There was then necessary to investigate the pharmacokinetic parameters of the drug prototype as a requirement to pre-clinical knowledge of it. This study aimed to obtain the pharmacokinetic profile of ruthenium (II) / tryptophan administered to rats intraperitoneally in a single dose by quantifying this substance in plasma using validated analytical methodology in UV-VIS spectrophotometer. The methodology consisted of the administration of the prototype rutrpII to 3 rats at a dose of 6 mg/kg, intraperitoneally. Samples of blood 1.0 ml were collected by cannulation of the left jugular vein with heparinized syringe, the intervals from 0 to 9 hr. After centrifugation the plasma was frozen at -20 until the time of analysis. The bioanalytical method to quantify rutrpII was developed and validated in UV-VIS at a wavelength of 417 nm. From the construction of the concentration versus time curve, the kinetic parameters were calculated (Software WinNonlin 5.0 (Pharsight ™) Results were: Tmax = 8 h, Cmax = 169.86 mg/mL, t1/2 = 1.04 ± 0.02 h, ClT/F = 1.32 ± 0.05 mL/min/kg, Vd/F = 1,98 ± 0,05 L/kg. The developed and validated bioanalytical method was suitable for the detection and quantification of RutrpII in rat plasma. The values of pharmacokinetic profiles found, and extrapolated on allometric scaling allow us to understand that the compound studied showed a slow tissue distribution profile and / or was eliminated slowly (Vd low and low value CLT), due to a possible affinity between RutrpII and plasma proteins. This affinity may be explained by the similarity of their chemical structure with iron, enabling it to be transported by biomolecules such as transferrin, albumin or any other, and having as a target the DNA of cancer cells. In general, the compounds of ruthenium (II) / amino acids may be promising drugs for the amino acids present in the complex, may facilitate the recognition of the complex as a whole by DNA, and thus less toxic. / O complexo de rutênio(II)/aminoácido (rutrpII) demonstrou alta atividade antineoplásica contra carcinoma de mama murino (tumor de Ehrlich) in vitro e in vivo, sendo que neste último, aumentou a média de sobrevida dos animais. Fez-se necessária então a investigação dos parâmetros farmacocinéticos deste protótipo de fármaco como requisito ao conhecimento pré-clínico do mesmo. O objetivo deste trabalho foi obter o perfil farmacocinético do rutênio(II)/triptofano administrado a ratos, via intraperitoneal, em dose única por meio da quantificação desta substância em plasma utilizando metodologia analítica validada em espectrofotômetro UV-VIS. A metodologia consistiu na administração do protótipo rutrpII a 3 ratos em dose de 6 mg/kg, por via intraperitonal. Foram coletadas amostras de 1,0 mL de sangue, por canulação da veia jugular esquerda, com seringa heparinizada, nos intervalos de tempo de 0 a 9 h. Após centrifugação, o plasma foi congelado a -20ºC até o momento da análise. O método bioanalítico de quantificação do rutrpII foi desenvolvido e validado em espectrofotômetro UV-VIS no comprimento de onda de 417 nm. A partir da construção da curva de concentração versus tempo, foram calculados os parâmetros cinéticos (Software Winnonlin 5.0 (Pharsight™)). Os resultados encontrados foram: Tmax= 8h, Cmax= 169,86 µg/mL, t(1/2) =1,04 ± 0,02 h,ClT/F = 1,32 ± 0,05 mL/min/kg, Vd/F = 1,98 ± 0,05 L/kg. O método bioanalítico desenvolvido e validado foi adequado para a detecção e quantificação do RutrpII em plasma de rato. Os valores dos perfis farmacocinéticos encontrados, e extrapolados em escala alométrica, permitem entender que o composto estudado apresentou um perfil lento de distribuição tecidual e/ou foi eliminado lentamente (baixo Vd e baixo valor de CLT), devido a uma possível afinidade entre o RutrpII e as proteínas plasmáticas. Esta afinidade pode ser explicada pela semelhança de sua estrutura química com o ferro, possibilitando que ele seja transportado por biomoléculas como a transferrina, albumina ou alguma outra, e tendo como alvo, o DNA da células cancerosas. De maneira geral os compostos de rutênio(II)/ aminoácidos podem ser fármacos promissores pois os aminoácidos presentes nos complexos, podem facilitar o reconhecimento do complexo como um todo pelo DNA, sendo assim menos tóxicos.

