Previous issue date: 2013 / A inflamacao neurogenica e um dos primeiros eventos de cura. Apos um estimulo nociceptivo na pele, ocorre a liberacao de neuropeptideos (NP 's) u substancias pro-inflamatorias u que sao produzidas pelos neuronios do ganglio da raiz dorsal e secretadas nas terminacoes nervosas da pele, provocando a fase inflamatoria da cicatrizacao das feridas. A expansao da pele e amplamente utilizada em cirurgia plastica como um recurso para ganho real de pele para rotacao de retalho e, assim, cobrir grandes defeitos decorrentes da perda de pele na remocao de tumores, acidentes de varias origens e reconstrucao de partes do corpo humano; mas nada se sabe sobre a acao dos neuropeptideos CGRP e SP nessas areas de tecido cutaneo expandido. OBJETIVO: Investigar os neuropeptideos SP e CGRP na pele cutanea expandida. METODOS: 28 ratos Wistar, separados em quatro grupos (n = 7): Grupo CO - (expansor vazio); Grupo EN - (expansao normal, 15 dias); Grupo ME - (meia expansao, 11 ml) e o Grupo EI - (expansao instantanea no dia da coleta). Depois de anestesia geral, cada grupo apresentou um expansor de pele de 22 ml de capacidade e o Grupo CO apenas o expansor vazio; o Grupo EN, o expansor inflado com metade da capacidade (11 ml) no implante e uma semana depois, inflado com mais 11 ml; Grupo ME igual ao grupo EN, mas com apenas metade da capacidade do expansor (11 ml) imediatamente apos o implante e o Grupo EI, expansor sendo insuflado com a capacidade total (22 ml) no dia da coleta. Em seguida, cada rato foi submetido a morte induzida, indolor, antes da coleta de amostras, que foram padronizadas quanto ao volume e area, utilizando-se dermatomo eletrico fixado em 500 μm e punch de oito milimetros. Esta amostra foi enviada para a quantificacao de CGRP e SP por Western blotting. RESULTADOS: Em relacao a presenca de NPÆs na pele de ratos, diminuiu a concentracao do neuropeptideo SP, sendo a maior concentracao do grupo CO, e tendo a seguir o grupo ME, o grupo EI e o grupo EN; o neuropeptideo CGRP obteve valores semelhantes na avaliacao de CGRP-5kDa (neuropeptideo efetivo) e a manutencao das concentracoes dos grupos CO, ME e EI. Houve uma queda acentuada na concentracao do grupo EN. CONCLUSAO: A lenta e progressiva expansao da pele inibe a liberacao de neuropeptideos SP, reduzindo a resposta inflamatoria independente do volume observado / The neurogenic inflammation (IN) is one of the first events of healing.
Before a nociceptive stimulus in the skin, there is release of neuropeptides (NP's) skin pro-inflammatory substances that are produced by the neurons of the dorsal root ganglia and secreted nerve endings in the skin, triggering the inflammatory phase of wound healing. The expansion skin, widely used in plastic surgery as a resource to gain real skin to skin flap rotation and thus cover large skin defects arising out of the loss is the removal of
tumors, accidents of various origins and reconstructions of parts of the
human body, but nothing is known about the action of neuropeptides
CGRP and SP in these skin areas expanded. OBJECTIVE: To evaluate the
effect of expanding the skin secretion of NP's SP and CGRP in the skin of
mice. METHODS: 28 Wistar rats divided into 4 groups (n = 7): group C
(empty Expander), EN group (Normal Expansion 15days), group (Half
Expansion) and group EI (Instant Expansion on the collection day). After
general anesthesia, each group has introduced a skin expander of 22ml
capacity with the CO group only without the expander breathes it in no
time, EM Group expander inflated with half capacity (11ml) after
introducing a week later inflated with 11ml more, equal to group ME but
with only half the capacity of the expander (11ml) and nothing else, and EI
group expander being inflated with total capacity (22ml), waited a minute
before the death of the animal and data collection . Next, each mouse was
subjected to induced death painless and a skin sample from each mouse
with standardized area and volume were taken from the center line incision
with the aid of electric dermatome and 8mm dermal punch. This sample
was sent for quantification of CGRP and SP by Western Blotting and
followed for statistical analysis. RESULTS: In relation to the presence of

NP's in the skin of rats in relation to neuropeptide SP was decreasing
concentration being the highest concentration of CO Group and so on with
the following GE Group ME and EI, as the neuropeptide CGRP have
similar values in the evaluation of CGRP-5kDa (actual measure) and
maintenance of the concentrations of CO Groups, ME and EI, as a sharp
drop in the concentration EN group (normal expansion). CONCLUSION:
It was observed that the slower expansion less concentration of
neuropeptides soon denervation can be observed. Thus, the slow and
progressive cutaneous expansion inhibits the release of neuropeptides
