Detection of delta 9-Tetrahydrocannabinol and metabolites in the meibomian lipids of tear samples through LC-MS/MS

Δ9-Tetrahydrocannabinol (THC) is the most psychoactive substance out of the over 80 active cannabinoids. Due to its psychoactive and impairing properties, detection and quantitation is important to determine impairment levels of individuals. With an increased use of recreational marijuana, the risk of Driving Under the Influence of Drugs (DUID) is steadily increasing. Current legislation outlaws driving under the influence of Marijuana however there exists limitations with current methods of detection of drug analyte. Δ9-Tetrahydrocannabinol, 11-Hydroxy- THC, and 11-nor-carboxy-THC, were used in detection because these analytes are produced in the metabolism of THC. Since THC is very lipid soluble, it is present in lipid rich environments in the body. Due to the lipid rich nature of meibomian fluid, a component of tears, and the presence of Fatty Acid Binding Protein (5) FAPB5, a protein known to bind to cannabinoids, tear fluid could be used as a less-invasive biological matrix to test for the presence of THC and its metabolites. This project optimized a collection of tear fluid, along with a simple buffer extraction, to create a method suitable for direct injection using LC-MS/MS. Collection was completed by BVI Weck-Cel® Sterile Cellulose strips, measuring approximately 2 x 20 mm, and placed in Thompson eXtreme PVDF 0.2 𝜇m, pre-slit, red cap, filter vials containing Quantisal buffer solution for extraction. All analysis and calibrations were completed with fortified matrix standards with concentrations ranging from 0.25 - 250 ng/mL. Validation was consistent with American Academy of Forensic Sciences Academy Standards Board (ASB) Standards of Forensic Toxicology Standard 036, First Edition 2018. Tear samples were collected from volunteer patients according to Institutional Review Board (IRB) standards before and after administration of Marijuana. Samples were collected approximately 30 minutes post administration in order to capture tears when the analyte is most potent in the body. Samples and calibration standards were analyzed using Liquid Chromatography Tandem Mass Spectrometry (LC-MS/MS) with the QSight® 220 CR LC-MS/MS and the Halo® C18 3.0x50 mm (2.7 µm) column. Limit of Detection (LOD) and Limit of Quantitation (LOQ) for THC was calculated at 0.25 ng/mL. Limit of Detection of THCOOH was detected at 0.25 ng/mL and Limit of Quantitation was calculated at 1 ng/mL. Upon analysis of Patient Samples, it was determined that THC and metabolites could be detected and quantitated in tear fluid. However, it is noted that insufficient sample volume in collection of this type of sample is an issue that leads to poor quantitation and should be optimized in future research.

This study is an examination of the theatre project at Tsa La Gi, a Cherokee cultural center in Tahlequah, Oklahoma. The thesis is organized into three areas: the drama, the theatre design, and the production techniques. Chapter I reports the process of the formulation of Trail of Tears and analyzes its success. Chapter II describes and interprets the process of the design of the physical theatre. Chapter III reports the techniques used in play production at Tsa La Gi and interprets their effects. Chapter IV presents conclusions about the success of the theatre project. This report accepts evidence that the theatre project at Tsa La Gi is a highly successful one, both economically and artistically.

The Effects of the Handwriting Without Tears Program on the Handwriting of Students in Inclusion Classrooms

Many handwriting programs are currently used in schools, but little research has been conducted on their effectiveness. A quasi-experimental non-equivalent comparison group pretest posttest design examined effects of the Handwriting Without Tears program with special and general education students enrolled in inclusion classrooms. Two experimental classes received instruction with the HWT method while two comparison classes received instruction using traditional methods. Handwriting performance was measured using the Minnesota Handwriting Assessment. One-way analysis of covariance tested the differences between rates of handwriting improvement for experimental and comparison groups while controlling for pretest scores. Students in the experimental classes showed no statistically significant improvement in overall handwriting skill compared to the classes receiving traditional handwriting instruction. However, the HWT program was found to be effective in improving the areas of size (p = .008) and spacing (p = .014) within a 10-week period, regardless of educational status or gender.

Orientador: Luiz Claudio Di Stasi / Resumo: A Doença Inflamatória Intestinal é uma doença considerada multifatorial com etiologia ainda pouco elucidada que compreende duas principais formas clinicas, denominadas como Retocolite Ulcerativa e a Doença de Crohn que induzem quadros crônicos de inflamação em diferentes regiões do trato gastrointestinal, com períodos de exacerbação e remissão dos sintomas. Os tratamentos farmacológicos disponíveis para a doença apresentam sérios efeitos colaterais e alto custo, o que faz com que o desenvolvimento de estratégias complementares de prevenção e tratamento seja uma importante meta na terapia da DII. A busca por estratégias complementares de prevenção e tratamento, como o uso de alimentos funcionais ricos em fibras e compostos bioativos, é uma perspectiva promissora, visto que as fibras têm sido associadas à promoção de efeitos prebióticos com consequente proteção ao processo inflamatório intestinal. Coix lacryma-jobi L., é uma planta cuja semente é rica em fibras e são amplamente utilizadas tanto na culinária quanto na medicina tradicional chinesa onde são indicadas para o tratamento de diversas doenças. The pharmacological treatments available for the disease have serious side effects and high cost, which makes the development of complementary prevention and treatment strategies an important goal in IBD therapy. The search for complementary treatment and prevention strategies, such as the use of functional foods rich in fiber and bioactive compounds, are a promising prospect, since the fibers have been associated with the promotion of prebiotic effects with consequent protection of the inflammatory bowel process.
