Nitrification of Landfill Leachate by Biofilm Columns

Landfill leachate characteristics vary depending on the operation type of the landfill and the age of the landfill. At landfills operated as bioreactors, where leachate recirculation is practiced, leachate ammonia nitrogen concentrations may accumulate to extremely higher levels than during single pass leaching, thereby requiring treatment before final discharge to a receiving system (Onay, 1998). Usually several physical/chemical wastewater treatment technologies are used to treat the leachate. In most cases the COD and BOD are treated, and then nitrification is performed in a separate sophisticated ex situ system. The additional costs of these systems can be very high. The use of a readily available media for in situ nitrification should be considered a prime objective to avoid extra costs. The possibility of removing ammonia nitrogen from bioreactor landfill leachate using trickling filter biofilm technology was studied in four laboratory scale reactors filled with four different types of packing media. The different packing media were examined to see which media is the most efficient at supporting ammonia removal biofilms. The highest efficiency was achieved by a packing media consisting of pine wood chips. The effects of varied concentration loading, varied hydraulic loading, and nitrification inhibitors were studied. Varied ammonia concentration did not have a huge impact on the ammonia removal rates (77-87%) in the reactor with pine wood media. The ammonia removal rates showed a strong dependence on hydraulic loading rate with the lowest loading rate producing the highest removal rates. Landfill leachate from the Middle Peninsula Landfill in Glens, Virginia was determined not to contain nitrifying inhibitors. Using a wood media filter chip and a low hydraulic loading rate was determined to be the best method to remove ammonia nitrogen from landfill bioreator leachate. / Master of Science

Nitrogen Removal And Operations Improvement In Pond Wastewater Treatment Systems

Study of wastewater treatment ponds at full-scale compared the areal total ammonia nitrogen (TAN) removal rates and specific TAN removal rates of high-rate algal ponds (HRPs) to the TAN removal rates of facultative ponds. The outer high-rate algal pond (HRPO) demonstrated superior specific and areal TAN removal rates compared to the inner HRP and the two facultative ponds. Solids return into the HRPO for a portion of the study period yielded increased volatile suspended solids content but no noticeable increase in TAN removal rate. Nitrification modeling for the HRPO tested multilinear regression, multilinear regression on every second observation of the data set, and a nonlinear Michaelis-Menten regression. The multilinear regression on the full data set explained the most variance with an R2 = 55.9% and the following significant (p-value < 0.05) variables: solar insolation, temperature, and ambient TAN concentration. Wastewater treatment pond systems are used worldwide as a method for affordable solids and nutrient removal, but these systems can be less predictable due to their reliance on biological processes. This thesis project recorded various water quality parameters and nitrogen species concentrations on a weekly basis from July 2020 to April 2022 to compare pond performance and identify process improvements. Accurately modeling a pond’s performance will better allow operators to save on aeration and coagulation costs while still meeting effluent goals.

Optimization of Biological Nitrogen Removal From Fermented Dairy Manure Using Low Levels of Dissolved Oxygen

