Wide Hybridization, Genomic, and Overwintering Characterization of High-Biomass Sorghum Spp. Feedstocks

The federally mandated 36 billion gallons a year production goal for "advanced biofuels" by 2022 has created a demand for lignocellulosic feedstocks that are inexpensive to produce. The current lack of market development for lignocellulosic feedstocks incentivizes the development of versatile biomass products with greater end-use possibilities, as in either a forage or bioenergy system. High-biomass, perennial grasses offer dual-use potential in either forage or biofuel systems. In 2009 and 2010 controlled pollinations were made to evaluate the efficiency of producing interspecific hybrids between homozygous recessive iap/iap and Iap/- Sorghum bicolor (L.) Moench, cultivated sorghum, and three S. halepense (L.) Pers., johnsongrass, genotypes. The iap/iap genotype removes reproductive barriers to alien pollen in S. bicolor and aids in wide hybridization. Total seed set, germinable seed set, and hybrid production were significantly higher using the iap/iap genotype. The iap/iap S. bicolor genotype is a valuable tool available to plant breeders for the creation of wide hybrids with S. halepense. In a related study a bulked segregant analysis was conducted using bulked samples of S. bicolor, typical flowering S. halepense, non-flowering S. halepense, and putative triploid hybrids of the two species to identify unique markers for each bulk and to evaluate S. bicolor genetic material introgression into the non-flowering S. halepense genome. Thirty-nine and 23 markers were found to be unique to the S. bicolor and typical flowering S. halepense bulks, respectively. These unique markers could be used in a breeding program to identify interspecific hybrids. Alleles at fifteen markers were found in both the S. bicolor and non-flowering S. halepense bulks but not in typical flowering S. halepense and may help explain the non-flowering phenotype. In 2010 and 2011 a study was conducted to investigate the rhizome composition of 11 genotypes of Sorghum species and its relationship to overwintering. Genotype, environment, and sampling date had significant effects on rhizome metabolite concentrations. Overwintering capacity was related to fructans and crude protein concentrations and NIRS (Near Infrared Spectroscopy) was effective at estimating these values. This information can be used to screen for stronger perennial parents to be used in future breeding programs.

Trait Correlation and Confirmation of QTLs for Rhizome Growth and Over-wintering in Sorghum

A growing world population drives an ever-increasing need for food and energy. These challenges, along with depletion of water and fossil fuel resources, call for improvements in crop production systems and the cultivars used within them. Perennial cropping systems present an attractive solution to many of these problems. A greater understanding of the genetic control of over-wintering ability within crop species is one way to begin the process of making perennial cropping systems a possibility. In this study an F3:F4 family derived from a cross between Sorghum bicolor (L.) Moench and S. propinquum (Kunth) Hitchc. segregating for rhizome production was phenotyped in both field and greenhouse environments for traits relating to rhizomatousness and over-wintering. Several statistical models were created to correlate rhizome growth and over-wintering. A known rhizome quantitative trait locus (QTL) region was saturated with SSR markers and the QTL interval was reduced from previous estimates of about 16 Mb or 7 cM to 12 Mb or 2 cM, a 25% or 71% reduction in physical or linkage distance respectively. Two previously unidentified QTL regions associated with over-wintering were also identified. Our results also support the hypothesis that rhizome growth is important and possibly necessary for over-wintering in Sorghum.