Optical Characterization and Energy Simulation of Glazing for High-Performance Windows / Optisk karakterisering och energisimulering av smarta fönster

This thesis focuses on one important component of the energy system - the window. Windows are installed in buildings mainly to create visual contact with the surroundings and to let in daylight, and should also be heat and sound insulating. This thesis covers four important aspects of windows: antireflection and switchable coatings, energy simulations and optical measurements. Energy simulations have been used to compare different windows and also to estimate the performance of smart or switchable windows, whose transmittance can be regulated. The results from this thesis show the potential of the emerging technology of smart windows, not only from a daylight and an energy perspective, but also for comfort and well-being. The importance of a well functioning control system for such windows, is pointed out. To fulfill all requirements of modern windows, they often have two or more panes. Each glass surface leads to reflection of light and therefore less daylight is transmitted. It is therefore of interest to find ways to increase the transmittance. In this thesis antireflection coatings, similar to those found on eye-glasses and LCD screens, have been investigated. For large area applications such as windows, it is necessary to use techniques which can easily be adapted to large scale manufacturing at low cost. Such a technique is dip-coating in a sol-gel of porous silica. Antireflection coatings have been deposited on glass and plastic materials to study both visual and energy performance and it has been shown that antireflection coatings increase the transmittance of windows without negatively affecting the thermal insulation and the energy efficiency. Optical measurements are important for quantifying product properties for comparisons and evaluations. It is important that new measurement routines are simple and applicable to standard commercial instruments. Different systematic error sources for optical measurements of patterned light diffusing samples using spectrophotometers with integrating spheres have been investigated and some suggestions are made for how to avoid such errors.

Monitoramento da coluna total de ozônio e a ocorrência de eventos de influência do buraco de ozônio antártico sobre o sul do Brasil / Monitoring of total ozone column and the occurrence of the influence of antarctic ozone hole events over southern Brazil

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES / The present PhD thesis analyzes the results obtained from the Total Ozone Column (TOC) monitoring conducted in the Southern Space Observatory - SSO (29.26 ° S, 53.48 °W and 488 m altitude) between 1992 and 2014 by three successive Brewer spectrophotometers (# 081, # 056 and # 167). First, the Brewer measurements were compared with TOMS (Total Ozone Mapping Spectrometer) and OMI (Ozone Monitoring Instrument) satellites obtaining excellent agreement. In addition, was determined that the seasonal TOC variability is dominated by the annual cycle, with a minimum of ~ 260 DU in April and a maximum of ~ 295 DU in September. The Quasi-Biennial Oscillation (QBO) is the main mode of interannual variability being approximately in antiphase with the QBO index. Next, 58 events of the influence of the Antarctic Ozone Hole on the SSO station was identificate in the period between 2005 and 2014. This events occurred on average 5.8 ± 3.51 times per year, with a mean reduction of TOC by Brewer' of -7.04 ± 2.97% and by OMI of -7.66 ± 3.11 respectively. Analyzing the ozone profiles from AURA/MLS (Microwave Limb Sounder) satellite, the average isentropic level for maximum reduction is 644.68 ± 158.59 K, with a mean reduction of 15.39 ± 6.47%, being October the month of greatest occurrence (18 events). The events were separated into three categories: TOC intensity reduction, height of the ozone reduction lamina in AURA/MLS satellite and the dynamic characteristic of Polar Filament or Polar Tongue through the application of Dybal (Dynamical Barrier Localization) code in the potential vorticity (PV) fields from MIMOSA (Modélisation Isentrope du transport Mésoéchelle de l'Ozone Stratosphérique par Advection) model. The events with a dynamic feature of Polar Tongue (20,68%) occurred more frequently in October, with medium intensity and in the medium stratosphere, while events of dynamic characteristic of Polar Filament (79,31%) occurred more frequently in September, also with medium intensity and in the medium stratosphere. / A presente Tese de Doutorado analisa os resultados obtidos a partir do monitoramento da Coluna Total de Ozônio (CTO) realizado no Observatório Espacial do Sul – OES (29,26 ° S, 53,48 ° e 488 m de altitude) entre 1992 e 2014 através de três sucessivos espectrofotômetros Brewer (# 081, # 056 e # 167). Primeiramente, as medidas dos Brewers foram comparadas com observações dos instrumentos de satélite TOMS (Total Ozone Mapping Spectrometer) e OMI (Ozone Monitoring Instrument), obtendo elevados valores de R2 (0,88 e 0,93, respectivamente). Além disso, foi determinado que a variabilidade sazonal da CTO é dominada pelo ciclo anual, com um mínimo de ~260 DU em abril e um máximo de ~295 DU em setembro. A Oscilação Quasi-Bienal (QBO) é o principal modo de variabilidade interanual estando aproximadamente em antifase com o índice QBO. Em seguida é apresentada a identificação da ocorrência de 58 eventos de Influência do Buraco de Ozônio Antártico sobre a estação do OES no período entre 2005 e 2014, os quais ocorreram em média 5,8 ± 3,5 vezes por ano, com redução média da CTO de Brewer e OMI de -7,04±2,9% e 7,66±3,1 respectivamente. Analisando os perfis de ozônio do satélite AURA/MLS (Microwave Limb Sounder), observa-se que o nível isentrópico médio para a máxima redução é de 644,6±158,5 K com redução média de 15,3±6,4%, sendo outubro o mês de maior ocorrência (18 eventos). Os eventos foram separados em categorias: Intensidade da redução da CTO, altura da camada de redução no perfil do ozônio do satélite AURA/MLS e a característica dinâmica de Filamento Polar ou Língua Polar através da aplicação do código Dybal (Dynamical Barrier Localisation) nos campos de vorticidade potencial (PV) do modelo MIMOSA (Modélisation Isentrope du transport Mésoéchelle de l'Ozone Stratosphérique par Advection). Os eventos com característica dinâmica de Lingua Polar (20,68%) ocorreram com mais frequência no mês de outubro, com intensidade média e na média estratosfera, enquanto que eventos de característica dinâmica de Filamento Polar (79,31%) ocorreram com mais frequência no mês de setembro, também com media intensidade e na média estratosfera.

