Fatty Acids Profiles of Yellow Perch (Perca flavescens) in Lakes of the Outaouais Region with and without Largemouth Bass (Micropterus salmoides) and Smallmouth Bass (Micropterus dolomieu)

Fatty acids (FAs) are used as trophic markers in aquatic food web studies, but few studies have quantified individual variability in FAs profiles over several sites in a range of conditions. I investigated whether FAs profiles of yellow perch (YP), Perca flavescens, vary with body size and between lakes with and without largemouth (Micropterus salmoides), and smallmouth bass (Micropterus dolomieu), the most common and abundant piscivores in lakes of the region. I analyzed the FAs of YP as well as zooplankton, benthic invertebrates, and prey fish collected from eight lakes where bass were either present or absent in the Outaouais region over the summer of 2016. I compared the growth rate of YP between the lakes and the YP in lakes without bass exhibited a slower growth rate. I also compared the FA signatures of YP using redundancy analysis (RDA). 23 FAs could be identified and quantified. FAs profiles were dominated by palmitic- (16:0), oleic- (18:1), stearic- (18:0), and palmitoleic acid (16:1). The RDA analysis based on FAs profiles of YP revealed variation along two main gradients (the presence of bass and the date of capture). The first two eigenvectors accounted for 42.1% of the variation (RDA1=27.6% and 2=14.6%). Arachidonic (20:4) and docosatrienoic (22:3) were the most correlated FAs with RDA1. Due to the sampling period, it was impossible to determine if the observed effects were due to the date of capture, the presence of bass, or a change in metabolism, but the last two were deemed as the most plausible explanations. It was concluded that the utility of FA signatures to quantify diet in natural environments is limited and that FAs might be more successful as markers in primary consumers and other lower trophic levels. It is recommended that a combination of FAs, stable isotopes, and modelling should be used in the future.

The historical development and a structural analysis of the Yellow River piano concerto

The Yellow River Piano Concerto was adapted from the famous Yellow River Cantata, a grand choral work composed by Xian Xinghai in 1939, during the Japanese invasion of China. It is a very famous and important work in China and exhibits strong nationalistic characteristics. The Cantata inspired the entire Chinese nation during their defense against the largest and most brutal genocide in human history. It has since become a symbol of heroism and solidarity of all Chinese people around the world. The Yellow River Piano Concerto was composed by a group of Chinese composer-pianists in 1969 led by Yin Chengzong. All the biographies of the composers of both the Cantata and the Piano Concerto are supplied in this dissertation. The Piano Concerto consists of four movements: The Song of The Yellow River Boatmen, Ode to the Yellow River, The Wrath of the Yellow River and Defend the Yellow River. Each movement has been analyzed in terms of structure. Disease and Hygiene in the Construction of a Nation: The Public Sphere, Public Space, and the Private Domain in Buenos Aires, 1871-1910

The maturation of the public sphere in Argentina during the late nineteenth and early twentiethcenturies was a critical element in the nation-building process and the overall development ofthe modern state. Within the context of this evolution, the discourse of disease generatedintense debates that subsequently influenced policies that transformed the public spaces ofBuenos Aires and facilitated state intervention within the private domains of the city’sinhabitants. Under the banner of hygiene and public health, municipal officials thusEuropeanized the nation’s capital through the construction of parks and plazas and likewiseutilized the press to garner support for the initiatives that would remedy the unsanitaryconditions and practices of the city. Despite promises to the contrary, the improvements to thepublic spaces of Buenos Aires primarily benefited the porteño elite while the efforts to rootout disease often targeted working-class neighborhoods. The model that reformed the publicspace of Buenos Aires, including its socially differentiated application of aesthetic order andpublic health policies, was ultimately employed throughout the Argentine Republic as theconsolidated political elite rolled out its national program of material and social development.

Sequence Analysis of Maize Yellow Stripe3 Candidate Genes

The work presented here focuses on the molecular mechanism of phytosiderophore secretion in graminaceous plants. In maize, yellow stripe3 (ys3) is a mutant that is deficient in its ability to secrete iron-chelating compounds of the mugineic acid family known as phytosiderophores. Phytosiderophores are specific to grasses and are used for the acquisition of iron. Genetic linkage mapping of the ys3 locus lead to a region of interest on chromosome 3 defined by marker UMC1773. The sequence of eleven candidate genes (GRMZM2G390345, GRMZM2G390374, GRMZM2G342821, GRMZM5G800764, GRMZM2G502560, GRMZM5G849435, GRMZM2G105766, GRMZM5G876835, GRMZM2G036976, GRMZM2G502563, miR167g) revealed several small deletions and point mutations within the coding regions of four candidate genes (GRMZM2G390345, GRMZM2G342821, GRMZM2G105766 and GRMZM2G502560). To further investigate these candidate genes, knockout (Mutator) lines were used in an effort to recapitulate the ys3 phenotype. I characterized a GRMZM2G390345 mutant and determined that the mutation in GRMZM2G390345 did not cause the ys3 phenotype. For the three remaining candidate genes, GRMZM2G342821, GRMZM2G105766 and GRMZM2G502560 further analysis is needed. The closest homolog of OsTOM1 (an effluxer of muginieic acid) GRMZM2G603306 which, lies ~7 Mbp outside the linked marker, has also been suggested to be yellow stripe3. Preliminary sequence analysis uncovered a 4 bp insertion within the 9th exon of this gene that shifts the reading frame and leads to a premature stop codon. Three of the genes analyzed remain plausible candidates for yellow stripe3.

