Ovlivnění odpovědi rostlin na teplotní stres modulovanými hladinami cytokininů - fenomická a protemická analýza

Cytokinins are important group of phytohormones regulating many physiological processes ranging from cell division to programmed cell death. This thesis is focused on effects of cytokinin levels in response to heat stress in Arabidopsis thaliana. Analysis of transgenic plants with regulated expression of ipt and HvCKX showed that cytokinins and their optimal levels play important role in the morphological alterations induced by heat stress. Seedlings with increased and decreased levels of cytokinins exhibit inhibition of petioles growth, decreased length of blades of true leaves and reduced leaf area. To obtain insights into molecular events underlying early response to heat stress LC-MS analysis of whole proteom was performed. Analysis revealed 57 differentialy regulated proteins in response to heat stress in Columbia ecotype. On the cellular level, most of the proteins were located in cytosol (47 %) nebo plastids (32 %). Coparative analysis between wild-type seedlings and seedlings with decreased level of cytokinins confirmed 31 proteinInfluencing plant responses to temperature stress modulating cytokinin levels - fenomic and proteomic analysiss regulated by cytokinins in response to heat stress. Among these proteins, desarurase 7 and Tudor SN1 protein were previously found as important factors in response to heat stress.

Vliv ošetření substrátu a teplotních podmínek na vývoj kultury Pleurotus eryngii a Pleurotus nebrodensis / Influence of the substrate treatment and the temperature on development of the Pleurotus eryngii and Pleurotus nebrodensis culture

Objective of this diploma thesis was to select the most appropriate composition and thermal treatment of a substrate and the optimal temperature for planting and development of the sporocarps of mushrooms Pleurotus eryngii and Pleurotus nebrodensis. Within the objective of this thesis there was a hypothesis that different levels of the thermal treatment of a substrate have an impact on the colonization of such substrate by mycelium and subsequent crop of sporocarps Pleurotus eryngii and Pleurotus nebrodensis and that within those selected strains there can be found a strain which tolerates temperatures in a grow room above 15 °C. Straw pellets, beech sawdust, spruce sawdust and Douglas tree sawdust were selected as substrates for the growth of sporocarps P. eryngii and P. nebrodensis. Some substrates were enhanced by a wheat bran or coniferous sawdust was fermented without and with 5% fugate. Obtained data were statistically processed using the Statistica (StatSoft) programme by the ANOVA method. Statistical assessment of results is presented graphically in the thesis. Results of the experiment proved that Pleurotus eryngii and Pleurotus nebrodensis have the highest growth of their sporocarps on the substrate from fermented coniferous sawdust with added fugate. The results further shown that the mushrooms had higher growth when they were on the substrate without a wheat bran compared to the substrate with a wheat bran, which was even more prone to its contamination. Optimal temperature for planting and development of the sporocarps Pleurotus eryngii was determined to be 12 - 15 °C. There were significant differences in the fructification of P. eryngii and P. nebrodensis which produced only minimal crop. Duration of a growing cycle of Pleurotus eryngii was 72 - 134 days. Development of sporocarps lasted 11 - 21 days.

Mikroklima porostu řepky a pšenice

The purpose was clarification of vertical stratification of temperature and humidity in winter rape and winter wheat canopy and their comparison with the data from standard station. Air temperature and relative humidity was monitored in wheat and rape canopy in Žabčice during the main growth season in 2010 and 2011 years. Automatic sensors were positioned at three levels (on the ground, at the effective height and at 2 meters above the ground). According to crop developmental stage period was divided to three (four) parts. Air temperature and relative humidity in vertical profile of canopy differed significantly in dependence on year, wheat or rape developmental stage and time of day. The differences in vertical stratification of air temperature and relative humidity were pronounced especially during the light part of the day. In this time the temperature in ground part of canopy was significantly lower and the relative humidity was significantly higher. Air temperature measured in wheat or rape canopy was compared with data from standard meteorological stations, also. The results can be used in precision of prediction methods of some harmful agent's occurrence.

