The Amburana cearensis (AllemÃo) A.C. Smith known as cumaru is a species widely used for restoration of the landscape. The aim of this study was to investigate the germination and seedling growth of cumaru depending on the weight of the seeds under conditions of light, temperature, drought stress, as well as to determine the seed imbibition curve, internal morphology, chemical composition and quantification of the coumarin. Initially seeds were individually weighed and separated into three weight classes (light, medium and heavy), these being submitted to the determination of water content, thousand seeds weight and germination tests in two light conditions (presence and absence ) and six schemes temperatures (20, 25, 30, 35, 40, and 20-30 ÂC). In addition to these tests, it was determined the imbibition curve of each weight class and made assessment of drought stress tolerance under different potentials(-0.2, -0.4, -0.6, -0.8 and -1,0 MPa). The experiments were arranged in a completely randomized design in four replications for each treatment. For the internal morphology of seeds were used in cytochemistry usual techniques for the identification of the main structures and substances reserves. To visualize the coumarin in the seeds by NMR spectroscopy was used. The optimum conditions for seed germination occurred at 30 ÂC, which are insensitive to light and vigor seeds for light and medium. The best conditions for seedling growth occurred with the light and medium seeds at 25 and 30 ÂC and in the presence of light, with temperatures of 35 and 40 ÂC harmful. Light and medium seeds showed the same pattern of water absorption, while not reach the heavy phase III of the curve. The decrease in the water potential of the substrate affect the germination and growth of seedlings from seed medium and heavy compared to the light, and from -0.6 MPa in a condition strictly limiting seedling development. The chemical constituents present in the seeds of A. cearensis are quantitatively different depending on the weight of the seeds, and the heavy characterized by a high content of lipids. The NMR spectroscopy falls greater proportion of coumarin in seed extract heavy and medium, possibly affecting ecophysiological needs of the species A. cearensis. / Diante das necessidades de recuperaÃÃo de Ãreas degradadas à crescente o interesse na propagaÃÃo de espÃcies florestais nativas, o que demanda informaÃÃes bÃsicas sobre as suas caracterÃsticas germinativas e ecofisiolÃgicas. A Amburana cearensis (AllemÃo) A.C. Smith conhecida como cumaru à uma espÃcie bastante utilizada para recomposiÃÃo da paisagem. O objetivo deste trabalho foi estudar a germinaÃÃo e o crescimento de plÃntulas de cumaru em funÃÃo do peso das sementes sob diferentes condiÃÃes de temperatura, luz, estresse hÃdrico, bem como determinar nas sementes a curva de embebiÃÃo, morfologia interna, composiÃÃo quÃmica e identificaÃÃo da cumarina. Inicialmente as sementes foram pesadas individualmente e separadas em trÃs classes de peso (leves, mÃdias e pesadas), sendo estas, submetidas à determinaÃÃo dos teores de Ãgua, peso de mil de sementes e a testes de germinaÃÃo em duas condiÃÃes de luz (presenÃa e ausÃncia) e seis regimes de temperatura (20, 25, 30, 35, 40 e 20-30ÂC). AlÃm destes ensaios, foi determinado a curva de embebiÃÃo de cada classe de peso e avaliaÃÃo da tolerÃncia ao estresse hÃdrico sob diferentes potenciais (-0,2, -0,4, -0,6, -0,8 e -1,0 MPa). Os experimentos foram dispostos em delineamento inteiramente casualizado com quatro repetiÃÃes para cada tratamento. Para a morfologia interna das sementes foram utilizadas tÃcnicas usuais em citoquÃmica para a identificaÃÃo das principais estruturas e substÃncias de reservas. Para a visualizaÃÃo da cumarina nas sementes foi utilizada a espectroscopia por RMN. As condiÃÃes Ãtimas para a germinaÃÃo das sementes ocorreu na temperatura de 30ÂC, sendo estas insensÃveis à luz e um maior vigor para sementes leves e mÃdias. As melhores condiÃÃes para o crescimento das plÃntulas ocorreram com as sementes leves e mÃdias nas temperaturas de 25 e 30ÂC e na presenÃa de luz, sendo as temperaturas de 35 e 40 ÂC prejudiciais. Sementes leves e mÃdias apresentam o mesmo padrÃo de absorÃÃo de Ãgua, enquanto as pesadas nÃo atingem a fase III da curva. O decrÃscimo do potencial hÃdrico do substrato prejudica a germinaÃÃo e o crescimento de plÃntulas oriundas de sementes mÃdias e pesadas em comparaÃÃo Ãs leves, sendo a partir de -0,6 MPa uma condiÃÃo estritamente limitante na formaÃÃo de plÃntulas. Os constituintes quÃmicos presentes nas sementes de A. cearensis sÃo quantitativamente diferenciados em funÃÃo do peso das sementes, sendo as pesadas caracterizadas por um elevado teor de lipÃdios. A espectroscopia por RMN releva uma maior proporÃÃo de cumarina no extrato de sementes pesadas e mÃdias, que possivelmente afetam as necessidades ecofisiolÃgicas da espÃcie A. cearensis.

