Sequential and Localized Implicit Wavelet Based Solvers for Stiff Partial Differential Equations

This thesis explains and tests a wavelet based implicit numerical method for the solving of partial differential equations. Intended for problems with localized small-scale interactions, the method exploits the form of the wavelet decomposition to divide the implicit system created by the time discretization into multiple, smaller, systems that can be solved sequentially. Included are tests of this method on linear and non-linear problems, with both its results and the time required to calculate them compared to basic models. It was found that the method requires less computational effort than the high resolution control results. Furthermore, the method showed convergence towards high resolution control results.

Dimensions of biculturalism: the development of bicultural identity orientation scale (BIOS)

Four studies (focus groups, exploratory and confirmatory analysis, and follow-up interviews) were designed to investigate people's experiences of biculturalism, particularly as they related to ethnic identity, identity conflict and integration, and hybridity, and to develop a new instrument that would better tap at the concept of biculturalism. Previous literature conducted by Benet-Martinez (2002, 2005) identified two relevant axes for bicultural identity: conflict-harmony and distance-overlap, while Yip (2005), Phinney (1991) and Noels (2004) emphasized the role of context for biculturals. We identified five interrelated dimensions, which provide a better understanding of the experiences of biculturalism. These dimensions are conflict (a perceived discord between the two cultures), monocultural orientation (the desire to be part of only one of the two cultures), flexibility (the alternation of behaviours and attitudes depending on the context), compatibility (perceived congruence between the two cultures) and hybridity (the blend of the two cultures to create one). A new instrument, the Bicultural Identity Orientation Scale, was developed based on the literature review and the anecdotal evidence provided by the participants. The instrument showed validity and reliability. The implications and future directions are discussed in light of the findings.

Epi-CHO, an episomal expression system for recombinant protein production in CHO cells

The current project is to develop a transient expression system for Chinese Hamster Ovary (CHO) cells based on autonomous replication and retention of plasmid DNA. The expression system, named Epi-CHO comprises (1) a recombinant CHO-K1 cell line encoding the Polyoma (Py) virus large T-Antigen (PyLT-Ag), and (2) a DNA expression vector, pPy/EBV encoding the Py Origin (PyOri) for autonomous replication and encoding the Epstein-Barr virus (EBV), Nuclear Antigen-1 (EBNA-1) and EBV Origin of replication (OriP) for plasmid retention. The CHO-K1 cell line expressing PyLT-Ag, named CHO-T was adapted to suspension growth in serum-free media (EXCELL-302) to facilitate large scale transient transfection and recombinant (r) protein production. PyLT-Ag-expressed in CHO-T supported replication of PyOri-containing plasmids and enhanced growth and r- protein production. A scalable cationic lipid based transfection was optimised for CHO-T cells using LipofectAMINE-2000??. Destabilised Enhanced Green Fluorescence Protein (D2EGFP) and Human Growth Hormone (HGH) were used as reporter proteins to demonstrate transgene expression and productivity. Transfection of CHO-T cells with the vector pPy/EBV encoding D2EGFP showed prolonged and enhanced EGFP expression, and transfection with pPy/EBV encoding HGH resulted in a final concentration of 75 mg/L of HGH in culture supernatant 11 days following transfection.
