The main objective of this diploma thesis is compare the production efficiency of special types of feed for African catfish in experimental conditions in a recirculating system with biological treatment of water. It was tested four different types of floating feed, differing in the proportion of main components, the chemical composition and determining - for catfish (CatCo GROWER - 12 EF, CatCo SELECT - 13 EF and CatCo GROWER - 13 EF), respectively salmonid fish species (Dibaq Trout Evolution). The primary outcome indicators was the growth rate, individual weight (and it's variability), feed conversion ratio, the cost of feed consumed per unit of growth and product quality, evaluated according the average dress-out percentage of skinless fillets, organoleptic assessment and chemical composition of flesh.

Technologie zpracování a kvalita masa uzené makrely / Processing technology and quality of meat smoked mackerel

The technological process of smoked mackerel (Scomber scombrus) manufacture was described in detail with a focus on quality of muscle. The smoked mackerel brined in 6%, 12% and 15% of brine was monitored by texture profile analysis (TPA) using a cylindrical probe P 75 and spherical probe P 1S. Textural properties of muscle, such as hardness, cohesiveness, springiness and chewiness were evaluated. With increasing concentrations of brine muscle hardness and chewiness significantly decreases. In texture measuring, using a cylindrical probe P 75, in 6% of brine approximately 10.4 % higher hardness can be achieved, about 1 % higher chewiness and about approximately 5 % lower springiness expect, than when brined in 12% of brine. When brined in 15% of brine about 6.7 % lower hardness, about approximately 11.2 % lower chewiness and about 6.5 % lower springiness expect, than when brined in 12% of brine. When using a spherical probe P 1S is at brined in 6% of brine about approximately 6.5 % higher hardness and about approximately 11 % lower chewiness that brined in 12% of brine. When brined in 15% of brine about approximately 13 % lower springiness and approximately 21.7 % lower chewiness expect, than when brined in 12% of brine. To a lesser extent springiness was influenced, cohesiveness with increasing concentrations of salt in brine was unchanged. Texture measurements were supplemented by microbiological analysis of the total count of microorganisms (TCM). This analysis confirmed that with increasing concentrations of salt brine the count of microorganisms decreases (on 103 using 6% of brine and on 102 using 12% and 15% of brine) as a result of reduced water activity (aw).

Úloha perivaskulární tukové tkáně v rozvoji kardiovaskulárních onemocnění / Role of perivascular fat tissue in the development of cardiovascular diseases

Abnormal vascular smooth muscle cell (VSMC) proliferation is thought to play an important role in the pathogenesis of atherosclerosis. Adipocytes produce several paracrine bioactive substances that can affect VSMC growth and migration. Our study focused on the ability of epicardial adipocytes to produce bioactive substances together with studying of direct effect of these substances on the VSMC proliferation rate. The gene expression of human cytokines (IL-6, IL-8, IL-18, RANTES and MCP-1) and adipokines (leptin and adiponectin) was measured in primary cell lines of epicardial and visceral adipocytes, both in undifferentiated and mature statuses. Moreover, adipokine production (IL-6, IL-8, MCP-1, VEGF and adiponectin) in conditioned media obtained from above mentioned primary cell cultures of adipocytes was measured by a Luminex assay. The VSMC proliferation rate was measured after co-culturing with CM obtained from primary cell cultures of adipocytes. The epicardial preadipocytes showed an increased expression of IL-8 (3,25-fold, p<0,05) compared with visceral preadipocytes. The expression of the adiponectin in epicardial preadipocytes was markedly decreased in comparison of the expression in visceral preadipocytes (p< 0,0001).
