Previous issue date: 2015-03-05 / Objective: Assess the impact of prematurity on telomere length, a cell aging marker, in children born with very low birth weight.
Methods: We conducted a controlled cross-sectional study evaluating 91 children between 7 and 12 years, been 46 former very low birth weight premature and 45 controls. The relative quantification of the size of telomeres (T/S) was performed by polymerase chain reaction (PCR) in peripheral blood leukocytes. Information on perinatal variables, socioeconomic status and stressful life events were obtained. The linear regression model was used for statistical analysis, both in univariate and multivariate analysis.
Results: There was no significant difference in the leukocyte T/S ratio among premature infants and controls (p = 0.841), even after adjustment for possible predictor variables. Univariate analysis showed no statistically significant association between T/S ratio and sex, age, parental age, smoking during pregnancy, education of the caregiver and the head of the family and stressful events that occurred last year. Among preterm infants, there was no relationship between the T/S ratio and birth weight below 1,000 g (?=0.133, 95% CI= -0.141 to 0.406; p=0.341)), gestational age below 30 weeks (?=0.030, 95% CI= -0.209 to 0.270; p=0.34) or bronchopulmonary dysplasia (? = 0.213, 95% CI = -0.166 to 0.591; p=0.15).
Conclusions: The results indicate that there is no relationship between preterm birth or very low birth weight and the leukocyte telomere length at school age. However, the findings need to be replicated, preferably in longitudinal studies. / Objetivo: Avaliar o impacto da prematuridade no comprimento dos tel?meros, um marcador de envelhecimento celular, em crian?as nascidas com muito baixo peso.
M?todos: Realizou-se um estudo transversal controlado, avaliando 91 crian?as entre 7 e 12 anos, sendo 46 ex-prematuros de muito baixo peso e 45 controles. A quantifica??o relativa do tamanho dos tel?meros (T/S) foi realizada por rea??o de polimerase de cadeia (PCR) em leuc?citos do sangue perif?rico. Foram obtidas informa??es sobre vari?veis perinatais, condi??o socioecon?mica e eventos de vida estressores. Na an?lise estat?stica, utilizou-se o modelo de regress?o linear, tanto no modelo de an?lise univariada quanto multivari?vel.
Resultados: N?o houve diferen?a significativa na raz?o T/S dos leuc?citos entre as crian?as prematuras e controles (p=0,841), mesmo ap?s ajuste para poss?veis vari?veis preditoras. A an?lise univariada n?o mostrou associa??o estatisticamente significativa entre a raz?o T/S e sexo, idade, idade parental, tabagismo na gesta??o, escolaridade do cuidador e do chefe da fam?lia e eventos estressores ocorridos no ?ltimo ano. Entre os prematuros, n?o houve rela??o entre a raz?o T/S e peso de nascimento abaixo de 1.000 g (?=0,133, IC95%= -0,141 a 0,406; p=0,341), idade gestacional abaixo de 30 semanas (?=0,030, IC95%= -0,209 a 0,270; p=0,34) ou displasia broncopulmonar (?=0,213, IC95%= -0,166 a 0,591; p=0,15).
Conclus?es: Os resultados indicam que n?o h? rela??o entre nascimento prematuro ou muito baixo peso ao nascer e comprimento dos tel?meros nos leuc?citos na idade escolar. Entretanto, os achados precisam ser replicados, preferentemente em estudos longitudinais.
