Analýza čtenářské úrovně žáků 3. ročníku ZŠ se zaměřením na různé metody čtení / Analysing Third-year Primary Pupils´ Reading Skills in Relation to the Reading Method

The diploma thesis deals with the reading level of pupils in the third year, who in the first year learned to read by different methods. The thesis is divided into theoretical and research parts. The theoretical part deals with the characteristics of pupils at the younger school age, the development of reading skills, the teaching of early reading by the analytical-synthetic, genetic and Sfumato methods. Furthermore, it defines reading literacy, and refers to international research surveys, types and qualities of reading, diagnosis of reading, reading defects and their correction. The research part focuses on the diagnosis of the reading performance of pupils in the 3rd grade of primary school. The reading results are evaluated according to the criteria based on the theoretical part of the thesis.

Odhad genetické variance užitkových vlastností kapra obecného s cílem předpovědět potenciál selekčního šlechtění v rybničních podmínkách chovu

The main aim of this thesis was to study the genetic variation of several performance traits under pond management conditions and to estimate future perspective of sustainable selective breeding program in common carp. In the present study, Amur mirror carp (AM) and Hungarian synthetic mirror carp (HSM) were used. Most of the studied performance traits showed sufficient genetic variation (h2 = 0.12 1.0), suggesting a good potential for genetic improvement of the traits through selective breeding. It was shown that selection of common carp under pond management conditions should be optimally applied after the second overwintering. Winter survival is often a bottleneck for common carp production. As a result, the genetic background of winter survival and traits that might be correlated to the survival was studied. Main focus was given to the muscle fat and body weight and their dynamics through winter period and successive third growing period. It was found that selection for i) lower fat content before and after winter, ii) lower decrease in muscle fat content and/or body weight during winter, iii) higher condition factor may lead to better survival during both winter and third growing period and growth of fish during the third growing season. Unfortunately, direct selection for slaughter yields is impossible on live breeding candidates. Therefore, morphological predictors that can be measured in vivo are considered as an interesting alternative. As a result, external and internal measures were combined on 1553 fish by linear regression to predict log-log residuals (Logr) of slaughter yields. It was found that the accuracy of the prediction of slaughter yields may be solid. From the genetic point of view, model-predicted (h2 = 0.48 0.63) and even individual predictors (h2 = 0.34 0.72) of slaughter yields were highly heritable and favourably genetically correlated to the Logr yields. Hence, selection on trait predictors has an interesting perspective for genetic improvement of slaughter yields in common carp. Furthermore, genetic parameters of fillet fatty acid content and performance traits in market size common carp cultured under semi-intensive pond conditions were estimated. For flesh FA composition analysis 158 individuals were processed. Heritability estimates of total muscle fat, some FA groups and most performance traits were moderate to high (0.23 0.62). Interestingly, genetic correlations showed that genetic improvement of growth via selective breeding under Central European pond conditions without changing the production technology would very likely negatively affect carp flesh quality with respect to FA composition. Finally, restriction-site-associated DNA sequencing (Rad-seq) was used to identify and genotype single nucleotide polymorphism (SNP) markers for subsequent parentage assignment, construction of a medium density genetic map (12,311 SNPs), and testing of efficiency of marker-assisted (MAS) and genomic selection (GS) for growth and Koi herpes virus (KHV) resistance. No genome-wide significant QTL was identified for growth. However, genomic prediction of its breeding value may outperform the traditional pedigree-based prediction, resulting in an 18% improvement in prediction accuracy. On the other hand, genome-wide significant QTL affecting resistance to KHV was identified on linkage group (LG) 44 explaining approximately 7% of the additive genetic variation. The presented results add further evidence supporting the application of selective breeding in common carp cultured even under traditional pond management conditions. However, it is evident that the rearing technology will need suitable modifications. In addition, real economic impact of selective breeding on carp culture should be verified by calculation of realized heritabilities, real genetic gains and the total fish biomass yield of genetically improved stocks from a pond area unit.

In further parts, emissions created during the cycle of production and distribution of biofuel are characterized. Last part brings comparison of biofuels from different points of view, and recommendation of the alternative ways of production of gaseous and liquid biofuels from renewable resources of energy.

