Thermal properties of starch from transgenic isolines of wheat differing in starch surface components

The presence of puroindoline proteins on the starch surface is the biochemical marker for wheat hardness. Near-isogenic samples over expressing puroindolines have been used to assess the effect of wheat hardness on final product characteristics. The objective of this study was to determine differences among starch isolated from near-isogenic samples and to investigate the role starch surface components play in pasting. The use of near-isogenic samples over expressing puroindolines combined with the use of two methods of starch isolation (batter and dough) was an effective means to create samples with varied amounts of surface components. Starch thermal properties were characterized and surface proteins and lipids were quantified. Starch isolated from hard wheat cultivars presented more similarities with starch isolated from its soft near-isogenic line when a dough method was used than when a batter method was used. Starch from soft experimental lines isolated using a batter method showed increased MVA peak viscosity, breakdown and swelling power. Increased levels of LysoPC in starch isolated from hard wheat cultivars or soft experimental lines by dough method could have complexed with amylose and restricted granule swelling. Thereby, decreasing peak viscosity, breakdown and swelling power.

Gaseous swelling and release in nuclear fuels during grain growth

A model of the generation and release of fission gas, as well as the total swelling over time, was created. It uses an ideal spherical fuel grain with a time-dependent radius. UO2 and quasi-homogeneous SBR MOX fuels were simulated with this model, and the results were compared to a fixed grain radius model of gaseous swelling. Gaseous swelling and fission gas release were calculated for temperatures from 1600 K to 2200 K. The grain growth of UO2 was found to decrease the time needed to saturate the intergranular boundaries as compared to simple diffusion without grain growth. Small temperatures increased the time required for saturation, as did small rates of grain growth. Gaseous swelling was within the range of values found by experimental data.

Polymer Gels: Kinetics, Dynamics Studies and Their Applications as Biomaterials

The polymer gels especially hydrogels have a very special structure and useful features such as unusual volume phase transition, compatibility with biological systems, and sensitivity to environmental stimuli (temperature, pH value, electric field, light and more), which lead to many potential applications in physical and biochemical fields. This research includes: (1) the theoretical and experimental studies of polymer gels on swelling kinetics, spinodal decomposition, and solution convection in gel matrix; (2) applications of polymer gels in wound dressing, tissue-simulating optical phantom and gel display. The kinetics of gel swelling has been theoretically analyzed by considering coupled motions of both solvent and polymer network. Analytical solutions of the solvent and the network movement are derived from collective diffusion equations for a long cylindrical and a large disk gel. Kinetics of spinodal decomposition of N-isopropylacrylamide (NIPA) polymer gel is investigated using turbidity and ultrasonic techniques. By probing movement of domains, a possible time-dependent gel structure in the spinodal decomposition region is presented. Theoretical studies of solution convection in gel matrix have been done and more analysis on dimensionless parameters is provided. To enhance the drug uptake and release capacity of silicone rubber (SR), NIPA hydrogel particles have been incorporated into a SR membrane. This SR/NIPA composite gel has promising attributes for wound dressing and other uses. Tissue-simulating optical phantom has been synthesized and studied using NIPA solution trapped inside a hydrogel. Polymer gels with engineered surface patterns were implemented. NIPA gel deposited on the surface of an acrylamide gel can be used as responsive gel display. A dynamically measurement technique of local shear modulus and swelling ratio of gel is presented based on an engineered periodic surface pattern as square array.

Étude du risque de développement d'une réaction sulfatique interne et de ses conséquences dans les bétons de structure des ouvrages nucléaires / Study of risk of developing a delayed ettringite formation and its consequences in concrete of nuclear structures

