Avaliação in vitro da permeabilidade cutânea da rutina em emulsões cosméticas / In vitro evaluation of rutin cutaneous permeability from cosmetic emulsions

A rutina é empregada como antioxidante e na prevenção da fragilidade capilar. Pode ser veiculada em emulsões tópicas adequadas para atingir o local de ação. Estudos de penetração in vitro através da pele humana seria a situação ideal, entretanto, há dificuldades de sua obtenção e manutenção de sua viabilidade. Entre os demais modelos de membrana, a muda de pele de cobra apresenta-se como estrato córneo puro, fornecendo barreira similar ao humano e é obtida sem a morte do animal. Os objetivos desta pesquisa foram: (1) desenvolver e avaliar a estabilidade de emulsões cosméticas, contendo rutina e promotores de penetração cutânea, tais como, uréia (U), isopropanol (IP) e propilenoglicol (PG); (2) avaliar a liberação da referida substância ativa das emulsões e; (3) avaliar a penetração e a retenção cutânea in vitro da rutina da formulação de melhor desempenho. Emulsões foram desenvolvidas com rutina a 5,0% p/p e U, IP e PG, associados ou não e em proporções distintas, segundo planejamento fatorial com dois níveis com ponto central. Quantificou-se a rutina das emulsões por espectrofotometria a 361,0 nm, método previamente validado. A liberação da rutina nas formulações foi realizada em células de difusão vertical com membrana de acetato de celulose e água destilada e álcool etílico absoluto 99,5% (1:1), como fluido receptor. O experimento foi conduzido em um período de seis horas, a 37,0 ±. 0,5 °C e agitação constante de 300 rpm.>f. emulsão de melhor desempenho quanto à liberação foi estudada quanto à estabilidade (Testes de Estabilidade Acelerada). Para o estudo de penetração e retenção cutânea da rutina dessa formulação foi utilizada muda de pele de cobra de Crotalus durissus. Empregou-se o método espectrofotométrico validado a 410,0 nm para a quantificação da rutina após liberação, penetração e retenção cutânea. Todas as emulsões foram consideradas adequadas após desenvolvimento das formulações. A uréia (isolada e em associação com IP e PG) e o isopropanol (isolado e em associação com PG) influenciaram negativamente a liberação da rutina das emulsões em diversos parâmetros. A rutina liberada e acumulada da formulação contendo PG a 5,0% p/p possuiu valor de 648,80 ±. 53,01 &#181g/cm2. Fora do esperado, a preparação contendo o número maior de promotores (U 5,0% p/p, IP 5,0% p/p e PG 5,0% p/p) resultou em liberação de menor magnitude igual a 419,76 ±. 17,98 &#181g/cm2. A presença do PG apresentou-se mais eficiente na liberação da rutina, mas não na sua penetração através da muda de pele de C. durissus, retendo 0,931 ± 0,0391 µg de rutina/mg de muda de pele de cobra. Nas condições de armazenamento a 25,0 ±2,0 °C; 5,0 ±0,5 °C e 45,0 ±. 0,5 °C, a emulsão com PG e rutina apresentou-se quimicamente estável durante 30 dias. De acordo com os resultados, a emulsão contendo PG apresentou liberação mais expressiva da rutina, no entanto, não ocorreu a penetração cutânea, mas apenas sua retenção no estrato córneo de C. durissus. A preparação manteve-se estável em todas as condições de armazenamento. / Rutin is employed as antioxidant and to prevent the capillary fragility and, when incorporated in cosmetic emulsions, it must target the action site. In vitro cutaneous penetration studies through human skin is the ideal situation, however, there are difficulties to obtain and to maintain this tissue viability. Among the membrane models, the shed snake skin presents itself as pure stratum corneum, providing barrier function similar to human and it is obtained without the animal sacrifica. The objectives of this research were: (1) development and stability evaluation of cosmetic emulsions containing rutin and penetration enhancers, as urea (U), isopropanol (IP) and propylene glycol (PG); (2) release evaluation of the mentioned active substance from the emulsions and; (3) evaluation of rutin in vitro cutaneous penetration and retention from the emulsion of the best performance. Emulsions were developed with rutin 5.0% w/wand U, IP and PG, associated or not according to factorial design with two levels and central point. Active substance on the formulations was quantified by a validated spectrophotometric method at 361.0 nm. Rutin release from emulsions was performed in vertical diffusion cells with cellulose acetate membrane and distilled water and ethanol 99.5% (1:1), as receptor fluid. The experiment was conducted for six hours, at 37.0 ± 0.5 °c with constant stirring of 300 rpm. Formulation with best profile of rutin release had its stability studied by the Accelerated Stability Assays. Rutin cutaneous penetration and retention from the mentioned emulsion was performed with shed snake skin from Crotalus durissus. Spectrophotometry at 410.0 nm, previously validated, determined the active substance after release and cutaneous penetration/retention. Ali emulsions were considered apparently stables after development. Urea (isolated and associated with IP and PG) and isopropanol (isolated and associated with PG) have influenced negatively the rutin release in several parameters. Emulsion with PG 5.0% w/w presented rutin released and accumulated equal to 648.80 ± 53.01 µg/cm2. Unexpectedly, the formulation containing all enhancers (U 5.0% w/w, IP 5.0% w/w and PG 5.0% w/w) has decreased the amount released of the active substance (419.76 ± 17.98 µg/cm2). Emulsion with PG presented more adequate for rutin release, but PG did not provide rutin cutaneous penetration through C. durissus skin, retaining 0.931 ± 0.0391 &$181;g rutin/mg shed snake skin. The referred formulation was chemically stable for 30 days after they have been stored at 25.0 ± 2.0 °c, 5.0 ± 0.5 °c and 45.0 ± 0.5 °C. In conclusion, emulsion with PG provided rutin release more expressively, although, it has not been verified the active cutaneous penetration, but only its retention on the Crotalus durissus stratum corneum. Formulation was stable in all storage conditions.

