Systémové píky v elektromigračních systémech s komplexujícími činidly / System peaks in elektromigration systems with complexing agents

Capillary zone electrophoresis (CZE) is a widely used analytical method. CZE is described theoretically very well and there are many simulation programs, which enable one to predict results of electrophoretic separations, and alternatively to study phenomena taking place during the electrophoretic separation in detail. The CZE method is not only an analytical method, but is often used for determination of many physical parameters of compounds, such as stability constants or complex mobilities. Among methods most often used for determination of complexation parameters belongs the affinity capillary electrophoresis (ACE). Its alternative is the vacancy affinity capillary electrophoresis (VACE). Whereas by the ACE method the stability constant is determined from the dependence of the analyte effiective electrophoretic mobility on the background electrolyte (BGE) composition, in the case of the VACE system peaks are used for this purpose. In this work the legitimacy of using system peaks in the VACE method for determination of stability constants was investigated. Several approaches dealing with the concentrating of complexing agent in the peak area were compared, both for the ACE and the VACE method. Este trabalho inicia com um estudo monográfico do comportamento químico do íon tiocianato, do ácido isotiociânico e uma rápida revisão da literatura sobre a reação uranilo/tiocianato. Foi feita uma rápida apreciação sobre os métodos de cálculos de constante de estabilidade e uso de métodos indiretos. A estabilidade dos complexos de tiocianato de mercúrio (II) foi estimada através de medidas potenciométricas com a finalidade de verificar a ordem de variação de seu ñ, dentro de larga faixa de concentração de tiocianato, com opropósito de utilizar este sistema como indicador de ligante livre em estudos de equilíbrios. A formação do ácido isotiociânico foi estudada por um método indireto, utilizando Hg(SCN)x2-x/Hg como sistema indicador para [SCN-]. Este estudo foi feito a 25°C e em força iônica 2,0 M e 4,0 M, obtendo-se: βHNCS = 0,135 ± 0,0070 (em força iônica 2,0 M). βHNCS = 0,168 ± 0,0069 (em força iônica 4,0 M). sendo: (ver fórmula no PDF). Estes valores de constante foram obtidos através de aproximações sucessivas, envolvendo cálculos iterativos das concentrações livres das espécies reagentes seguindo do tratamento da bem conhecida função Fo (X). Nas medidas potenciométricas também foi estimado ΔEj causado pela presença de [H+], com base na variação de ESO\' de um eletrodo de Cl-, AgCl/Ag, trabalhando-se em condições semelhantes. O sistema uranilo/tiocianato foi também estudado potenciometricamente e da mesma maneira, em meio fortemente ácido, onde a concentração de HNCS formado deve ser considerada. Foi determinada a formação de cinco espécies sucessivas: β1 = 11,4 ± 0,06 K1 = 11,4 β2 = 49,0 ± 0,82 K2 = 4,30 β3 = 88,4 ± 3,25 K3 = 1,80 β4 = 127,4 ± 4,42 K4 = 1,44 β5 = 91,4 ± 1,82 K5 = 0,72. Evidências da existência da sexta espécie foram encontradas, mas seu valor apresentou elevada incerteza. Foram feitas medidas espectrofotométricas em comprimentos de onda entre 280 e 300 nm para o urânio em meio de concentrações crescentes de tiocianato. O sistema apresenta forte absorção, com absortividades molares médias maiores que 7700 1.mol.cm-1, o que é de interesse analítico. A combinação das constantes de formação com os dados espectrofotométricos levou às absortividades molares individuais dos complexos. / This thesis start with a monographic study on the chemical behaviour as thiocyanate ion, the iso-thiocyanic acid and a revision on the uranyl-thiocyanate reaction. A short and critical comment on methods on computation of stability constants and use of indirect methods was included. The stability of mercury(II)/thiocyanate complexes was estimated by potentiometric measurements in order to check the range of its ñ variations, at a wide thiocyanate ion concentration, in order to use this sistem as indicator of the free ligand concentrations in equilíbria studies. The formation of iso-thiocyanic acid wasstudied by an indirect potentiometric method by using Hg(SCN)x2-x/Hg as an indicator system for [SCN-]. The system present strong absorption, with an average molar absorbtivity higher than 7.700 1.mol.cm-1, which is interesting for analytical purposes. The combination of the formation constants with the spectrophotometric data lead to individual molar absortivities of the complexes.

Determinação das constantes de estabilidade dos complexos formados entre os aminoácidos cisteína, N-acetilcisteína e lisina com chumbo em solução aquosa / Determination of stability constants for lead complexes with cysteine, N-acetylcysteine and lysine in aqueous solutions

