Reassessment of the statistical power of published controlled clinical trials. / CUHK electronic theses & dissertations collection

Background. The randomized controlled clinical trial is currently the most scientific method for evaluating the effect of medical interventions. The sample size of a trial is crucial for reliably estimating the effect. However, many clinical trials may not be sufficiently large in size to detect the effect of interventions assessed. Previous studies of the statistical power, a relative measure of the largeness of a study, were normally small, mainly examined trials with a statistically insignificant result and were flawed because of the biased or purely hypothetical estimate of the effect for the computation of the power. By using meta-analysis, we conducted this study with improved methods for estimating the power and included a larger number of trials. / Findings. A total of 2,923,912 patients from 2,872 clinical trials from 466 systematic reviews were included in the analyses of this thesis. Of the 466 systematic reviews, 24% (113) were identified from the five journals and the remaining 76% (353) were from the Cochrane Library. 1,000 trials and 1,583,204 patients were obtained from 113 systematic reviews identified in the journals, in which 13.7% (95% C.I.: 11.6%, 15.8%) of trials had a sufficient power and the overall power was 34.0% (95% C.I.: 33.7%, 34.3%). 1,872 trials and 1,340,708 patients were obtained from 353 systematic reviews identified in the Cochrane Library, in which 16.7% (95% C.I.: 15.0%, 18.4%) of trials had a sufficient power and the overall power was 37.8% (95% C.I.: 37.6%, 38.0%). We identified trials from systematic reviews of clinical trials with binary outcomes published in five medical journals and the Cochrane Database of Systematic Reviews. We analyzed the power of trials with a significant result as well as those with an insignificant result. In estimating the power, we used the combined odds ratio of the meta-analysis as the estimate of the effect for trials from systematic reviews with a statistically significant result and a relative risk reduction of 25% for trials from systematic reviews with a statistically insignificant result. In addition to use of the conventional method to estimate the power, we also developed a new "counting method" that does not need any assumption about the effect. 1990 - 2008 Fertility factors in the disctrict of Most in 2000-2010 period Abstract The objective of this study was to find causes of regional fertility differences and context with social and economic indicators. Then to find situation of district of Most in regional comparison and social and economic causes of fertility developement in this district in 2000-2010 period. In first descriptive part there was made the comparative analysis of age structure and fertility indicators in Czech republic, Ústí region and district of Most. The result is different values of the fertility indicators (intensity and timing) and also younger age structure. In the second part were found social and economic factors of regional fertility differences and factors in district of Most with multivariate statistical methods. The cluster analysis confirmed the differences of district of Most and border of North Bohemia too. For use factor analysis and canonical correlation analysis the conclusion was the important factor of regional differences is especially education, but also availability and quality of housing, economic level. Para a avaliação das características do solo relacionadas com o potencial uso dos solos, assim como para a avaliação da fertilidade, as análises químicas e físicas de rotina são os métodos convencionalmente usados. Estes são bastante custosos e demorados o que tem representado no Brasil uma dificuldade no seu uso por parte de pequenos agricultores, além da aplicabilidade da agricultura de precisão no manejo de solos. Atualmente a pedometria está fornecendo a possibilidade de incorporar em ciência do solo técnicas bastante sofisticadas que podem ajudar a diminuir o custo na obtenção da informação e compreender melhor o funcionamento dos processos do solo. Entre os tópicos mais recentes que estão incluídos na pesquisa relacionada com pedometria está a espectroscopia de reflectância. Embora se tenha demonstrado que uma grande quantidade de atributos podem ser estimados a partir da resposta espectral do solo via sensoriamento, ainda não se têm atingido níveis de acurácia ótimos em relação às metodologias convencionais, sobretudo no referente a atributos químicos. Para tanto, o presente trabalho foi desenvolvido com a finalidade de responder basicamente os seguintes questionamentos: a. Existem faixas espectrais específicas das bases trocáveis ou se estas podem mudar em função do argilomineral fornecedor da capacidade de troca de cátions?; b. A calibração de modelos usando unicamente algumas faixas espectrais específicas pode melhorar o desempenho destes?; c. Qual é a influência dos níveis de acurácia dos modelos espectrais sobre mapas construídos com atributos estimados a partir destes?; d. Como os tamanhos dos grupos de amostras de calibração influenciam a acurácia dos modelos?; e. Como a calibração de atributos relacionados com o intemperismo podem auxiliar no mapeamento de classes de solo? / The routine soil analysis is traditionally used on the evaluation of soil attributes related to land use potential, and the assessment of fertility. It is costly and time consuming, making it inaccessible for small farmers, and hampering the applicability of precision agriculture on soil management in Brazil. Currently, pedometrics is providing the possibility of incorporating in soil science sophisticated techniques that can help to reduce the cost of obtaining information and improve the understanding about how several soil processes works. One of the more recent topics on pedometrics is the soil reflectance spectroscopy. Through the soil reflected energy is possible to infer several soil properties, although optimum accuracy levels in the spectral estimation of soil attributes have not yet reached. In this sense, the aim of this study was basically answer the following questions: a. The exchangeable bases have specific spectral bands or the spectral responses of theses depends on the clay mineral?; b. the calibration of models by using only some specific spectral bands may improve the prediction performance?; c. What is the influence of the accuracy of prediction models on maps constructed with predicted soil attributes?; D. How calibration set size affect the accuracy of the models?; e. How the calibration of models for prediction of soil attributes related to soil weathering may assist the digital soil mapping?