Nuclear magnetic resonance is a non-invasive way to observe material properties on a molecular level. Magnetic resonance imaging is an important diagnostic tool in medicine. Molecules of several metabolites in muscle tissue show similar interactions as molecules partially oriented in orienting media. These interactions could provide new information about processes in vivo, this can serve for diagnostics of metabolism. New insight into the function is gained by observation of metabolites in orienting media. Observable anisotropic interactions in muscle tissue in vivo could be used for diagnostic purposes. Anisotropic NMR interaction of solvent as a new method for observation of phase transition of hydrogel with temperature change or change of solvent composition. Use of magnetic resonance imaging in slices to observe the collapse of polyacrylamide in water-acetone mixtures. Use of diffusion-weighted magnetic resonance imaging to observe phase transition of PNIPAM-based semi-interpenetrating polymer.

Structure, Flexibility, And Overall Motion Of Transmembrane Peptides Studied By NMR Spectroscopy And Molecular Dynamics Simulations

Nuclear magnetic resonance (NMR) spectroscopy was used to determine the structure of transmembrane (TM) segment IX of the Na+/H+ exchanger isoform 1 (NHE1) in dodecylphosphocholine micelles. Studying isolated TM segments in this fashion constitutes a well-established "divide and conquer" approach to the study of membrane proteins, which are often extremely difficult to produce, purify, and reconstitute in full-length polytopic form. A similar approach was combined with NMR spin relaxation experiments to determine the peptide backbone flexibility of NHE1 TM VII. The combined NMR structural and dynamics studies are consistent with an important role for TM segment flexibility in the function of NHE1, a protein involved in apoptosis and myocardial disease. The study of the rhomboid protease system is also described from two perspectives: 1) I attempted to produce several TM constructs of the substrate spitz or a related construct and the production and purification are described in detail; and 2) I present coarse-grained molecular dynamics simulation results for the E. coli rhomboid ecGlpG and a spitz TM construct. Spitz appears to preferentially associate with rhomboid near TMs 1 and 3 rather than the proposed substrate gate at TM 5. The two proteins primarily interact at the termini of helices rather than within the hydrocarbon core of the bilayer. Finally, I present a detailed analysis of coarse-grained molecular dynamics simulations of the fibroblast growth factor receptor 3 TM domain dimerization. Specifically, algorithms are described for analyzing critical features of wild-type and G380R mutant constructs. The G380R mutation is the cause of achondroplasia, the most common form of human dwarfism. The results suggest that the proximity of a residue to the dimer interface may impact the severity of the mutant phenotype. Strikingly, heterodimer and mutant homodimer constructs exhibit a secondary dimer interface which may explain the increased signaling activity previously reported for the G380R mutation--the helices may rotate with the introduction of G380R. The unifying theme of this work is the 'study of membrane proteins' using complementary techniques from structural biology and computational biochemistry.