A pilot scale nitrogen (N) removal system was constructed and operated for approximately 365 days and was designed to remove inorganic total ammonia nitrogen (TAN) from solids-separated dairy manure. An anaerobic fermenter, upstream of the N removal reactor, produced volatile fatty acids (VFAs), to be used as an electron donor to fuel denitrification, and TAN at a COD:N ratio of 18:1. However, sufficient amounts of non-VFA COD was produced by the fermenter to fuel the denitrification reaction at an average NO3- removal rate of 5.3 ± 2 mg/L NO₃--N. Total ammonia N was removed from the fermenter effluent in an N removal reactor where a series of aerobic and anoxic zones facilitated aerobic TAN oxidation and anoxic NO₃- and NO₂- reduction. The minimum dissolved oxygen (DO) concentration allowing for complete TAN removal was found to be 0.8 mg/L. However, TAN removal rates were less than predicted using default nitrifying kinetic parameters in BioWin®, a biological modeling simulator, which indicated the presence of a nitrification inhibitor in fermented dairy manure. Furthermore, an N balance during the aerobic zone indicated that simultaneous nitrification-denitrification (SND) was occurring in the aerobic zone of the N removal reactor and was most apparent at DO concentrations below 1.3 mg/L. A series of nitrite generation rate (NGR) experiments confirmed the presence of an inhibitor in fermented dairy manure. A model sensitivity analysis determined that the most sensitive ammonia oxidizing bacteria (AOB) kinetic parameters were the maximum specific growth rate, , and the substrate half saturation coefficient, . Nitrifying inhibition terms of competitive, non-competitive, mixed competitive, and un-competitive were applied to the growth rate equation in BioWin® but an accurate representation of the observed TAN removal rates in the pilot scale system could not be found by adjusting the kinetic parameters alone. Reducing the default BioWin® hydrolysis rate by approximately 50% produced a more accurate calibration for all inhibition terms tested indicating that the hydrolyization of organic N in dairy manure is less than typical municipal waste water. / Master of Science

A transition state physiochemical model predicting nitrification rates in soil-water systems

Transition state theory was applied to the nitrification process in soil-water systems, and a computerized, theoretical rate model was developed to include NH₄⁺ and 0₂ concentrations, pH, temperature, moisture content, and local differences in nitrifying capacities of Nitrosomonas bacteria. The model was restricted to enriched calcareous soils thus simplifying the application of basic physicochemical principles. Experimental rate data from an agricultural and a native desert soil provided verification of a zero order reaction for nitrification with respect to NH₄⁺ concentrations above a certain saturation level, as previously reported. The saturation concentration in soils was found to be about 1.0 to 5.0 ppm. A theoretical linear relationship between activation energy and ionic strength was confirmed by application of the above data. However, each local population of nitrifiers tended to display different values for the slope and intercept of the linear relationship. The structure of the activated complex for NH₄⁺ oxidation to NO₂⁻ was determined to be more like NH2OH or NH₄⁺ than NO₂⁻. As a first approximation, the NH₂OH activated complex was included in the rate model. The equation form for the equilibrium between the reactants and the activated complex was found to differ from the stoichiometric reaction between NH₄⁺ and O₂ to form NH₂OH. The equilibrium expression was found to be more closely approximated by the relationship, 2 NH₄⁺ + O₂ ≶ (ACTIVATED COMPLEX) + + H⁺. A method was developed to compute soil pH values as a function of moisture content. Verification was obtained by using data obtained from the agricultural and native desert soils, including cases where samples were acidified. The calculated pH values were used in the nitrification rate model. Further verification of the model was obtained using data from the literature for two soils from the Northern Great Plains. Data pairing of observed and predicted rates for these soils yielded R values of 0.944 and 0.940. The rate model was programmed in FORTRAN IV computer language and designed to operate in conjunction with existing computer models. Thus, this relatively sophisticated model may be applied to field simulation studies with a minimum of adaptive procedures. The model should aid in obtaining reliable predictions of NO₃⁻ formation and movement under a wide range of field conditions.

Do cluster roots contribute to the costs of carbon and nitrogen metabolism during variations in phosphate supply in the legume Lupinus albus

