MicroRNAs (miRNAs) are small non-coding RNAs with a length of 18 to 25 nucleotides playing a pivotal role in post-transcriptional regulation of gene expression. There are miRNAs whose expression is limited to a certain tissue type and diseases which are characterized by a unique miRNA expression profile. I assumed spontaneous preterm birth (PTB) and preterm prelabor rupture of membranes (PPROM) would be characterized by a unique miRNA expression profile. I observed the gene expression of 15 placental specific miRNAs (miR-512-5p, miR-515-5p, miR-516b-5p, miR-517-5p, miR-518b, miR-518f-5p, miR-519a-5p, miR-519d-3p, miR-519e- 5p, miR-520a-5p, miR-520h, miR-524-5p, miR-525-5p, miR-526a and miR-526b-5p) in placental tissue of pacients with PTB, PPROM and women with term in labor pregnancies (FG). PTB group consisted of 24 pacients, PPROM group of 75 pacients and FG group of 20 pacients. Quantitative real-time PCR was used to quantify gene expression. In the group of PTB pregnancies I identified 3 significantly upregulated miRNAs (miR-516b-5p, miR-519d-3p and miR-524-5p) and 4 miRNAs (miR-518b, miR-519a-5p, miR-520h and miR-526a) with a trend to upregulation compared to controls (FG). The aim of this thesis is an overall assessment of the hydrogeological conditions of the Český Brod Permian Carboniferous and overall the balance of upper groundwater aquifer in the northern part of the Český Brod Permian Carboniferous using a mathematical model. For the realization of the model was used ZABAGED data provided by the Czech Office for Surveying, Mapping and Cadastre, which is located in Prague 8 Kobylsích. The mathematical model was created using computer applications such as FeFlow 5. 2, ArcGIS for Desktop 10. 1 and Surfer 10. 7. 972. The result of this work is to evaluate the overall balance of precipitation, surface water and groundwater in the upper aquifer north of Český Brod Permian Carboniferous, respectively, in the hydrologic basin of the Upper and Middle Elbe, in the hydrological basin fourth Order river Vyrovka (HEIS.VUV.cz). The evaluation was done by comparing the data obtained from the studied opinions and based on data obtained from calculations of the mathematical model. Title: Spontaneous activity of lower limbs in a vertical position in the aquatic environment Objectives: The aim of this study was to determinate whether spontaneous activity of lower limb in the aquatic environment enable to determinate preferred swimming stroke of an individual. Also, whether identified asymmetry of the lower limbs during treading water depend on passive range of hip joints. Methods: Spontaneous activity of lower limbs in a vertical position with a hands and head out of water was recorded on video. Passive intra/extrarotion, flexion, extension, abduction in hip joints were measured using mechanical two-arm goniometer. Qualitative analysis of spontaneous activity of lower limbs recorded on video was conducted using Kinovea 0.8.15. Results: According to the results of our work, it can not be said that the preferred swimming stroke can be determined on the basis of the spontaneous activity of the lower limbs in a vertical position in the aquatic environment. We can assume that the individual chose the pattern of treading water according to an energetically economical way while minimizing resistive forces and not in pursuance of preferred swimming stroke. The aim of this thesis is to monitor how the amount of water (water activity) affects the amount of microorganisms is selected packaged meat products from the point of view standards, which specify their specific requirements for the health safety of these products. The first part of this work describes various types of packaging materials of meat products, the microorganisms that occur in them and the definition of water activity. The second part describes the actual measurement of water activity and the measurement of the occurrence of microorganisms and coliforms, from selection of suitable meat products, their types of packaging, sample preparation and evaluation of measured methods. The results showed, that the activity of water affects the occurrence of microorganisms and coliforms, in that the higher the activity of water, the higher microorganisms presence.

