Anne Lister (1791-1840), a provincial gentlewoman of Shibden Hall, near Halifax in the West Riding of Yorkshire, produced between 1806 and her death a four-million-word diary documenting her daily life, intimate thoughts, ambitions, sexual and emotional adventures with women and her musings on the nature of her sexuality. About a sixth is written in cipher. Lister scholarship so far has focused mainly on the social and cultural implications of her writings. This thesis, however, examines the diary as a literary text and considers Lister's deployment of literary forms and structures, strategies and conventions in producing a sense of self. It explores her relationship to contemporary ideas of authorship and to notions of public and private, and investigates the impact of reading in the autobiographical writing of Lister and her circle and the significance of the cipher in her social and sexual self-representation. It asks whether her literary production helped accommodate her self-representation as a traditional country gentlewoman with her unconventional sexuality. The linking theme throughout the thesis, bringing together the many different aspects of Lister's self-fashioning, is the significance of literary considerations in her diary and letters. It begins the work of investigating the literary structures and strategies of her writings, and offers a fresh perspective on this remarkable work of literary self-construction.

Mathematical modelling of cell population dynamics in the colonic crypt with application to colorectal cancer

Colorectal cancer has the third highest mortality and incidence rates of all cancers worldwide, but the prognosis for long-term survival is good if diagnosed early. It is a well-characterised disease, and is initiated in colonic crypts which line the colon wall. The aim of this thesis is to use mathematical modelling to describe the heavily regulated cell renewal cycle in the crypt to determine the key features of the system kinetics, and help to explain the initiation of tumourigenesis. The dynamics of a single colorectal crypt is considered using a compartmental approach, which accounts for populations of stem, transit-amplifying and fullydifferentiated cells. A number of different model formulations are derived, and their validity and suitability are discussed. Two mechanisms are presented that could regulate the growth of cell numbers and maintain homeostasis (equilibrium), and it is illustrated how a model can describe both regulated and unregulated growth, with cancer-driving cells deriving from stem and/or transit cells. This model is used to explain the long lag phases observed in carcinogenesis, which occur between periods of rapid tumour expansion, before unlimited growth in cell numbers ensues. Significantly, it is found that, contrary to general belief, the proportion of cancer-driving cells in the exponential growth phase of a tumour may vary depending on tumour type. The process of cells accumulating mutations is also examined by considering both a stochastic individual cell-based model and an analytic approach. Finally, an ordinary differential equation model is shown to be valid by considering a simplified description of a one-dimensional spatial model, and the latter is used to consider the effect of changing the crypt shape. The suitability of this modelling approach to tracking stem cells in a niche, as well as mutant cell clones as they propagate in the crypt, is also discussed.

Diskkrypteringsprestanda i GNU/Linux

This thesis compares the impact on disk performance in a GNU/Linux  environmentwith three encryption algorithms: AES,  Serpent and Twofish in three different implementations: DM-crypt, Loop-AES and Truecrypt. For all three algorithms a key length of 256 bits is used. The thesis shows that the least performance impact during data encryption, and thus file writing, is reached by using AES or Twofish encryption implemented in DM-crypt or TrueCrypt. The thesis shows that some data operations with a sufficiently low processor utilization barely affects disk performance at all if encrypted using the optimal implementation and algorithm. It is also discovered that the performance impact during data decryption, or file reading, can be minimized by using the most efficient implementation and algorithm. The best results are met with the AES or Twofish cipher, regardless of implementation. An important conclusion that  is  drawn is that it is hard to determine a superior encryption solution for all purposes. However, by reviewing and examining the collected data from all aspects of disk performance the AES implementation in TrueCrypt is, with small marginals, determined to be the most optimal. / Undersökningen jämför hur krypteringsalgoritmerna AES, Serpent och Twofish implementerade i DM-crypt, Loop-AES och TrueCrypt påverkar diskprestandan i en GNU/Linux-miljö. Samtliga krypteringsalgoritmer tillämpas med en nyckellängd på 256 bitar. Undersökningen visar att högst skrivhastighet och således krypteringshastighet uppnås med algoritmerna AES och Twofish i DM-crypt eller Truecrypt. Krypteringens påverkan på diskprestanda är beroende av vilka typer av operationer som utförs och hur hög processorsysselsättningsgraden är. Vid  simplare  I/O-operationer har  den mest optimala  krypteringslösningen knappt någon påverkan på diskprestandan överhuvudtaget. Undersökningen visar också att påverkan av prestanda vid dekryptering är minst vid tillämpning av AES eller Twofish oavsett implementering. En viktig slutsats som dras är att det inte finns en överlägsen krypteringslösning för samtliga användningsområden. Genom att sammanställa och granska insamlad data gällande samtliga aspekter beträffande diskprestanda fastställs TrueCrypts implementation av AES till att vara den mest optimala, trots små marginaler.