Thesis (MSc)--Stellenbosch University, 2013. / ENGLISH ABSTRACT: The generally low concentrations of P and N in the soil, causes most plants to experience nutrient deficiency during their life cycle. Lupins can rely on both cluster roots and nodules for P acquisition and biological nitrogen fixation (BNF) respectively. The legume Lupinus albus is able to survive under low nutrient conditions, because it has two specialized belowground organs for the acquisition of N and P. In this regard, cluster roots increase P uptake and root nodules acquire atmospheric N2 via biological nitrogen fixation (BNF). Although these organs normally tolerates low P conditions, very little is known about their physiological and metabolic flexibility during variations in P supply. Furthermore, the resource allocation (C, N and P) between cluster roots and nodules has also been largely understudied. The aim of this investigation was therefore to determine the resource allocation, physiological and metabolic flexibility of these organs during variations in P supply. Although variation on P supply had no effect on the total biomass, there were significant differences in specialised below-ground organ allocation to cluster roots and nodule formation. Cluster root formation and the associated C-costs increased during low P supply. In contrast to the cluster root decline at high P supply, there was an increase in nodule growth allocation and corresponding C-costs. Since cluster roots were able to increase P acquisition under low P conditions, this below-ground investment may also have benefited the P nutrition of nodules. These findings provide evidence that when lupins acquire N via BNF in their nodules, there may be a trade-off in resource allocation between cluster roots and nodules. The short-term elevated P supply, caused an increased allocation of C and respiratory costs to nodules, at the expense of cluster roots. This alteration was also reflected in the increase in nodule enzyme activities related to organic acid synthesis, such as Phosphoenol-pyruvate Carboxylase (PEPC), Pyruvate Kinase (PK), Malate Dehydrogenase (NADH-MDH) and Malic Enzyme (ME). In cluster roots, the elevated P conditions, caused a decline in these organic acid synthesizing enzymes. This suggests that during short-term elevated P supply, there is a great degree of physiological and metabolic flexibility in the lupin nutrient acquiring structures. / AFRIKAANSE OPSOMMING: Die algemeen lae konsentrasies van fosfaat en stikstof in die grond , veroorsaak dat die meeste plante voedingstekorte ervaar tydens hul lewensiklus. Kvävestatus och risk för nitrifikation i två avverkade skogsområden i Halland

