Defeito bioquímico em fator V, fator V de Leiden, em paciente jovem com trombose venosa profunda : estudo de caso

Um fator precipitante como o trauma pode desencadear a trombose venosa profunda (TVP) em pacientes jovens portadores da mutação FVR506Q ou Fator V de Leiden, a causa mais comum de trombofilia hereditária. Foi estudado um paciente de 15 anos, SBV, sexo masculino, que aos 13 apresentou TVP após trauma e seus pais assintomáticos. A família refere que os avós paternos do paciente morreram por conseqüência de trombose. Foram utilizados kits reagentes para atividade de Proteína C (PC), Proteína S (PS), Antitrombina III (AT III), determinação do fibrinogênio, Resistência à Proteína C Ativada (RPCA), Tempo de Protrombina com Atividade Enzimática e Tempo de Tromboplastina Parcial Ativada (IL); e kits comerciais para purificação do DNA (GENTRA), e pesquisa do FVL e do FII G20210A por PCR. O paciente e seu pai apresentaram RPCA e a mutação FVR506Q em heterozigoze. Os demais testes foram normais. Não foi identificada a mutação da protrombina G20210A na família. A mãe de teve todos os resultados normais. O FVL representa menor risco para trombose que outros defeitos como deficiência de PC, PS, ou AT III. No entanto, quando associado com outro distúrbio congênito ou adquirido, o risco para TV aumenta significativamente. S.B.V. teve pelo menos três fatores: o FVL, o exercício forçado e o trauma, que parecem ter tido papel desencadeador da TVP neste caso. Seu pai tem a mutação do FVL e forte história familiar, é recomendável que receba profilaxia em situações de risco, e o acompanhamento prospectivo. O trabalho indicou a necessidade de mais pesquisas em bioquímica da Hemostasia, do conteúdo gênico brasileiro pela sua grande diversidade étnica, das interações genéticas entre si e com o meio ambiente e suas manifestações fenotípicas

Padronização, otimização e caracterização bioquímica/biofísica da expressão de NPP1 e Anexina V / Standardization, optimization and biochemical/biophysical characterization of the expression of NPP1 and Annexin V

O processo de biomineralização óssea é a deposição de cristais de fosfato de cálcio, na forma de hidroxiapatita formando o tecido ósseo. São mediados pelas vesículas da matriz (MVs) que são liberadas no local específico do início da biomineralização. As MVs contêm altas concentrações de íons Ca2+ e fosfato inorgânico (Pi), proporcionando um microambiente adequado para a formação inicial e propagação dos cristais de hidroxiapatita. Para que isso ocorra corretamente são necessárias diversas proteínas/enzimas, bem como microambientes com condições específicas. Neste trabalho uma atenção especial foi dada a duas proteínas presentes nas MVs: Anexina V (AnxA5) e a nucleotídeo pirofosfatase/fosfodiesterase-1 (NPP1). A Anexina V é responsável pela formação de um canal de cálcio por meio da associação desta proteína tanto com a face externa quanto interna da membrana das MVs. A NPP1 possui a função principal de hidrolisar adenosina trifosfato (ATP), formando adenosina monofosfato (AMP) e pirofosfato (PPi). Assim, experimentos de expressão de Anexina V e NPP1 foram realizados com o intuito de iniciar os estudos de interação entre as duas proteínas, por meio de reconstituição em lipossomos. A expressão de Anexina V foi realizada em células E. coli BL21(DE3) e induzida por isopropil -D-1-tiogalactopiranosídeo (IPTG); para purificação, três procedimentos foram necessários, utilizando coluna de níquel, coluna Desalting e coluna de troca iônica (Mono-Q). Experimentos de dicroísmo circular foram realizados com amostras de Anexina V após purificação e mostraram que todas as amostras apresentavam estruturas em -hélice. Pelo método da gota pendente foi estudada a interação de Anexina V com íons Ca2+ (10 mM) em monocamadas constituídas por dipalmitoil fosfatidilcolina (DPPC) e de uma mistura de composição lipídica 9:1 de DPPC e dipalmitoil fosfatidilserina (DPPS). Os dados obtidos mostraram alta afinidade de Anexina V por monocamadas constituídas de (9:1) DPPC:DPPS na presença de íons Ca2+. A expressão de NPP1 foi realizada com transfecção por meio de eletroporação do DNA recombinante em células COS-1, e seleção com antibiótico G418 após 24 horas de cultivo. Amostras de fração de membrana controle e NPP1 recombinante foram preparadas após 60 horas de cultivo celular e foi observada atividade catalítica na amostra de fração de membrana da NPP1. Todas as amostras de expressão, tanto de Anexina V e NPP1, foram analisadas por eletroforese em gel de poliacrilamida. A padronização da Anexina V foi obtida com sucesso, porém com relação à NPP1, experimentos ainda devem ser realizados a fim de padronizar a obtenção desta proteína recombinante em quantidade suficiente para continuar os estudos. / The bone biomineralization process is the deposition of calcium phosphate crystals in the form of hydroxyapatite forming the bone tissue. They are mediated by matrix vesicles (MVs) that are released at the specific site of the onset of biomineralization. MVs contain high concentrations of Ca2+ ions and inorganic phosphate (Pi), providing a suitable microenvironment for the initial formation and propagation of hydroxyapatite crystals. To occur properly, several proteins/enzymes are needed, as well as microenvironments with very particular conditions. In this work, special attention should be given to two proteins present in the MVs: Annexin V (AnxA5) and nucleotide pyrophosphatase/ phosphodiesterase (NPP1). Annexin V is responsible for the formation of a calcium channel through the association of this protein with both the outer and the inner face of the MVs membrane. NPP1 has the main function of hydrolyzing adenosine triphosphate (ATP), adenosine monophosphate (AMP) and pyrophosphate (PPi).Thus, experiments of expression of Annexin V and NPP1 were performed with the aim of initiating the interaction studies between the two proteins, through reconstitution in liposomes. Annexin V expression was performed in E.coli BL21 (DE3) and the cells were induced by isopropyl -D-1-thiogalactopyranoside (IPTG); for purification, three procedures were required using a nickel column, a Desalting column and an ion exchange column (Mono-Q). Circular dichroism experiments were performed with Annexin V samples after purification and showed that all samples contain -helix structures. Using the pendant drop method, the interaction of Annexin V with (10 mM) Ca2+ ions was studied in monolayers composed of dipalmitoyl-phosphatidyl-choline (DPPC) and a mixture of lipid composition 9:1 DPPC and dipalmitoyl-phosphatidyl-serine (DPPS). Data showed high affinity of Annexin V by monolayers constituted of (9: 1) DPPC:DPPS in the presence of Ca2+ ions. NPP1 expression was performed with transfection by electroporation of the recombinant DNA into COS-1 cells and selection with G418 antibiotic after 24 hours of culture. Samples of the control membrane fraction and recombinant NPP1 were prepared and the activity of the membrane fraction of NPP1 was observed in the samples. All expression samples, both AnxA5 and NPP1, were analyzed by polyacrylamide gel electrophoresis. The standardization of Annexin V has been obtained with success, but regarding NPP1, experiments have yet to be performed to standardize the production of this recombinant protein and to obtain enough quantity to continue the study.