La réaction sulfatique interne (RSI) dans les ouvrages en béton est une pathologie susceptible de se développer lorsque des conditions particulières portant sur la composition du béton, les conditions thermiques au jeune âge et l'environnement sont réunies. Ce phénomène est attribué à la formation d'ettringite différée qui provoque un gonflement du matériau et une fissuration dans la structure. Deux types de béton sont concernés par cette pathologie : les bétons préfabriqués traités thermiquement et les bétons coulés en place dans les pièces massives. Bien que largement étudiée depuis une dizaine d'années, la RSI n'est pas à ce jour parfaitement connue du fait de la complexité du phénomène au niveau des mécanismes mis en jeu, des paramètres influents et de ses conséquences tant à l'échelle microscopique qu'à l'échelle de l'ouvrage. Ce travail vise donc à apporter des éléments de réponse à plusieurs interrogations concernant cette pathologie, en se focalisant sur l'étude de l'impact de certains facteurs intervenant lors de la réaction et sur l'application nucléaire. De nombreuses études expérimentales ont été ainsi mises en œuvre. Une large étude est consacrée à évaluer le risque de développement de la RSI dans une enceinte de confinement d'une centrale nucléaire, du fait de la présence de structures massives dans ce type d'ouvrages. Ces travaux ont permis de mettre en évidence dans quelle mesure la pathologie influe sur les caractéristiques mécaniques et sur les propriétés de transfert du béton, et ainsi de vérifier si les exigences de sûreté attendues par ces structures sont mises en cause. Ensuite, nous nous sommes intéressés à étudier l'impact de l'hygrométrie du béton sur le développement de la RSI. Nous avons pu identifier le couplage qui existe entre l'humidité environnante et le gonflement. Le rôle joué par la lixiviation des alcalins a été également mis en évidence. Des suivis des propriétés de transfert ont été menés et confrontés aux gonflements observés. Enfin, nous nous sommes intéressés à caractériser le rôle joué par le squelette granulaire sur la cinétique et l'amplitude du gonflement par RSI. Les paramètres étudiés concernent la taille et la fraction volumique des inclusions granulaires. De plus, un exemple d'application d'une modélisation numérique mésoscopique du gonflement est proposé / Delayed ettringite formation (DEF) in concrete structures is a pathology that can develop when special conditions on the concrete composition, the thermal conditions at the young age and the environment are met. This phenomenon provokes swelling of the material and cracking in the structure. It affects two types of concrete : the concrete heat-treated and the concrete cast in place in massive parts. Although many studies were done before in order to better understand this pathology, the DEF is still not well known. This is due to its complex mechanism, the influential parameters and its consequences on microscopic and structural scales. For that purpose, the thesis work was designed in order to better understand this pathology. Experimental studies were done to evaluate the impact of certain factors during the reaction, by focusing on nuclear application. An important part of this study was dedicated to assess the risk of DEF development in a nuclear power plant, and to understand how this pathology affects the mechanical characteristics and transfer properties of the concrete. Then, we have studied the impact of the hygrometry on the development of DEF. This has lead to identify a relation between environmental humidity and swelling. We have also examined the role of alkali leaching. A follow up study of the transfer properties was also done and was confronted to the observed swelling. Finally, we were interested in the characterization of the aggregates effect on the kinetics and the amplitude of DEF swelling. So, we have examined the parameters related to the size and the volume fraction of granular inclusions. Furthermore, an application of a mesoscopic numerical modeling of swelling is proposed

Rôle de la microstructure des sols argileux dans les processus de retrait-gonflement : de l’échelle de l’éprouvette à l’échelle de la chambre environnementale / Role of microstructure of clayey soils in the shrink-swell process : from specimen scale to environmental chamber scale