Materials such as asphalt, polymers or the Earth's crust tend to behave in a way that can be described neither with a model of viscous fluid, nor a model from solid mechanics. There are indeed models capable of capturing these so called viscoelastic phenomena far better, but they are based on the presumption of constant temperature. In many cases, e.g. in the glass industry or in geophysics, the properties of a viscoelastic material strongly depend on temperature. That is why it is precisely these changes that need to be described. There are viscoelastic models used in practice that take into account the material parameters' dependence on temperature, however, they do not consider the viscoelastic nature of the material when describing the temperature evolution. The objective of this thesis is to derive thermodynamically consistent viscoelastic models with temperature dependent parameters and the appropriate evolution equation for temperature, implementation of the models and computing simple test simulations. Numerické simulace teplotního a vlhkostního pole ve dřevě v prostředí se zvýšenou teplotou

The thesis deals with numerical simulation of temperature field with internal heat sources and experimental verification of this model in an environment where the high temperatures are applied (during thermal modification of wood). The work was further extended by analysis of coupled heat and monture transfer in the wood. Designed mathematical models are solved numerically using software COMSOL Multiphysics based on the finite element analysis (FEM). The chemical reactions occur due to decomposition of woodconstituents (cellulose, hemicelluloses and lignin) are taken into account in present study. These reactions are usually not solid in known models. The results of numerical simulations of temperature field, including pyrolysis model, are compared with experimental measurements for verification of the proposed model. Contribution of this thesis is the possible application of the proposed model for the design and optimization of thermal modification Schedule with respekt to wood species and sample dimensions.

Tato práce je zaměřena na zlepšení tepelného managementu mikroelektroniky implementací materiálů na bázi uhlíku, přesněji grafitových fólií, do tepelné architektury mikroelektroniky. Práce začíná vysvětlením současných metod chlazení a výzev v oblasti mikroelektroniky. Poté přechází k běžně používaným materiálům - hliníku a mědi a přidává do výběru i uhlíkové alotropie. Ve druhé části tato práce obsahuje několik příkladů použití pyrolytické grafitové fólie v tepelném managementu mikroelektroniky, prokazuje jeho použitelnost a analyzuje přínosy pro šíření tepla, tepelnou vodivost při dodržení elektrické izolace a možné využití ve flexibilní elektronice.

Title: Comparison of thermal comfort of sleeping bag according to EN 13537 Objectives: Objectives of the proposed work was to measure the temperature inside the sleeping bag during sleeping. Then to compare it with the information given on the labels of sleeping bags. Methods: This work has a experimental character. In our work we have used methods of experiment and field study. These methods were used to sample 7 sleeping bags from various manufacturers. Results: The information reported by manufacturers always do not correspond to real use of sleeping bags. EN 13537 has primarily orientation character. For the choice of sleeping bags we do not recommend to follow according only to the EN 13537. The thesis analyzes the temperature field in construction details in an envelope of a wooden house. The fundamental task is a comparison of steady and unsteady calculation taking into account or disregarding the material properties depending on temperature. The partial task was to design a construction of envelope for wooden building and on the basis of this construction to create 2-D geometric models in the software Comsol Multiphysics which is based on the finite element method. For the calculation of the temperature field is applied boundary condition of the third order with actual values of the exterior temperature in winter. To maximize the credibility of simulation software are included the equation expressing the dependence on the temperature of material constants of timber and mineral wool. As the final result to the temperature field numerical models of the peripheral wall between stationary and non-stationary plot is analyzed mutual description of the behavior of thermal processes. In the results are further clarified the cause of the construction physical phenomena.

This thesis deals with the determination of temperature dependence of motorcycle oils. A review of literature contains knowledge of fluid mechanics, partition information, use, appropriate selection and analysis of lubricants including their production and historical development. It is also characterized by a chapter dealing with the recycling of lubricants, including legal claims needed to be placed on the market. There is also a chapter on the lubricating circuit of motorcycles. The practical part of the detailed specification used oil samples, including technical parameters motorcycles from which they were taken. The data needed for evaluation was obtained by measurement with a rotary viscometer DV2T and apparatus for measuring the density Densito 30 PX. The values so obtained were graphically using Microsoft Excel 2010 for application of a suitable mathematical model.
