached cotyledonary segments) withstood rapid (flash) drying to a water content of c. 0.28 g gˉ¹; however, the use of chemical cryoprotectants, singly or in combination, before flash-drying was lethal. Rapid cooling rates were detrimental to axes flash-dried to 0.28 g gˉ¹, with no explants showing shoot production after exposure to -196ºC and -210ºC. Ultrastructural examination revealed that decompartmentation and loss of cellular integrity were associated with viability loss after rapid cooling to cryogenic temperatures, although lipid bodies retained their morphology regardless of the thawing temperature employed. Furthermore, analysis of the lipid composition within embryos of L. kirkii revealed negligible amounts of capric and lauric acids, suggested to be the medium-chained saturated fatty acids responsible for triacylglycerol crystallisation when lipid-rich seeds are subjected to cryogenic temperatures. Hence, lipid crystallisation was not implicated in cell death following dehydration, exposure to cryogenic temperatures and subsequent thawing and rehydration. Rapid rehydration of embryonic axes of L. kirkii by direct immersion in a calcium-magnesium solution at 25ºC for 30 min (as apposed to slow rehydration on moistened filter paper or with rehydration in water) was associated with highest survival post-dehydration. Cooling at 1ºC minˉ¹ and 2ºC minˉ¹ facilitated survival of 70 and 75% respectively of axes with attached cotyledonary segments at 0.28 g gˉ¹ after exposure to - 70ºC. Viability retention of 40 and 45% were recorded when embryonic axes with attached cotyledonary segments were cooled at 14 and 15ºC minˉ¹ to temperatures below -180ºC. However, no axes excised without attached cotyledonary segments produced shoots after cryogenic exposure. The use of slow cooling rates is promising for cryopreservation of mature axes of L. kirkii, but only when excised with a portion of each cotyledon left attached. / Thesis (M.Sc.)-University of KwaZulu-Natal, Westville, 2011.