Le processus de retrait-gonflement des sols argileux est à l'origine des dommages au bâti durant les périodes de sécheresse. Dans cette thèse, les processus de retrait-gonflement de deux formations de sol argileux en France qui sont les Argiles Plastiques du Sparnacien et l'argile d'Héricourt du Lias sont étudiés à différentes échelles et différents états (intact, remanié, compacté et traité à la chaux) en considérant spécialement le rôle de la microstructure. Pour ce faire, une partie de la recherche est réalisée sur la caractérisation des sols étudiés pour analyser leurs propriétés minéralogiques, microstructurales, géotechniques et hydriques par rapport à leur propriété de retrait-gonflement. La deuxième partie de la recherche est consacrée à l'analyse des processus de gonflement, de retrait, de fissuration des sols au cours d'un cycle simple ou de cycles répétés d'humidification-séchage à l'échelle de l'éprouvette. Les processus de retrait-gonflement de sol à une échelle plus grande sont étudiés dans la troisième partie en réalisant les essais d'infiltration et d'évaporation dans une chambre environnementale. Les résultats obtenus mettent en évidence les différences de comportement au retrait-gonflement pour différents états des sols étudiés. Les minéraux argileux ainsi que le quartz, les carbonates et des hydrates, mais aussi la microstructure des sols contrôlent ces phénomènes de retrait-gonflement lors des échanges hydriques. Tous les changements de volume de sol argileux au cours de gonflement-retrait sont gouvernés principalement par la famille de pores inter-agrégats naturels. Ces résultats sont apportés par les analyses microstructurales porosimétriques et au MEB. / The shrink-swell process of clayey soils is causing damage to the structures during periods of drought. In this thesis, the shrink-swell process of two clayey soils in France that are the Plastic Clays of Sparnacian age and Héricourt clay of Lias is studied at different scales and different conditions (intact, remoulded, compacted and lime treated) by insisting on the role of microstructure. To do this, a part of the study is carried out on the characterisation of soils studied to analyse their mineralogical, microstructural, geotechnical and hydraulic properties in relation with their shrink-swell property. The second part of the study focuses on the analysis of swelling, shrinkage, cracking processes of soil during a single cycle or repeated wetting-drying cycles at specimen scale. The shrink-swell process of soil at a larger scale is studied in the third part by performing infiltration and evaporation tests in the environmental chamber. The results highlight the difference in the shrink-swell behavior at different conditions of soils studied and emphasize the importance of the presence and nature of clay minerals also quartz, hydrates and microstructure of soil to the shrinkage and swelling during water exchange. All changes in the volume of clayey soil during shrinkage and swelling are mainly governed by natural inter-aggregate pores, which are showed by microstructural analyses of MIP and SEM tests.

Drenagem linfática manual no pós-operatório de enxerto ósseo alveolar: uma nova abordagem para a redução do edema facial / Manual lymphatic drainage after alveolar bone grafting: a new approach to reduce facial swelling

OBJETIVO: Determinar a efetividade de manobras padronizadas de drenagem linfática manual (DLM) na redução do edema facial, na distância interincisal máxima ativa e no quadro álgico de pacientes submetidos à cirurgia de Enxerto Ósseo Alveolar (EOA). METODOLOGIA: Esta pesquisa analisou 51 indivíduos com fissura labiopalatina reparada, entre 10 e 15 anos de idade, submetidos ao EOA no Hospital de Reabilitação de Anomalias Craniofaciais-USP, divididos em 2 grupos, grupo tratamento (n=29, 12 homens e 17 mulheres) e grupo rotina (n= 22, 15 homens e 7 mulheres). No grupo tratamento foram aplicadas manobras de DLM padronizadas diferentemente daqueles do grupo rotina. Foram realizadas nos dois grupos avaliações do edema facial por meio das medidas da linha 1 (distância da base da asa nasal ao tragus) e linha 2 (distância da base da asa nasal à implantação inferior do pavilhão auricular), na hemiface operada nos períodos pré-operatório (Pré), segundo (2ºPO) e quarto pós-operatório (4ºPO). Também foram realizadas as medidas da distância interincisal máxima ativa nos mesmos períodos. A Escala Analógica da Dor (EAD) foi aplicada no 1º, 2º, 3º e 4º períodos pós-operatórios. Adicionalmente, no grupo tratamento, após a terapia de DLM, foram realizadas perguntas sobre dor, relaxamento e sono. RESULTADOS: Houve redução do edema facial no grupo tratamento do 2ºPO para o 4ºPO o que não ocorreu no grupo rotina. As medidas da distância interincisal apresentaram aumento entre o 2ºPO e 4ºPO nos dois grupos estudados. Na avaliação da EAD, o grupo tratamento mostrou ausência de dor no 3ºPO enquanto o grupo rotina somente no 4ºPO. No grupo tratamento, a totalidade dos indivíduos relataram que a dor diminuiu, e que se sentiram mais relaxados após a DLM, nos 3 períodos avaliados. Relataram também em muitos casos que a DLM teve efeito facilitador na indução ao sono durante a DLM. CONCLUSÕES: As manobras padronizadas de DLM aplicadas da maneira proposta produziram redução significante do edema facial, aumento da distância interincisal máxima ativa, redução do quadro álgico no grupo tratamento comparativamente ao grupo rotina. Esses dados reforçam a hipótese de que a DLM proposta foi efetiva contribuindo de maneira importante na recuperação do paciente submetido à cirurgia de EOA. / OBJECTIVE: To determine the effectiveness of standardized manual lymphatic drainage (MLD) maneuvers in reducing facial swelling, maximum interincisal active distance and pain of patients undergoing surgery for Alveolar Bone Grafting (ABG). METHODS: This study examined 51 individuals with repaired cleft lip and palate, aged 10 to 15 years old, who underwent ABG at the Hospital for Rehabilitation of Craniofacial Anomalies- USP, divided into two groups: treatment group (n = 29, 12 men and 17 women) and routine group (n = 22, 15 men and 7 women). In the treatment group MLD standardized maneuvers were performed differently from those of the group routine. In both groups evaluation of facial swelling was performed by measuring the distance from the ala nasi to the tragus (line 1) and the distance from the ala nasi to the inferior portion of the ear auricle (line 2), in the operated side, preoperatively, and at the second and the fourth postoperative (PO) day. The maximum active interincisal distance was measured during the same periods. The Pain Analog Scale (PAS) was applied at the first, second, third and fourth days after ABG. Additionally, patients from the treatment group, were asked about pain, relaxation and sleep after MLD therapy. RESULTS: There was a reduction of facial edema in the treatment group from the 2nd to 4th PO what was not observed the routine group. Interincisal distance increased between the 2nd and 4th PO in both groups. PAS evaluation has shown that patients from the treatment group present no pain already at 3rd PO. In the routine group this occurred only at 4th PO. All the subjects from treatment group reported decrease of pain, and patients reported felt more relaxed after the MLD, at the three postoperative evaluated. Many patients reported that MLD had a facilitative effect on inducing sleep during MLD. CONCLUSIONS: The standardized MLD maneuvers applied in the proposed manner produced significant reduction of facial swelling, increase of the maximum active interincisal distance, reduction of the pain in the treatment group as compared to the routine group.These Abstract data reinforce the hypothesis that the proposed MLD is a effective procedure for the recovery of the patients undergoing ABG surgery.

