Bipolar disorder is a severe psychiatric disorder, multifactorial and highly disabling whose progression has been associated with apparently irreversible cognitive impairments. Is characterized by alternating episodes of mood (mania / hypomania and depression) that vary in intensity, duration and frequency. The objective of the present study was to evaluate the behavioral effects of acute treatment with olanzapine in animals chronically treated with methylphenidate. In order to do that, adult male Wistar rats were divided into two groups, animals that received acute administration of olanzapine in doses of 2mg/kg (n = 40) and 5mg/kg (n = 40). For the characterization of the model of mania, animals received methylphenidate (2,5mg/kg) administered intraperitoneally for seven consecutive days. The results show that olanzapine has antimanic effect observed in the number of crossing, rearings and groomings in the open-field habituation test, similar between the 2mg/kg and 5mg/kg. However, the acute administration of olanzapine, more significantly at a dose of 2mg/kg, showed amnesic effect, represented by lower recognition rate observed in the test object recognition. É caracterizado por episódios alternados de humor (mania/hipomania e depressão) que variam em intensidade, duração e frequência. O objetivo do presente estudo foi avaliar os efeitos comportamentais do tratamento agudo de olanzapina em animais tratados cronicamente com metilfenidato. Para isso, ratos Wistar machos adultos foram divididos em dois grupos, animais que receberam administração aguda de olanzapina nas doses de 2mg/kg (n=40) e de 5mg/kg (n=40). Para a caracterização do modelo de mania, os animais receberam metilfenidato (2,5mg/kg), administrado por via intraperitoneal, durante sete dias consecutivos. Os resultados mostraram que a olanzapina exerce efeito antimaníaco observado no número de cruzamentos, movimentos de exploração e de auto-limpeza avaliados no teste de habituação ao campo aberto, de forma semelhante entre as doses de 2mg/kg e 5mg/kg. No entanto, a administração aguda de olanzapina, de forma mais expressiva na dose de 2mg/kg, apresentou efeito amnésico, representado pelo menor índice de reconhecimento observado no teste de reconhecimento de objetos. O conjunto destes resultados sugere que não existe associação entre efeitos indicadores de mania a partir da administração de metilfenidato e o melhor desempenho no teste de memória em animais experimentais que receberam tratamento agudo de olanzapina.