A pesquisa intitulada Motivação Criadora e Recepção Estética no Espetáculo Re-Sintos da Muovere Companhia de Dança tem a seguinte indagação: Como operam as motivações criadoras e o processo de recepção de um espetáculo de dança contemporânea? O espetáculo escolhido para investigar o processo de criação e recepção traz o cruzamento das fronteiras entre a dança e o teatro, seja pela composição do elenco de bailarinos e atores, seja pela direção geral e cênica ter sido realizada, respectivamente, por Jussara Miranda e Jezebel de Carli, uma artista da dança e outra do teatro. Re-Sintos apresenta como características: a ênfase da não neutralidade facial; a utilização de elementos sonoros, ora produzidos pelos bailarinos, ora acompanhados pela trilha musical; a utilização de cenário, o uso de narrativa fragmentada; a utilização de diálogo com questões do cotidiano; por fim, valoriza a plasticidade do movimento corporal como principal fio condutor. Como modo de sistematizar e organizar a leitura, esta pesquisa é dividida em três capítulos: O capítulo I analisa como acontece a articulação entre a dança e o teatro no espetáculo Re-Sintos. O capítulo II verifica o que envolve o processo de recepção e como o espetáculo se oferece ao exercício receptivo. O capítulo III examina e apresenta o modo como opera a produção e a recepção do espetáculo Re-Sintos a partir de algumas características aproximativas do rizoma de Gilles Deleuze e Félix Guatarri. As características presentes no Re-Sintos retratam um modo de condução do trabalho, no qual se evidencia a presença de um caráter investigativo que tem como efeito a multiplicidade. Essa multiplicidade compreende as conexões, as rupturas e as contaminações consentidas pelo olhar questionador dos produtores num processo dinâmico de territorialização, desterritorialização e reterritorialização da criação. Quanto à recepção estética, observa-se que cada espectador articula os conhecimentos prévios, os sentidos plurais e paradoxais em linhas de segmentaridade e linhas de desterritorialização. Como a percepção produz constantemente conexões e desdobramentos, ao criar novos nexos e exigir sem cessar outros agenciamentos, os vínculos entre as motivações criadoras e a recepção estética são rizomáticos e não permitem ao espectador a identificação completa das fontes ou origens. / The research entitled Creative Motivation and the Aesthetic Reception at Re- Sintos Performance of Muovere Dance Company has a question: How creative motivations and receptive process operate in a contemporary dance performance? The performance chosen to investigate the process of creation and reception crosses the borders between dance and theater, either by the casting composition by dancers and actors or by the general and scenic direction done, respectively, by Jussara Miranda and Jezebel de Carli, a dance artist and a theater artist. Re-Sintos has as characteristics: the emphasis of the non-facial neutrality; the utilization of sound elements, be it produced by the dancers or accompanied by the musical track; the scenery presentation; the use of the fragmented narrative; the utilization of dialogue with daily subjects; at least, values the corporal movement plasticity as its principal thread. As a way to systematize and organize the reading, this research is divided in three chapters: Chapter I analyses how the articulation between dance and theater happens at the Re-Sintos performance. Chapter II verifies what involves the reception process and how the performance offers itself to the receptive exercise. Chapter III examines and presents the way how the production and the reception of the Re-Sintos performance operate from some approximate characteristics from Gilles Deleuze and Félix Guattarri's rhizome. THE EFFECT OF COLLECTION AND ESTABLISHMENT SEASON, PLANTING ORIENTATION AND ATTACHED CULMS ON THE SURVIVAL AND GROWTH OF ARUNDINARIA GIGANTEA (WALT.) MUHL. (GIANT CANE) RHIZOME PROPAGULES

Arundinaria gigantea (Walt.) Muhl. (giant cane) is a native North American bamboo. The mass assemblages of giant cane stems (culms) can form monotypic stands called "canebrakes" with historic distribution encompassing floodplains of the southeastern United States. Canebrake ecosystems have served as an important habitat for a variety of mammalian, avian, and reptilian species due in part by the protective nature the dense culms provide. Also, giant cane also can serve as an effective riparian zone buffer for the protection of water quality. Land conversion and altered disturbance regimes have reduced cane to 2% of historical accounts. Consistently, there is great interest in restoring canebrake ecosystems. However, canebrake restoration efforts face difficulties such as infrequent seeding and low viability, limited availability of seedlings or rhizome planting stock, and inefficient establishment and management techniques. To address these problems, two studies were designed to further develop giant cane propagation thereby aiding management efforts intended in restoring canebrake ecosystems. Study 1 (greenhouse) objective was to compare the survivability and growth responses of bare rhizomes and rhizomes with attached and trimmed culms with different planting orientation. Study design was a randomized complete block with approximately 20 rhizomes of each propagule type (bare rhizomes, rhizomes with culms trimmed to 3cm and rhizomes with culms trimmed to 20cm) planted in two orientations (buried flat or angled and partially exposed) in each of 4 benches (N=239). Propagule type and planting orientation were determined randomly. After six months, propagule survival was 86% and was independent of orientation (chi square 1df = 1.56, p=0.212) and propagule type (chi square 2df =3.88, p=3.88). There was an interaction between planting orientation and propagule type for the number of new rhizomes and culms, above ground biomass, but not for newly formed rhizome diameter or cumulative culm and rhizome length. Rhizome propagules with attached trimmed culms produced, on average, one more rhizome and were 71 cm longer than newly formed rhizomes from the bare rhizome propagules. Planting orientation had no effect on any measured character of long culmed propagules, burying the short-culmed or bare rhizomes tended to reduce growth responses. However, among exposed propagules, growth responses tended to be similar. Study 2 (field-scale) objective was to determine if genotype (3 collection sources) and collection season/ planting season (C/P) (fall/fall, fall/spring, spring/spring) affect survival and growth of giant cane. Study design was a randomized complete block design with between 12 and 20 bare rhizomes per each of 3 collection sources (subplots) planted in each of 3 rows (collection season/ planting season main plots) blocked 6 times across 2 sites (N=2086). Location of collection sources within subplots and C/P within plots were randomly chosen. Rhizomes were planted in rows using a tree planter. Mean survival of cane plants after one growing season was similar at each site with a mean of 11.1%. Survival was dependent on collection source and C/P seasons. Survival ranged from a high of 38.3% for the spring/spring planted Upper Cache River source to 0.4% for two of the other 9 treatment combinations. Collecting and planting rhizomes in the spring for two of the three collection sources produced the highest percent survival compared to stock collected in the fall then planted or stored until spring. These results suggest the importance of collection source, collection season, planting season, propagule morphology and orientation on the survival and new growth of giant cane in southern Illinois.