Este trabalho apresenta os resultados obtidos no desenvolvimento da pesquisa \"Estudo do potencial expansivo dos sedimentos argilosos da Formação Guabirotuba na região do Alto Iguaçu- PR\". Foram realizados ensaios laboratoriais para a caracterização e previsão do comportamento expansivo dos materiais, tais como: Análises granulométricas, Limites de Consistência e demais Índices Físicos, Difração de Raios - X, Análise Térmico Diferencial, Capacidade de Troca de Cátions, além de Expansão Livre e Pressão de Expansão. Para estas investigações foram utilizadas 51 amostras deformadas e 11 blocos de material indeformado. A análise desses ensaios e de informações básicas do meio físico, permitiram a avaliação de áreas que apresentam diferentes potenciais à expansão, permitindo a elaboração da Carta de Predisposição à Ocorrência de Problemas Expansivos. / This work presents the results obtained in the development of the research \"Study of the swell potential of Guabirotuba Formation clayey sediments in the area of Alto Iguaçu - PR\". Laboratorial tests, such as: Grain Size Analysis, Liquid and Plastic Limits and others Index Tests; X-Ray Diffractometry, Differential Thermal Analysis, Cation Exchange Capacity, besides Free Swelling and Axial Swelling Stress, were executed for the characterization and prediction of the studied material swelling behavior. Fifty - one disturbed samples and eleven blocks of undisturbed material were used for these investigations. The analysis of the test results and the basic information of the environment allowed the evaluation of are as that present different swell potentials, and the elaboration of the Chart of Predisposition to the Occurrence of Swell Problems.