'Foreign' books for English readers : published translations of navigation manuals and their audience in the English Renaissance, 1500-1640

Although there has been an increasing interest in the study of Renaissance translations and the early world of print, the history of navigation and exploration has not been the subject of any such in-depth bibliographical research. This thesis identifies and analyses a corpus of translated navigation manuals and related works that were printed in England between 1500 and 1640. The context is sketched by defining the different areas of maritime writing found in Renaissance England. Although English contributions were particularly strong in such topics as the mathematical side of navigation, the technical instruments and the debates about magnetism and compass variation, publications of manuals and sailing directions were scarce. This thesis reveals that such knowledge was imported from continental Europe through translation. Forty-three translations out of seven different source languages are discussed from a book-historical perspective to establish what their source text was, how they came to England and who was responsible for translating and publishing them. Such information was obtained, in part, from a study of the paratexts, in particular the translators’ and publishers’ dedications and addresses to the reader, which show the reason and purpose of the translations, the methods employed and particular problems encountered, as well certain linguistic and rhetorical characteristics. One work is selected as a case-study for in-depth research, namely Martin Cortés’s Breue compendio de la sphera y de la arte de nauegar (1551) and its translation by Richard Eden, The Arte of Navigation (1561), which went through ten editions and became the model for English navigation manuals. Finally, by turning to the agents involved in the production and dissemination of these translations, particularly the printers and booksellers, and establishing the connections between them, this thesis reveals intricate social networks and sheds new light on certain aspects of the fields of navigation, translation and print.

Progress Toward the Total Synthesis of the Highly Selective Cytotoxic Natural Product, Maoecrystal V

Strategies to synthesize the natural product maoecrystal V have been investigated. The initial strategy involved a tandem Michael-aldol reaction to form the [2.2.2] bicyclic core of maoecrystal V. This proposed route was not successful. A modified route to maoecrystal V, inspired by studies on the aldol ring closure reactions, enabled the synthesis of a complex intermediate that allowed for the formation of the core structure. Further elaboration of this key intermediate afforded the methodology to form four of the five rings in maoecrystal V. Additionally, this key intermediate allowed for further modifications that can potentially be an entry point toward an enantioselective synthesis of maoecrystal V that intersects with the initial synthesis of the racemic compound.

"So barabarous a practice" : Cornish wrecking, ca. 1700-1860, and its survival as popular myth

The popular myth of Cornish wrecking is well-known within British culture, but there has not been a comprehensive, systematic inquiry to separate out the layers of the myth from the actual practices. This study rectifies this omission by examining wrecking activity as reported in popular sources and traditional tales; deconstructing the most widely believed elements; illuminating the complexity of the practices; and investigating the process of myth-making which sustained the image of the wrecker in popular consciousness. It suggests that violent wrecking was not nearly as widespread and invidious as popular histories allow. The coastal populace had their own popular morality, including the use of mediation and constraint, which allowed them to practise wrecking, salvage, and lifesaving activities simultaneously. They did not condone all forms of wrecking; thus it cannot be deemed a 'social crime'. Wreckers did not escape conviction because of local resistance to centralised authority, but as a result of the complex legal practices of discretion that were incorporated into the eighteenth century English criminal justice system. The role of the lord of the manor was also more complex; their relationship with the coastal populace was based on reciprocity as well as antagonism. However, the tightening of governmental control and increasing bureaucratisation in the Victorian period resulted in the loss of customary wreck rights for both the coastal inhabitants and the local elites. At the same time, the press and pulpit were the primary conduits for establishing and popularising the wrecker stereotype through symbolic violence and moral panics. The stereotype became reflexive, touted as an accurate description in Victorian histories, and thus burying the reality of wrecking under accretions of moralising discourse. Therefore, the process of historical 'beach combing' across the disciplinary boundaries has revealed wrecking as a multi-faceted, sophisticated cultural practice and cultural construct.