As it was a descriptive study, the data source used was the curricular information available in CNPq Lattes Platform, from Professors and students of these programs. In this study, we proposed to give an overview about the profile and scientific publications of the graduates, in order to identify factors that affect the scientific production, the continuity of the academic formation, the choice of research lines, and the professional career in Universities. We also aimed to infer whether the Professors?? academic factors, with Doctoral or PhD degree titles, the acquisition of a scholarship, the authorship of international publications, or the membership in teaching staff/ or researchers of first class Universities, have positive effects on the trajectory of graduated students. We verified a high academic productivity in the applied sample, through the general Lotka index, and the Price??s Elitism Law. Multiple-imputation approaches to haplotypic analysis of population-based data with applications to cardiovascular disease

[Truncated abstract] This thesis investigates novel methods for the genetic association analysis of haplotype data in samples of unrelated individuals, and applies these methods to the analysis of coronary heart disease and related phenotypes. Determining the inheritance pattern of genetic variants in studies of unrelated individuals can be problematic because family members of the studied individuals are often not available. For the analysis of individual genetic loci, no problem arises because the unit of interest is the observed genotype. When the unit of interest is the linear combination of alleles along one chromosome, inherited together in a haplotype, it is not always possible to determine with certainty the inheritance pattern, and therefore statistical methods to infer these patterns must be adopted. Due to genotypic heterozygosity, mutliple possible haplotype configurations can often resolve an individual's genotype measures at multiple loci. When haplotypes are not known, but are inferred statistically, an element of uncertainty is thus inherent which, if not dealt with appropriately, can result in unreliable estimates of effect sizes in an association setting. The core aim of the research described in this thesis was to develop and implement a general method for haplotype-based association analysis using multiple imputation to appropriately deal with uncertainty haplotype assignment. Regression-based approaches to association analysis provide flexible methods to investigate the influence of a covariate on a response variable, adjusting for the effects of other variables including interaction terms. ... These methods are then applied to models accommodating binary, quantitative, longitudinal and survival data. The performance of the multiple imputation method implemented was assessed using simulated data under a range of haplotypic effect sizes and genetic inheritance patterns. The multiple imputation approach performed better, on average, than ignoring haplotypic uncertainty, and provided estimates that in most cases were similar to those observed when haplotypes were known. The haplotype association methods developed in this thesis were used to investigate the genetic epidemiology of cardiovascular disease, utilising data for the cholesteryl ester transfer protein gene (CETP), the hepatic lipase (LIPC) gene and the 15- lipoxygenase (ALOX15) gene on a total of 6,487 individuals from three Western Australian studies. Results of these analyses suggested single nucleotide polymorphisms (SNPs) and haplotypes in the CETP gene were associated with increased plasma high-density lipoprotein cholesterol (HDL-C). SNPs in the LIPC gene were also associated with increased HDL-C and haplotypes in the ALOX15 gene were associated with risk of carotid plaque among individuals with premature CHD. The research presented in this thesis is both novel and important as it provides methods for the analysis of haplotypic associations with a range of response types, while incorporating information about haplotype uncertainty inherent in populationbased studies. These methods are shown to perform well for a range of simulated and real data situations, and have been written into a statistical analysis package that has been freely released to the research community.