Pedogênese de Vertissolos em ambientes de formação contrastantes / Pedogenesis of Vertisols in contrasting environments formation

Os Vertissolos apresentam alto potencial para a produção agrícola e para estocar carbono. No entanto, diversos aspectos sobre sua gênese, fundamentais para qualidade e sustentabilidade do solo, permanecem incompreendidos. As várias lacunas sobre suas características particulares estão relacionadas ao incomum teor de alumínio trocável; a manifestação de feições morfológicas e vérticas em uma matriz inorgânica dominada por fases cauliníticas; e a preservação e estoque de carbono em condições climáticas de semiárido e temperado. Dessa forma, os objetivos deste estudo foram (i) investigar a gênese de um Vertissolo ácido, bem como os fatores e mecanismos da geração dessa acidez, (ii) compreender o papel dos interestratificados no desenvolvimento das feições vérticas em Vertissolo com predomínio de caulinita na fração argila, e (iii) investigar os fatores e mecanismos que promovem a gênese de Vertissolos Ebânicos sob diferentes condições climáticas no Brasil. Amostras de seis perfis de Vertissolos foram utilizadas, sendo quatro Ebânicos e dois Háplicos, ambos localizados entre as regiões nordeste, sudeste e sul do Brasil. Análises morfológica, físicas, químicas, mineralógicas e isotópicas foram realizadas. Na modelagem dos difratogramas foi utilizado o programa Newmod. Para identificação dos grupos funcionais orgânicos do solo realizou-se a ressonância magnética nuclear da fração ácido húmico, e a subtração espectral. Todos os Vertissolos apresentaram textura de argilosa a muito argilosa, consistência seca variando de dura a extremante dura e consistência molhada entre muito plástica e muito pegajosa. Os valores de pH variaram entre ácido e alcalino. Foram eutróficos, com predomínio de cálcio no complexo de troca na maioria dos perfis. O teor de carbono reduziu em profundidade, não mascarou o potencial de contração e expansão, e promoveu melhorias na agregação, principalmente nos horizontes superficiais. Houve autigênese de minerais, os minerais primários predominaram nas frações areia e silte, e na fração argila espécies de esmectitas dioctaedrais. Independente da espécie esmectítica e do tipo de vegetação (plantas C3 e/ou C4) houve maior diversidade de grupos funcionais nos horizontes superficiais. O mecanismo de estabilização do carbono nos Vertissolos Ebânicos ocorreu por meio da interação entre as fases inorgânicas e orgânicas, mediada por pontes de cátions polivalentes não metálicos e metálicos, e também por interação eletrostática entre os constituintes inorgânicos e orgânicos. A manifestação das feições morfológicas e vérticas no Vertissolo com predomínio de caulinita na fração argila foi gerada pelos interestratificados caulinita-montmorilonita resultantes da perda da lâmina tetraedral das esmectitas via transformações no estado sólido. A acidez no Vertissolo ácido foi gerada pela hidrólise e dissolução das fases minerais mediados por um clima passado mais úmido e continuado pelo clima atual úmido, sendo os teores de alumínio trocável naturais. Nesse contexto, esse estudo contribui com avanços na gênese, mineralogia e mecanismos de estabilização do carbono no solo, os quais são temáticas relevantes desse milênio e contribui a outras áreas cientificas. / Vertisols have high potential for agricultural production and for carbon stocking. However, several aspects about its genesis, fundamental to soil quality and sustainability, remain misunderstood. The various gaps on its particular characteristics are related to the unusual exchangeable aluminum content; the manifestation of morphological and vertic features in an inorganic matrix dominated by kaolinitic phases; and the preservation and stocking of carbon in semi-arid and temperate climatic conditions. Thus, the aims in this study were to (i) investigate the genesis of an acidic Vertisol, as well as the factors and mechanisms of the acidity generation, (ii) understand the role of the interstratified clay minerals in the development of vertic features in a kaolinitic Vertisol, and iii) investigate the factors and mechanisms that promote the genesis of Pellic Vertisols under different climatic conditions in Brazil. Samples of six profiles of Vertisols were used, being four Pellic and two Haplic, both located between the northeast and southern regions of Brazil. Morphological, physical, chemical, mineralogical and isotopic analyzes were performed. In the modeling of the diffractograms, the Newmod program was used. For the identification of the organic functional groups of the soil, the nuclear magnetic resonance of the humic acid fraction and the spectral subtraction were performed. All the Vertisols presented a texture of clayey to very clayey, dry consistency ranging from hard to extremely hard and wet consistency between very plastic and very sticky. The pH values varied between acid and alkaline. They exhibit high base saturation, with the predominance of calcium in the exchangeable complex in most of the profiles. The carbon content reduced in depth; did not overlap the shrinking-swelling potential; and promoted improvements in aggregation, especially in the upper horizons. There was autigenesis of minerals, primary minerals predominated in the sand and silt fractions, and in the clay fraction species of dioctahedral smectites. Regardless of the smectic species and the type of vegetation (C3 and/or C4 plants), there was a greater diversity of functional groups in the superficial horizons. The mechanism of carbon stabilization in the Pellic Vertisols occurred through the interaction between inorganic and organic phases, mediated by bridges of non-metallic and metallic polyvalent cations, and by electrostatic interaction between inorganic and organic constituents. The manifestation of the morphological and vertic features in the kaolinitic Vertisol was generated by the interstratified kaolinite-montmorillonite resulting from the loss of the tetrahedral sheet of smectites by solid-state transformations. The acidity in the acid Vertisols was generated by the hydrolysis and dissolution of the mineral phases mediated by a humid past climate and continued by the current humid climate, being the exchangeable aluminum contents natural. In this context, this study contributes to advances in the genesis, mineralogy, and mechanisms of soil carbon stabilization, which are relevant themes of this millennium and contribute to other scientific areas.