Para a fabricação de filtros ópticos com característica espectral crítica, utiliza-se a tecnologia de filmes finos multicamadas, a qual exige o monitoramento e controle de espessura dos filmes com alta precisão. O sistema tradicional de monitoramento de espessura de filmes, baseado em cristal ressonante, não atende aos requisitos de precisão e repetibilidade do processo exigidos para a fabricação deste tipo de filtro. Já o monitoramento óptico, por sua vez, é um sistema que atende aos requisitos de fabricação destes filtros. Neste trabalho apresenta-se o desenvolvimento de um sistema automático de monitoramento óptico banda larga, para fabricação de filtros com filmes finos multicamadas, que se baseia na utilização de equipamentos ópticos de uso geral disponíveis no mercado. Para realizar análises de desempenho referentes exclusivamente ao algoritmo de automatização, um ambiente de simulação foi preparado, no qual os dados fornecidos pelo equipamento de aquisição de características ópticas durante a deposição efetiva foram substituídos por dados gerados por um algoritmo de simulação de crescimento do filme. Deste modo, o algoritmo de automatização e seus sistemas de segurança puderam ser exercitados. As simulações mostraram que o sistema proposto possui potencial para tornar mais preciso e repetitivo o processo de fabricação de filtros, de modo que atendam aos requisitos das aplicações especiais. / Optical filter manufacturing with critical spectral characteristics uses multilayer thin film technology, which demandas film thickness monitoring and control with high precision. Traditional monitoring systems based on oscillating crystal do not fulfill requirements of accuracy and process repeatability for manufacturing such type of filter. On the other hand, optical broadband monitoring system fulfills the manufacturing requirements to produce these filters. This work presents the development of a broadband optical monitoring system for multilayer thin film filters production, based on general optical equipment commercially available. In order to make the performance analysis related to automation algorithm, a simulation environment has been prepared, where the data supplied by optical data acquisition equipment has been replaced by simulating signals of film growing. Thus the automation algorithm and security systems could be worked. Simulations showed that the system has potential to improve the accuracy and repeatability of the manufacturing process so that the produced filters are able to fulfill the requirements of special applications.

Shear controlled orientation effects with injection mouldings produced by the SCORIM process