Conselho Nacional de Desenvolvimento Científico e Tecnológico / In this study we investigated the interaction between lead and the amino acid cysteine, lysine and N-acetylcysteine by testing long-term contact with ion exchanger resins and also the determination of stability constants. The importance of investigating the presence of lead in solutions for parenteral nutrition is related to the risk of internal contamination of these solutions due to the affinity between amino acids that compose these solutions and lead present in plastic packaging. The amino acids according to their relative affinities can act as carriers of metal in living organisms. The long term tests with adsorbents in form of Pb2+ showed that the higher extraction of lead occurs at amino acid that has a greater affinity for the metal and this is evidenced by the value of stability constant calculated, i.e., there was a direct relationship of the values of the calculated stability constants. Kinetic effects were not significant in this study, since the trials were of long duration. Tests with complexing agents such as EDTA and citric acid, that have known stability constants, were also performed for comparison purposes. The quantification of lead in long term tests was performed by atomic absorption spectrometry with flame and electrothermal atomization. The stability constants were obtained by potentiometry by processing the data through the software BEST®. Titrations in aqueous solutions with constant ionic strength and temperature showed that the predominant species in solution have molar ratio of 1:1 (L: M) to N-acetyl-cysteine and lysine and 1:1 and 2:1 for cysteine. The main species found for cysteine were Cys2Pb and CysPbOH with log K = 1.61 and 24.00, respectively, for N-acetylcysteine N-Acetyl-Cys2Pb, N-Acetyl-Cys2PbH and N-Acetyl-CysPbOH with log K =-1.55, 14.52 and 21.49 respectively, and for lysine LysPb with log K = 9.69. The interaction of amino acids studied with lead was also detected by the tests made with the raw material of the packaging of parenteral nutrition solutions through the analysis by AAS. / Neste trabalho investigou-se a interação entre chumbo e os aminoácidos cisteína, N-acetilcisteína e lisina por meio de ensaios de contato de longa duração com resina de troca iônica e estudos para determinar as constantes de estabilidade dos complexos formados entre eles. A importância de investigar a presença de chumbo em soluções de nutrição parenteral relaciona-se ao risco de contaminação interna destas soluções em decorrência da afinidade entre os aminoácidos que compõem as soluções e o chumbo presente nas embalagens plásticas. Os aminoácidos em função de suas afinidades relativas podem atuar como transportadores do metal nos organismos vivos. Os resultados dos ensaios de contato com resina na forma Pb2+ mostraram que a maior extração do chumbo ocorre pelo aminoácido que possui maior afinidade pelo metal e isto é comprovado pelo valor da constante de estabilidade calculada, ou seja, houve relação direta dos valores das constantes obtidas através do cálculo das mesmas para os aminoácidos estudados com a taxa de extração do chumbo da resina pelos ligantes (aminoácidos). O efeito cinético, estabelecido pelas leis de velocidades não importaram nesse estudo, uma vez que os ensaios foram de longa duração. Ensaios com agentes complexantes, como EDTA e ácido cítrico, com constantes de estabilidade já conhecidas, também foram realizados para fins comparativos. A quantificação de chumbo nos ensaios de longa duração foi realizada por espectrometria de absorção atômica (AAS) com atomizações por chama e eletrotérmica. As constantes de estabilidade foram obtidas por potenciometria com o tratamento dos dados através do programa computacional BEST®. Titulações potenciométricas, de soluções aquosas com força iônica e temperatura constantes, mostraram que as espécies predominantes em solução são de razão molar 1:1 (L:M) para N-acetilcisteína e lisina e de 1:1 e 2:1 para cisteína. As principais espécies encontradas foram para cisteína, Cys2Pb e CysPbOH com log K = 1,61 e 24,00 respectivamente, para N-acetilcisteína N-Acetil-Cys2Pb, N-Acetil-Cys2PbH e N-Acetil-CysPbOH, com log K = -1,55; 14,52 e 21,49 respectivamente, e para lisina LysPb, com log K = 9,69. A interação dos aminoácidos estudados com o chumbo também foi constatada pelos ensaios feitos com a matéria-prima das embalagens das soluções de Nutrição Parenteral através da análise por AAS.

Surface Complexation Modelling of the Adsorption of Cd(II), Cu(II), and Ni(II) to the Roots of Triticum turgidum

The goal of this study was to characterize the binding sites on the surface of wheat roots, Triticum turgidum, involved in the adsorption of protons and metals, and quantify the thermodynamic constants needed for a surface complexation model to predict metal binding. The adsorption of protons, Cd(II), Cu(II), and Ni(II) to the root surface as a function of pH and ionic strength in single metal exposure scenarios was quantitatively described using potentiometric titrations, batch metal adsorption experiments, and the least squares fitting program FITEQL. Model predictions from single metal exposures were compared to measured metal adsorption concentrations when roots were exposed to binary and ternary combinations of the metals. Proton dissociation was a function of three discrete monoprotic acid sites on the root surface with log proton dissociation constants of -4.50, -6.23, and -7.37 respectively, upon which varied ionic strength had no effect. The total proton binding capacities for the three sites were 2.58 x 10-4, 1.29 x 10-4, and 2.58 x 10-4 M, respectively. Metal complexation was best described by a two-site model having conditional stability constant log values of 3.04 and 3.30 for Cd(II), 3.21 and 3.25 for Cu(II), and 2.83 and 2.84 for Ni(II) at ionic strength 0.01M. At ionic strength 0.1 M the conditional stability constants log values were 2.37 and 3.36 for Cd(II), 3.11 and 2.56 for Cu(II), and 2.18 and 3.00 for Ni(II). When roots were exposed to binary or ternary mixtures of the metals, the two monoprotic acid single metal model did not provide ideal fits to the data indicating that adsorption in a metal mixture scenario cannot be considered additive and is dependent on the combination of metals present in the exposure environment. The experimentally determined proton dissociation constants and metal stability constants could be used in commercial geochemical speciation programs such as Visual MINTEQ to predict metal adsorption to plants. / Natural Sciences and Engineering Research Council of Canada, The Mining Association of Canada, Ontario Power Generation, Environment Canada.