Engineering the phase behaviour of high performance inkjet colorants

Dyes for inkjet printing are typically of the chromonic type. Chromonic mesophases have gained considerable attention as a well-defined group of lyotropic mesogens with different properties from conventional amphiphiles. While extensive research has been dedicated to the field of surfactant liquid crystals, structural and aggregation studies of chromonics have only emerged as a topic of interest within the last few years. The liquid crystalline structures in aqueous solutions of commercial Cu - phthalocyanine and black dyes have been examined using a combination of optical microscopy, UV-vis spectroscopy, nuclear magnetic resonance, wide- and small-angle X-ray diffraction and electronic paramagnetic resonance with a view to examining the phase behaviour of the chromonic mesophases formed over a broad range of concentrations and temperatures. These studies were performed in order to resolve outstanding problems concerning structural properties of these systems. Optical microscopy allowed us to identify the liquid crystalline phases and to construct the phase diagram. The observations show that both of these dyes form nematic mesophase above 15% wt / wt% dye. The small-angle diffraction data confirmed that the nematic phase for the black dye is maintained throughout the 16-25% composition range. A further increase in concentration leads to the formation of the hexagonal phase. The Cu – phthalocyanine dye also formed a nematic phase at low concentrations, with the aggregates undergoing a phase transition to an orientationally ordered chromonic liquid crystal phase at high dye concentration. These studies showed that this ordered phase possessed hexagonal symmetry. The wide-angle X-ray results demonstrated that aggregation involved π-π stacking of the molecules into columns. An additional reflection at ca. 6.8Å was observed for the black dye, which is believed to arise from “head – to – tail” packing of the molecules within the aggregates (a similar phenomenon observed in other azo dyes, e.g. Edicol Sunset Yellow).The densities of both dyes were measured over the studied range of concentrations. This enabled us to calculate the parameters of the aggregates within the hexagonal mesophase. A comparison between the area of the molecule and the cross-section of the aggregates showed that the aggregates of both dyes were the unimolecular stacks.

O metaboloma é definido como a coleção qualitativa e quantitativa de todos os metabólitos de baixa massa molecular presentes nas células, os quais participam de reações bioquímicas necessárias para a manutenção, crescimento e fisiologia celular. A avaliação metabolômica permite o delineamento do processo bioquímico de sistemas a fim de ampliar o entendimento de como as patologias se manifestam. A Espectroscopia de Ressonância Magnética Nuclear (RMN) é usada para investigar uma variedade de processos biológicos em diversos sistemas. A magnética aplicada a células e biópsias de tecido intacto de melanoma murino B16F10 contribuiu para a caracterização bioquímica de biomarcadores das diferentes fases de progressão tumoral do melanoma murino B16F10. Os resultados obtidos neste estudo permitiram a identificação de 33 metabólitos possíveis, envolvidos na carga lipídica e no metabolismo secundário da via glicolítica, que favorecem o crescimento e a progressão tumoral. A presença da taurina, prolina, serina, fenilalanina, que aumentaram quantitativamente, são possíveis marcadores da invasão, progressão e metastatização. A análise quantitativa desses metabólitos mostrou diferença significativa em 11 compostos, dos quais 9 estão diretamente envolvidos na expressão das respostas proliferativas, de morte celular e angiogênese, nos diferentes períodos de crescimento do melanoma B16F10, avaliados neste estudo. Desta forma, os achados obtidos neste estudo, quando associados no futuro a outros fatores, poderão ser úteis no diagnóstico e auxiliar na escolha terapêutica alvo com maior especificidade e menores efeitos colaterais. RMN pode ter um importante impacto na monitorização de metabólitos em células e tecidos tumorais, possibilitando a detecção mais precoce de tumores malignos, em suma, através da combinação de métodos de ressonância magnética. / The metabolome is defined as the qualitative and quantitative collection of all low weight molecular metabolites in cells that participate in biochemical reactions necessary for the maintenance, growth and physiology of cell. The metabolomic evaluation allows the design of systems of biochemical process in order to broaden the understanding of how diseases manifest. The Nuclear Magnetic Resonance Spectroscopy (NMR) is used for investigating a variety of biological processes in several systems. The magnetics applied to cell and tissue biopsies intact murine melanoma B16F10 contributed to the biochemical characterization of biomarkers of different stages of tumor progression of murine melanoma B16F10. The results obtained in this study allowed the identification of 33 potential metabolites involved in lipid content in the secondary metabolism of the glycolytic pathway, which promote growth and tumor progression. The presence of taurine, proline, serine, phenylalanine, which quantitatively increased, is possibly markers of invasion and metastasis progression. Quantitative analysis of these metabolites showed a significant difference in 11 compounds, of which 9 are directly involved in the expression of proliferative responses, cell death and angiogenesis in different periods of growth of B16F10 melanoma, evaluated in this study. Thus, the findings from this study, when associated in the future with other factors, may be useful in the diagnosis and may assist in choosing therapeutic target with greater specificity and fewer side effects. NMR may have a significant impact on monitoring metabolites in tumor cells and tissues, allowing for earlier detection of malignant tumors; in short, by combining MRI methods.