The aim of this master thesis is to design and check a bearing structure of The multifunctional center in Jihlava. A bulding is partly ground flooring. The structure follows an L-curve in a plan with the biggest axial lenght of 40 m. A roofing is in a shed shape and the pitch of the roof is 5,73°. The highest point of the bearnig structure is 11,5 m. The structure has various frame systems, which is a truss, a gross-cross section beam with an inner column in the middle of a span and a gross-cross section beam without an inner column. The structure is designed mainly from timber members, a glulam and solid timber. A steel part of structure consists of the metal fasteners and rods.

In the 2nd floor is an apartment with terrace and facilities for the officers of club. The house is made from brick technology of ceramic blocks. The building is covered with shed roof and vegetation.

This thesis describes the design of a project Family Winery VILARO in Velké Bílovice. It consists of two objects. The first is a guesthouse with a restaurant and the second is a production building with a tasting room. The guesthouse is submitted in the stage of building design. Production facility, a winery, is discussed further as the implementation project. The winery is located in a slightly sloped terrain. It has one partially cellared ground floor. The roof is atypical, one part is sloped triple-coated shed roof, second part is flat with stabilizing layer. The support systém mainly consists of brick blocks, ferroconcrete walls, ferroconcrete boards, ferroconcrete beam ceiling and prestressed concrete ceiling panels Spiroll. The project assumes using of typical building materials.