<p>Nitrogen deposition leads to environmental damage in areas where the nitrogen deposition is high. Southwest Sweden receives an annual nitrogen deposition of up to 20 kg N/ha. Nitrogen that is not assimilated by the vegetation is accumulated in the soil, which may lead to nitrogen saturation and an elevated risk of nitrogen leaching. Nitrogen leaching from forest areas in southern Sweden has proven to be higher than from agricultural areas, which have been thought to be the main contributors to elevated nitrogen levels in rivers and lakes. The amount of nitrogen that leaches depends on the fraction of the total nitrogen in the soil that consists of nitrate, since nitrate is easily transported through the soil. Nitrogen leaching increases after clear-cutting since the uptake by vegetation is greatly reduced.</p><p>In this study the soil chemistry of two clear-cut spruce stands in Halland, in southwest Sweden, has been analyzed. A previous study in these areas has indicated higher nitrate concentrations in the groundwater in one of the areas and thus a greater nitrogen leaching. Nitrogen deposition in the two areas is estimated to be the same and therefore the soil chemistry has been analyzed to evaluate if differences in the soil can have resulted in differences in the nitrate concentration in the groundwater. The hypothesis is that the area with higher nitrate concentrations in the groundwater has properties more favorable for nitrification, which would be especially apparent in a lower carbon to nitrogen ratio. The pH and storage of nitrate, ammonium, total carbon, total nitrogen and exchangeable cations was analyzed in soil samples from both areas. In addition, a study of stand properties and previous use of the areas was made.</p><p>The analyses performed in this study indicate that the soil properties could not explain the differences in nitrate concentration in the groundwater. The differences found between the areas were that the area with lower nitrate concentrations in the groundwater had higher nitrate and ammonium concentrations and higher pH in the soil. The reason for this might be that the soil in this area has larger capacity to bind elements. The fact that the soil samples were sampled during different seasons probably had a major effect on these results. The history and stand properties of the two areas were similar. According to site index one area was more fertile, which benefits nitrification. This fact was not confirmed by the analyses, but it probably caused the nitrification rate to be higher in this area.</p> / <p>Det stora nedfallet av föroreningar i form av kväve i framförallt sydvästra Sverige leder till försurning och övergödning av både skogsmarker och vattendrag. Kvävedepositionen i södra Sverige kan uppgå till 20 kg N/ha och år. Det kväve som inte tas upp av vegetationen ackumuleras i marken som efterhand riskerar att bli kvävemättad vilket ökar risken för kväveutlakning. Kväveutlakningen från svenska skogsmarker har visat sig kunna vara i samma storleksordning som från jordbruksmarker som anses vara en av de största orsakerna till ökande kvävehalter i sjöar och vattendrag. Kväveutlakningens storlek beror till stor del på hur stor del av markens kväveförråd som finns i den oorganiska formen nitrat eftersom den är en lättrörlig jon. Efter skogsavverkning ökar nitratutlakningen betydligt på grund av att det finns väldigt lite vegetation kvar som tar upp kväveföreningarna.</p><p>I den här studien har markkemin i två avverkade granskogsområden i Halland undersökts. Tidigare studier i dessa två områden har visat att halterna av nitrat var högre i grundvattnet i det ena området än i det andra, och således var även nitratutlakningen större i detta område. Eftersom kvävedepositionen anses vara densamma i de två områdena har markemin undersökts för att finna skillnader där som kan förklara de olika nitratnivåerna i grundvattnet. Hypotesen var att det ena området skulle ha mer gynnsamma förhållanden för nitrifikation, framförallt en lägre C/N-kvot. Markens förråd av nitrat- och ammoniumkväve, totalkol, totalkväve, utbytbara katjoner samt pH undersöktes i båda områdena. En studie av områdenas historia gjordes även för att kunna utesluta skillnader där som kunde ge upphov till skillnaden i nitrat i grundvattnet.</p><p>Slutsatsen som kan dras från studien är att de analyserade egenskaperna inte visade på mer gynnsamma förhållanden för nitrifikation i det ena området än i det andra. Ingen skillnad i C/N-kvot uppmättes. Skillnaderna som hittades var att området med lägst halter av nitrat i grundvattnet hade högst halter av oorganiskt kväve i marken och högre pH. Att nitrathalterna var högre i grundvattnet men inte i marken kan ha sin förklaring i att marken i det ena området har större förmåga att binda till sig kväve vilket gör att det inte lika lätt kommer ut i grundvattnet. Antagligen spelade det stor roll för dessa resultat att jordproven tagits vid olika årstider. Undersökningen av tidigare användningsområde i skogarna visade att de hade en tämligen liknade historia. Det ena området har enligt ståndortsindex högre bördighet vilket gynnar nitrifikation. Detta framgick dock inte av analyserna men är antagligen orsaken till de högre nitrathalterna i grundvattnet i detta område.</p>

Nitrate sources and cycling at the Turkey Lakes Watershed: A stable isotope approach