Formins are proteins involved in regulation and construction of actin filaments of eucaryotic organism. They parcipitate in regulating cytokinesis, polar tip growth, and thus participate in development of whole organisms. There are 2 classes of formins in Arabidopsis thaliana. Both classes include FH1 and FH2 domains (formin homology 1 a 2). Class I formins have N-terminal transmembrane domain, unlike class II formins. Some formins of class II have a N-terminal PTEN domain (Phosphatase and Tensin Homolog). Sequence analyses predicted membrane binding via phosphatase or C2 subdomain of PTEN. This thesis was focused on the formin AtFH14, specifically its PTEN domain. Based on predicted sequence, a DNA fragment encoding the PTEN domain was amplified, sequenced and cloned to destination vectors for YFP and EOS phusions. Marked protein was visualized by transient expression in Nicotiana benthamiana. Stably transformed Arabidopsis lines were prepared for stably expression of protein. The tagged protein was localized in cortical cytoplasm, cytoplasmatical strands, probably in nuclear membrane or perinuclear cytoplasm, as well as in peculiar "folicle-like" structures that might be due to binding of PTEN at the periphery of some membrane organelles. Differentiating Microtus Xanthognathus and Microtus Pennsylvanicus Lower First Molars Using Discriminant Analysis of Landmark Data

The distinct ecological requirements of Microtus xanthognathus (yellow-cheeked vole or taiga vole) and M. pennsylvanicus (meadow vole) warrant accurate discrimination of their remains in studies of paleoecology and past biogeographical shifts. An occlusal length of the lower 1st molars (ml) that is >3.2 mm for M. xanthognathus is the method most frequently used to separate these 2 taxa in archaeological and paleontological samples. However, these measurements alone are unreliable because some specimens of M. pennsylvanicus overlap smaller individuals of M. xanthognathus in size. Therefore, I created and tested a morphometric technique that discriminates Recent lower 1st molars (mis) of M. pennsylvanicus from those of M. xanthognathus, and is applicable to other taxa (both modern and fossil). Despite overlapping occlusal length, my discriminant function based on landmark data correctly classified 100% (n = 53) of Recent m1s from the 2 taxa and 97.7% (43 of 44) of (assumed) m1s of M. pennsylvanicus from an archaeological site from about AD 1200 in central Nebraska. This landmark scheme is applicable to fossil and modern Microtus worldwide. © 2006 American Society of Mammalogists.

Agreement and Screening Accuracy Between Physical Therapists Ratings and the Ӧrebro Musculoskeletal Pain Questionnaire in Screening for Risk of Chronic Pain During Musculoskeletal Evaluation

Introduction: Identifying patients at risk for chronic musculoskeletal pain can inform evaluation and treatment decisions. The ability of physical therapists to assess patients’ risk for chronic pain without use of validated tools has been questioned. The Ӧrebro Musculoskeletal Pain Questionnaire (OMPQ) is used to determine risk for chronic pain. Methods: The aim of this pragmatic study was to prospectively quantify the agreement between physical therapists’ assessment of patients’ risk for chronic symptoms compared to the OMPQ. Patients were asked to complete the OMPQ during the initial visit. Physical therapists, blinded to OMPQ risk classification, carried out their usual patient assessment procedures. The physical therapists rated patients as either high or low risk for chronic pain based on their clinical assessment. Agreement between therapist and OMPQ was determined using Cohen’s Kappa (κ) and screening accuracy compared clinician risk to the OMPQ risk classification (reference standard) by way of contingency table analysis. Results: Ninety-six (96) patients’ risk classifications and 15 corresponding physical therapists’ risk estimates were available for analysis. The OMPQ identified a 47% prevalence for high risk of chronic pain. Agreement (κ and 95% confidence interval) between physical therapist rating and OMPQ was slight, κ = 0.272 (0.033–0.421), p = .026. Therapists’ sensitivity and specificity (95% CI) for determining risk classifications were 60.0% (44.3–74.3) and 62.8% (48.1–75.6), respectively. The positive and negative likelihood ratios (95% CI) were 1.61 (1.05–2.47) and 0.64 (0.42–0.97). Discussion: The use of validated self-report questionnaires are recommended to supplement clinician prognosis for patients at risk of chronic musculoskeletal pain.

Comparisons of Five DNA Repair Pathways Between Elasmobranch Fishes and Humans

Although DNA repair capacity has been correlated with lifespan in terrestrial vertebrate species, it remains unknown how evolutionarily conserved the process is across all vertebrate taxa. In particular, chondrichthyan fishes have lifespans that range from 3-350 years and they are evolutionarily separated from modern humans Homo sapiens by approximately 400 million years. We hypothesized that chondrichthyan fishes would show significant homology in nuclear excision repair (NER) genes with humans, and that the expression of NER genes will correlate with the lifespan of the respective assessed species. For this study, DNA repair gene homology and expression was performed on the nurse shark Ginglymostoma cirratum (n=3) and yellow stingray Urobatis jamaicensis (n=3). The five main NER pathways were analyzed and compared to see the differences in both elasmobranch species, then compared with human foreskin fibroblast samples (n=3). RNA sequencing was used to determine the extent of gene expression in each species, comparing the read counts in each gene and comparing between the two species. The elephant shark Callorhinchus milii reference genome was used to align the nurse shark and yellow stingray samples. Homology of each gene of the NER pathways was assessed by the NCBI BLAST software. Results show that the MMR pathway has all the significant genes in higher frequencies in the nurse shark than in human. Within elasmobranchs in the five DNA repair pathways, the longer-lived species (nurse shark) has a significant higher gene expression than shorter-lived species (yellow stingray). Genes involved in the NER and BER pathways showed significantly lower expression in elasmobranch than in humans. However, there were significantly higher expression of more genes for the HR and MMR pathways in elasmobranchs than in humans.