Basic of this Diploma thesis is Civil construction in Kostelec nad Orlici, which is in a part of city, where is planning off-grid community housing. Part of off-grid community housing is considered a partially sistainable civil construction, serving both for cultural use through the club, for commercial purposes such as various salons (hairdresser, massage or shop), as well as for sports activities with the possibility of small refreshments, such as bouldering, climbing wall and exercise hall. Four sustainable (off-grid) familly houses, which aren´t the subject of this thesis, are considered. Object SO01 – Civil construction is designed as a partially basement, two floors. The shape of the object is designed as several blocks with different height levels. From a material point of view it is a wooden building from the system of large-format laminated wood panels (CLT). The basement is designed with a waterproof reinforced concrete so-called white tub and prefabricated ceiling panels SPIROLL. A facade is made of wooden cladding, cement-fiber boards or thin-layer silicone plaster. A roof of the object is partially flat (vegetation and walkable) and a shed roof with a slope of 5°. A dance hall and the rest of the club + exit is located in the basement. A sanitary facilities, a technical room and a main entry with reception is also located in the basement. Right in the middle of the building is a climbing wall that runs across all floors. To the right of the climbing wall is a shop, exercise hall and staircase. To the left side are a sanitary facilities, a boulder (low climbing wall) and separate staircase for a office space on the 2nd floor. Behind the climbing wall is a bistro with entry for a terrace and entry for the club. There are some establishments like a hairdresser, a cosmetic salon, a tattoo salon and a massage salon on the 2nd floor. There are an entrance to a terrace above the bistro and a staircase to an attic and to second terrace as well on the 2nd floo

Detached houses are made of load bearing timber frame with cast-in-situ hempcrete, building of facilities is made of reinforced concrete with precasted blocks. All buildings are cellarless. The subject of this thesis was to develop project documentation for building construction. It is a new four-storey house with restaurant, situated on the outskirts of the village Bridges at Jablunkova. The building is brick, has a two-level gable and shed roof, consisting of wood roof system. Slope of the parcel on which the house is located, allows the building of the plant into the ground. Because of this it is possible to enter it, either the main entrance to the first floor or building around and over the summer terrace come directly to the restaurant on the second floor. Above it are two more floors for hotel guests only, a total of 10 accommodation units with its own bathroom.

The topic of the masters thesis is newly built sports center in Broumov on the outskirts of the historic center. The proposal was placed high demands on appearance. There is a non-traditional stone facade with a combination of wood ventilated facades. The combination of wood and stone construction gives a historic feel and fit and existing buildings, which will carry the dominant built-up area. The new building is set into the sloping terrain with a flair for existing buildings. The building is part sunk into the ground to reduce the height to the surrounding area. The sports center is divided into three dilatations objects: Technical facilities, sports hall, bowling with squash and gym.

This diploma thesis deals with the design of a hotel with a restaurant at the base the highest peak of the White Carpathians, Veľká Javorina. The area is located near the White Carpathians Protected Landscape Area and the Cetuna Natural Monument. The location has the possibility of using year-long and seasonal activities such as downhill and cross - country skiing or ice skating on frozen ponds, cycling in the new built-up cycle paths, hiking or walking on educational trails, horse riding, swimming in nature or visiting a family mini-zoo with the possibility of personal contact with animals. The potential of the concerned area is still used mainly by domestic, persons with the possibility of accommodation by acquaintances or owners of seasonal cottages and houses due to insufficient short-term accommodation. It was the main imulse for the design of accommodation. A piece of land on the border of the cadastral area of Dolné Bzince and Hrubá Strana of the village of Bzince pod Javorinou was selected for the building construction, due to its satisfactory position, orientation and also for the possibility of its evaluation. The building is designed as a separate-standing object onto an undeveloped territory of the municipality, which was used for agricultural purposes. It's been a long time it lies on ice, decays and its value decreases with expanding overgrown trees. The land is slightly sloping, oriented to southwest. The building has three main entrances and secondary entrances for smooth operation of the hotel. There is a day (leisure) part placed on the 1st floor, also a part for accommodation employees and immobile clients, parts for hotel management, cleaning zone and restaurant. On the 2nd floor there is an accommodation part with 19 rooms and a library. Into basement is a sport and relaxation part, which consist of fitness center and wellness center.