Injection moulding using the process of Shear Controlled Orientation Injection Moulding (SCORIM) to enhance the aesthetic characteristics of plastics was investigated. Unsightly surface weld lines were successfully removed from highly reflective aluminium flake pigmented plastics by the application of a single macroscopic SCORIM shear when used in series with Bright Surface Moulding (BSM). A gonio spectrophotometer (GSP) was used for the quantitative characterisation of the Al flake pigmented mouldings as a measure of surface reflectivity and preferred angle of reflection. The different directional properties of surface reflectivities to either side of a conventional weld line are unacceptable, but were successfully reoriented approximately uniformly with the use of SCORIM and BSM moulding (i. e. SBM) used in series. SBM therefore provided an acceptable quality of surface finish for mouldings originally containing a weld line, without deterioration of mechanical properties. Indeed, some improvements in mechanical properties were observed. Translucent two-colour mouldings were used to successfully demonstrate the flow paths taken by sheared material during the application of macroscopic shears. The use of intermittent shearing to encapsulate shear oriented material in the solidifying layers, manifested original and profound aesthetic effects. This resulted from mixing the two colours and was reproducible and widely variable. The morphology of isotactic polypropylene (iPP) processed in this way and examined by light and electron microscopy revealed how only one or two intermittent shears were required to orient a large volume of the moulding in the shear direction. Moreover, U-shaped flow paths demonstrated that the easiest shear route was close to the mouldings edges, an observation supported by x-ray analysis. The addition of Al flake pigment was found to act as a heterogeneous nucleant for ß-spherulites. This acted as a suitable marker for the clear identification of the displaced weld interface using polarised light microscopy, of filled and unfilled iPP. y-phase was identified with the use of only one or two intermittent shears which reflects an increase in molecular alignment and consequent improved mechanical properties. The intensity of the y-phase increased with the volume of material sheared. Strong evidence was also obtained of a linear relationship between the logarithm of the time lapse between two intermittent shears and the corresponding values of a-phase index, crystallinity index and percentage crystallinity. The values of each increasing proportionally with the length of time used. Microhardness characterisation revealed anisotropy within SCORIM samples consistent with preferred orientation and increased modulus in the shear direction. The skin layers were characterised as the softest region through the thickness of SCORIM mouldings. The results of this work were used to provide the basis of a computer simulation of the SCORIM process under development at the University of Wales Swansea.

Alfredo Volpi (1896 - 1988) é um dos mais importantes pintores brasileiros contemporâneos. Sua extensa obra foi marcada, principalmente, por composições cromáticas e abstrações geométricas. Foi um artista que dava importância muito grande à técnica e aos materiais, tendo desenvolvido uma maneira própria e sistemática de trabalhar: confeccionava suas próprias telas, tintas e até pigmentos, com terras naturais. Analisou-se ao todo 11 pinturas a têmpera em diversos suportes (tela, aglomerado de madeira e mural), 10 delas realizadas na década de 1950, período em que a obra do artista passou por uma grande transformação, do figurativismo ao abstracionismo e do uso de tintas industriais à base de óleo para a técnica medieval da têmpera. As características de opacidade, saturação cromática e presença material do pigmento sobre a superfície da tela, inerentes a esta técnica pictórica, tornam-se elementos tão importantes em sua obra, quanto o uso da cor na formação do espaço. Foram realizados estudos de cor e composição química dos materiais e levantamentos de informações sobre seus procedimentos técnico-artísticos a partir de métodos analíticos complementares não destrutivos e pesquisa em diversas fontes, pois entende-se que estes elementos são chaves importantes para o entendimento mais profundo de sua obra. O espectrofotômetro foi utilizado para medição da cor com base em curvas espectrais e coordenadas numéricas de referência dentro de um espaço de cor uniforme. Técnicas de imageamento por luz visível, fluorescência visível por radiação ultravioleta e reflectografia de infravermelho próximo foram utilizadas para levantar informações sobre o processo criativo do artista, detectar locais de intervenções de restauro e observar o grau de fluorescência de cada área cromática das pinturas como auxiliar na identificação de pigmentos. A técnica de EDXRF (Espectroscopia de Fluorescência de Raios X por Energia Dispersiva) foi importante para caracterização química dos materiais. O MEV-EDS (Microscópio Eletrônico de Varredura com Espectroscopia por Dispersão de Energia de Raios X) forneceu informações sobre composição química e imagens da superfície dos materiais presentes nas pinturas murais. A partir da integração destas informações pôde-se identificar uma parte da rica paleta deste pintor, documentar as cores das pinturas no presente, identificar ou dar indicações da composição dos pigmentos e da base de preparação das pinturas e iluminar partes da história de cada uma das obras, como presença de esboços feitos à grafite, pinturas anteriores encobertas, pinceladas e maneiras de misturar a tinta e documentar seu estado atual de conservação. As ferramentas utilizadas se mostraram úteis na caracterização das pinturas, revelaram informações inéditas sobre a obra deste artista e levantaram novas indagações. As informações obtidas podem ser o início de um banco de dados sobre sua obra. / Painter Alfredo Volpi (1896 - 1988) is one of the most important contemporary Brazilian artists. His extensive work, mostly characterised by chromatic compositions and geometric abstractions, is technically unique. The artist attributed great importance to craft and materials, developing his own systematic way of working. Among the paintings analysed were 11 temperasin various supports (canvas, hardboard panel and mortar). Of the selection, 10 paintings were made around 1950, when the artist\'s work underwent a major transformation, from figuration to abstraction, from the use of industrial oil-based paints to the medieval technique of tempera. This technique, known for its opacity, colour saturation and material presence of pigments on the canvas,became as important as the colour itself in the shaping of the artist\'s new spatial relation. It is understood that these elements are important keys to a deeper understanding of his work, both technically and artistically. The present investigation was carried out making use of non-destructive complementary analytical techniques. The aim was to study the chromatic and chemical composition characteristics of his paintings and gather information on the technical procedures the artist may have used. The spectrophotometer was used to measure the colour based on the spectral curves and numerical reference coordinates within a uniform colour space. Techniques of visual examination with visible light, visible fluorescence by ultraviolet radiation and near-infrared reflectography were used to gather information about the artist\'s creative process. These techniques also helped detect restoration patches and identify pigments. The EDXRF (Energy Dispersive X-Ray Fluorescence) technique was important for chemical characterization of materials, while the SEM-EDS (Scanning Electron Microscope with Energy Dispersive X-Ray Spectroscopy) provided surface images and information on the chemical composition of the material present in the murals. From the integration of these techniques it was possible to identify a part of the rich palette of Volpi´s paintings; to document the present aspects of the colours; to recognize or give indications of pigment and ground compositions and to shed light on parts of each of the works background, such as the presence of graphite sketches, covered earlier paintings and brushstrokes as well as to document the current condition of the works under study. The tools used have proven useful in characterizing the paintings, revealing new information about the work of the artist and raising new questions. The information obtained may be the beginning of a database on his oeuvre.