Hydrologické sucho v pramenných oblastech Krušných hor / Hydrological drought in headwater areas of the Ore Mountains

Changes in the hydrological cycle are one of the expected impacts of climate change. Until recently, Central European mountain ranges have not been considered to be affected by water scarcity. Nevertheless, recent years have suggested an increasing risk of drought occurrence also in these regions, which have a major impact on the water supply to rivers that drain them. The master thesis deals with the evaluation of hydrological drought in the headwater areas of three catchments in the Ore Mountains for the period from 1967 to 2018. The main aim is to compare the of hydrological drought characteristics in the catchment areas of upper Svatava River, upper Rolava River and Načetínský brook based on available hydrological and climatic data. In connection with this, the work aims to detect the long-term trends of drought occurrence in selected basins. Furthermore, the emphasis is given on the evaluation of natural conditions, especially the historical and current land cover changes in the study catchments. The results point to a significant change in the seasonality of the occurrence of hydrological drought in the second half of the studied period, when the streamflow deficits concentrate between August and October. COMPARATIVE STUDY OF FLUVIAL LAKES IN FLOODPLAINS OF THE ELBE, LUŽNICE AND SVRATKA RIVERS Petra Havlíková ABSTRACT The aim of the thesis was to specify key differences in chemistry and biota (zooplankton communities) among fluvial lakes in three regions of Czech Republic: "střední Polabí" (central part of the Elbe River on the territory of Bohemia), "Horní Lužnice" (the upper part of the Lužnice River on the territory of Bohemia), and the Svratka River near Milovy (upper part of the Svratka River). The 10 studied lakes of the three regions differ in size, geology, shading, the influence of the river, and the level of anthropogenic impact. The following hypotheses were tested: 1) The chemical composition of the water in fluvial lakes is significantly different in different areas (floodplains). In the central Elbe River floodplain, there are the highest values of conductivity and concentrations of organic matter and nutrients. This dissertation thesis focuses on the removability of algal organic matter (AOM) by coagulation during water treatment and also on the influence of AOM on the coagulation of other substances present in source water. Special emphasis is put on the description of coagulation mechanisms. The effectiveness of AOM removal by coagulation was investigated by coagulation tests performed with optimized doses of coagulants (aluminium or ferric sulphate) under different pH values. Peptides and proteins contained in cellular organic matter of cyanobacterium Microcystis aeruginosa were used in the experiments since they have been previously reported to disturb the coagulation process. Moreover, peptides and proteins underwent coagulation experiments together with kaolin particles, representing clay particles in turbid waters, in both the presence and absence of coagulants to investigate the effect of AOM on the coagulation of turbid waters. To enable the description of coagulation mechanisms, AOM were characterised in terms of charge, functional groups, molecular weight and ability to form dissolved complexes with coagulant metals. The bankruptcy problem involves the distribution of perfectly divisible goods. Particular attention is paid to the situations, where the amount of goods available is not enough to cover the demand. An example of real life situations that can be solved using various bankruptcy rules may be a division of a heritage or when a company goes bankrupt and its estates are sold to satisfy interested parties' claims. This paper introduces to the problem a linear structure of the participants, meaning that participants are now satisfied one after another in a preset order. It applies the equal awards (CEA) and the equal losses (CEL) solutions on the revised problem. Since their axiomatization is no longer valid, both solutions are extended and new characterizations are given in the thesis. The thesis contains a series of original proofs for both extended solutions and whole problem is examined in the setting of the well-known river sharing problem.

In this thesis, water quality and the assessment of anthropogenic pollution in the sediments of the middle course of the Elbe River oxbow lakes Kozelská and Vrť were studied. It is widely accepted that the oxbow lakes are extremely significant ecosystems, However, a large amount of contaminated material may deposit in these lakes. The pollution probably comes from industrial sources of contamination from the second half of the 20th century. In addition, the oxbow lakes show the development of the riverbed, and contribute to the stability of the river ecosystem. The research of Lake Kozelská was chosen especially to its proximity to the chemical factory Spolana in Neratovice, which used to be the biggest source of pollution of the Elbe River. The research included bathymetric measurements, regular observations of hydrological regime, monthly analysis of chemical and physical parameters of water in the period from December 2016 to November 2017, and marginally microscopic analyses of phytoplankton and zooplankton species too. The next part of this research included grain analysis and determination of metal and arsenic concentrations in the sediment fraction of 20µm using Aqua Regia leaching and total decomposition as well.