Molecular Level Characterization and Mobility of Radionuclide-Carrying Natural Organic Matter in Aquatic Environments

Radionuclides, 129I and 239,240Pu, are major products or by-products of nuclear fission and among the top risk drivers for waste disposal at the Savannah River Sites (SRS) and Rocky Flats Environmental Technology Sites (RFETS), respectively, due to their perceived mobility in the environment, excessive inventory, toxicity, and long half-life. The objective of this study is to investigate the role of natural organic matter in retarding or facilitating the migration of 129I and 239,240Pu in the Department of Energy (DOE) sites. Measurements of 127I and 129I in humic acids (HAs) and fulvic acids (FAs) obtained by five successive alkaline, two glycerol and one citric acid-alkaline extractions, demonstrated that these extractable humic substances (HS) together account for 54-56 percent and 46 percent of the total 127I and 129I in the soil, respectively. The variations among 127I and 129I concentrations, isotopic ratios (129I/127I), chemical properties of all these humic substances indicated iodine was bound to a small-size aromatic subunit (~10 kDa), while the large-size subunit (~90 kDa), which likely linked the small-size unit through some weak chemical forces, determined the relative mobility of iodine bound to organic matter. Soil resuspension experiments simulating surface runoff or stormflow and erosion events were conducted with soils collected from SRS. Results showed that 72-77 percent of the newly-introduced I- or IO3- were irreversibly sequestered into the organic-rich soil, while the rest was transformed into colloidal and dissolved organo-iodine by the soil. The resulting iodine remobilization contradicts the conventional view that considers only I- or IO3- as the mobile forms. Quantitative structure analysis by 13C DPMAS NMR and solution state 1H NMR on these humic substances indicate that iodine is closely related to the aromatic regions containing esterified products of phenolic and fomic acid or other aliphatic carboxylic acids, amide functionalities, quinone-like structure activated by electron-donating groups (e.g., NH2) or hemicelluloses-lignin-like complex with phenyl-glycosidic linkage. The micro-molecular environment, such as the hydrophobic aliphatic periphery hindering the active aromatic cores and the hydrophilic polysaccharides favoring its accessibility towards hydrophilic iodine species, play another key role in the interactions between iodine and SOM. NMR spectra of the colloidal organic Pu carrier which can potentially be released from the soil during the surface runoff or stormflow showed Pu was transported, at sub-pM concentrations, by a cutin-derived soil degradation products containing siderophore-like moieties and virtually all mobile Pu.

A análise quantitativa desses metabólitos mostrou diferença significativa em 11 compostos, dos quais 9 estão diretamente envolvidos na expressão das respostas proliferativas, de morte celular e angiogênese, nos diferentes períodos de crescimento do melanoma B16F10, avaliados neste estudo. Desta forma, os achados obtidos neste estudo, quando associados no futuro a outros fatores, poderão ser úteis no diagnóstico e auxiliar na escolha terapêutica alvo com maior especificidade e menores efeitos colaterais. RMN pode ter um importante impacto na monitorização de metabólitos em células e tecidos tumorais, possibilitando a detecção mais precoce de tumores malignos, em suma, através da combinação de métodos de ressonância magnética. / The metabolome is defined as the qualitative and quantitative collection of all low weight molecular metabolites in cells that participate in biochemical reactions necessary for the maintenance, growth and physiology of cell. The metabolomic evaluation allows the design of systems of biochemical process in order to broaden the understanding of how diseases manifest. The Nuclear Magnetic Resonance Spectroscopy (NMR) is used for investigating a variety of biological processes in several systems. The magnetics applied to cell and tissue biopsies intact murine melanoma B16F10 contributed to the biochemical characterization of biomarkers of different stages of tumor progression of murine melanoma B16F10. The results obtained in this study allowed the identification of 33 potential metabolites involved in lipid content in the secondary metabolism of the glycolytic pathway, which promote growth and tumor progression. The presence of taurine, proline, serine, phenylalanine, which quantitatively increased, is possibly markers of invasion and metastasis progression. Quantitative analysis of these metabolites showed a significant difference in 11 compounds, of which 9 are directly involved in the expression of proliferative responses, cell death and angiogenesis in different periods of growth of B16F10 melanoma, evaluated in this study. Thus, the findings from this study, when associated in the future with other factors, may be useful in the diagnosis and may assist in choosing therapeutic target with greater specificity and fewer side effects. NMR may have a significant impact on monitoring metabolites in tumor cells and tissues, allowing for earlier detection of malignant tumors; in short, by combining MRI methods.