Estudo do comportamento à expansão de materiais sedimentares da Formação Guabirotuba em ensaios com sucção controlada

Este trabalho tem como objetivo principal estudar a influência das variações de sucção no comportamento à expansão dos materiais sedimentares da Formação Guabirotuba, aflorantes na região metropolitana de Curitiba-PR. Foram coletadas amostras indeformadas em cinco pontos característicos dos materiais com maior potencial expansivo na região. As amostras foram, inicialmente, submetidas a ensaios de caracterização para determinação da composição mineralógica da fração fina, da textura, da estrutura, dos índices físicos, dos limites de consistência, da capacidade de troca de cátions e da distribuição de poros. As curvas de retenção de água foram determinadas por ensaios em câmara de pressão, pelo método do papel de filtro e do dessecador de vácuo. Os ensaios de pressão de expansão foram conduzidos em amostras nas quais a sucção foi imposta mediante o uso de soluções salinas. O umedecimento para provocar a expansão foi realizado tanto por inundação da amostra com água, em ensaios edométricos convencionais, quanto por transferência de vapor, utilizando-se um edômetro, similar ao de Esteban e Sáez (1988), especialmente construído para ser utilizado nessa pesquisa. Os resultados dos ensaios edométricos convencionais mostraram pressões de expansão crescentes com aumento das sucções impostas, atingindo valores de até 4189 kPa para as sucções altas, a amostra mais expansiva. Em contrapartida, os resultados provenientes do edômetro com controle de sucção não acusaram pressão de expansão, nos mesmos materiais e para as mesmas faixas de sucção. Atribui-se este fato às diferenças no processo e na velocidade de umedecimento do material. Constatou-se também neste trabalho que os valores de pressão de expansão, para sucções menores que 1000 kPa são insignificantes nos materiais estudados. Observou-se ainda, a partir de correlações entre atributos mineralógicos e as pressões de expansão, que existe acentuada influência da mineralogia na previsão do potencial expansivo quando a sucção é baixa, porém, quando a sucção cresce, a previsibilidade a partir da mineralogia diminui significativamente / The main objective of this thesis is to study the influence suction variations of the expansive behavior of sedimentary materials from Guabirotuba Formation, that occours from Curitiba - PR metropolitan area. Undisturbed samples were collected from five characteristic sites with high swelling potential in the region. Different characterization tests were carried out in all the samples in order to determinate clay mineralogy, texture, structure, physical properties, Atterberg limits, cation exchange capacity and pore distribution. The soil-water characteristics curves were obtained by means of pressure plate, the filter paper technique and the desiccator test. The swelling pressure tests were conducted in samples which the suction was imposed using saline solutions. The wetting of samples to produce swelling was carried out, not only flooding the samples with water in conventional oedometer tests, but also, with vapour transference, using a oedometer similar to Esteban and Saéz (1988), especially made to be used in this research. The results from the conventional oedometer tests showed that the swelling pressure increases with the suctions imposed to the sample, yielding values up to 4189 kPa for the most expansive sample. On the other hand, the oedometer test results with suction controled did not show swelling pressures, in the same materials and for the same range of suction. This behavior is attributed to differences in the process and in the rate of sample wetting. It was also evidenced that swelling pressure, for lower suctions than 1000 kPa are insignificant. From some correlations between mineralogical attributes and swelling pressure were observed that there is a higher influence of the mineralogy in the swell prediction when the suction is low, however, when the suction increases the swell prediction is less affected by mineralogy