The aim of this thesis is to analyse the linguistic awareness of children at the first and second grade of elementary school in both reading methods, i.e. the analytic-synthetic and the genetic method of reading. In the theoretical part of the thesis, the critical analysis of the terms "linguistic awareness" and "linguistic sense" is done and further the approaches and traditions used abroad are compared to actual situation in the Czech Republic. In the analytic part of the thesis, the research project focused on the comparison of two tests of linguistic awareness is developed. The first method is the traditional Linguistic Sense Examination (Žlab, 1992), used not only for the diagnosing of the specific learning disorders, but also for diagnosing of bilingual children, children from minority groups and children with hearing or mental disabilities. The second method is the new Linguistic Awareness Test (Kucharská, Šmejkalová, under the preparation) prepared in the cooperation with Department of Czech Language and the Department of psychology PedF UK. In the research project, there were 40 schoolchild from the first and second grade of elementary school, 20 of them in the analytic-synthetic method of reading and 20 of them in the genetic method of reading. The effectiveness of teaching synthetic phonics to EFL students Abstract This diploma thesis deals with the effectiveness of systematic and explicit Synthetic Phonics teaching methods in the EFL learning environment. The theoretical part of the text investigates the similarities and differences between teaching Synthetic and Analytic Phonics. Whether synthetic phonics is essential not only for native English speakers, but also for EFL students is examined. Furthermore, it introduces the changes and development in phonics teaching in a historical context. The practical section describes the test preparation and presents how the research methodology was applied. It also examines the data collected from testing four groups of Prague primary school children who have different experience of phonics. Last but not least, the empirical section presents the results of 60 students' readings and analyses their performances concluding with an assessment as to whether explicit Synthetic Phonics teaching instruction helps EFL students in pronouncing words or not. Hodnocení hospodářské politiky pomocí syntetické kontrolní metody / Evaluating Economic Policy Using the Synthetic Control Method

The doctoral thesis consists of three essays that address the application of synthetic control method to various economic policy intervention. In the first essay I evaluate the quantitative effects of the Czech National Bank's commitment to keep the Koruna from appreciating that were put in place in 2013. I find that the commitment helped decrease unemployment substantially. The effect on overall output is also strongly positive, almost 2 percentage points for growth in 2015. The effect of the commitment on inflation is positive but not statistically significant at standard levels. In the second essay I focus on the impact of joining the EU on the Czech agriculture. The results show that the Czech Republic would have had a higher food index if it had not entered the CAM and CAP. Moreover, I show that the CAP and CAM had different impacts on farms in the Czech Republic and Bulgaria, which have the most comparable agriculture according to the results of the synthetic control method. In the final essay I estimate how the UK financial markets would have evolved if the Remain camp had won the referendum. The results suggest that there would not have been any significant change in the development of the FTSE 100 Index in the medium to long term if there had not been a referendum. This thesis deals with perfusion analysis using DCE-MRI (Dynamic contrast-enhanced magnetic resonance imaging). DCE-MRI is commonly used for microcirculation evaluation mainly in oncology and in recent years also in cardiology. The theoretical overview focuses on the issue of pharmacokinetic modeling and the estimation of perfusion parameters using selected models. The experimental part describes research software PerfLab and then it is aimed at the proposed program for synthetic data generation. Simulated data obtained under ideal conditions and in the presence of noise were used to compare models for the accuracy and reliability of DCE-MRI analysis.

This Diploma thesis focuses on the study of penetration of selected phenolic substances contained in extract from black elder (Sambucus nigra) via modelling systems represented by two types of synthetic membranes and a real system represented by a biological membrane. As a biological membrane, we have chosen skin from pig´s earlobe. Experiments of transdermal absorption and absorption through synthetic membranes were conducted through vertical (Franz´s) cells. Selected substances contained in the extract from black elder were rutin and chlorogenic acid. The main focus of this work was to determine how readily, or if at all, these two selected phenolic substances penetrate the skin and selected synthetic membranes. The percentage of absorbed amount of the substance was determined by HPLC analysis.

This study is part of a project which aims to understand evolution of genetic code together with structural and functional analysis of prebiotic proteins. The repertoire of amino acids in the first proteins was probably developing in time and it influenced the development of structure and function of today's proteins. First amino acid alphabet was apparently only half of the size of present alphabet, which contains twenty amino acids. These ten amino acids were probably prebiotically available from endogenous and exogenous sources. This work includes cell-free expression and purification of two randomized protein libraries (containing approximately 1011 variants) with various amino acid composition and following comparison of their propensity to form secondary (using circular dichroism) and tertiary (using proteolytical analysis of sequences) structures. First library contains only ten probably prebiotically available amino acids; second library contains all twenty amino acids in today's genetic code. This project could help us understand benefits of genetic code expansion in terms of developing structure in protein sequences.