Defining and predicting species-environment relationships : understanding the spatial ecology of demersal fish communities

[Truncated abstract] The aim of this research was to define key species-environment relationships to better understand the spatial ecology of demersal fish. To help understand these relationships a combination of multivariate analyses, landscape analysis and species distribution models were employed. Of particular interest was to establish the scale at which these species respond to their environment. With recent high resolution surveying and mapping of the benthos in five of Victoria's Marine National Parks (MNPs), full coverage bathymetry, terrain data and accurate predicted benthic habitat maps were available for each of these parks. This information proved invaluable to this research, providing detailed (1:25,000) benthic environmental data, which facilitated the development and implementation of a very targeted and robust sampling strategy for the demersal fish at Cape Howe MNP. The sampling strategy was designed to provide good spatial coverage of the park and to represent the park's dominant substrate types and benthic communities, whilst also satisfying the assumptions of the statistical and spatial analyses applied. The fish assemblage data was collected using baited remote underwater stereo-video systems (stereo- BRUVS), with a total of 237 one-hour drops collected. Analysis of the video footage identified 77 species belonging to 40 families with a total of 14,449 individual fish recorded. ... This research revealed that the statistical modelling techniques employed provided an accurate means for predicting species distributions. These predicted distributions will allow for more effective management of these species by providing a robust and spatially explicit map of their current distribution enabling the identification and prediction of future changes in these species distributions. This research demonstrated the importance of the benthic environment on the spatial distribution of demersal fish. The results revealed that different species responded to different scales of investigation and that all scales must be ix considered to establish the factors fish are responding to and the strength and nature of this response. Having individual, continuous and spatially explicit environmental measures provided a significant advantage over traditional measures that group environmental and biological factors into 'habitat type'. It enabled better identification of individual factors, or correlates, driving the distribution of demersal fish. The environmental and biological measures were found to be of ecological relevance to the species and the scale of investigation and offered a more informative description of the distributions of the species examined. The use of species distribution modelling provided a robust means for the characterisation of the nature and strength of these relationships. In addition, it enabled species distributions to be predicted accurately across unsampled locations. Outcomes of the project include a greater understanding of how the benthic environment influences the distribution of demersal fish and demonstrates a suite of robust and useful marine species distribution tools that may be used by researcher and managers to understand, monitor, manage and predict marine species distributions.

Quasar host galaxies at intermediate and high redshifts

<p>Quasars form one of the most energetic phenomena in the universe, and can be traced out to very large redshifts. By studying the galaxies which host the active nuclei, important insights can be gained into the processes that trigger and maintain the quasar powerhouse. The evolution rate of the quasar population is furthermore similar to that of ordinary galaxies, which implies a connection between black hole accretion and star formation in the host galaxies. While the properties of quasar host galaxies at low redshift have become better constrained in recent years, less is known about hosts at earlier cosmic epochs. In addition, though radio-quiet quasars are by far more common than their radio-loud counterparts their host galaxies have not been studied to the same extent, in particular not at higher redshifts.</p><p>An imaging campaign of a large sample of quasars at intermediate redshift (0.4 < z < 0.8) was carried out at optical wavelengths using the Nordic Optical Telescope, and is studied in this thesis together with two smaller samples. The joint material forms more than half of the total number of observed sources in this redshift interval and increases the number of resolved radio-quiet hosts at z>0.4 considerably. The morphology and mean magnitudes are found to be similar for radio-loud and radio-quiet host galaxies. Both types of host are shown to have optical colours as blue as those of present-day late-type spirals and starburst galaxies, which is likely the result of ongoing star formation.</p><p>With increasing redshift, observations of host galaxies become more difficult. High spatial resolution can be achieved with adaptive optics, but the variation of the point spread function in the near-infrared wavelength band which is most suited for detection is large and rapid. A statistical approach to the problem of characterizing the point spread function has been developed, making use of simulated objects which are matched to the different atmospheric conditions. Bright, compact host galaxies showing signs of merging and interaction were detected in this way for three quasars at z~2.2, which were observed with the ESO 3.6 m telescope. The method is not restricted to host galaxy analysis but can be utilized in other applications as well, provided that the underlying extended source can be described by an analytical model. </p>
