The topic of semestral thesis is methods to detect selection in DNA sequences. In the begining of the thesis we will describe molecular evolution. It will be written what made the evolution and how the evolution is shown. Moreover there are gen mutations and mechanisms of diffuse and fixation. It will be defined what pozitive, negative and neutral selection is. The thesis is focused on evolution distance of synonymous and nonsynonymous substitution. There will be described three methods – Nei-Gojobori, Li-Wu-Luo and Comeron. All these methods will be described with mathematic formulas. There will be statistic test to decide what kind of selection ti is – there will be used z-test. In the practical part, there will be information about developed software what counts selection pressure from sequences from databazes in format GenBank and it shows parts where selection is. The software will be used for two data sets with two different genetic codes. The result will be discussed. We will discuss results of all three methods of selection pressure and influence of input parametrs.

The first part of this work is about history of infusion technology, specially about recent developments in electronic infusion devices. Then, there are described basic principles of infusion pumps and syringe pumps. The second part introduces safety features of infusion technology. An occlusion alarm is one of them. There is described a method of measurement that verifies the functionality of this alarm in two instruments of different producers with various settings. Data obtained by this method are statistically processed and the results discussed.

Anesthetics cause profound alterations in respiratory and cardiovascular systems. Our experiments demonstrated that different anesthetics caused different changes in blood pressure regulating components. The role of particular BP regulating systems was disclosed by their selective inhibition - sympathetic nervous system blocked by pentolinium (peripheral ganglionic blockade), renin-angiotensin system by captopril (angiotensin converting enzyme blocker) and nitric oxide production by L-NAME (nitric oxide synthase blocker). Components of blood pressure regulating mechanisms in conscious normotensive Wistar rats and spontaneously hypertensive rats were compared with four different groups of anesthetized rats by pentobarbital, ketamine-xylazine, chloralose-urethane and isoflurane. Each anesthesia caused different hemodynamic changes. If hemodynamic conditions should be similar to conscious rats, the most suitable anesthetic is pentobarbital. L-serine-induced effects represent endothelium-derived hyperpolarizing factor (EDHF)-mediated response, which is a type of endothelium-dependent regulation of vascular tone, independent of nitric oxide and prostacyclin production. The study is focused on advanced life support (ALS) performed by rescue team physicians in an out-of-hospital setting. The first part of the study analyzes diagnostic possibilities and correctness of assumed aetiology of the sudden cardiac arrest during cardiopulmonary resuscitation (CPR) in the field. It introduces an original method of "Crosscheck Tables" and applies this method to 211 cases of CPR provided by physicians of the Emergency Care Service. The study demonstrates that significantly misleading conclusions can result from a global analysis of a set of cases. It stresses importance of diagnostic analyses applied to individual cases. The second part of the study concentrates on changes in PETCO2 level as related to the return of spontaneous circulation (ROSC - Return of Spontaneous Circulation) as opposed to the circulation fully dependent on chest compressions. The study demonstrates that in constantly ventilated patients undergoing CPR in an out-of-hospital setting, PETCO2 is significantly higher (about 10 mmHg) after ROSC than before ROSC. It demonstrates that a sudden increase in PETCO2 exceeding 10 mmHg is likely to indicate the moment of ROSC. Methods are evaluated teoreticly as well as practically by companing results of experimental testing. Thesis also includes description of created model which evaluates acoustic measurement acording to the definitionof point method. The model is designed for measuring chain of Engineering Test Institute, Public Enterprise (SZU).

This thesis focuses on the design of new high pressure injection system application on diesel engine. The design came from company Zetor’s demands. For this application was chosen system Common Rail supplied by company Bosch. Assignment was separated into several tasks. Research of suitable components for low pressure section, design of waste system and high pressure injection tubes design including injector clamping. Due to injectors used in this system was necessary to redesign cover for valves and even new system to lead wires from outside of the engine to injectors. Injector is attached by screw connection, which is composed from screw and clamping jaw. These parts are subjected to an analytic calculations. Clamping jaw of injector is even subjected to a FEM analysis, which focuses on fatigue life. Finally, the drawings were documented.

Main goal of this work is familiarization with basic features of absorption heat pumps and it’s growing importance on the market. In the first part of the work there is a research for clarifying facts associated with such pumps. In the introduction there are descriptions of basic information from the field of heating industry and its connections to our systems. Next there is a brief analysis of price changes in electricity and heat in the last five years. Main part of the analytical section is a description of the used absorption system. Second part of the work is focused on the calculation of the absorption system itself. First there is calculated internal and external part of the device according to the parameters from GE Power s.r.o. After this step there are parametric studies based on changes of key parameters of the default device for a demonstration of the behavior of a heating pump. In the end of the work there is a integration of the unit into the heating cycle for variations including flue gas condenser and using a cooling water from the turbine condenser.

The thesis deals with a description and a static and dynamic analysis for an existing construction of lookout tower. The steel watchtower is located in Město Albrechtice and is made up of two lattice towers connected by a bridge. Main point was to create calculation models for the purpose of dynamic and static analysis, in the software SCIA Engineer 19.1, which is based on the principle of finite element method. Attention was paid to load created by winds according to Eurocode 1 and Eurocode 3, glaze and rime ice load and dynamic coefficient calculation. The thesis also includes an assesment of selected parts of the construction according to Eurocodes.