O uso de alimentos funcionais contendo componentes como fibras e compostos antioxidante tem sido empregado como estratégia terapêutica em enfermidades crônicas, pois são considerados eficazes na modulação do sistema imune e da microbiota colônca, controlando os processos da inflamação intestinal. O rizoma de Typha angustifolia L. possui alto conteúdo de fibras, fração de parede celular como celulose e a 11.25% de glutamina e 4.85% de glicina em suas folhas, os quais juntamente com a cisteína atuam como precursores da síntese de glutationa. O objetivo do estudo foi investigar a atividade antiinflamatória da suplementação de farinha do rizoma de T. angustifolia L. (FRT) na dieta nas doses de 5%, 10% e 20%, isoladas e em associação a prednisolona a 2mg/Kg na colite experimental induzida por ácido trinitrobenzosulfônico (TNBS). O dano colônico foi avaliado macroscopicamente, microscopicamente e bioquimicamente, quantificando o conteúdo colônico de glutationa (GSH) e atividade da mieloperoxidade (MPO) e da fosfatase alcalina (FA), além de seu efeito prebiótico, na concentração de 20%, sobre bactérias Ácido-Láticas em amostras fecais. Adicionalmente foi avaliado á partir de extrato etanólico a 70% da FRT a atividade antioxidante in vitro por lipoperoxidação em membrana de cérebros de ratos e detecção fitoquímica qualitativa do grupo de saponinas, taninos, flavonóides e cumarinas. A suplementação de FRT na dose de 10% diminuiu significantemente o dano colônico evidenciado macroscopicamente, microscopicamente e bioquimicamente, com contenção nos níveis de GSH e diminuição nas atividades da MPO e FA... / Inflammatory bowel disease (IBD) is a collective term to the group of chronic inflammatory disorders with relapses in the gastrointestinal tract, defined mainly by two types, ulcerative colitis and Crohn's disease. The use of functional foods containing ingredients such as fiber and antioxidant compounds have been used as a therapeutic strategy for chronic diseases because they are considered effective in the modulation of the immune system and colonic microbiota, controlling the processes of intestinal inflammation. The rhizome of Typha angustifolia L. has a high fiber content, cell wall fraction as cellulose and 11.25% of glutamine and 4.85% glycine in the leaves, which together with cysteine act as precursors of glutathione synthesis. This study investigate the anti-inflammatory activity of the diet supplementation of T. angustifolia L. (FRT) rhizome at doses of 5%, 10% and 20%, alone and in combination with prednisolone 2 mg/kg in experimental colitis induced by trinitrobenzosulfônico acid (TNBS). The colonic damage was evaluated macroscopically, microscopically and biochemically by quantifying the colonic glutathione (GSH) content, mieloperoxidade (MPO) and alkaline phosphatase (AP) activity, and in addition their prebiotic effect, at concentration of 20% on acid-lactic bacteria in fecal samples. Additionally the antioxidant activity was evaluated from FRT ethanol extract 70% by lipoperoxidation in vitro in membrane brains of mice and phytochemical qualitative detection of saponins, tannins, flavonoids and coumarins group. Dietary supplementation of FRT at dose 10% significantly decreased the colonic damage observed macroscopically, microscopically and biochemically, with counteraction of colonic GSH and decreased MPO and FA activity. Complicating, considering, connecting: Rhizomatic philosophizing in music education

abstract: This philosophical inquiry explores the work of philosophers Gilles Deleuze and Félix Guattari and posits applications to music education. Through the concepts of multiplicities, becoming, bodies without organs, smooth spaces, maps, and nomads, Deleuze and Guattari challenge prior and current understandings of existence. In their writings on art, education, and how might one live, they assert a world consisting of variability and motion. Drawing on Deleuze and Guattari's emphasis on time and difference, I posit the following questions: Who and when are we? Where are we? When is music? When is education? Throughout this document, their philosophical figuration of a rhizome serves as a recurring theme, highlighting the possibilities of complexity, diverse connections, and continual processes. I explore the question "When and where are we?" by combining the work of Deleuze and Guattari with that of other authors. Drawing on these ideas, I posit an ontology of humans as inseparably cognitive, embodied, emotional, social, and striving multiplicities. Investigating the question "Where are we?" using Deleuze and Guattari's writings as well as that of contemporary place philosophers and other writers reveals that humans exist at the continually changing confluence of local and global places. In order to engage with the questions "When is music?" and "When is education?" I inquire into how humans as cognitive, embodied, emotional, social, and striving multiplicities emplaced in a glocalized world experience music and education. In the final chapters, a philosophy of music education consisting of the ongoing, interconnected processes of complicating, considering, and connecting is proposed. Complicating involves continually questioning how humans' multiple inseparable qualities and places integrate during musical and educative experiences. Considering includes imagining the multiple directions in which connections might occur as well as contemplating the quality of potential connections. Connecting involves assisting students in forming variegated connections between themselves, their multiple qualities, and their glocal environments. Considering a rhizomatic philosophy of music education includes continually engaging in the integrated processes of complicating, considering, and connecting.