O MEV-EDS (Microscópio Eletrônico de Varredura com Espectroscopia por Dispersão de Energia de Raios X) forneceu informações sobre composição química e imagens da superfície dos materiais presentes nas pinturas murais. A partir da integração destas informações pôde-se identificar uma parte da rica paleta deste pintor, documentar as cores das pinturas no presente, identificar ou dar indicações da composição dos pigmentos e da base de preparação das pinturas e iluminar partes da história de cada uma das obras, como presença de esboços feitos à grafite, pinturas anteriores encobertas, pinceladas e maneiras de misturar a tinta e documentar seu estado atual de conservação. As ferramentas utilizadas se mostraram úteis na caracterização das pinturas, revelaram informações inéditas sobre a obra deste artista e levantaram novas indagações. As informações obtidas podem ser o início de um banco de dados sobre sua obra. / Painter Alfredo Volpi (1896 - 1988) is one of the most important contemporary Brazilian artists. His extensive work, mostly characterised by chromatic compositions and geometric abstractions, is technically unique. The artist attributed great importance to craft and materials, developing his own systematic way of working. Among the paintings analysed were 11 temperasin various supports (canvas, hardboard panel and mortar). Of the selection, 10 paintings were made around 1950, when the artist\'s work underwent a major transformation, from figuration to abstraction, from the use of industrial oil-based paints to the medieval technique of tempera. This technique, known for its opacity, colour saturation and material presence of pigments on the canvas,became as important as the colour itself in the shaping of the artist\'s new spatial relation. It is understood that these elements are important keys to a deeper understanding of his work, both technically and artistically. The present investigation was carried out making use of non-destructive complementary analytical techniques. The aim was to study the chromatic and chemical composition characteristics of his paintings and gather information on the technical procedures the artist may have used. The spectrophotometer was used to measure the colour based on the spectral curves and numerical reference coordinates within a uniform colour space. Techniques of visual examination with visible light, visible fluorescence by ultraviolet radiation and near-infrared reflectography were used to gather information about the artist\'s creative process. These techniques also helped detect restoration patches and identify pigments. The EDXRF (Energy Dispersive X-Ray Fluorescence) technique was important for chemical characterization of materials, while the SEM-EDS (Scanning Electron Microscope with Energy Dispersive X-Ray Spectroscopy) provided surface images and information on the chemical composition of the material present in the murals. From the integration of these techniques it was possible to identify a part of the rich palette of Volpi´s paintings; to document the present aspects of the colours; to recognize or give indications of pigment and ground compositions and to shed light on parts of each of the works background, such as the presence of graphite sketches, covered earlier paintings and brushstrokes as well as to document the current condition of the works under study. The tools used have proven useful in characterizing the paintings, revealing new information about the work of the artist and raising new questions. The information obtained may be the beginning of a database on his oeuvre.
