Metabolomics investigation of microbial cell factories

The stream of new technological advancements and their integration into the field of microbiology have contributed significantly towards our understanding of life in the micro-scale world, making the fields of microbiology and biotechnology shine like never before. Since 1980, the recombinant protein-based therapeutics industry has become one of the fastest growing sectors in the biopharmaceutical market. Nearly 30% of commercially available recombinant proteins are produced in Escherichia coli, making this species one of the most commonly used bacterial expression systems for the production of recombinant biotherapeutics. However, when it comes to the production of enzymes and bioactive secondary metabolites (antibiotic, antifungal, antiviral and immunosuppressant), Streptomyces species remain the major producer within this sector. Meeting the high demand for such products requires a clear and in-depth understanding of the bioprocesses involved to achieve high yield and quality products, whilst keeping the process industrially attractive. It is generally accepted that the metabolome, as a down-stream process to the genome and proteome, may provide a clearer picture of a biological system. Thus, in this thesis a series of metabolomics approaches were adopted to obtain a deeper insight into the metabolic effects of recombinant protein production in E. coli and Streptomyces lividans. Furthermore, a Geobacter-based biomagnetite nanoparticle production system which displayed a prolonged lag phase upon scale-up was investigated by employing metabolic profiling and fingerprinting approaches combined with multivariate analysis strategies, to identify growth-limiting metabolites. The results of this analysis identified nicotinamide as the growth limiting metabolite. Nicotinamide-feeding experiments confirmed the above findings, leading to improved biomass yield whilst restoring the lag phase to bench-scale level. Raman and Fourier transform infrared spectroscopies combined with stable isotopic probing strategies were also employed to demonstrate the application of metabolic fingerprinting in providing detailed biochemical information for quantitative characterisation and differentiation of E. coli cells at community and single-cell levels. The single-cell approach proved promising, offering detailed biochemical information and perhaps accompanying other cultivation-free approaches such as metagenomics for further future investigations. It is hoped that the advances made in these studies have proved the potential applications of metabolomics strategies to aid the optimisation of microbially-driven bioprocesses.

A new method to achieve lithic use-wear discrimination using laser scanning confocal microscopy

My study sought to acquire quantitative data from the surface of lithic tools and use that data to discriminate tools used on different contact materials. An experimental archaeological wear production method was conceived, whereby I and several volunteers produced wear on chert, heat-treated chert, and obsidian flakes by using those flakes on several contact materials. The flakes were then analyzed using a laser scanning confocal microscope, which recorded three-dimensional surface data from each tool. The data was analyzed using cluster analysis to find the ideal combination of parameters which correctly discriminated the flakes based on use-wear data. After finding acceptable parameters which grouped flakes appropriately through cluster analysis, those groups were subjected to a discriminant analysis. Each analysis returned a p-value under .05, meaning that the clustering based on the parameters Sq and Sfd produced by the cluster analysis was not random, but indicative of these variables' ability to discriminate lithic use-wear. Formal variation in lithic projectile armatures: re-interpreting points from Tabun Cave, Israel

Lithic projectile armatures represent a significant innovation over thrusted spears in the subsistence strategies of hominins. Previous researchers have disagreed over the timing of the appearance of projectile weapons in the archaeological record (Brooks 2006; Shea 2006). To discover when projectile technology first appears in the Levant, I have compared tip cross-sectional areas, weights, and tip penetrating angles (three variables useful for discriminating between projectile and thrusting weapons) of pointed Blades, Levallois points, and Mousterian points with analogs from known and suspected chipped stone projectile points. Results indicate that pointed Blades from Tabun and Skhul caves are statistically indistinguishable from other suspected projectile point types. Levallois and Mousterian points from Tabun and Skhul are also statistically indistinguishable from suspected projectile types when the lower halves of the populations are compared. Consequently, I conclude that points from Tabun and Skhul caves fall within the known and suspected range of variation of projectile point morphology. / by David E. The major objective has been to create tools for analyzing the reasoning regarding the school the different actors involved - pupils, teachers, school and politicians show. A subordinate aim was to explore relevance in this connection of eight studies carried out by the author 1979 – 1999 concerning three basic areas: the pedagogical processes at the school level, at the teacher education level and at the level of school change. I conclude that a recurring question is that of the relevance and the sense of meaningfulness experienced by those involved in different educational processes. In one of the eight empirical studies, termed the compulsory-school teacher study (1997), an approach to analyzing what teacher trainees find relevant in their work in schools is presented, involving two basic concepts, those of the event space and the relational space. In considering these concepts in terms of Alfred Schutz' theory of the lifeworld, I rename the relational space the space of contemporaries. A new study is also reported in this thesis – Teachers and schoolpoliticians in the pedagogical process of school change, the LoP-study. In considering the earlier eight studies in conjunction with the LoP-study, I find the studies to have dealt with personal processes and societal processes as well as pedagogical processes in how the parties involved reason in talking about the school. Three different aspects of the processes are distinguished: meta-aspects, connected with the aims a given process has; core aspects, connected with what one concretely does; and approach aspects, pertaining to how the persons involved relate to each other and to the process. In analyzing the LoP- interviews carried out with use of the tools developed – enabling different processes and aspects of these to be analyzed – I find marked individual differences in the patterns of different aspects of the processes involved. The width of the event space and of the space of contemporaries on the part both of the teachers and of the politicians were found to vary considerably. A conclusion drawn is that further development of the tools created would be worthwhile. It also seems possible to apply these tools to other areas, such as those of the scientific community with its research processes and of the political community with its steering processes.

The subject of this diploma thesis is the design development of marketing communication to enhance brand competitiveness in company focused on fast moving consumer goods. Concretely the Knorr, food brand of Unilever company. The first part provides a theoreti-cal basis for the processing of the issue. In the practical part I have assessed the current marketing communication by using inter alia SWOT, PEST and Porter's analysis. Based on analysis of the current situation, I propose to improve marketing communications, including analysis of time, costs and risk.