Marie, Ontario, Canada. <span style="mso-spacerun: yes">?? </span> <p class=MsoNormal><span style="mso-spacerun: yes">?????? </span><span style="mso-spacerun: yes">????????????</span>Bulk precipitation collected biweekly at the TLW from 1995 to 2000 had nitrate isotope values that ranged from +42. 4 to +80. 4&permil; for <span style='font-family:Symbol'>d</span><sup>18</sup>O and -6. 3 to +2. 8&permil; for <span style='font-family:Symbol'>d</span><sup>15</sup>N. <span style="mso-spacerun: yes">?? </span>An incubation experiment indicated that the isotopic composition of atmospheric nitrate was not compromised by collection methods whereby unfiltered bulk precipitation samples remain in the collector for up to two weeks. <span style="mso-spacerun: yes">?? </span> <p class=MsoNormal><span style="mso-spacerun: yes">?????????????????? </span>The first direct measurement of the isotopic composition of microbial nitrate produced <i>in situ</i> was obtained by eliminating precipitation inputs to three forest floor lysimeters and subsequently watering the area with a nitrate-free solution. <span style="mso-spacerun: yes">?? </span>Microbial nitrate had <span style='font-family:Symbol'>d</span><sup>18</sup>O values that ranged from +3. 1 to +10. 1&permil; with a mean value of +5. 2&permil;, only slightly higher than values predicted based on the <span style='font-family:Symbol'>d</span><sup>18</sup>O-H<sub>2</sub>O of the watering solution used. <span style="mso-spacerun: yes">?? </span><span style='font-family:Symbol'>d</span><sup>18</sup>O values of soil O<sub>2</sub> (+23. 2 to +24. 1&permil;) down to a depth of 55cm were not significantly different from atmospheric O<sub>2</sub> (+23. 5&permil;) and therefore respiratory enrichment of soil O<sub>2</sub> did not affect the <span style='font-family:Symbol'>d</span><sup>18</sup>O values of microbial nitrate produced at the TLW. <span style="mso-spacerun: yes">?? </span> <p class=MsoNormal><span style="mso-spacerun: yes">?????????????????? </span>Nitrate export from two undisturbed first-order stream basins was dominated by microbial nitrate, with the contribution of atmospheric nitrate peaking at about 30% during snowmelt. <span style="mso-spacerun: yes">?? </span>Clear-cutting of catchment 31 in 1997 resulted in elevated nitrate concentrations, reaching levels that exceeded the drinking water limit of 10 mg N/L. <span style="mso-spacerun: yes">?? </span>Isotopic analysis indicated that the source of this nitrate was predominantly chemolithoautotrophic nitrification. <span style="mso-spacerun: yes">?? </span>The <span style='font-family:Symbol'>d</span><sup>18</sup>O values of microbial nitrate in stream 31 progressively increased during the post-harvest period due to an increase in the proportion of nitrification that occurred in the summer months. <span style="mso-spacerun: yes">?? </span>Despite drastic alteration of nitrogen cycling in the catchment by the harvest, <span style='font-family:Symbol'>d</span><sup>15</sup>N-nitrate values in shallow groundwater did not change from the pre-harvest. <span style="mso-spacerun: yes">???? </span>Denitrification and plant uptake of nitrate in a small forested swamp in catchment 31 attenuated 65 to 100% of surface water nitrate inputs following harvest, reducing catchment-scale nitrate export by 35 to 80%.

The use of nitrogen timing and nitrification inhibitors as tools in corn and wheat production in Kansas

The current seven billion people on earth are expected to reach nine billion by 2050 with resulting demands on world food production. In addition, the quality of our environment is being impacted by human activities, including agricultural production and crop fertilization. Nitrogen (N) management is the process of applying N fertilizers in a way to maximize use of N by crops, while minimizing loss to the environment. It is becoming imperative, as a means of increasing crop yields and food supplies, while reducing input usage, and minimizing the impact of N fertilization on the quality of our environment, that improved N application practices be identified and utilized. The objectives for this study were to compare the timing of anhydrous ammonia (AA) fertilizer N applications, fall and spring, with and without two different nitrification inhibitors (NI) as possible tools to enhance yield and Nitrogen Use Efficiency (NUE) in corn (Zea mays) and winter wheat (Triticum aestivum L.) in Kansas. Two different nitrification inhibitors were tested as alternatives, N-Serve (nitrapyrin) produced and marketed by Dow AgroSciences, and an experimental product under development by Koch Agronomic Services LLC. Three differing rates of the experimental product were used to assist in determining the optimal rate for this product. The study was conducted over two growing seasons, 2012 and 2013, which differed significantly in rainfall, rainfall distribution, and resulting NUE. Experiments were established at three sites for both crops in both years, on sites/soils selected for differing potentials for N loss, and mechanisms of N loss. One site was established at the Kansas State University Agronomy North Farm (N Farm), where yield potential was high, and N loss potential was low. A second site was established under irrigation at the Kansas River Valley Experiment Field near Topeka, KS (KRV), on a coarse silt loam soil with high potential for N loss through leaching. The third site was established at the East Central Kansas Experiment Field near Ottawa KS (ECK), on a clay pan soil with a high potential for denitrification loss. Weather conditions together with soil characteristics played a major role in the performance of N timing applications and impacted the response to the use of the inhibitors. In low N loss environments such as the N Farm, fall applications of AA to increase spring time-availability for producers showed minimal negative effects on yield or NUE. When combined with a nitrification inhibitor in the fall, performance was similar to spring application for both corn and wheat. At the KRV site leaching loss or potential loss from fall application was high for corn and wheat in both years, however little impact on NUE with NI use was observed. At the high ECK denitrification site, there was only one N loss potential event leading to inhibitor performance at Ottawa in corn in 2013.