Determinacao das caracteristicas de intumescimento de adjuvantes

O intumescimento é um dos fatores decisivos que influenciam o comportamento de desintegração de formas farmac6uticas compactadas e, consequentemente, o perfil biofarmacêutico de substancias ativas. Entre os diversos adjuvantes empregados para promover a desagregação daquelas formas farmac6uticas nas suas partículas primarias originais, são utilizadas varias substancias, abrangidas pelo conceito de desintegrantes intumescedores. A escolha do adjuvante mais adequado nestes casos pode ser vista como uma atividade que exige o gasto considerável de tempo e de custos elevados. Além disto deve ser levada em consideração a existência de vários produtos similares no mercado. A questão da equival6ncia representa portanto um desafio importante na fase de formulação. Com o objetivo de aumentar os conhecimentos nesta área, foi desenvolvido equipamento para a avaliação dinâmica do intumescimento de pês. Inicialmente foram considerados fatores de construção do aparelho, tais como a porosidade, espessura e área do filtro de vidro sintetizado. Foram tamb6m avaliadas variações metodológicas, entre as quais a temperatura de realização do teste e o meio liquido empregado. Croscarmelose, um superdesintegrante, foi utilizada como adjuvante-modelo no teste do equipamento. A alteração do meio liquido de ensaio foi o fator que causou a maior influ6ncia sobre os resultados, embora a espessura dos filtros de vidro sintetizado tenha causado alguma alteração quer no tempo como no volume final de intumescimento. intumescimento. Já a mudança de temperatura não provocou diferenças significativas nas observações. Numa segunda fase foi investigada a influência deste desintegrante sobre as características de intumescimento de quatro celuloses microcristalinas (CMCr) isoladas ou adicionadas com um lubrificante. Para tanto elaborou-se um planejamento fatorial 4 X 2 X 2. A presença do desintegrante, como esperado, provocou aumento do volume final de intumescimento das CMCr, que apresentaram, no entanto, comportamentos diferenciados frente ao adjuvante. Já a presença do lubrificante conduziu a resultados totalmente dependentes do tipo de CMCr testada. A análise comparativa entre estes produtos demonstrou que somente num caso houve simitaridade de comportamento. / Swelling is one of the decisive factors influencing the disintegrant behavior of compacted pharmaceutical dosage forms and consequently the biopharmaceutical profile of drug substances. Among the various excipients used to promote the disintegration of such dosage forms in its originally primary particles, several substances, classified as swelling disintegrants, are employed. The choice of the most appropriate excipient in this case can be a time and cost consuming enterprise. Additionally it must be considered that the same substance can be delivered in the market by different producers. The question of equivalence represents also an important challenge in the formulation phase. In order to provide more knowledge in this area an apparatus to analyze the dynamic swelling of powder admixtures was developed. Construction factors such as filter porosity, thickness and area were considered. Methodological variants including essay temperature and liquid medium were also evaluated. Croscarmelose, a superdisintegrant, was used as model excipient to test the self built equipment. The change of the test fluid was the determinant factor on the results, although the thickness of the sintered glass filter showed some influence on both swelling volume and swelling time of this adjuvant. As a second step, the influence of this disintegrant on the swelling behavior of four microcrystalline celluloses (MCC) alone or added with a lubricant were investigated by using a 4 X 2 X 2 factorial design. The presence of the disintegrant was able to increase the swelling volume. Both final swelling volume and time were dependent of the MCC type. The lubricant showed different influences on the four tested MCC. A comparative study demonstrated that only in one case these products presented similarly swelling behavior.