O principal objetivo desse trabalho foi desenvolver uma nova configuração de biofiltro aerado submerso utilizado no pós-tratamento do efluente de reator UASB, capaz de realizar nitrificação e desnitrificação em um único sistema. Em busca de bases operacionais dessa nova configuração, a pesquisa foi inicialmente conduzida com três reatores seqüenciais. Esses reatores foram dispostos nas seqüências I e II, respectivamente. Os resultados obtidos com esses sistemas seqüenciais levaram a concepção do projeto da nova configuração do biofiltro proposto (BF definitivo). Os resultados da seqüência I mostraram alto potencial para conversão de nitrogênio amoniacal e total, e também alta eficiência na remoção da matéria orgânica carbonácea. Esse sistema, porém, não apresentou potencial para desnitrificação. Na seqüência II foi realizada recirculação entre dois reatores do sistema (anóxico e aeróbio) para aumento de tal potencial. O maior potencial de desnitrificação, nessa seqüência, foi observado para razão de recirculação, Rc, igual a 2,65 e com o uso de 40%, em volume, de esgoto sanitário bruto como fonte de carbono. Sob essas condições operacionais, para concentração média afluente ao sistema igual a 33,74 mg de \'N-\'NH IND.3\'/L as concentrações efluentes médias de \'N-NH IND.3\', \'N-NO IND.2\'POT.-\' e \'N-NO IND.3\'POT.-\' foram, respectivamente, iguais a 0,16, 0,0026 e 9,72 mg/L. Os resultados do BF definitivo mostraram que a nova configuração proposta é viável como unidade de pós-tratamento de efluente de reator UASB, promovendo nitrificação e desnitrificação em um único sistema, além de alta eficiência de remoção da matéria orgânica. Em todas as fases dessa pesquisa, praticamente, todos os resultados obtidos atenderam ao padrão de lançamento de nitrogênio amoniacal estabelecido pela legislação ambiental. / The main objective of this work was developing a new configuration of submerged aerated biofilter used in UASB reactor post-treatment, with nitrification and denitrification in a single system. Searching operational bases of this new configuration, this research was initially driven by three sequential reactors. These reactors were arranged in the sequences I and II, respectively. The results obtained with these sequential systems took the conception of the project of the new configuration of the proposed biofilter (definitive BF). The results of the sequence I showed high ammoniacal and total nitrogen conversion potential, and also high carbonaceous organic matter removal efficiency. This system, however, not presented potential for denitrification. In the sequence II recirculation between two reactors (anoxic and aerated) of the system was made for such potential increasing. The higher denitrification potential, in this sequence, was observed with recirculation reason, Rc, equal to 2,65 and using 40%, in volume, of raw sanitary sewage as carbon source. Under these operational conditions, for average affluent concentration of 33,74 mg of \'N-NH IND.3\'/L the average effluent concentrations of \'N-NH IND.3\', \'N-NO IND.2\'POT.-\' e \'N-NO IND.3\'POT.-\' were, respectively, equal to 0,16, 0,0026 and 9,72 mg/L. The results of the definitive BF showed that the new proposed configuration is feasible as effluent UASB post-treatment unity, with nitrification and denitrification in a single system, besides high organic matter removal efficiency. Practically in all the phases of this research, all the results obtained attended to the launch standard of ammoniacal nitrogen established by the environmental legislation.
